Abstract

本发明涉及医学领域,公开了一种用于小型动物肝脏淋巴细胞分离的肝脏灌注方法,将动物处死后,经后腔静脉进行灌注;本发明方法操作简便、快速,循环血液排除充分,且不需要特殊设备,实验成本低,能够达到理想的肝灌注效果,适合于大多数实验室在进行小鼠等小型动物的肝脏淋巴细胞分离时采用,具有良好的推广应用价值。

技术领域
本发明涉及医学领域,特别涉及用于小型动物肝脏淋巴细胞分离的肝脏灌注方法。
背景技术
正常的肝组织由动脉和门静脉供血,其中70%-85%的供血来源于门静脉。门静脉收集来自胃、小肠、大肠(直肠后部除外),胰和脾的血液,经肝门入肝,在肝内反复分支汇入窦状隙,最后集合为数支肝静脉汇入后腔静脉。
肝脏淋巴细胞与肝脏疾病关系密切,可能参与了肝炎、肝硬化、肝癌等诸多肝脏疾病的发生与发展,此外,与自身免疫性疾病和器官移植免疫等也有一定关系。要深入研究肝脏淋巴细胞在上述疾病中的表型变化和意义,前提是要建立有效的肝脏淋巴细胞分离方法。影响肝脏淋巴细胞分离的因素较多,其中肝脏的预处理最为重要。国外常用门静脉肝灌注法以排除过客淋巴细胞的影响,但是该技术较难操作,特别是当实验动物为小鼠等体型较小的动物时,由于其门静脉很细,解剖结构位置较深,需仔细剥离周围韧带,操作难度很大,而且穿刺针不易固定,容易脱落和刺破门静脉,经常会出现穿刺针刺入小叶分支静脉而导致各叶肝产生大小不等的盲区(即有血液残留的区域)的情况,由此造成循环血液排除不充分,后续分离得到的肝脏淋巴细胞中混杂有过客淋巴细胞,从而对实验结果的准确性造成明显影响;此外,门静脉肝灌注法还需要恒流泵等灌流装置以及外科显微镜等,操作比较繁琐、耗时。
发明内容
有鉴于此,本发明的目的在于提供一种用于小型动物肝脏淋巴细胞分离的肝脏灌注方法,可以克服门静脉肝灌注法在小鼠等小型实验动物体上操作时存在的缺陷,适用于小型实验动物的肝脏淋巴细胞分离,可以充分排除肝内循环血液以避免过客淋巴细胞的干扰,同时操作简便、快速、成本低。
为达到上述目的,本发明采用如下技术方案:
用于小型动物肝脏淋巴细胞分离的肝脏灌注方法,将动物处死后,经后腔静脉进行肝脏灌注。
进一步,所述肝脏灌注方法是将动物脱颈处死后,消毒,解剖腹部,暴露后腔静脉,将穿刺针自后腔静脉远端刺入,缓慢注入灌注液至可见肝脏表面毛细血管网,肝脏由深红色褪色至无灌注盲区为止。
本发明的有益效果在于:
本发明的肝脏灌注方法与门静脉肝灌注法比较,具有如下优点:(1)由于后腔静脉较门静脉粗,分离及穿刺操作非常简单,操作时间比门静脉肝灌注法缩短近1小时;(2)远离门静脉进行充分灌注,循环血液排除充分,可以避免未灌注完全的盲区的存在;(3)可以普通的小儿头皮针及一次性注射器替换恒流泵等灌流装置,可克服恒流泵安装复杂、间断性灌流的缺点,而且降低实验成本,简化操作步骤。研究证明,本发明的肝脏灌注方法能够达到理想的肝灌注效果,适合于大多数实验室在进行小鼠等小型动物肝脏淋巴细胞分离时采用,具有良好的推广应用价值。

具体实施方式
以下将参照附图,对本发明的优选实施例进行详细的描述。
本发明的用于小型动物肝脏淋巴细胞分离的肝脏灌注方法,以已经死亡的动物为实施对象,经后腔静脉进行灌注。
本实施例是将Balb/c小鼠脱颈处死后,用湿润的酒精棉球擦身消毒,解剖腹部,将胃肠移向左侧,暴露后腔静脉,将接有一次性注射器(体积为15ml)的4号小儿头皮针自后腔静脉远端刺入,并用眼科弯镊夹住以固定,缓慢注入预冷的Hank’s平衡盐溶液至可见肝脏表面毛细血管网为止,实验过程中可见肝脏逐渐膨胀,并由深红色逐渐褪色,最后整个肝脏呈均匀土黄色,基本无盲区存在。
参见图1-图5,图1为小鼠门静脉和后腔静脉解剖示意图,从图可知,门静脉为分支静脉,很细,且解剖位置较深(图中是将肝脏贴向膈面以暴露肝底),故悬空穿刺且视野较暗,操作极为不便;而后腔静脉为主静脉,很粗浅,将肠管翻向右侧即可见,且其紧贴腹后壁,穿刺固定非常容易;图2为小鼠灌注前的肝脏解剖示意图,可见小鼠灌注前的肝脏呈深红色;图3为小鼠经后腔静脉肝灌注后的肝脏解剖示意图,可见小鼠灌注后的肝脏呈均匀土黄色;图4为经后腔静脉肝灌注后分离出的小鼠完整肝脏,可见各叶肝均灌注良好,无盲区存在;图5为经门静脉肝灌注后分离出的小鼠完整肝脏,方框所指区域即为未灌注完全的盲区。
最后说明的是,以上实施例仅用以说明本发明的技术方案而非限制,尽管通过参照本发明的优选实施例已经对本发明进行了描述,但本领域的普通技术人员应当理解,可以在形式上和细节上对其作出各种各样的改变,而不偏离所附权利要求书所限定的本发明的精神和范围。

2.权利要求1所述的用于小型动物肝脏淋巴细胞分离的肝脏灌注方法,其特征在于:将动物脱颈处死后,消毒,解剖腹部,暴露后腔静脉,将穿刺针自后腔静脉远端刺入,缓慢注入灌注液至可见肝脏表面毛细血管网,肝脏由深红色褪色至无灌注盲区为止。
