发明领域
本发明涉及合成具有聚合物壳的空心球(hollow spheres)的方法。此外,本发明涉及空心球和这些球在超声成像或超声药物递送中的用途。
发明背景
超声作为诊断成像技术用于临床装置中。其相对便宜且更加重要的是,不采用电离辐射。在超声成像中,声波经换能器(transducer)传输至生物组织中。取决于涉及的组织和流体的声学性质,声波可被完全或部分地反射或吸收。反射的声波通过接收换能器检测,并被进一步处理以形成被研究的组织的图像。通过组织的声学性质的相对差异,确定图像对比度。尽管超声技术在进步,但是主要器官如肝、脾、肾、前列腺和脉管系统中恶性肿瘤和疾病的成像和检测仍然存在技术挑战。
为了优化能够在超声成像中获得的图像质量,已经开发了造影剂(contrast agents)。现有的超声造影剂被设计为是声学活性的,并且包括例如采用类脂、聚合物或蛋白质壳稳定的气体填充的微泡的组成。固体、液体和气体的声学性质,特别是声阻抗是不同的。这些声阻抗和可压缩性方面的明显变化导致在固体、液体和气体之间的界面处更加强烈的声波反射,导致超声图像强度的升高。气体填充的超声造影剂通过压缩和膨胀还对超声波起反应,导致产生可选择性检测的特定共振频率。此外,在造影剂可破裂而释放可检测的气体,和在造影剂中包含药物,局部释放治疗剂的情况下,造影剂能够采用超声脉冲活化。其它递送模式包括通过新释放的气泡的相互作用改变周围的组织,和暂时提高血管壁对从造影剂单独结合或给予的药物的渗透性。
中空的以聚合物为壳的试剂可由乳化过程合成,其中形成油填充的聚合物包覆的胶囊(capsule),然后采用冷冻干燥法除去核。这类合成路线的一个例子公开于Le等的文章(IEEE,proceedings of the annual northeast bioengineering conference Issue,2003年3月22-23日,315-316页)中,其公开了通过调节乳液聚合物微胶囊制备方法合成纳米胶囊。为制备油填充的胶囊,将油或其它疏水性添加剂添加至聚合物溶液中,该聚合物溶液是在该聚合物的仅与水轻度混溶的良溶剂如二氯甲烷中。然后将该溶液在包含稳定剂的水相中乳化。将所述良溶剂在加工期间除去。结果,乳液滴中聚合物的溶解性将下降,并随后从富含油的相中相分离,从而形成包含油的聚合物壳的胶囊。为通过乳化法生产中空核,将包封的疏水性添加剂或油通过冷冻干燥除去。通常将稳定剂如聚乙烯醇(PVA)添加至水相中,以控制粒度和防止聚集。稳定剂的添加通常必须过量,从而需要后续纯化和/或洗涤步骤,导致降低的工艺产率。
发明概述
本发明的一个目的在于改进用于具有声学性质的空心球合成的已知方法,以实现快速且方便的制备。
根据本发明,该目的通过包含以下步骤的方法实现:
提供包含以下的溶液:
-具有疏水性嵌段和亲水性嵌段的第一聚合物(1),
-不与水混溶的第二聚合物(2),
-疏水性化合物,
-水混溶性有机溶剂,
将所述溶液与水溶液混合
除去水混溶性有机溶剂
冷冻干燥以除去疏水性化合物。
空心球的直接沉淀通过将聚合物溶液与水溶液混合来引发。所述有机溶剂将立即均匀分布于水相中,导致疏水性化合物填充的聚合物球的形成。将形成所述球的壳的聚合物上亲水性嵌段的引入在不使用添加剂的情况下促进了稳定化。这种亲水性嵌段的一个例子为大亲水性基团例如聚乙二醇(PEG)。
由于不存在必须要除去的稳定剂如PVA和自动获得所述球的PEG化的(pegylated)外表面,所以根据本发明的方法与常规乳化过程相比更加快速和更加方便。相对于不具有这些嵌段的颗粒,该PEG化的表面还导致了增加的颗粒循环时间。
根据另一个实施方案,所述亲水性有机溶剂选自丙酮、四氢呋喃、二甲基亚砜、二甲基甲酰胺、乙腈、二醇醚和二甲基乙酰胺或其组合。这些溶剂是本发明方法中所用聚合物的良溶剂,是水混溶性的。所述溶剂从所述混合物中的除去可通过蒸发便利地进行促进。
