技术领域

背景技术
分子对于细菌生长的解偶联活性可以很好地通过细菌能量失衡促使的氧气过度消耗表示。
对于在废水处理装置(随后用法语缩写STEP表示)中的应用,分子的这种解偶联活性是让人感兴趣的,当其能够在生产源头显著减少活性污泥的的情况下。
在废水处理中,生物质和活性污泥的生产来源于对废水中营养物质的消耗。通过呼吸过程,营养物质被氧化并释放能量,该能量可以被微生物在细胞分裂的范围内使用。现在,通过氧化磷酸化现象,营养物质的消耗引起细胞膜中的质子流;该流将建立质子梯度,其本身操作允许从ADP+P合成ATP(三磷酸腺苷)的质子泵。ATP在细胞进程(包括细胞分裂)中向细胞提供能量。
如果这种能量释放可以避免,则通过抑制能量生成,将导致生物质生产的降低。解偶联相当于抑制形成ATP形式的能量供应。解偶联剂降低碳消耗的能量产率,而增加碳氧化为CO2的比例。解偶联因而用更少的生物质生成和更高的氧气消耗来表示。
在废水处理期间产生的细菌生物质的移除是昂贵的,并因而在生物质减少使得移除成本降低。
发明内容
本发明的一个目的是提供解偶联分子,其效率通过在源头(即在城市废水处理装置的曝气池)的生物污泥的生产下降了至少30%来测量。
本发明的另一个目的是提供解偶联分子,其效率与参照分子THPS(四羟甲基硫酸磷鎓)相当,甚至更好,THPS对于降低生物污泥生产的效率已经在专利申请WO 2004/113236中展示。
本发明的另一个目的是提供替代THPS的解偶联分子,其非生物降解性和生物降解性比THPS更慢,但是所述分子并不是不可降解的(不可降解可引起环境问题)。
最后,本发明的另一个目的是提供解偶联分子,其毒物学的特征和生态毒理的特征对于在STEP中使用后者是令人满意的并且是足够的。

优选地,所述含水体系是用于处理工业或城市污水的废水处理装置。这种设施回收来自工业进程(例如,造纸、食品工业、化学工业)和/或住所和办公楼宇以及类似设施的废水,通过使用微生物在有氧、缺氧进程(例如脱硝)中,消耗有机污染物并使得该水适于再利用或将其弃排入环境中。
本发明因此提供了控制在含水体系中细菌生物质生长的方法,包括向含水体系中加入或让其接触有效量的如上定义的解偶联剂。
具体实施方式
在优选的实施方式中,本发明提供用于控制含水体系中细菌生物质生长的方法,该方法包括使细菌生物质与有效量的如上定义的解偶联剂直接接触。为了应用这个方法,建议在有限时间内加入最大体积的活化污泥,使之与解偶联剂接触,以使后者获得最优效率。
因此,对于在实验室的生物小规模试验,在水中解偶联剂与细菌生物质的直接接触通过称作“瞬时给药”或者“瞬时混合”的快速混合(flash mixing)而完成。
已发现如果解偶联剂是简单地直接加入到含有污泥的生物反应器中,则试剂的效率大幅度降低,因为解偶联剂能够与存在于生物反应器中的其它材料相互作用并且有机磷鎓衍生物的作用被大幅度降低。
此外,所述的有机磷鎓衍生物的有效量可以表示为对于每克存在于含水体系污泥中的固体(以干物质或DM表示),为0.1-100mg,优选为0.5-100mg/g,例如1-50mg/g,比如2-10mg/g。
下面的实施例阐明本发明而不是限制其范围。
实施例1:
步骤a)三羟甲基膦溶液的合成
在安装有减压蒸馏的、事先在氮气下惰化的充分搅拌的反应器中,装入:
滴定80%THPC(四(羟甲基)氯化磷)而含有150gTHPC(0.63mol)的溶液;
将混合物冷却至5-15℃;
在3小时内将385g 8%苏打溶液倒入并控制温度在5-15℃;
在倾倒结束后,将反应混合物保持在10℃下12小时;

