Die vorliegende Erfindung betrifft Kristalle aus einem pharmazeutisch akzeptablen Salz von Citalo-pram 1-[3-(Dimethylamino)propyl]-1-(4-fluorphenyl)-1,3-dihydro-5-isobenzofurancarbonitril. Hintergrund der Erfindung
Citalopram ist ein wohl bekanntes Antidepressivum, das die folgende Struktur hat:

Es ist ein selektiver, zentral wirkender Serotonin-(5-Hydroxytryptamin; 5-HT)-Wiederaufnahmehemmer, der entsprechend antidepressive Aktivitäten aufweist.
Citalopram wurde zuerst in DE 2 657 013 offenbart, entsprechend US-PS 4 136 193. Diese Patentveröffentlichung beschreibt die Herstellung von Citalopram durch ein Verfahren und umreisst ein weiteres Verfahren, das zur Herstellung von Citalopram verwendet werden kann. Das hergestellte Citalopram wurde in kristalliner Form als Oxalat-, Hydrobromid- bzw. Hydrochloridsalz isoliert. Ausserdem wurde die Citalopram-Base als Öl erhalten (Siedepunkt 175 DEG C/0,03 mmHg). Die Veröffentlichung umreisst ebenfalls die Herstellung von Tabletten, die Salze von Citalopram enthalten. Citalopram wird als Hydrobromid bzw. Hydrochlorid in den Verkehr gebracht.
Die Herstellung von kristalliner Citalopram-Base wird in der gleichzeitig anhängigen DK 200 000 402 offenbart. Diese Patentveröffentlichung beschreibt die Herstellung von kristalliner Citalopram-Base und die Verwendung von kristalliner Citalopram-Base als Zwischenstufe in der Reinigung von rohem Citalo-pramhydrobromid zu reinem Citalopramhydrobromid. Die Veröffentlichung umreisst ebenfalls die Herstellung von Tabletten, die Citalopram-Base enthalten.
Citalopram wird in einer Reihe von Ländern als Tablette vermarktet, die durch Verpressen von granuliertem Citalopramhydrobromid, Lactose und anderen Zusatzstoffen hergestellt wird.
Es ist wohl bekannt, dass die Herstellung von Tabletten mit einer reproduzierbaren Zusammensetzung erfordert, dass alle trockenen Bestandteile gute Fliesseigenschaften haben. Für den Fall, dass der Wirkstoff gute Fliesseigenschaften besitzt, können Tabletten durch Direktverpressen der Bestandteile hergestellt werden. Jedoch ist in vielen Fällen die Teilchengrösse des Wirkstoffs klein, der Wirkstoff ist kohäsiv oder hat schlechte Fliesseigenschaften.
Ausserdem werden sich Wirkstoffe mit einer geringen Teilchengrösse, die mit Zusatzstoffen mit einer grösseren Teilchengrösse vermischt werden, typischerweise segregieren oder während des Tablettierungsverfahrens entmischen.
Das Problem der geringen Teilchengrösse und schlechten Rieselfähigkeit wird herkömmlich durch Vergrösserung der Teilchengrösse des Wirkstoffs gelöst, gewöhnlich durch Granulierung des Wirkstoffs entweder allein oder in Kombination mit einem Füllstoff und/oder mit anderen herkömmlichen Tablettenbestandteilen.
Ein solches Granulierungsverfahren ist das "Nass"-Granulierungsverfahren. Bei der Verwendung dieses Verfahrens werden die trockenen Feststoffe (Wirkstoffe, Füllstoffe, Bindemittel etc.) vermischt und mit Wasser oder einem anderen Benetzungsmittel (z.B. einem Alkohol) befeuchtet, und Agglomerate oder Granalien werden aus den angefeuchteten Feststoffen aufgebaut. Das nasse Bearbeiten ("wet massing") der Masse wird fortgesetzt, bis eine gewünschte homogene Teilchengrösse erreicht worden ist, worauf das granulierte Produkt getrocknet wird.