在一个优选实施方案中,第一聚合物(1)包括选自聚交酯、聚己内酯、聚氰基丙烯酸酯和前述之一的共聚物、聚乙交酯的共聚物或它们的任意组合的组。更优选地,所述聚合物具有小于10000,优选为1000-6000的嵌段长度。当空心球用于人和医疗用途如超声成像中时,生物可降解性是期望的。
根据一个优选实施方案,第二聚合物(2)包括烷基或氟化端基或它们的组合。在聚合物上烷基和/或含氟基团的引入导致空心球内产生疏水性,保证了憎水性能。这将在含水生物环境中稳定所述球。稳定的声学活性球可通过对于约1000-10000,更优选1000-5000的聚合物分子量使用具有8-14个碳原子的烷基得到。
可以设想采用聚合物的不同组合以及聚合物的不同改性水平。具有亲水性嵌段的聚合物可与疏水改性的聚合物组合。
根据本发明方法的另一个实施方案为步骤(a)的溶液还包括冷冻干燥后得以保留的疏水性化合物如油,和药学或诊断化合物。这种疏水性化合物的优选例子为具有至少16个碳原子的烷烃、类脂类、石蜡或油中的至少一种,更优选十六烷。这种添加促进了除气体外还包含溶解或细分散于保留的疏水性化合物中的药物的可生物降解空心球的形成。可通过采用超声脉冲来局部释放溶解于保留在可生物降解空心球内的化合物中的药物。
本发明的另一个实施方案为根据包括以下步骤的方法合成的具有聚合物壳的空心球:
提供包含以下的溶液:
-具有亲水性嵌段和疏水性嵌段的第一聚合物(1),
-不与水混溶的第二聚合物(2),
-疏水性化合物,
-水混溶性有机溶剂,
将所述溶液与水溶液混合
除去水混溶性有机溶剂
冷冻干燥以除去疏水性化合物。
这种空心球优选能够在施加诊断或治疗超声时破裂。
所述空心球的核与壳之间的比例由溶液中聚合物与疏水性化合物之间的关系决定。优选地,该比例为2∶1-1∶5w/w,更优选为1∶1-1∶4w/w。
优选地,具有亲水性嵌段的第一聚合物(1)与不与水混溶的第二聚合物(2)之间的比例在2∶1-1∶12w/w范围,更优选1∶3-1∶10w/w。应理解的是,添加具有疏水性或亲水性性能的额外聚合物,或添加同时具有亲水性和疏水性性能的一种聚合物落在本发明的范围内。
根据本发明该实施方案的空心球当施加超声脉冲时能够破裂。该行为导致了在超声脉冲对准的区域中能够与超声相互作用的游离气泡的形成,增强了药物递送的超声引导。以类似的方式,还可以在相同区域中释放药学或诊断化合物。这开启了控制递送的量和位置的可能。此外,新形成的气泡易于吸收入射的超声能量,然后作为次级声源再发射,增加局部超声能量吸收。进而,局部升高的超声能量沉积可增加局部组织免疫反应的刺激和增强声孔效应(sono-poration),并从而增强药物摄取。此外,局部超声能量吸收可通过包封的气体的量(即可冷冻干燥的化合物的量)和声学脉冲的活化振幅进行控制。对应于特定尺寸气泡的独特声学信号的出现允许识别活化释放以及理想地允许一定的释放药物浓度置信度。
所述空心球优选具有50-400nm的尺寸,以通过避免身体中的主要清除机制或抑制其速率来增加循环时间。
在本发明的另一个实施方案中,气态内容物(gaseous content)至少部分地被气体前体替代。根据该实施方案,所述球包含至少一种可相转变的疏水性化合物例如液态全氟化碳,如全氟己烷、全氟戊烷、全氟庚烷、全氟辛烷和全氟辛基溴化物。与气体填充的空心球相比,气体形成仅通过施加超声进行诱导。
在另一个实施方案中,所述空心球的壳包括靶向部分。通过将靶向部分或前靶向部分偶联至所述球,促进了所述球的选择性靶向。靶向部分的例子包括但不限于抗体或抗体片段、生物素/抗生蛋白链菌素连接体和化学正交偶合部分(chemical orthogonal coupling moieties)如用于Staudinger反应的膦和叠氮基团。
