Die Erfindung beinhaltet ein neues Verfahren zur Herstellung von Hypoxanthin.
Hypoxanthin findet sich in Muskeln, Blut und Harn als Bestandteil der Nucleinsäure vieler Organismen (Römpp's Chemie Lexikon 8. Auflage, Bd. 3, S. 1825) und wird in der Medizin z.B. für die Leberregeneration angewendet (Erhart, Ruschig, Arzneimittel Band 1; Verlag Chemie Weinheim, 1968, S. 807)
Zur Herstellung von Hypoxanthin wurden zahlreiche Synthesen beschrieben.
So geht aus der französischen Patentschrift FR 1 425 603 hervor, dass in einem ersten Schritt, durch Umsetzung von 2-Phenylazocyanacetamid mit Formamidinacetat und Weiterumsetzung mit Formamid, in Gegenwart von Natriumdithionit, Hypoxanthin in einer Ausbeute von 58% gewonnen werden kann.
Nachteilig bei diesem Verfahren ist aber, dass durch die Verwendung des Dithionits eine beträchtliche, nicht rezirkulierbare Salzlast anfällt. Da das Ausgangsprodukt zudem schwer zugänglich ist, ist eine Umsetzung dieses Verfahrens in die Technik nicht denkbar.
Die CA: 70 57 900f, japanische Patentpublikation 68-22 868 beschreibt die in situ Umsetzung von Cyanessigester mit Natriumnitrit und Natriumdithionit und die Weiterumsetzung mit Ammoniak und Orthoameisensäureester zu Hypoxanthin. Ausbeute 43%. Neben der ungenügenden Ausbeute ist wiederum die anfallende Salzlast, d.h. die damit verbundenen ökologischen Probleme, ein grosses Hindernis für eine Umsetzung dieses Verfahrens in den technischen Massstab.
Taylor et al., Tetrahedron Letters No. 12 (1959) S. 9-11 offenbaren ein Verfahren, worin Hypoxanthin in einer Ausbeute von 75% aus Acetamidocyanessigsäurester, das mit Ammoniak, Orthoameisensäureester und Ammoniumacetat umgesetzt wird, gewonnen werden kann. Von der guten Ausbeute und vom relativ gut zugänglichen Edukt her erscheint diese Synthese für die Umsetzung in den technischen Massstab interessant.
Die anfallende Salzlast sowie die in sehr hohem Überschuss eingesetzten Reaktanden, Ammoniak und Orthoameisensäureester, die in der Reaktionsmischung zum Teil unreagiert anfallen und entsorgt werden müssen, machen den Prozess aber vom ökologischen Standpunkt aus nicht praktikabel.
Die Aufgabe der Erfindung bestand daher darin, eine ökologisch vertretbare Synthese zur Herstellung von Hypoxanthin zu entwickeln, die neben einer guten Ausbeute auch eine Umsetzung in den technischen Massstab gewährleistet.
Diese Aufgabe konnte mit einem Verfahren nach Anspruch 1 gelöst werden.