Eine Alternative zum "Nass"-Granulierungsverfahren ist die "Schmelz"-Granulierung, die ebenfalls als "thermischplastisches" Granulierungsverfahren bekannt ist, worin ein niedrigschmelzender Feststoff als Granulierungsmittel verwendet wird. Zunächst werden die trockenen Feststoffe vermischt und erwärmt, bis das Bindemittel schmilzt. Wenn das Bindemittel verflüssigt ist und sich über die Oberfläche der Teilchen ausbreitet, werden die Teilchen aneinander haften und Granalien bilden. Das Bindemittel.verfestigt sich beim Abkühlen unter Bildung eines trockenen körnigen Produktes.
Die Nassgranulierung sowie Schmelzgranulierung sind energieintensive Grundverfahren, die eine komplizierte und teure Ausrüstung sowie technische Qualifikation erfordern.
Das zur Herstellung von Citalopramhydrobromid verwendete Verfahren resultiert in einem Produkt mit einer sehr geringen Teilchengrösse um 2 bis 20 mu m, das wie viele andere teilchenförmige Produkte mit einer geringen Teilchengrösse sehr schlechte Fliesseigenschaften hat. Um eine angemessene Dosierung des Citaloprams während des Tablettierens zu erreichen, wurde es daher als notwendig erachtet, ein Granulat aus Citalopram mit höherer Teilchengrösse und verbesserten Fliesseigenschaften herzustellen.
Die Citalopram-Tablette, die in den Handel gebracht wird, ist eine aus granuliertem Citalopramhydrobromid mit verschiedenen Arzneimittelzusatzstoffen hergestellte Tablette.
Angesichts der Tatsache, dass die Direktverpressung viel einfacher und billiger als die Verfahren ist, die eine Granulierung beinhalten, besteht ein Wunsch nach einem Verfahren zur Direktverpressung von Citalopramhydrobromid.
Die Hindernisse, die bislang die Direktverpressung von Citalopram-Tabletten verhindert haben, wurden nun nach umfassender Forschung im Labor überwunden.
Es wurde gefunden, dass grössere Teilchen, d.h. Teilchen mit einer mit der Grösse des Füllstoffs vergleichbaren Grösse, durch ein neues und erfinderisches Kristallisationsverfahren hergestellt werden können und dass diese Teilchen nützlich zur Herstellung von direktverpressten Tabletten sind. Eine genaue Dosierung in Kapseln kann ebenfalls mit solchen grossen Teilchen erfolgen.
Es wurde ebenfalls gefunden, dass Tabletten mit einer überraschend geringen Variation im Citalo-pram-Gehalt durch Direktverpressen von Citalopramhydrobromid, das eine signifikant kleinere Teilchengrösse als der Füllstoff hat, hergestellt werden können. Die genaue Dosierung in Kapseln kann ebenfalls trotz der geringen Teilchengrösse von Citalopram erreicht werden. Aufgaben der Erfindung
Es ist Aufgabe der Erfindung, grosse Kristalle eines pharmazeutisch akzeptablen Salzes von Citalo-pram bereitzustellen, die zur Verwendung in der Direktverpressung geeignet sind.
Eine weitere Aufgabe der Erfindung ist es, ein Verfahren zur Herstellung grosser Kristalle aus einem pharmazeutisch akzeptablen Salz von Citalopram bereitzustellen. Zusammenfassung der Erfindung
Die Erfindung umfasst damit u.a. Folgendes, allein oder in Kombination:
Kristalle aus einem pharmazeutisch akzeptablen Salz von Citalopram, geeignet zur Verwendung in einer festen Arzneiform, mit einer mittleren Teilchengrösse (Median) von wenigstens 40 mu m.
Ein Verfahren zur Herstellung von Kristallen aus einem pharmazeutisch akzeptablen Salz von Citalo-pram mit einer mittleren Teilchengrösse von wenigstens 40 mu m, die zur Verwendung in einer festen Arzneiform geeignet sind, worin eine Lösung eines pharmazeutisch akzeptablen Salzes von Citalopram in einem geeigneten Lösungsmittelsystem bei einer ersten Temperatur zuerst auf eine zweite Temperatur abgekühlt, dann durch Zugabe von Kristallen aus dem Citalopramsalz geimpft wird, gefolgt von einer Haltezeit bei der zweiten Temperatur und einem kontrollierten Abkühlen auf eine dritte Temperatur, worauf die Kristalle durch herkömmliche Fest/Flüssig-Trennungstechniken isoliert werden.