在另一个实施方案中,所述空心球包括药物活性化合物,从而所述球可起到药物载体的作用。特别地,所述药物活性化合物可选自抗体、蛋白质、siRNA、shRNA和pDNA。
在根据本发明的另一个实施方案中,所述空心球包括选自PET、SPECT或MRI试剂的成像剂(imaging agent)。通过将空心球的超声特性与额外的成像部分结合,有助于监视例如聚合物壳的降解途径或局部药物递送。

根据本发明的方法
基于现有技术,声学活性空心球可通过采用乳化技术生产。乳化依赖于不与水混溶或仅在非常有限的程度上与水混溶的溶剂,例如卤化溶剂如二氯甲烷。通常,需要添加剂以控制形成的颗粒的尺寸和防止在冷冻干燥期间颗粒聚集。这些方法通常导致尺寸在微米范围的空心球。
在本发明中,我们改进了合成具有声学性质的空心球的已知方法以实现快速且方便的制备,特别适于亚微米空心球的制备。以下详细描述可在本发明合成方法中辨认的步骤。
步骤(a)
在根据本发明方法的步骤(a)中,提供溶液,其包含具有亲水性嵌段的第一聚合物(1)、不与水混溶的第二聚合物(2)、疏水性化合物如烷烃或油和水混溶性有机溶剂。
所述聚合物或聚合物的组合用于形成空心球的壳。用于本发明方法的聚合物可包含不同构件(elements),如图1中示意性显示的那样。
本发明方法中使用的聚合物优选包含主要决定刚性(如结晶或玻璃态)壳的机械性质的合成或生物聚合物嵌段。这些嵌段可以是聚合物1和2这两者的一部分,并且在很大程度上有助于所述球的稳定性。符合这些要求的典型可生物降解聚合物为L或DL形式的聚丙交酯(PLA)或其共聚物、聚交酯、聚己内酯、聚氰基丙烯酸酯和上述之一的共聚物、聚乙交酯的共聚物,或它们的任意组合。这个或这些构件形成在中空颗粒的壳形成中占主导地位的聚合物的主链并在图1中由嵌段(A)示意性表示。相对低分子量的聚合物在壳中具有较少缠结,并且因此在施加超声时更易于导致壳破裂。
可使用具有不同改性情况的聚合物的混合物,在图1中示出3个实例。根据本发明,所述混合物中存在具有亲水性嵌段的聚合物。该优选大体积的(bulky)亲水性基团(由嵌段B示意性地表示)将位于由本发明方法形成的球的外侧。这种基团的使用稳定了所述球,并促进了直接沉淀。这类基团的一个例子为聚乙二醇。
第二聚合物包括疏水性部分。这种部分的例子为烷基基团或氟化基团。这些基团在图1中由嵌段C示意性地表示。不期望被任何理论束缚,据信在所述制备方法中,这些基团将朝向胶囊的核一侧取向,提供疏水性内部。通过采用这些疏水性嵌段,由于憎水性能,增强了所述空心球在生物环境中的稳定性。
具有亲水性嵌段的聚合物和疏水改性的聚合物的组合的使用在根据本发明的方法中是优选的。还可预期使用同时包含亲水性基团和疏水性部分的聚合物,如图1中下格所示。
空心球的核与壳之间的比例由溶液中聚合物与疏水性化合物之间的关系决定。优选地,该比例为2∶1-1∶5w/w,更优选1∶1-1∶4w/w。
优选地,具有亲水性嵌段的第一聚合物(1)和不与水混溶的第二聚合物(2)之比在2∶1-1∶12w/w范围,更优选1∶3-1∶10w/w。本领域技术人员理解,添加具有疏水性或亲水性性质的额外聚合物,或添加一种同时具有亲水性和疏水性性质的聚合物落在本发明的范围内。
不同于采用几乎不与水溶液混合的溶剂生产乳液,我们采用与水完全混合的亲水性有机溶剂。根据本发明水混溶性是指在水中在所有浓度的完全混溶性。这类溶剂的例子为丙酮、四氢呋喃、二甲基亚砜、二甲基甲酰胺、乙腈、二醇醚和二甲基乙酰胺。二醇醚包括二甲醚、三甘醇二甲醚和乙二醇二甲醚。由于这些溶剂也是聚合物的良溶剂,所以通过将所述溶液与水混合除去亲水性溶剂导致聚合物在疏水性溶剂液滴上的直接沉淀。
