Abstract
本发明公开了冬凌草甲素通过降解Bcr-Abl融合蛋白可用于治疗费城染色体阳性肿瘤,涉及冬凌草甲素在制备费城染色体阳性肿瘤治疗药物中的应用。经过大量研究发现,冬凌草甲素能够激活Bcr-Abl特异的泛素化降解途径,迅速和高效地降解Bcr-Abl癌蛋白,从而使费城染色体阳性的肿瘤细胞发生显著的生长抑制和不可逆转的细胞凋亡。在小鼠体内,冬凌草甲素同样能够降解Bcr-Abl蛋白,显著抑制费城染色体阳性肿瘤的生长。冬凌草甲素主要来源于我国传统中药冬凌草,对人体毒性较低,且易于制备、可应用性强。另外,本发明所揭示的冬凌草甲素作用机制与目前用于白血病临床治疗的一线药物STI-571(格列卫)完全不同,因此可作为一种治疗STI-571耐受的费城染色体阳性肿瘤的潜在药物。

背景技术
肿瘤是威胁人类健康的头号杀手。2008年全球新增肿瘤患者1270万,死亡760万,并且这些数字仍在持续增长中。随着现代医学的发展,人们逐渐认识到肿瘤的形成是由于基因突变造成的,由此开发出多种以基因为靶点的分子靶向疗法,并在肿瘤治疗中取得了显著成效。
染色体畸变是导致肿瘤细胞中基因突变的常见原因。1960年,两位美国科学家Peter Nowell和Dacid Hungerford在慢性髓性白血病(CML)病人的血液样品中发现了一个异常的染色体,由于是在美国费城(Philadelphia)发现的,故命名为费城染色体(Ph)。后来经染色体显带发现费城染色体是由9号染色体长臂(9q34)和22号染色体长臂(22q11)易位造成的,又被称为t(9; 22)(q34; q11)。费城染色体具有重要的临床意义:大约95%的慢性髓性白血病病例都是Ph阳性,因此可作为诊断的依据。另外,在急性髓性白血病、前B细胞和T细胞白血病/淋巴瘤中大约20%-30%的成人与2-10%的未成年患者中也能检测到费城染色体。
t (9; 22) (q34; q11)染色体易位产生一种致癌性的融合基因Bcr-Abl,该基因编码的融合蛋白产物具有异常增高的酪氨酸激酶活性,是导致慢性髓性白血病及其他Ph阳性肿瘤发生的首要原因。研究表明,Bcr-Abl融合蛋白还是造成Ph阳性肿瘤细胞对多种化疗药物不敏感的原因。在K562细胞及其它Ph阳性的肿瘤细胞中抑制Bcr-Abl蛋白,可以显著抑制和杀伤肿瘤细胞,还可以逆转这些肿瘤细胞对化疗药物的耐药性。
STI-571(格列卫)是靶向Bcr-Abl的酪氨酸激酶竞争性抑制剂,目前已经成为治疗慢性髓样白血病(CML)病人的一线用药,对于慢性期(CP)的CML病人具有良好的治疗效果。但是,对于加速期(AP)和急变期(BC)的病人,STI-571的治愈率很低,而且病人容易复发并最终演变成STI-571耐受的CML-BC。因此急需开发一些作用机制与STI-571不同的的抗肿瘤药物用于CML的治疗。
除了使用小分子抑制剂干扰癌蛋白的酶活性外,靶向癌蛋白的泛素化降解也是一种很好的分子靶向治疗策略。泛素化降解是细胞中重要的蛋白质质控系统,可以选择性降解细胞中不需要的蛋白质,例如一些短寿命的细胞周期调节蛋白、癌基因和抑癌基因产物以及变性变构蛋白等。在肿瘤细胞中,促进癌蛋白的降解对于逆转肿瘤恶性程度、改善治疗效果具有重要的意义。
冬凌草甲素是我国传统抗癌中药冬凌草中最主要的抗肿瘤活性成分,经鉴定属于贝壳杉烯类四环二萜类化合物。中国专利(申请号200410016891.6,公开日2005年9月14日)公开了冬凌草甲素在降解AML1-ETO融合蛋白中的应用,为开发具有t(8;21)染色体易位的白血病靶向治疗药物奠定了基础。另一中国专利(申请号201110413556.X,公开日2012年7月4日)公开了冬凌草甲素在制备FBW7抑癌蛋白激活剂中的应用,报道了冬凌草甲素通过激活FBW7介导的泛素化降解途径靶向降解癌蛋白c-Myc、cyclinE和mTOR的作用。关于冬凌草甲素在降解Bcr-Abl蛋白以及制备费城染色体阳性肿瘤治疗药物的应用还未见报道。

发明内容