Die Direktverpressung von Citalopram, einem Füllstoff und anderen pharmazeutisch akzeptablen Arzneimittelzusatzstoffen zu Tabletten hat den grossen Vorteil, dass die Granulierung und ein Trocknungsschritt vermieden werden. Ausserdem ist es nicht länger erforderlich, ein Bindemittel hinzuzugeben, da der Granulierungsschritt vermieden wird.
Wie hier verwendet bedeutet "Direktverpressung", dass die feste Arzneiform durch Verpressen einer einfachen Mischung aus dem Wirkstoff und Arzneimittelzusatzstoffen hergestellt wird, ohne dass der Wirkstoff einem intermediären Granulierungsverfahren unterworfen wurde, um ihn in einem grösseren Teilchen einzubetten und seine Fliesseigenschaften zu verbessern.
Wie hier verwendet bedeutet "Bindemittel" ein Mittel, das in Nass- oder Schmelzgranulierungsverfahren verwendet wird und als Bindemittel im granulierten Produkt wirkt.
Wie hier verwendet bedeutet "Teilchengrösseverteilung" die Verteilung von äquivalenten Kugeldurchmessern, bestimmt durch Laser-Beugung bei 1 bar Dispersionsdruck in einer Sympatec Helos-Ausrüstung. "Mittlere Teilchengrösse" bedeutet entsprechend den Median der Teilchengrössenverteilung.
Wie hier verwendet bedeutet "Rückflusstemperatur" die Temperatur, bei der das Lösungsmittel oder Lösungsmittelsystem bei Atmosphärendruck im Rückfluss steht oder siedet.
In einer Ausführungsform der vorliegenden Erfindung haben die Kristalle eines pharmazeutisch akzeptablen Salzes von Citalopram eine mittlere Teilchengrösse im Bereich von 40 bis 200 mu m, bevorzugt 45 bis 150 mu m und besonders bevorzugt 50 bis 120 mu m.
In einer bevorzugten Ausführungsform der vorliegenden Erfindung sind die Kristalle aus Citalopram-hydrobromid oder Citalopramhydrochlorid, bevorzugt Citalopramhydrobromid.
In noch einer anderen Ausführungsform der vorliegenden Erfindung werden die Kristalle aus einem pharmazeutisch akzeptablen Salz von Citalopram mit einer mittleren Teilchengrösse von wenigstens 40 mu m, die zur Verwendung in einer festen Arzneiform geeignet sind, aus einer Lösung aus einem pharmazeutisch akzeptablen Salz von Citalopram in einem geeigneten Lösungssystem kristallisiert. Das Lösungsmittelsystem kann einen oder mehrere Alkohole und gegebenenfalls Wasser umfassen, bevorzugt ist das Lösungsmittelsystem eine Mischung aus Methanol und Wasser, worin das Methanol:Wasser-Gewichtsverhältnis bevorzugt im Bereich von 5:1 bis 50:1 ist; besonders bevorzugt 10:1 bis 30:1 und am meisten bevorzugt 15:1 bis 25:1.
Das pharmazeutisch akzeptable Salz von Citalopram wird bevorzugt im Lösungsmittelsystem bei einer Temperatur im Bereich zwischen 50 DEG C und der Rückflusstemperatur des Lösungsmittelssystems gelöst, bevorzugt zwischen 60 DEG C und der Rückflusstemperatur und besonders bevorzugt zwischen 64 DEG C und der Rückflusstemperatur. Die verwendeten Mengen des pharmazeutisch akzeptablen Salzes von Citalopram und des Lösungsmittels entsprechen bevorzugt einem Gewichtsverhältnis von Lösungsmittel gelöster Stoff im Bereich von 0,5:1 bis 5:1, besonders bevorzugt 0,7:1 bis 2:1 und am meisten bevorzugt 0,9:1 bis 1,5:1.