所述疏水性化合物被包括以作为成壳聚合物的不良溶剂。这类物质的选择应使得其能够采用冷冻干燥除去。因此,待除去的化合物应能够变为固体,并在冷冻干燥条件下具有适当的蒸气压,从而可将其除去。优选地,该疏水性化合物为烷烃或油。环辛烷和环癸烷是适宜的例子,因为它们可采用冷冻干燥完全除去,从而所述球适于进一步处理。
步骤(b)
为制备与水的混合物,优选将所述溶液通过另一种形式的搅动/剪切力搅拌或混合。任选地包括进一步的混合处理。用于获得这样的混合物的适宜设备例如选自胶体磨、均化器、超声破碎器(sonicaters)。任选地,在这种处理前或后将该混合物加压通过玻璃过滤器。若需要,这种处理可重复多次。不期望被任何理论束缚,据信所述亲水性溶剂将立刻与水相混合,从而乳液内相中壳组合物的浓度升高到超过溶解性阈值,和在此时所述壳组合物将立即沉淀。
这种沉淀导致在包含所述疏水性溶剂的乳液内相(液滴)的表面处形成聚合物壳。据信一旦亲水性溶剂汽化,就产生壳组合物,其覆盖包含非溶剂和可能已在所述方法的较早阶段添加的任选的其它成分的核。
步骤(c)
在根据本发明方法的步骤(c)中,将亲水性溶剂从水相除去,例如在减压下。另一种除去亲水性溶剂的适宜方法为提高温度,例如将温度提高至25-35℃的温度,或简单地通过搅拌该混合物给定量的时间。
步骤(d)
通过采用具有亲水性嵌段的聚合物,不再需要添加稳定剂,导致步骤(d)的样品可被直接冷冻干燥,无需采用任何纯化步骤。通过冷冻干燥,除去了构成所述球的核的疏水性化合物,形成气体填充的或气体部分填充的球形颗粒。另外,优选聚合物的该非溶剂升华而非蒸发。所以,优选熔点不远小于0℃的非溶剂。也可考虑可在冷冻干燥过程中除去的其它非溶剂,其中据信,优选但并不总是绝对必要的是所述非溶剂在冷冻干燥过程中为固体。例如,可使用具有8-14个碳水化合物部分的烷烃或环烷烃。
迄今为止,尚不存在尺寸低于微米范围,特别是50-400nm的超声造影剂的有效合成路径。这已经妨碍了对超声对中空纳米胶囊的影响的理解。对于游离气泡,如果尺寸降至100或200nm,则共振频率增加到超过用于成像的范围。以聚合物为壳的试剂不仅共振,而且由超声诱导的壳破坏导致的气体逸出可表现出非常不同的频率相关性。由于它们的高Laplace压力和发生Ostwald熟化现象,以类脂为壳的纳米尺寸的试剂并不非常稳定。这导致小球的损失和大球的生长。
应该注意的是,在循环中来自纳米球的背景信号的缺失在一些药物递送方案中可能是有利的,其中对于检测,仅经活化的纳米球(气体已经从聚合物壳释放的那些)是所希望的。
超声造影剂被研究用于很多新应用如分子成像、药物递送及它们的组合。对于这些应用,需要约数小时的长循环时间以在感兴趣的区域得到有效的累积,这是用于其它模式的经靶向的造影剂(MRI试剂)和药物递送载体的情况。增加的循环时间可通过将试剂的尺寸降至远低于1um实现。根据本发明的方法促进了这些具有低于微米范围的尺寸,优选50-400nm的稳定空心球的合成。此外,立体亲水性嵌段如PEG通过降低所述球经RES的摄取增加了循环时间。
可制备具有与所述球直接结合的药物或造影剂的药物递送载体。这些药物或造影剂可结合于壳中,作为药物承载颗粒如脂质体附着于壳上,或包封于核中(在壳内)。如果药物结合于核中,则一个优选的选择是使它们溶解在不能被冷冻干燥的溶剂中。这类溶剂的例子为例如十六烷或类脂类例如甘油三酯如三辛酸甘油酯。将要包含在这样的体系中的药物优选为疏水性药物,紫杉醇是常见的例子。对于亲水性药物,将包含亲水性药物的脂质体附着于外壳上的选择是最优选的。
另一种可行途径是使冷冻干燥的纳米球再悬浮,然后在小瓶内或通过并置给予技术(连接至相同的静脉或动脉入口的不同注射泵)与期望的药物或生物制剂组合。该“邻近”递送技术利用血管壁和局部细胞渗透性的提高来联合给予药物或生物制剂。
