本发明涉及一种活体皮肤组织细胞聚合物,通过以下步骤制备:在正常皮肤上形成去表层的创面;用多锋刀纵横切划创面,切划深度为0.3-0.5mm之间,一边切划,一边收集刀具上的皮肤组织碎粒,将所收集的皮肤组织碎粒浸在含肝素的盐水中,洗去血凝块;随后将上述所有碎粒集中,放置于杯背底上,使用眼科剪将上述碎粒反复剪细,并反复冲洗,剔除带有的脂肪细粒;经过上述6-8次剪洗后,再将上述含有皮肤组织细胞的碎粒集中,即得到具有粘附性的皮肤组织细胞聚合物。本发明制备的皮肤组织细胞聚合物应用于疤痕修复技术,种植上去的细胞能够完全覆盖创面,形成正常或接近正常的皮肤,可以修复面部多个凹陷性疤痕和多条线状疤痕,不受疤痕数量的限制。

一种活体皮肤组织细胞聚合物的制备方法及其用途
技术领域
本发明涉及一种活体皮肤组织细胞聚合物的制备方法及其在疤痕修复技术中的用途。
技术背景
疤痕的形成原因是正常皮肤组织的缺陷不能完全愈合,由纤维组织替代修复创面而形成疤痕。常规治疗疤痕的方法如切缝法治疗疤痕,是将切除疤痕后两侧创缘拉拢缝合,因为皮肤量本来就不够,两侧拉拢后会更紧,医学上称为张力大,拆线后,切口线易裂开、增生,从而达不到祛疤目的;还有一种治疗疤痕的方法是植皮,该法的缺点之一是植上去的皮肤的颜色与周边皮肤差异大,四边留下缝合针迹疤,而且还会在取皮处还会留下更大的疤痕。
发明内容
本发明的目的是提供一种活体皮肤细胞组织细胞聚合物的制备方法,及将由该方法制备的活体皮肤细胞组织细胞聚合物应用于人体脸面部疤痕修复技术,可以达到很好地修复脸、面部小面积凹陷性疤痕、外伤后疤痕、外伤后粉尘染色及手术后刀疤的效果。
本发明所述的一种活体皮肤细胞组织细胞聚合物通过以下方法制备:
在正常皮肤上形成去表层的创面;用多锋刀纵横切划创面,切划深度为0.3-0.5mm之间,一边切划,一边收集刀具上的皮肤组织碎粒,将所收集的皮肤组织碎粒浸在含肝素的盐水中,洗去血凝块;随后将上述所有碎粒集中,放置于杯背底上,使用眼科剪将上述碎粒反复剪细,并反复冲洗,剔除带有的脂肪细粒;经过上述若干次剪洗后,再将上述含有皮肤组织细胞的碎粒集中,即得到具有粘附性的皮肤组织细胞聚合物。
上述聚合物含有皮肤真皮乳头层和表皮层及小部份真皮基底层的成份,含有大量的皮肤组织干细胞,其厚度相当于中厚皮片,因此这些聚合物在种植于去疤痕的创面上后,能长成正常皮肤,修复创面。上述制得的皮肤组织细胞聚合物通过下述方法应用于疤痕修复技术:
将通过上述方法制备的皮肤组织细胞聚合物平铺在上述切疤后的创面上,以稍高出周边皮肤平面为佳;盖上无菌油纱面,洒上50%葡萄糖针剂;外盖敷料。术后第十三天分层揭去全部敷料,可见创面平整,种植上去的皮肤组织聚合物全部成活,形成正常或接近正常皮肤。
将本发明制备的皮肤组织细胞聚合物应用于疤痕修复技术,既没有增加皮肤张力,不用缝合,所以不存在切口裂开增生,也不会有缝合线迹;其次,取的是很细小的皮肤组织细胞,取处能够完全正常愈合,不会留下疤痕,再者,切除疤痕后,种植上去的细胞能够完全覆盖创面。形成正常或接近正常的皮肤,还有一个重要优点是,可以修复面部多个凹陷性疤痕和多条线状疤痕,不受疤痕数量的限制。
具体实施方式
下面结合具体实施例对本发明进一步进行详细说明。
本发明实施例所述的活体皮肤细胞组织细胞聚合物通过以下方法制备:
首先沿疤痕边缘设计切口,局部麻醉后,切除全部疤痕,深度约1-2mm;根据疤痕大小确定需要填充量的多少,再确定提取皮肤组织细胞范围,用钢齿轮磨头磨削机在上述创面四周1-5cm范围内的正常皮肤上形成去表层的创面;随后用多锋刀纵横切划创面,深度为0.5mm,一边切划,一边收集刀具上的皮肤组织碎粒,将所收集的皮肤组织碎粒浸在含肝素的盐水中,洗去血凝块;随后将上述所有碎粒集中,放置于瓷杯背底上,使用眼科剪将上述碎粒反复剪细,并反复冲洗,剔除带有的脂肪细粒;经过上述6-8次剪洗后,再将上述含有皮肤组织细胞的碎粒集中,即得到具有粘附性的皮肤组织细胞聚合物。
将通过上述方法制备的皮肤组织细胞聚合物平铺在上述祛疤后的创面上,以稍高出周边皮肤平面为佳;盖上无菌油纱面,洒上50%葡萄糖针剂;外盖敷料。术后第三天开始干燥,术后第十三天分层揭去全部敷料,可见创面平整,种植上去的皮肤组织聚合物全部成活,形成正常或接近正常皮肤。

Claims (1)

1.一种活体皮肤组织细胞聚合物,其特征在于所述活体皮肤组织细胞聚合物通过以下步骤制备:
(1)使离体的,深度为0.3-0.5mm之间的正常皮肤的皮肤组织碎粒浸在含肝素的盐水中,洗去血凝块;
(2)随后将上述所有碎粒集中,放置于杯底上,使用眼科剪将上述碎粒反复剪细,并反复冲洗,剔除带有的脂肪细粒;
(3)经过上述6-8次剪洗后,再将上述含有皮肤组织细胞的碎粒集中,即得到具有粘附性的皮肤组织细胞聚合物。
