Abstract

本发明涉及一种健康查体粪便致病菌检测筛选增菌培养液,用于对健康人的粪便样品中携带致病菌的检验。该培养液可将数份健康查体者的粪便进行致病菌筛选检测。该增菌培养液配制简单、选择性强,与传统培养基比较,检出率高,污染小,临床应用效果满意,大大提高检测阳性率,试验方法简单、时间短、准确性高。

健康查体粪便致病菌检测筛选增菌培养液
技术领域本发明涉及一种健康查体粪便致病菌检测筛选增菌培养液,其主要成分是胰蛋白胨、乳糖、枸橼酸钠、去氧胆酸钠、胆盐、硫酸铝钾、磷酸氢二钾、磷酸二氢钾、水杨苷、土温-80、Triton X-100、甘氨酸、氯化钠、十二烷基硫酸钠、蒸馏水制备而成;用于对健康人的粪便样品中携带致病菌的检验。从业人员健康体检中,肝炎、痢疾、伤寒、肺结核、皮肤病(简称“五病”),伤寒、痢疾杆菌的检出率明显低,分析其原因,除了就业人群的带菌率可能处于一个很低的水平之外,也与检验方法有关,一方面采便方法影响了检验结果,由于多数人有早晨排便的习惯,上午查体的肛拭采到的粪便较少;另外培养方法也有一定的影响,如果不进行增菌培养而直接将大便标本划碟培养,可能由于正常的肠道菌生长能力强,将致病菌掩盖,也可以造成假阴性。如威海市监测到的2例副伤寒带菌者,第一次未增菌培养结果阴性,重复增菌后培养结果才显示阳性,证实为带菌者,为此完善检验方法,避免造成漏检是相当必要的。而任何一个粪便样品的增菌培养的实验,都必须有可靠的培养基,而培养基是人工配制的适合微生物生长繁殖或积累代谢产物的营养基质,就营养物质而言,一般不外乎碳源、氮源、无机盐、生长因子及水等几大类。在配制培养基时,根据各类微生物的特点,就可以配制出适合不同种类微生物生长发育所需要的培养基。培养基除了满足微生物所必需营养物质外,还要求有一定的酸碱度和渗透压。各类微生物对营养的要求不尽相同,因而培养基的种类繁多。培养基的制备程序方法,可因其种类不同而各异,即使同一种配方的培养基,因工艺和使用方法不同,也会有不同的效果。实验工作者一直希望能有一种能克服上述不足的新培养基,专用于健康查体粪便致病菌检测筛选的增菌液。该培养液可将数份健康查体者的粪便进行致病菌筛选检测。
背景技术本发明的目的在于提供一种健康查体粪便致病菌检测筛选增菌液,该培养基营养丰富,适宜肠道菌的测定;用于样品增菌培养,可将数份健康查体者的粪便进行致病菌筛选检测。也可用于中和样品增菌培养。配制简单、选择性强,与传统培养基比较,检出率高,污染小,临床应用效果满意.大大提高检测阳性率,试验方法简单、时间短、准确性高。
发明内容本发明的要点在于选择合适的组分,并进行合理混配,经加温、溶解、混合、冷却、分装、高温灭菌等工艺而成一种十二烷基硫酸钠中和增菌肉汤。
本发明选择的培养基配方如下(克/每升蒸馏水):胰蛋白胨20.0-22.0;乳糖5.0-5.5;磷酸氢二钾2.70-2.75;磷酸二氢钾2.70-2.75;枸橼酸钠0.4-0.5;去氧胆酸钠0.04-0.06;水杨苷0.5-0.6;土温-801.0-1.2;Triton X-1001.0-1.2;胆盐0.2-0.4;硫酸铝钾0.2-0.3;甘氨酸0.5-0.6;氯化钠5.0-5.5;十二烷基硫酸钠0.1-0.2;蒸馏水至1000mL。
胰蛋白胨对接种少量大肠菌群可刺激其生长和产气。需氧芽孢杆菌则全被抑制,可直接用于吲哚试验,做吲哚试验时,应预先加入少量乙醚并震摇后,再加上一层吲哚试剂,不在震摇,视二液接触面有无紫红色出现,若不加乙醚,则易形成持久的乳化而干扰吲哚试验,这是因存在十二烷基硫酸钠的关系。
本培养基含有枸橼酸钠和去氧胆酸钠,对革兰氏阴性菌有抑制作用,大肠杆菌,绿脓杆菌及变形杆菌,在接种6小时内生长缓慢,而痢疾杆菌可相对地得到增殖.。胆盐可抑制大多数革兰氏阴性杆菌,而对伤寒杆菌无抑制作用或抑制力很小;硫酸铝钾可激活伤寒杆菌活性,使生长数量大大增加十二烷基硫酸钠中和增菌肉汤,原材料易得,制作方便,基础成分加入营养丰富的牛肉浸粉、酵母浸膏粉,酵母浸膏粉主要为微生物生长繁殖提供可溶性含氮物质、核酸降解物、维生素及无机盐类等,以补充蛋白胨营养成分的不足部分。