本发明的目的在于针对现有技术中的上述不足,提供一种靶向泛素化降解癌蛋白Bcr-Abl的方法和试剂,以及该方法在制备Ph阳性肿瘤治疗药物中的应用。
本发明的另一目的在于提出一种诱导剂,在临床和基础研究中用于降解Bcr-Abl蛋白,从而应用于其生物学功能及相关信号通路的研究。
申请人经大量研究发现冬凌草甲素在体内和体外均能够激活Bcr-Abl特异的泛素化降解途径,迅速和高效地降解Bcr-Abl癌蛋白,从而使Ph阳性的肿瘤细胞发生显著的生长抑制和不可逆转的细胞凋亡。
对本发明的研究结果进一步阐述如下:
对Ph阳性肿瘤细胞K562,采用20μM冬凌草甲素处理24小时。免疫印迹检测Bcr-Abl蛋白的表达,发现冬凌草甲素能够降低Bcr-Abl和c-Abl蛋白的水平,而对Bcr蛋白没有影响,表明冬凌草甲素能够在Ph阳性细胞系中选择性作用Bcr-Abl蛋白家族中的癌蛋白成员。与Ph阴性肿瘤细胞HL-60相比,冬凌草甲素在Ph阳性肿瘤细胞K562和KU812中均能显著降低Bcr-Abl蛋白的水平。
采用0-20 μM冬凌草甲素处理K562细胞24小时,Bcr-Abl和c-Abl蛋白的降低具有显著的剂量依赖模式。在较低浓度(2.5-7.5 μM)冬凌草甲素诱导下,Bcr-Abl和c-Abl蛋白水平即发生显著降低;而10 μM以上浓度冬凌草甲素诱导下,Bcr-Abl蛋白几乎完全降解。表明采用冬凌草甲素诱导降解Bcr-Abl只需较低的剂量即可达到显著的效果。
冬凌草甲素对Bcr-Abl蛋白的降解非常迅速,20 μM冬凌草甲素处理2h即可完全降解Bcr-Abl。该作用持续时效较长且不易逆转,处理24小时后撤销冬凌草甲素,48小时内未见Bcr-Abl的表达回复。
采用qRT-PCR检测发现冬凌草甲素并不影响Bcr-Abl和c-Abl的mRNA水平。采用蛋白酶体抑制剂MG-132、ALLN和Lactacystin预处理细胞2小时,然后加入20 μM冬凌草甲素,可完全抑制冬凌草甲素对Bcr-Abl和c-Abl蛋白的降解。表明冬凌草甲素对Bcr-Abl和c-Abl的特异性降解是通过泛素化蛋白酶体途径实现的。由于泛素化蛋白酶体降解途径的高度选择性,这从另一个方面证明了冬凌草甲素对Bcr-Abl蛋白降解的专一性。
采用0-20 μM冬凌草甲素处理K562细胞和KU812细胞,均可引发不同程度的细胞生长抑制和凋亡,且与Bcr-Abl的降解呈正相关。10 μM以上浓度冬凌草甲素能够诱导显著的细胞凋亡。20 μM冬凌草甲素处理24小时,K562细胞和KU812细胞的凋亡率均大于50%,且细胞生长受到显著抑制,抑制率分别为68%和79%。冬凌草甲素处理K562细胞24小时后撤销药物诱导,发现在20 μM浓度处理组中细胞凋亡不可逆转,细胞凋亡率在药物撤销后48小时内仍逐渐增高。表明冬凌草甲素可通过降解Bcr-Abl融合蛋白,从而抑制Ph阳性肿瘤细胞的生长并诱导细胞凋亡。
在NOD-SCID小鼠后肢背部接种Ph阳性肿瘤细胞,待小鼠成瘤后采用冬凌草甲素(15 mg/kg)进行腹腔注射,连续注射12天。与对照组相比,冬凌甲素对小鼠体内肿瘤组织的生长具有显著的抑制作用,且能显著降低肿瘤组织内的Bcr-Abl和c-abl蛋白的表达水平。
本发明具有以下有益效果:
本发明提出了一种靶向降解Bcr-Abl融合蛋白的方法。使用冬凌草甲素处理Ph阳性细胞系和Ph阳性移植瘤小鼠,均能够选择性作用于Bcr-Abl蛋白家族成员中具有癌蛋白性质的成员(Bcr-Abl融合蛋白和c-Abl蛋白),从而抑制肿瘤细胞的生长并诱导凋亡。该方法利用了细胞内源的泛素化降解途径,具有反应灵敏迅速、作用时效长、不易逆转、应用方便等优点,而且特异性较高,对未发生基因重排的Bcr基因基本没有影响。这为靶向治疗Ph阳性肿瘤提供了新思路,并为在临床和基础研究中分析Bcr-Abl生物学功能及其相关信号通路提供了新途径。
本发明提供了一种靶向降解Bcr-Abl和治疗Ph阳性肿瘤的试剂,该试剂主要来源于中国传统草药冬凌草,是一种易于制备的天然药物,其应用剂量较低,对人体毒副作用性较小,可应用性强。另外,冬凌草甲素对Bcr-Abl的作用机制与STI-571不同,因此可能对STI-571耐受的费城染色体阳性病人也有较好的疗效。