Die Lösung aus einem pharmazeutisch akzeptablen Salz von Citalopram wird auf eine Temperatur, die Impftemperatur, im Bereich 20 bis 40 DEG C, bevorzugt 25 bis 35 DEG C abgekühlt, worauf sie mit Citalopram-Kristallen geimpft und bei der Impftemperatur für eine Haltezeit für das Kristallwachstum, im Bereich von 30 min bis 7 Tagen, bevorzugt 1 h bis 4 Tagen und besonders bevorzugt 12 bis 36 h gehalten wird. Nach der Haltezeit wird der Kristallisationsansatz allmählich in einer kontrollierten Weise von der Impftemperatur auf die Temperatur abgekühlt, bei der die Kristalle aus der Mutterlauge isoliert werden, wobei das allmähliche Abkühlen über eine Zeitspanne im Bereich von 5 min bis 6 h, bevorzugt 15 min bis 4 h und besonders bevorzugt 30 min bis 2 h durchgeführt wird.
Die Kristalle aus dem pharmazeutisch akzeptablen Salz von Citalopram werden bevorzugt aus der Mutterlauge bei einer Temperatur im Bereich von 0 bis 20 DEG C, besonders bevorzugt 5 bis 15 DEG C isoliert, wobei herkömmliche Trennungstechniken verwendet werden, z.B. Filtration.
Die kleinen Kristalle aus einem pharmazeutisch akzeptablen Salz von Citalopram, die in einer Ausführungsform der Erfindung verwendet werden, können gemäss den in US-PS 4 136 193 beschriebenen Verfahren hergestellt werden.
Die in einer Ausführungsform der Erfindung verwendeten Kristalle aus Citalopram-Base können gemäss dem im niederländischen Patent Nr. 1 016 435 beschriebenen Verfahren hergestellt werden.
Im Folgenden wird die Erfindung durch Beispiele veranschaulicht. Jedoch sollen die Beispiele die Erfindung bloss veranschaulichen und sollten nicht als Beschränkung aufgefasst werden. Beispiel 1 Kristallisation von Citalopramhydrobromid zu grossen Kristallen
Citalopramhydrobromid (200 g) wird in einer Mischung aus Methanol (200 g) und Wasser (20 g), bei 69 DEG C gelöst. Die Lösung wird auf 30 DEG C abgekühlt, mit Citalopramhydrobromid-Kristallen geimpft und für 24 h bei 30 DEG C gehalten, worauf sie innerhalb 1 h auf 10 DEG C abgekühlt wird. Die Kristalle werden durch Filtration isoliert, mit kaltem Methanol gewaschen und getrocknet. Die Teilchengrössenverteilung für die resultierenden Kristalle ist in Tabelle 1 aufgeführt. Beispiel 2 Kristallisation von Citalopramhydrobromid zu grossen Kristallen
Citalopramhydrobromid (12,0 kg) wird in einer Mischung aus Methanol (12,5 kg) und Wasser (1,2 kg) im Rückfluss gelöst. Die Lösung wird auf 30 DEG C abgekühlt, mit Citalopramhydrobromid-Kristallen (27 g) geimpft und für 16 h bei 30 DEG C gehalten, worauf sie innerhalb 1 h auf 10 DEG C abgekühlt wird. Die Kristalle werden durch Filtration isoliert, mit kaltem (10 DEG C) Methanol (3,5 kg) gewaschen und getrocknet. Die Teilchengrössenverteilung für die resultierenden Kristalle ist in Tabelle 1 aufgeführt. Beispiel 3 Kristallisation von Citalopramhydrobromid zu kleinen Kristallen
Citalopramhydrobromid (200 kg) wird in einer Mischung aus Methanol (170 l) und Aceton (680 l) bei 56 DEG C gelöst. Die Lösung wird auf 15 DEG C abgekühlt, mit Citalopramhydrobronid-Kristallen (50 g) geimpft, Hexan (1600 l) wird allmählich innerhalb von 60 min hinzugegeben, worauf die Suspension unter mässigem Rühren und Abkühlen für 8 h stehen gelassen wird. Die Kristalle werden durch Filtration isoliert, zuerst mit einer kalten (10 DEG C) Mischung aus Aceton (50 l) und Hexan gewaschen, dann mit kaltem (10 DEG C) Hexan (220 l) und getrocknet. Die Teilchengrössenverteilung für die resultierenden Kristalle ist in Tabelle 1 aufgeführt. Beispiel 4 Kristallisation von Citalopram als freie Base