在结合药物后,纳米泡的特定靶向同样是可行的。这可例如通过生物素抗生蛋白链菌素偶合剂促进。可合成生物素化的PLA-PEO,可向其添加抗生蛋白链菌素,然后连接生物素化的抗体或其片段。直接偶合也是可行的,例如采用硫酯。其它靶向试剂是可行的,但不限于抗体片段或化学正交反应对,例如膦和叠氮基团,以促进Staudinger连接或反应。
本发明包括通过纳米沉淀方法由在非卤化有机溶剂中的聚合物溶液不添加稳定剂方便地形成气体填充的球的方法,形成疏水性溶剂填充的以聚合物为壳的胶囊。该方法直接继之以采用冷冻干燥过程除去疏水性溶剂。
应理解,尽管对于本发明方法和空心球在这里已经讨论了优选实施方式、具体混合物和材料,但是在不脱离本发明的范围和精神的情况下,可实施形式和细节的各种改变、改进或组合。
实施例
合成
将平均分子量为2000-5000的疏水改性的PLLA(L-聚丙交酯)的丙酮溶液与分子量比为5000∶700的PLLA和PEO的嵌段共聚物或分子量比为1800∶2000的PLLA和PEO(聚氧化乙烯)的嵌段共聚物混合。将环癸烷添加至该溶液中,并将该溶液与水充分混合并施压通过1μm过滤器。通过在减压下在旋转烧瓶中蒸发除去丙酮。添加作为冷冻干燥添加剂的聚乙二醇,并如别处所述实施冷冻干燥。动态光散射测量显示冷冻干燥后200nm的流体动力学直径。在离心时观察到纳米泡上浮至管上部,差示扫描量热法表明已经除去了环癸烷。由于颗粒上浮至上部,这表明它们的密度低于1g/cm3,并且由于聚合物的密度为约1.25g/cm3,所以这表明气体与所述结构相关联。
表格显示了已经制备的许多组合物,包括冷冻干燥前和后通过动态光散射测量的平均泡尺寸,包括多分散指数,所有这些组合物在再悬浮后都显示出漂浮的颗粒,表明包含气体:
超声测量
采用在32周期脉冲长度使用的1.0MHz空穴换能器(Panametrices V392)建立聚焦声场。采用被动声波检测器检查经活化的微胶囊的行为。所述被动检测器由具有5MHz中心频率的宽带聚焦换能器(直径3.8cm,焦距5.1cm)(Panametrics V307)和宽带低噪声信号放大器(40dB)构成。使用3.0MHz的高通滤波器(TTE HB5-3M-65B)和10.7MHz的低通滤波器(MiniCircuits BLP-10.7),来除去来自空穴换能器的直接传输的衍射诱导的1.0MHz声学信号。使用数字示波器(Model LT374L,LeCroy,Chestnut Ridge,NY)采用20MHz取样频率数字化经放大的散射信号。
使用时间调制器(Four Channel Digital Delay/Pulse Generator;Standford Research Systems DG535)在2.0Hz的PRF(脉冲重复频率)同步声波检测器与活化超声脉冲。活化换能器水平安装在矩形测试室(20.2×20.2×9.6cm3)的侧壁上,而声波检测器垂直放置,并与空穴换能器成直角共焦排列。由于发射和接收换能器均为聚焦换能器,所以检测器仅对在所述两个换能器的小共焦区域内的球非常敏感。采用所述被动技术,微胶囊的活化阈值和活化后振动[=游离(逸出)气体泡的振动]或活化诱导的破坏可通过辨别收到的声学信号的波形和通过经由信号谱分析谐波和噪音产生来研究。采用LabView通过自动计数收到的被散射的信号,测量对于1.0MHz猝发声列(toneburst)的每100个声波作用微胶囊的活化事件计数(或相对活化率)。