具体实施方式
实施例1 冬凌草甲素在Ph阳性肿瘤细胞中高效、迅速和不可逆地降解Bcr-Abl蛋白
我们在Ph阳性肿瘤细胞系K562中检测冬凌草甲素对Bcr-Abl融合蛋白的影响。各细胞按3x105个/mL的密度接种于细胞培养板上,培养24小时至对数生长期后分别加入20 μM冬凌草甲素和药物溶剂DMSO处理。诱导24小时后收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。结果显示与对照DMSO处理相比,冬凌草甲素处理使K562细胞中的Bcr-Abl和c-Abl蛋白水平显著降低,几乎完全降解,而内对照GAPDH蛋白的水平没有明显变化(图1)。说明冬凌草甲素在K562细胞中能专一性降解Bcr-Abl蛋白家族中的癌蛋白成员。
另外,我们以Ph阴性肿瘤细胞系HL60为对照,在Ph阳性肿瘤细胞系K562和KU812中检测冬凌草甲素对Bcr-Abl融合蛋白的影响。细胞培养24小时后分别加入20 μM冬凌草甲素和DMSO,诱导2小时后收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。结果显示K562细胞和KU812细胞中Bcr-Abl表达水平较高,采用冬凌草甲素处理使K562细胞和KU812细胞中的Bcr-Abl蛋白水平显著降低,而内对照GAPDH蛋白的水平没有明显变化(图2)。而HL-60细胞中没有检测到Bcr-Abl蛋白的表达。说明冬凌草甲素在Ph阳性肿瘤细胞中能专一性降解Bcr-Abl蛋白家族中的癌蛋白成员。
接种K562细胞,培养24小时后加入0-20 μM梯度浓度的冬凌草甲素处理。诱导24小时后收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。Bcr-Abl和c-Abl蛋白的降低具有显著的剂量依赖模式:在2.5-7.5 μM冬凌草甲素诱导下,Bcr-Abl和c-Abl蛋白水平即发生显著降低;而10 μM以上浓度冬凌草甲素诱导下,Bcr-Abl蛋白几乎完全降解(图3)。
接种K562细胞,培养24小时后加入20 μM冬凌草甲素处理。于0-7小时收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。结果显示冬凌草甲素对Bcr-Abl的降解十分迅速,诱导2小时后即可完全降解Bcr-Abl蛋白(图4)。
接种K562细胞,培养24小时后加入0-20 μM的冬凌草甲素处理。诱导24小时后收集细胞用PBS洗去培养基中的药物,并用不含药物的新鲜培养基重新悬浮细胞,继续培养24-48小时。于相应时间点收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。20 μM冬凌草甲素处理组的细胞在撤销药物24-48小时后仍几乎检测不到Bcr-Abl蛋白的表达(图5),表明该浓度冬凌草甲素处理下Bcr-Abl蛋白的降解是不易逆转的。
实施例2 冬凌草甲素靶向泛素化降解Bcr-Abl蛋白
接种K562细胞,培养24小时后分别加入三种蛋白酶体抑制剂MG-132(M,10 μM)、ALLN(A,100 μM)或Lactacystin(L,25 μM)预处理2小时,再加入20μM冬凌草甲素诱导。2小时后收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。MG-132、ALLN和Lactacystin可完全抑制冬凌草甲素对Bcr-Abl和c-Abl蛋白的降解(图6),说明冬凌草甲素对Bcr-Abl和c-Abl的降解依赖于泛素蛋白酶体途径。
接种K562细胞和KU812细胞,培养24小时后分别加入蛋白酶体抑制剂MG-132预处理2小时,再加入20 μM冬凌草甲素诱导。2小时后收集细胞并用SDS法提取细胞总蛋白,进行免疫印迹检测。在两种细胞中MG-132可完全抑制冬凌草甲素对Bcr-Abl蛋白的降解(图2),说明在费城染色体阳性细胞中,冬凌草甲素均能靶向泛素化降解Bcr-Abl蛋白。