Citalopramhydrobromid (101 g) wird in Wasser (500 ml) und Toluol (500 ml) suspendiert. NaOH (60 ml, 5 N (aq)) wird hinzugegeben, und die Mischung (pH>10) wird für 15 min gerührt, bevor die Phasen getrennt werden. Die organische Phase wird mit Wasser (2 x 100 ml) gewaschen und durch ein Kissen aus Filterhilfe filtriert. Die flüchtigen Stoffe werden im Vakuum entfernt, und die Titelverbindung wird als Öl erhalten. n-Heptan (400 ml) wird hinzugegeben, und die Mischung wird auf 70 DEG C erwärmt. Beim Abkühlen bilden sich Kristalle. Die weissen Kristalle aus Citalopram-Base werden abfiltriert und bei Umgebungstemperatur über Nacht im Vakuum getrocknet. Tabelle 1:

Citalopramhydrobromid-Kristalle aus Beispiel 3 und ProSolv SMCC90 wurden bei 7 U/min für 10 min einem 100 l-Bohle PTM 200-Mischer vermischt. Magnesiumstearat wurde hinzugegeben und das Mischen für 3 min fortgesetzt.
25 kg der resultierenden Mischung wurden auf einer Tablettenpresse Fette P 1200/IC mit 30 Stationen, ausgerüstet mit rechteckigen, erhabenen, gekerbten Stempeln von 5,5 x 8 mm tablettiert (125 000 Tabletten/Stunde). Das Tablettenkerngewicht wurde auf 125 mg eingestellt. Die Nennausbeute betrug 200 000 Tabletten. Die Tablettenpresse wurde laufen gelassen, bis das Mischungsniveau gerade oberhalb der Zwangszufuhr war, d.h. die Tablettierung wurde so lange wie möglich fortgesetzt, um mögliche Segregationstendenzen in den letzten Mengen der Mischung zu identifizieren. Tabletteneigenschaften:

Zerbröckelbarkeit: Nicht erhältlich
Gewichtsvariation: 0,84% relative Standardabweichung (gemessen an 20 Tabletten)
Stempeladhäsion: Nicht beobachtet Citalopram-Gehalt in der Zusammensetzung während des Verpressens
Proben der Tabletten wurden während der Kompression entnommen, um die Segregationstendenz zu messen. Da es eine signifikante Grössendifferenz zwischen dem Wirkstoff, Citalopramhydrobromid, und dem inerten Füllstoff, ProSolv SMCC90, gibt, wie aus Tabelle 1 ersichtlich, würde man erwarten, dass die Komponenten ungleicher Grösse segregieren würden, d.h. sich entmischen würden, während des Übergangs vom Mischbehälter zum Füllschacht der Tablettenpresse oder während des Verbleibens im Füllschacht der Tablettenpresse während des Tablettierens.
Die Probenentnahme wurde 50-mal in regelmässigen Intervallen während des Tablettierens durchgeführt, entsprechend einer Probenentnahme bei jeweils 4000 hergestellten Tabletten. Zwei Tabletten wurden für jede Probe entnommen.
Die Tabletten wurden durch ein validiertes Verfahren unter Verwendung der UV-Absorption in einer wässrigen Lösung untersucht, wodurch insgesamt 100 Tabletten untersucht wurden. Die relative Standardabweichung im Citalopram-Gehalt betrug 1,6%.
Die Variabilität in der Tablettenfestigkeit ist überraschend gering hinsichtlich der geringen Teilchengrösse von Citalopramhydrobromid im Vergleich zum inerten Füllstoff.
