本发明属微生物动物细胞系领域。本发明为从经历过放疗、化疗和靶向药物治疗后的肺腺癌病人前臂转移灶切除物中培养成功的细胞系Am1010,保藏编号为CCTCC NO:C200940。所述细胞具有一下生物学特性:对多种化疗药物、放射线和靶向药物有抵抗性;染色体核型属超三倍体,染色体变异大,细胞DNA不稳定;所述细胞株是理想的人肺腺癌对放疗、化疗和靶向药物治疗抵抗机制的研究模型,在揭示人肺腺癌的耐放疗、耐化疗和耐靶向药物治疗的机制和抗肿瘤治疗药物筛选中有着广阔的应用前景。

技术领域
本发明涉及一株人的细胞系,特别是一株人肺腺癌放化疗靶向治疗后复发转移细胞株。
背景技术
肿瘤细胞系(株)是研究细胞癌变机理、肿瘤转移、肿瘤放疗和化疗敏感性分子基础等生物医学问题的重要材料,建立和鉴定不同的肿瘤细胞系是一项很有意义的工作。
本发明的目的是提供一株能够在体外多带培养的人肺腺癌放化疗靶向治疗后复发转移细胞株。
发明内容
本发明的人肺腺癌放化疗靶向治疗后复发转移细胞株Am1010已于2009年6月9日保藏在中国典型培养物保藏中心(其简称为CCTCC),保藏编号为CCTCC NO:C200940
本发明的人肺腺癌放化疗靶向治疗后复发转移细胞株Am1010是发明人从经历过放疗、化疗和靶向药物治疗后的肺癌病人前臂包块切除物中培养成功,经病理鉴定为肺腺癌转移。为上皮细胞,具有典型的肿瘤细胞生物学特性。它能在体外连续长期传代,在体外培养一年,传100多代,细胞仍然生长增殖活跃。
本发明细胞系经检测,其一般生物学特性表明该细胞系为多边形上皮样细胞,贴壁生长,接触性生长抑制消失,超三倍体核型,染色体范围60-110条,范围变异大,细胞DNA不稳定,并存在标记染色体。细胞群体倍增时间45小时,克隆形成率38.1±3.2%。本发明细胞系体外药物实验表明,其对多种化疗药、放射线和靶向药物有抵抗性。
人类全基因表达谱分析22k芯片分析表明,Am1010与来源于未经过放化疗初治肺腺癌病人的细胞进行对比,发现了2倍以上差异表达基因共有3122个,其中3倍以上差异表达基因1628个。对3倍以上的1628个基因表达数据采用MAS系统进行了Pathway和Go的统计分析。以下展示了差异表达基因所涉及到具有显著性意义的Pathway。这些pathway与与肿瘤发生、耐药、粘附、细胞骨架、转移有关。

具体实施方式
高糖DMEM培养基(Hyclone,Utah,USA)加20%(v/v)胎牛血清(Hyclone,Utah,USA)。培养温度为37℃,气体环境为5%CO2/95%空气,湿度为饱和湿度。细胞生长稳定,每3天更换培养液,每5天传代一次,常规冻存后复苏成活良好。

Claims (2)

1.一种人肺腺癌放化疗靶向治疗后复发转移细胞株,保藏编号为CCTCC NO:C200940。具体培养方法如下:
高糖DMEM培养基(Hyclone,Utah,USA)加20%(v/v)胎牛血清(Hyclone,Utah,USA)。培养温度为37℃,气体环境为5%CO2/95%空气,湿度为饱和湿度。细胞生长稳定,每3天更换培养液,每5天传代一次,常规冻存后复苏成活良好。
2.本发明细胞系体外药物实验表明,其对多种化疗药物、放射线和靶向药物有抵抗性;人类全基因表达谱分析22k芯片分析表明,Am1010与来源于未经过放化疗初治肺腺癌病人的细胞进行对比,2倍以上差异表达基因共有3122个,其中3倍以上差异表达基因1628个,其中最可能相关的十条pathway与肿瘤发生、耐药、粘附、细胞骨架、转移有关。