Die Aminocyanessigsäurederivate, insbesondere das vorzugsweise verwendete Aminocyanessigsäureamid oder der Aminocyanessigsäureethylester, bzw. deren Salze, sind grosstechnisch, gemäss der europäischen Patentanmeldung EP 342 616, zugänglich. Zweckmässig wird ein Formamidinsalz, wie z.B. das Formamidinhydrochlorid oder das Formamidinacetat, vorzugsweise aber das Formamidinacetat, eingesetzt.
Auf 1 Mol des Aminocyanessigsäurederivats werden zweckmässig 2 Mol bis 6 Mol, vorzugsweise 2,5 Mole bis 3,5 Mole Formamidin bzw. Formamidinsalz eingesetzt.
Zweckmässig wird die Umsetzung in Gegenwart eines polaren Lösungsmittels durchgeführt. Geeignete Vertreter davon sind, Acetonitril, Propionitril, Methylacetat, Ethylacetat oder nie dere aliphatische Alkohole wie z.B. Methanol, Ethanol, iso-Propanol, Methoxyethanol oder Ethylenglykol.
Liegt das Aminocyanessigsäurederivat in Salzform vor, wird zweckmässig zunächst auf übliche Weise, z.B. mit Ammoniak, in das freie Aminocyanessigsäurederivat überführt.
Die eingesetzten Lösungsmittel können daher bei der erfindungsgemässen Umsetzung Ammoniak, bzw. das betreffende Agens, zur Entfernung des Salzes enthalten.
Es hat sich als vorteilhaft gezeigt, die Umsetzung bei Temperaturen zwischen 100 und 220 DEG C, vorzugsweise zwischen 130 und 200 DEG C, durchzuführen. Je nach verwendetem Lösungsmittel stellt sich dabei ein Druck von 1 bis 20 bar ein.
Nach einer Reaktionszeit von üblicherweise 3 bis 7 Stunden kann das Hypoxanthin auf übliche Weise in guter Ausbeute und Reinheit aus dem Reaktionsgemisch gewonnen werden.
Beispiel 1
Herstellung von Hypoxanthin aus Aminocyanacetamid
1,5 g (15 mmol) Aminocyanacetamid wurden mit 5,64 g (52,5 mmol) Formamidinacetat in 15 g Ethanol abs. vorgelegt. Das Reaktionsgemisch wurde anschliessend in einem Autoklaven während 6 Stunden bei 151 DEG C und einem Druck von 5 bis 7 bar gehalten, dann abgekühlt und am Rotationsverdampfer eingeengt. Der Rückstand wurde mit Ethanol abs. aufgeschlämmt, filtriert und anschliessend getrocknet.
Ausbeute 1,6 g = 78%.
Beispiel 2
Herstellung von Hypoxanthin aus Aminocyanessigsäureethylester
1,92 g (15 mmol) Aminocyanessigsäureethylester wurden mit 4,83 g (45 mmol) Formamidinacetat in 15 g Ethanol abs. und 0,76 g Ammoniak abs. vorgelegt. Das Reaktionsgemisch wurde anschliessend im Autoklaven während 6 Stunden bei 150 DEG C und einem Druck von 4 bis 7 bar gehalten, dann abgekühlt und am Rotationsverdampfer eingeengt. Der Rückstand wurde mit Ethanol abs. aufgeschlämmt, filtriert und anschliessend getrocknet.
Ausbeute 1,35 g = 66,1%.
Beispiel 3
Herstellung von Hypoxanthin aus Aminocyanessigsäuremethylester
4,30 g (15 mmol) Aminocyanessigsäuremethylester-tosylat wurden in 50 ml Acetonitril gelöst. Durch diese Lösung wurde während 15 Minuten NH3 durchgeleitet, wobei ein weisser Niederschlag von Ammoniumtosylat ausfiel.
Das Gemisch wurde filtriert, das Filtrat eingeengt und getrocknet. Der erhaltene Aminocyanessigsäuremethylester wurde in 20 ml Ethanol gelöst und mit 4,70 g (45 mmol) Formamidinacetat in einen Autoklav gegeben.
Unter Rühren wurde das Gemisch auf 150 DEG C aufgeheizt. Nach 5 Stunden wurde abgekühlt, eingedampft, mit 50 ml Ethanol aufgeschlämmt, filtriert, der Filterrückstand mit Ethanol gewaschen und getrocknet.
Man erhielt 1,38 g Hypoxanthin.
Ausbeute: 68%
Beispiel 4
Herstellung von Hypoxanthin aus Aminocyanessigsäurebenzylester
0,92 g (2,5 mmol) Aminocyanessigsäurebenzylester-tosylat wurden in 20 ml Acetonitril gelöst.
Durch diese Lösung wurde während 15 Minuten NH3 durchgeleitet, wobei ein weisser Niederschlag von Ammoniumtosylat ausfiel. Das Gemisch wurde filtriert, das Filtrat eingeengt und getrocknet.
Der erhaltene Aminocyanessigsäurebenzylester wurde in 15 ml Ethanol gelöst und mit 0,80 g (7,5 mmol) Formamidin-acetat in einen Autoklav gegeben.
Unter Rühren wurde das Gemisch auf 150 DEG C aufgeheizt. Nach 5 Stunden wurde abgekühlt, eingedampft, mit 30 ml Ethanol aufgeschlämmt, filtriert, der Filterrückstand mit Ethanol gewaschen und getrocknet.
Man erhielt 0,12 g Hypoxanthin.
Ausbeute: 35%

Numerous syntheses have been described for the production of hypoxanthine.