Eine mögliche Erklärung für dieses überraschende und vorteilhafte Ergebnis kann sein, dass die Tendenz zur Segregation zwischen kleinen Citalopram-Kristallen und grösseren Füllstoffteilchen in besonderer Weise durch die schlechten Fliesseigenschaften der kleinen Kristalle ausgeglichen wird. Beispiel 6 Durch Direktverpressen von grossen Citalopramhydrobromid-Kristallen hergestellte Tablette

Kristalle aus Citalopram-Base aus Beispiel 4 wurden durch eine Sieböffnung von 0,3 mm gesiebt und mit ProSolv SMCC90 für 3 min in einem Turbula-Mischer vermischt.
Magnesiumstearat wurde hinzugegeben und das Mischen für 30 s fortgesetzt.
Die Tabletten wurden auf einer Einzelstempel-Tablettiermaschine Korsch EK0 hergestellt. Tabletteneigenschaften:

This patent publication describes the production of crystalline citalopram base and the use of crystalline citalopram base as an intermediate in the purification of crude citalopram hydrobromide to pure citalopram hydrobromide. The publication also outlines the manufacture of tablets containing citalopram base.
Citalopram is marketed in a number of countries as a tablet made by compressing granulated citalopram hydrobromide, lactose and other additives.
It is well known that the production of tablets with a reproducible composition requires that all dry ingredients have good flow properties. In the event that the active ingredient has good flow properties, tablets can be produced by pressing the components directly. However, in many cases the particle size of the active ingredient is small, the active ingredient is cohesive or has poor flow properties.
In addition, active ingredients with a small particle size, which are mixed with additives with a larger particle size, will typically segregate or separate during the tabletting process.
The problem of small particle size and poor flowability is conventionally solved by increasing the particle size of the active ingredient, usually by granulating the active ingredient either alone or in combination with a filler and / or with other conventional tablet ingredients.
One such granulation process is the "wet" granulation process. Using this method, the dry solids (active ingredients, fillers, binders, etc.) are mixed and moistened with water or another wetting agent (e.g. an alcohol), and agglomerates or granules are built up from the moistened solids. Wet massing of the mass is continued until a desired homogeneous particle size has been reached, whereupon the granulated product is dried.
An alternative to the "wet" granulation method is the "melting" granulation, which is also known as the "thermoplastic" granulation method, in which a low-melting solid is used as the granulating agent. First, the dry solids are mixed and heated until the binder melts. When the binder is liquefied and spreads over the surface of the particles, the particles will stick together and form granules. The binder solidifies on cooling to form a dry granular product.
Wet granulation and melt granulation are energy-intensive basic processes that require complicated and expensive equipment and technical qualifications.
The process used to produce citalopram hydrobromide results in a product with a very small particle size around 2 to 20 μm, which like many other particulate products with a small particle size has very poor flow properties. In order to achieve an adequate dosage of the citalopram during tableting, it was therefore considered necessary to produce granules from citalopram with a larger particle size and improved flow properties.
The commercialized citalopram tablet is a tablet made from granulated citalopram hydrobromide with various drug additives.
Given that direct compression is much easier and cheaper than the methods that involve granulation, there is a desire for a method for direct compression of citalopram hydrobromide.
The obstacles that previously prevented direct compression of citalopram tablets have now been overcome after extensive research in the laboratory.
Larger particles, i.e. Particles with a size comparable to the size of the filler, can be produced by a new and inventive crystallization process and that these particles are useful for the production of directly compressed tablets. Precise dosing in capsules can also be done with such large particles.
It has also been found that tablets with a surprisingly small variation in the citalo pram content can be produced by directly compressing citalopram hydrobromide, which has a significantly smaller particle size than the filler. The exact dosage in capsules can also be achieved despite the small particle size of citalopram. Objects of the invention
It is an object of the invention to provide large crystals of a pharmaceutically acceptable salt of Citalo-pram which are suitable for use in direct compression.
Another object of the invention is to provide a method for producing large crystals from a pharmaceutically acceptable salt of citalopram. Summary of the invention

The direct compression of citalopram, a filler and other pharmaceutically acceptable pharmaceutical additives into tablets has the great advantage that granulation and a drying step are avoided. In addition, it is no longer necessary to add a binder since the granulation step is avoided.