实施例3:
步骤a)三羟甲基膦溶液的合成
在安装有减压蒸馏的、事先在氮气下惰化的充分搅拌的反应器中,装入滴定80%THPC(四(羟甲基)氯化磷)而含有100g THPC(0.42mol)和水(50g)的溶液;
将混合物冷却至5-15℃;
在3小时内倒入228g 8%苏打NaOH溶液并控制温度在5-15℃;
在倒入结束后,保持在10℃12小时;
在温度15-30℃和压力小于10mbar下,通过蒸馏水/甲醛混合物移除生成的甲醛;加入786g的乙醇并在相同的条件下进行蒸馏以耗尽甲醛;
观察到NaCl的沉淀。
实施例4
步骤b)和c):磷鎓(式(1),n=6,m=0或1)的合成
将实施例3中制得的反应物引入安装有减压蒸馏的、事先在氮气下惰化的充分搅拌的反应器中,控制温度在45-55℃;
在30分钟内倒入111.8g 1,6-二碘代己烷并控制温度在45-55℃;
所得的反应介质是两相的并且观察到盐的沉淀(NaI/NaCl);
在15%THPC存在下,反应混合物的温度保持在50℃下30小时;
在10℃的温度下,加入30g 8%NaOH;
温度设为40℃;
倒入14.5g 1,6-二碘代己烷;
反应介质在50℃保持12小时;
将反应混合物冷却至10℃,并随后在多孔玻璃砂上过滤;
在15-30℃温度下和压力<10mbar下,通过蒸馏水/甲醛混合物移除生成的甲醛;并加入400g乙醇在相同的条件下进行蒸馏以耗尽甲醛;
在10℃的温度下,加入15g 8%NaOH;
温度设为40℃;
倒入10g的1,6-二碘代己烷并同时保持反应混合物在50℃下12小时;
将反应介质冷却至10℃并随后在多孔3玻璃砂上过滤;
在15-30℃温度下和<10mbar的压力下,通过蒸馏水/甲醛混合物移除生成的甲醛;并随后加入350g的水并在相同的条件下进行蒸馏以耗尽甲醛;
所得的产物是浅黄色透明溶液,从其中排除327.7g的重量。
排除的水相用乙酸乙酯(AcOEt 350g)洗涤三次;
洗涤后的水相在旋转蒸发仪中脱溶剂并得到284g浅黄色溶液,具有如下的核磁共振分析:

实施例5:
为了在呼吸计量法中评价解偶联化学试剂促使的污泥生产的减少(为了测量氧气的过量消耗,解偶联剂的一个典型特征),使用了oxytop测试技术,其包括合成介质和对解偶联剂特别敏感的菌株。这些模型菌株(特别是菌株Schinellagranuli)是STEP的活性污泥的特定代表,因为它们是在曝气池中从后者分离得到的。
所用的方法和设备是在作为文献引用的WO 2004/112236的实施例3中公开的那些。在这个方法学中,菌株Schinella granuli被用作接种体(其与WO2004/113236实施例3中用的活性污泥不同)。
在筛查测试中得到的结果汇总于下面的表1中。它们被表示为相对于对照(不含有任何解偶联剂)的解偶联百分比。这意味着对于在0.05mg/l的Ex2(上面的实施例2),相对于不含有任何解偶联剂的对照,我们具有18%的氧气过度消耗。对于3ppm的额定浓度,THPS(四(羟甲基)硫酸磷鎓)在oxitop呼吸计量法测试中产生的解偶联效力为16±8%,就解偶联因子为[10-22]而言,THPS的置信区间为95%,这些数值是在26次Oxitop测试中得到的结果计算的。

