本发明涉及一种鳃叶状触角的暗黑鳃金龟的触角电位测定方法,该方法为:将已自基部切下的暗黑鳃金龟触角,继续切下鳃叶状的端部后,将闭合的3片鳃叶用昆虫针分离开,在记录电极上缠上铂金丝以增加触角长度。
Description
技术领域
本发明涉及利用触角电位技术,具体来说是摸索一种鳃叶状触角的暗黑鳃金龟的触角电位测定方法。
背景技术
蛴螬是一种地下害虫,对多种草坪、作物和果树造成毁灭性危害。暗黑鳃金龟是危害花生、地瓜、玉米等作物的的优势种,占地下害虫70%以上。近年来,遭受暗黑鳃金龟危害的花生一般减产20%,严重的减产60%~70%,甚至绝收,同时花生的品质也明显下降,秕仁、空壳大量增加。暗黑鳃金龟已成为制约花生生产的最主要因素,也是目前广大农民种植花生最忧心的问题。据我国山东、河南、安徽、河北、辽宁等省连续五年的跟踪调查,每年因暗黑鳃金龟危害造成的花生产量损失都在1.5亿公斤以上。防治蛴螬已成为我国花生生产迫在眉睫的重大课题,防治形势刻不容缓!
地下害虫地上治,对于在地下完成幼虫生活史的暗黑鳃金龟而言是最安全的防治手段。土壤施药不仅对药效的残效期和剂型有很高的要求,而且对花生和土壤的农药残留影响比地上害虫类要高的多,因此利用暗黑鳃金龟成虫晚上出土交配、取食的习性,对成虫进行诱杀,起到安全高效的目的。利用性诱剂诱杀害虫在国外防治林木害虫是很成熟的手段之一,触角电位技术是记录昆虫触角在被施加不同刺激物时的生理变化,用于研究昆虫的电生理反应,在本项目中用于研究暗黑鳃金龟触角对性信息素的反应,可为确定候选化合物、鉴定性信息素活性组分提供理论依据,为进一步筛选性信息素各组分的组合、最佳剂量、最佳配比的研究奠定基础。
在以往的报道中,有将鳃叶状的端部小部分切除和将鳃叶状的端部完全切除,仅留鞭节部分两种方法,本研究将这三种方法同时进行,来确定哪种方法最好。
发明内容
有鉴于此,本发明的主要目的在于提供一种鳃叶状触角的暗黑鳃金龟的触角电位测定方法。
为达到上述目的,本发明的技术方案是这样实现的:
选用健壮的暗黑鳃金龟雄虫为青岛莱西市山东花生研究所实验基地人工捕捉;二氯甲烷(进口色谱纯);巴斯德管(200℃,2h烘干);剪好的滤纸片(0.5cm×2cm);刺激样品;刀片;眼科剪;导电胶;昆虫针;微量进样器;触角电位仪(产自荷兰Syntech公司,金属电极)。
将从1头健壮的暗黑鳃金龟雄虫基部用眼科剪迅速切下的触角,选用3种触角处理方法,分别为:将鳃叶状的端部小部分切除;将鳃叶状的端部完全切除,仅留鞭节部分;将鳃叶部分切下小部分端部,将闭合的3片鳃叶分离开。
因暗黑鳃金龟触角较短,需要在记录电极缠上铂金丝变相增加触角长度。
具体实施方式
下面结合具体实例对本发明作进一步详细的描述。
选用三种方法处理触角,结果如下:
测定可知前两种方法所得结果处理和对照数值无显著差异,统计无意义。以对异亮氨酸甲酯母液的EAG反应值为例,附上三组3方法所测得的数值如下:

上表中7∶1的二元混合物的电生理净反应值与粗提液的相当,可知该比例为这两种组分的最佳配比。由此可知方法三是一种较适合测定暗黑鳃金龟类触角电位的方法。

1.一种鳃叶状触角的暗黑鳃金龟的触角电位测定方法,其特征在于:将已自基部切下的暗黑鳃金龟触角,继续切下鳃叶状的端部后,将闭合的3片鳃叶用昆虫针分离开,在记录电极上缠上铂金丝以增加触角长度。
2.如权利要求1所述鳃叶状触角的暗黑鳃金龟的触角电位测定方法,其特征在于:将鳃叶状的端部小部分切除。