水杨苷、土温-80、Triton X-100、甘氨酸中和样品中磺胺类药物及消毒剂的抑菌作用;十二烷基硫酸钠破坏样品中存在的四环素、金霉素、新霉素、多黏菌素及链霉素的抑菌作用。
本发明产品的制备方法是:
1)、各成分按比例混合于水中,加热溶解,矫正pH至7.8,分装于试管中,每管10mL,121℃15min高压灭菌备用。
2)、质控菌株为产气大肠杆菌(ATCC13048)、大肠埃希菌(ATCC25922)、鼠伤寒沙门菌(ATCC14028)、金黄色葡萄球菌(ATCC25923)与相应合格对照培养基平行进行细菌生长的灵敏度试验,应符合要求。
本发明具有以下优点:(1)健康查体粪便致病菌检测筛选增菌液,营养丰富、配制简单;(2)选择性强、检出率高、污染小,可将数份健康查体者的粪便进行致病菌筛选检测,临床应用效果满意;(3)大大提高检测阳性率,方法简单、时间短、准确性高。(4)灵敏度高、特异性强、稳定性好。
具体实施方式下面结合实施例对本发明作进一步的详细说明:
按照下表所列数据(克、ml/每升蒸馏水)及其所述步骤进行配置:
配方一:
胰蛋白胨20.0-22.0;乳糖5.0-5.5;磷酸氢二钾2.70-2.75;磷酸二氢钾2.70-2.75;枸橼酸钠0.4-0.5;去氧胆酸钠0.04-0.06;水杨苷0.5-0.6;土温-801.0-1.2;TritonX-1001.0-1.2;胆盐0.2-0.4;硫酸铝钾0.2-0.3;甘氨酸0.5-0.6;氯化钠5.0-5.5;十二烷基硫酸钠0.1-0.2;蒸馏水至1000mL。
配方二:
胰蛋白胨21.0-22.0;乳糖5.2-5.5;磷酸氢二钾2.73-2.75;磷酸二氢钾2.72-2.75;枸橼酸钠0.42-0.5;去氧胆酸钠0.05-0.06;水杨苷0.55-0.6;土温-801.1-1.2;TritonX-1001.1-1.2;胆盐0.3-0.4;硫酸铝钾0.25-0.3;甘氨酸0.55-0.6;氯化钠5.2-5.5;十二烷基硫酸钠0.15-0.2;蒸馏水至1000mL。
配方三:
胰蛋白胨20.0-21.0;乳糖5.0-5.3;磷酸氢二钾2.70-2.73;磷酸二氢钾2.70-2.74;枸橼酸钠0.4-0.45;去氧胆酸钠0.04-0.05;水杨苷0.5-0.55;土温-801.0-1.1;TritonX-1001.0-1.1;胆盐0.2-0.3;硫酸铝钾0.2-0.25;甘氨酸0.5-0.55;氯化钠5.0-5.3;十二烷基硫酸钠0.1-0.15;蒸馏水至1000mL。

1.本发明涉及一种健康查体粪便致病菌检测筛选增菌培养液,其特征在于具有如下配方(克/每升蒸馏水):胰蛋白胨20.0-22.0;乳糖5.0-5.5;磷酸氢二钾2.70-2.75;磷酸二氢钾2.70-2.75;枸橼酸钠0.4-0.5;去氧胆酸钠0.04-0.06;水杨苷0.5-0.6;土温-801.0-1.2;Triton X-1001.0-1.2;胆盐0.2-0.4;硫酸铝钾0.2-0.3;甘氨酸0.5-0.6;氯化钠5.0-5.5;十二烷基硫酸钠0.1-0.2;蒸馏水至1000mL。
2.一种权利要求1所述健康查体粪便致病菌检测筛选增菌培养液的制备方法,其特征在于具有如下的步骤:
1)、各成分按比例混合于水中,加热溶解,矫正pH至7.8,分装于试管中,每管10mL,121℃15min高压灭菌备用;
2)、质控菌株为产气大肠杆菌(ATCC13048)、大肠埃希菌(ATCC25922)、鼠伤寒沙门菌(ATCC14028)、金黄色葡萄球菌(ATCC25923)与相应合格对照培养基平行进行细菌生长的灵敏度试验,应符合要求。