对于声学活化测量,将包含1±0.1mg纳米泡的各样品瓶采用5ml空气饱和的去离子水复原。将10μL量的该悬浮液注入用空气饱和的水填充的3.4L槽中,使之静置整夜,以最小化微小气泡的存在。在采用1MHz、32周期超声猝发声列的声波作用产生纳米泡的声学活化的同时,根据来自经活化的纳米泡的散射信号的3-8MHz之间的高谐波含量,检测各声学事件。
图2给出了样品mPEG(2000)PLA(1800)/PLA-PFO的活化曲线的例子,具有1∶3的聚合物/环癸烷比例,以及1∶9的PEG化对疏水改性pla。将声学事件计数作为在1MHz的以MPa为单位的负声压振幅(或机械指数)的函数做图,其它组合物表现出类似的性质(将在再次测量它们中的一些后添加)。负声压振幅是纳米泡解体和随后气体从破裂的纳米泡释放(所谓“活化”)的直接原因。另外,在具有P4-2探针的HDI-5000超声扫描器上观察到这些纳米泡的强烈活化(作为当在1.2的MI暴露于超声脉冲时的明亮闪光)。
体内实验
常规超声造影剂的循环时间通常低于30分钟。为显示通过本发明方法合成的球的延长的循环,实施以下实验。形成加载有荧光染料尼罗红的球,采用环辛烷与十六烷的1∶1混合物形成核,通过冷冻干燥从其除去环辛烷级分。将样品在盐水中复原,并将100μl眼球后注入采用异氟烷麻醉的C57BL/6小鼠中。注射后4和7小时,采集血液样品并杀死动物。取肝、肺、脾、肾和心脏并称重。添加Triton X100(250μl,在水中3%),然后添加750μl水。将所述器官用超声破碎器(Sonicator Heat Systems W375细胞粉碎器)粉碎,冷冻干燥并采用异丙醇提取尼罗红。离心后,测量荧光(激发570nm,检测630nm,截止590nm)。由这些值和样品(器官或血液)重量,确定尼罗红的相对量。
测定4和7小时后的分布,各五只小鼠,显示主要存在于肝脏和血液中。7小时后肝脏与血液之比更高,表明发生缓慢的肝脏累积,然而,即使在7小时后,在循环中仍然存在显著部分。
Claims (14)
1.用于合成具有聚合物壳的空心球的方法,其包括以下步骤:
提供溶液,该溶液包含:
-具有亲水性嵌段和疏水性嵌段的第一聚合物(1),
-不与水混溶的第二聚合物(2),
-疏水性化合物,
-水混溶性有机溶剂,
将所述溶液与水溶液混合得到混合物
从所述混合物除去所述水混溶性有机溶剂
冷冻干燥以从所述混合物除去所述疏水性化合物。
2.根据权利要求1的方法,其中所述亲水性有机溶剂选自包括丙酮、四氢呋喃、二甲基亚砜、二甲基甲酰胺、乙腈、二醇醚和二甲基乙酰胺或它们的任意组合的组。
3.根据权利要求1-2任意一项的方法,其中所述第一聚合物(1)选自包括聚丙交酯、聚己内酯、聚氰基丙烯酸酯、和前述之一的共聚物、聚乙交酯的共聚物或它们的任意组合的组。
4.根据权利要求1-3的方法,其中所述第二聚合物(2)包括烷基端基或氟化端基。
5.根据权利要求1-4任意一项的方法,其中所述混合物包括在步骤(d)后保留的疏水性化合物,和药学或诊断化合物。
6.根据权利要求5的方法,其中所述保留的疏水性化合物选自包括以下的组:具有至少16个碳原子的烷烃,类脂,石蜡或油。
7.根据前述权利要求任意一项合成的具有聚合物壳的空心球。
8.根据权利要求7的空心球,具有50-400nm的尺寸。
9.根据权利要求7和8任意一项的空心球,其在施加超声波时能够释放气态内容物。
10.根据权利要求7-9任意一项的空心球,其中所述气态内容物至少部分地被气体前体替换。
11.根据权利要求7-10任意一项的空心球,其中所述球的聚合物壳包括靶向部分。
12.根据权利要求7-11任意一项的空心球,其中所述球包含药物活性分子。
13.根据权利要求7-12任意一项的空心球,包括选自PET、SPECT或MRI试剂的成像剂。
14.通过权利要求1-6任意一项的方法得到的或权利要求7-13限定的空心球在超声成像或超声触发的药物递送中的用途。