观察到在实施例2中合成的磷鎓衍生物在相对窄的浓度范围给出了好的效率。
这个浓度范围经常表现出在低浓度下解偶联效果的活化,随之是轻微的抑制以及随后在约5ppm(额定浓度)获得毒性。
观察到解偶联效果就像用THPS获得的那些(16±8%,使用3ppm的THPS、26次Oxitop获得的平均值)的相同数量级。实施例2的二磷鎓与三磷鎓的混合物在包括在0.1-2ppm之间的相对低的浓度范围内给出可重复的和显著的解偶联作用。最优浓度似乎位于1ppm,具有15%的数量级。
实施例6
在Chemostat生物小规模试验中检测有机磷鎓衍生物对活性污泥的减少。
连续用Ex2(上面的实施例2)的分子处理的合成污水给料的生物小规模试验表现出显著的减少污泥生产的解偶联作用。生物小规模试验具有5升的体积并保持通风;每天进行后者的清除(除了周末)以获得大约7天的泥龄。生物小规模试验是从St-Fons城市STEP(法国)中的活性污泥接种。污泥的短的适应期后,用实施例2分子的处理是根据也公开在作为文献引用的WO2004/113236的实施例1中的快速混合方法进行的。
实验中,根据快速混合方法注入解偶联产物的总时期持续了3个月,给出下面的结果:
与未处理的对照相比,观察到化合物Ex2对污泥生产的减少具有好的效率,尤其是在2.3ppm,观察到略微大于20%的减少。
补充的实验,这次使用上面实施例2的化合物Ex2的两个不同等级,此后命名为EX21和EX22,经过1个月的注入期给出下面的结果:
化合物EX22表现出对减少污泥生产27.6%和减少生长率26%的好的效率(已证明在这个浓度下,其没有改变污泥的纯化能力)。
Claims (15)
2.根据权利要求1所述的衍生物,其特征在于n是包括在5-10之间的数值,m是0或者1,并且
Y是氯离子、硫酸根离子、磷酸根离子或者溴离子。
3.根据权利要求1或2所述的衍生物,其特征在于n是8并且m是0或1。
4.控制含水体系中细菌生物质生长的方法,包括向所述含水体系中加入有效量的选自如权利要求1-3任一项定义的有机磷鎓衍生物的解偶联剂或者使所述有机磷鎓衍生物的解偶联剂与所述含水体系接触。
5.根据权利要求4所述的方法,其特征在于所述解偶联剂的有效量是包括在0.001-100mg/l之间。
6.根据权利要求5所述的方法,其特征在于所述解偶联剂的有效量是包括在0.005-50mg/l之间。
7.根据权利要求6所述的方法,其特征在于所述解偶联剂的有效量是包括在0.01mg/l-30mg/l。之间
8.根据权利要求7所述的方法,其特征在于所述解偶联剂的有效量是包括在0.1mg/l-10mg/l之间。
9.根据权利要求8所述的方法,其特征在于所述解偶联剂的有效量是包括在0.5mg/l-7.5mg/l之间。
10.根据权利要求9所述的方法,其特征在于所述解偶联剂的有效量是包括在1mg/l-5mg/l之间。
11.根据权利要求4所述的方法,其特征在于对于存在于所述含水体系污泥中的以干物质表示的每克固体,所述解偶联剂的有效量为0.1mg-100mg。
12.根据权利要求11所述的方法,其特征在于对于存在于所述含水体系污泥中的以干物质表示的每克固体,所述解偶联剂的有效量为1mg-50mg。
13.根据权利要求12所述的方法,其特征在于对于存在于所述含水体系污泥中的以干物质表示的每克固体,所述解偶联剂的有效量为2mg-10mg。
14.根据权利要求4-13任一项所述的方法,其特征在于所述含水体系是处理工业或城市污水的废水处理装置。
15.制备如权利要求1所定义的有机磷鎓衍生物的方法,其包括下面的步骤:
a)根据下面的反应流程图,在惰性气氛下使四(羟甲基)氯化磷鎓与强碱例如苏打反应,移除甲醛,以获得三羟甲基膦:
其中X代表阴离子;
b)使α,ω,二卤代亚烷基与步骤a)得到的反应物反应并获得相应的亚烷基二磷鎓
c)根据下面的反应流程图,任选将步骤b)得到的亚烷基二磷鎓与过量的二卤代亚烷基和三羟甲基膦反应,释放出甲醛,以生成三磷鎓或多磷鎓:
X、Y、n和m具有如上面式(1)给出的定义。