As used herein, "direct compression" means that the solid dosage form is made by compressing a simple mixture of the active ingredient and drug additives without the active ingredient being subjected to an intermediate granulation process to embed it in a larger particle and improve its flow properties.
As used herein, "binder" means an agent that is used in wet or melt granulation processes and acts as a binder in the granulated product.
As used herein, "particle size distribution" means the distribution of equivalent sphere diameters, determined by laser diffraction at 1 bar dispersion pressure in Sympatec Helos equipment. "Average particle size" means according to the median of the particle size distribution.
As used herein, "reflux temperature" means the temperature at which the solvent or solvent system will reflux or reflux at atmospheric pressure.
In one embodiment of the present invention, the crystals of a pharmaceutically acceptable salt of citalopram have an average particle size in the range from 40 to 200 μm, preferably 45 to 150 μm and particularly preferably 50 to 120 μm.
In a preferred embodiment of the present invention, the crystals are made of citalopram hydrobromide or citalopram hydrochloride, preferably citalopram hydrobromide.
In yet another embodiment of the present invention, the crystals are made from a pharmaceutically acceptable salt of citalopram having an average particle size of at least 40 microns suitable for use in a solid dosage form, from a solution of a pharmaceutically acceptable salt of citalopram in one suitable solution system crystallizes. The solvent system can comprise one or more alcohols and optionally water, preferably the solvent system is a mixture of methanol and water, in which the methanol: water weight ratio is preferably in the range from 5: 1 to 50: 1; particularly preferably 10: 1 to 30: 1 and most preferably 15: 1 to 25: 1.
The pharmaceutically acceptable salt of citalopram is preferably dissolved in the solvent system at a temperature in the range between 50 ° C. and the reflux temperature of the solvent system, preferably between 60 ° C. and the reflux temperature and particularly preferably between 64 ° C. and the reflux temperature. The amounts of the pharmaceutically acceptable salt of citalopram and the solvent preferably correspond to a weight ratio of solvent-solute in the range from 0.5: 1 to 5: 1, particularly preferably 0.7: 1 to 2: 1 and most preferably 0, 9: 1 to 1.5: 1.
The solution of a pharmaceutically acceptable salt of citalopram is cooled to a temperature, the inoculation temperature, in the range from 20 to 40 ° C., preferably 25 to 35 ° C., whereupon it is inoculated with citalopram crystals and at the inoculation temperature for a holding time for crystal growth , in the range of 30 minutes to 7 days, preferably 1 hour to 4 days and particularly preferably 12 to 36 hours. After the holding time, the crystallization batch is gradually cooled in a controlled manner from the seeding temperature to the temperature at which the crystals are isolated from the mother liquor, the gradual cooling over a period in the range from 5 min to 6 h, preferably 15 min to 4 h and particularly preferably 30 min to 2 h.
The crystals of the pharmaceutically acceptable salt of citalopram are preferably isolated from the mother liquor at a temperature in the range of 0 to 20 ° C, particularly preferably 5 to 15 ° C, using conventional separation techniques, e.g. Filtration.
The small crystals of a pharmaceutically acceptable salt of citalopram used in one embodiment of the invention can be made according to the methods described in U.S. Patent 4,136,193.
The crystals of citalopram base used in one embodiment of the invention can be prepared according to the process described in Dutch Patent No. 1,016,435.