实施例3 冬凌草甲素抑制Ph阳性肿瘤细胞的生长并诱导不可逆转的细胞凋亡
接种K562细胞,培养24小时后加入0-20 μM的冬凌草甲素处理。诱导24小时后采用MTT检测细胞存活率,同时收集细胞通过AnnexinV-FITC/PI染色检测细胞凋亡。在不同浓度冬凌草甲素处理下,K562细胞和KU812细胞发生不同程度的生长抑制和细胞凋亡,并呈现出明显的剂量依赖效应。20 μM冬凌草甲素处理24小时,K562细胞和KU812细胞的生长受到显著抑制(图7A,图8A),存活率分别仅为32%和21%,而且发生显著的细胞凋亡,凋亡率均大于50%(图7B,图8B)。
接种K562细胞,培养24小时后加入0-20 μM的冬凌草甲素处理。诱导24小时后收集细胞用PBS洗去培养基中的药物,并用不含药物的新鲜培养基重新悬浮细胞,继续培养24-48小时。于相应时间点收集细胞通过AnnexinV-FITC/PI染色检测细胞凋亡。在20 μM浓度处理组中细胞凋亡不可逆转,凋亡率在药物撤销后48小时内仍逐渐增高(图7B),这与Bcr-Abl融合蛋白的不可逆降解相一致。
实施例4 冬凌草甲素在K562移植瘤小鼠体内对Bcr-Abl的降解作用
小鼠移植瘤模型的建立:收集培养至对数生长期的K562细胞,计数活细胞。在5-6周龄的NOD-SCID小鼠的右后肢背部皮下注射6.67x106个K562细胞,并观察小鼠成瘤情况。
冬凌草甲素处理:当瘤块体积达到200 mm3时,将小鼠随机分成两组:第一组为对照组,应用助溶剂(2% DMSO)0.07 mL/10g腹腔注射;第二组为治疗组,应用冬凌草甲素(15 mg/kg体重)腹腔注射,连续注射12天。每隔 2天测量一次瘤块的大小和小鼠的体重,治疗组小鼠的肿瘤比对照组生长缓慢(P<0.05),而两组小鼠的体重没有明显差别。开始给药后的第 13 天,处死小鼠并解剖瘤块,可见治疗组小鼠的肿瘤大小比对照组明显减小。
Bcr-Abl表达检测:将小鼠肿瘤组织切成约1 cm3,取适量于RIPA蛋白裂解中匀浆并充分裂解,离心取上清,使用BCA试剂盒进行蛋白定量。取120 μg总蛋白进行免疫印迹,以GAPDH为内参,分别检测Bcr-Abl和c-Abl的蛋白水平。与对照组相比,治疗组中Bcr-Abl和c-Abl的蛋白水平均发生显著降低(图9)。
Claims (9)
1.一种靶向降解Bcr-Abl蛋白的试剂,其特征在于其主要成分为冬凌草甲素,或以冬凌草甲素为先导化合物的具有环外亚甲基环戊酮结构的四环二萜类化合物。
2.如权利要求1所述的试剂,其特征在于冬凌草甲素的浓度为2.5-20 μM。
3.如权利要求1所述的试剂在哺乳动物肿瘤细胞的实验研究中作为诱导剂降解Bcr-Abl和c-Abl蛋白的应用。
4.如权利要求3所述的应用,其特征在于所述的哺乳动物为人类、小鼠或大鼠。
5.冬凌草甲素或以冬凌草甲素为先导化合物的具有环外亚甲基环戊酮结构的四环二萜类化合物在制备费城染色体阳性肿瘤治疗药物。
6. 如权利要求5所述的应用,其特征在于所述的费城染色体(Ph)阳性肿瘤包括Ph阳性的慢性髓性白血病、Ph阳性的急性髓性白血病、Ph阳性的急性淋巴性白血病和Ph阳性的前B细胞、T细胞淋巴瘤。
7.如权利要求5所述的应用,其特征在于使用2.5-20 μM及以上浓度的冬凌草甲素处理费城染色体阳性的K562细胞和KU812细胞,2-24小时后可引起Bcr-Abl蛋白的泛素化降解,使细胞生长受到抑制并且诱导细胞凋亡。
8.如权利要求5所述的应用,其特征在于以15mg/kg冬凌草甲素的剂量连续注射携带费城染色体阳性肿瘤的小鼠,12天后可引起肿瘤组织中Bcr-Abl蛋白的降解,抑制肿瘤的生长。
9.如权利要求5所述的应用,其特征在于冬凌草甲素可专一性地降解费城染色体阳性肿瘤的病源蛋白Bcr-Abl,以及其同一蛋白家族的成员c-Abl蛋白。