The invention is illustrated below by examples. However, the examples are intended to illustrate the invention only and should not be construed as a limitation. Example 1 Crystallization of Citalopram Hydrobromide to Large Crystals
Citalopram hydrobromide (200 g) is dissolved in a mixture of methanol (200 g) and water (20 g) at 69 ° C. The solution is cooled to 30 ° C., inoculated with citalopram hydrobromide crystals and kept at 30 ° C. for 24 hours, after which it is cooled to 10 ° C. within 1 hour. The crystals are isolated by filtration, washed with cold methanol and dried. The particle size distribution for the resulting crystals is shown in Table 1. Example 2 Crystallization of Citalopram Hydrobromide to Large Crystals
Citalopram hydrobromide (12.0 kg) is refluxed in a mixture of methanol (12.5 kg) and water (1.2 kg). The solution is cooled to 30 ° C., inoculated with citalopram hydrobromide crystals (27 g) and kept at 30 ° C. for 16 hours, after which it is cooled to 10 ° C. within 1 hour. The crystals are isolated by filtration, washed with cold (10 ° C.) methanol (3.5 kg) and dried. The particle size distribution for the resulting crystals is shown in Table 1. Example 3 Crystallization of Citalopram Hydrobromide to Small Crystals
Citalopram hydrobromide (200 kg) is dissolved in a mixture of methanol (170 l) and acetone (680 l) at 56 ° C. The solution is cooled to 15 ° C., inoculated with citalopram hydrobronide crystals (50 g), hexane (1600 l) is added gradually over the course of 60 min, whereupon the suspension is left to stand with moderate stirring and cooling for 8 h. The crystals are isolated by filtration, first washed with a cold (10 ° C.) mixture of acetone (50 l) and hexane, then with cold (10 ° C.) hexane (220 l) and dried. The particle size distribution for the resulting crystals is shown in Table 1. Example 4 Crystallization of Citalopram as Free Base
Citalopram hydrobromide (101 g) is suspended in water (500 ml) and toluene (500 ml). NaOH (60 ml, 5 N (aq)) is added and the mixture (pH> 10) is stirred for 15 min before the phases are separated. The organic phase is washed with water (2 x 100 ml) and filtered through a cushion of filter aid. The volatiles are removed in vacuo and the title compound is obtained as an oil. n-Heptane (400 ml) is added and the mixture is warmed to 70 ° C. Crystals form on cooling. The white crystals of citalopram base are filtered off and dried in vacuo at ambient temperature overnight. Table 1:

Citalopram hydrobromide crystals from Example 3 and ProSolv SMCC90 were mixed at 7 rpm for 10 min in a 100 l screed PTM 200 mixer. Magnesium stearate was added and mixing continued for 3 minutes.
25 kg of the resulting mixture were tabletted on a Fette P 1200 / IC tablet press with 30 stations, equipped with rectangular, raised, notched punches of 5.5 × 8 mm (125,000 tablets / hour). The tablet core weight was adjusted to 125 mg. The nominal yield was 200,000 tablets. The tablet press was run until the mix level was just above the forced feed, i.e. tableting was continued for as long as possible to identify possible segregation tendencies in the last batches of the mixture. Samples of the tablets were taken during compression to measure the tendency to segregate. Since there is a significant difference in size between the active ingredient, citalopram hydrobromide, and the inert filler, ProSolv SMCC90, as can be seen in Table 1, one would expect that the components of different sizes would segregate, i.e. would separate during the transition from the mixing container to the filling shaft of the tablet press or while remaining in the filling shaft of the tablet press during tableting.
Sampling was carried out 50 times at regular intervals during tableting, corresponding to a sampling for every 4000 tablets produced. Two tablets were taken for each sample.
The tablets were examined by a validated method using UV absorption in an aqueous solution, whereby a total of 100 tablets were examined. The relative standard deviation in the citalopram content was 1.6%.
The variability in tablet strength is surprisingly small with regard to the small particle size of citalopram hydrobromide compared to the inert filler.
A possible explanation for this surprising and advantageous result can be that the tendency towards segregation between small citalopram crystals and larger filler particles is compensated in a special way by the poor flow properties of the small crystals. Example 6 Tablet Made by Direct Compression of Large Citalopram Hydrobromide Crystals

Citalopram hydrobromide crystals from Example 2 and ProSolv SMCC90 were mixed. Magnesium stearate was added and mixing continued.
Tablets (125 mg nominal weight) were made.

Crystals of citalopram base from Example 4 were sieved through a sieve opening of 0.3 mm and mixed with ProSolv SMCC90 for 3 min in a Turbula mixer.
Magnesium stearate was added and mixing continued for 30 s.