Taylor et al., Tetrahedron Letters No. 12 (1959) pp. 9-11 disclose a process in which hypoxanthine can be obtained in a yield of 75% from acetamidocyanacetic acid ester which is reacted with ammonia, orthoformate and ammonium acetate. In terms of the good yield and the relatively easily accessible starting material, this synthesis appears to be interesting for implementation on an industrial scale.
The resulting salt load as well as the reactants, ammonia and orthoformic acid esters used in a very high excess, which accumulate in the reaction mixture in part unreacted and have to be disposed of, make the process impractical from an ecological point of view.
The object of the invention was therefore to develop an ecologically justifiable synthesis for the production of hypoxanthine which, in addition to a good yield, also ensures implementation on an industrial scale.
This object could be achieved with a method according to claim 1.

The aminocyanacetic acid derivatives, in particular the preferably used aminocyanacetic acid amide or the aminocyanacetic acid ethyl ester, or their salts, are commercially available, according to the European patent application EP 342 616. A formamidine salt such as e.g. the formamidine hydrochloride or the formamidine acetate, but preferably the formamidine acetate.
2 mol to 6 mol, preferably 2.5 mol to 3.5 mol, of formamidine or formamidine salt are expediently used per mol of the aminocyanacetic acid derivative.
The reaction is expediently carried out in the presence of a polar solvent. Suitable representatives thereof are acetonitrile, propionitrile, methyl acetate, ethyl acetate or any other aliphatic alcohols such as e.g. Methanol, ethanol, iso-propanol, methoxyethanol or ethylene glycol.
If the aminocyanacetic acid derivative is in salt form, it is expedient first of all to use the usual method, e.g. with ammonia, converted into the free aminocyanacetic acid derivative.
In the reaction according to the invention, the solvents used can therefore contain ammonia, or the agent in question, for removing the salt.
It has proven advantageous to carry out the reaction at temperatures between 100 and 220 ° C., preferably between 130 and 200 ° C. Depending on the solvent used, a pressure of 1 to 20 bar is established.
After a reaction time of usually 3 to 7 hours, the hypoxanthine can be obtained from the reaction mixture in a customary manner in good yield and purity.

4.30 g (15 mmol) methyl aminocyanacetate tosylate were dissolved in 50 ml acetonitrile. NH3 was passed through this solution for 15 minutes, during which a white precipitate of ammonium tosylate precipitated out.
The mixture was filtered, the filtrate was concentrated and dried. The aminocyanacetic acid methyl ester obtained was dissolved in 20 ml of ethanol and placed in an autoclave with 4.70 g (45 mmol) of formamidine acetate.
The mixture was heated to 150 ° C. with stirring. After 5 hours, the mixture was cooled, evaporated, slurried with 50 ml of ethanol, filtered, the filter residue was washed with ethanol and dried.
1.38 g of hypoxanthine were obtained.
Yield: 68%
Example 4
Preparation of hypoxanthine from benzyl aminocyanacetate
0.92 g (2.5 mmol) benzyl aminocyanacetic acid tosylate were dissolved in 20 ml acetonitrile.
NH3 was passed through this solution for 15 minutes, a white precipitate of ammonium tosylate precipitating out. The mixture was filtered, the filtrate was concentrated and dried.
The benzyl aminocyanacetic acid obtained was dissolved in 15 ml of ethanol and placed in an autoclave with 0.80 g (7.5 mmol) of formamidine acetate.
The mixture was heated to 150 ° C. with stirring. After 5 hours, the mixture was cooled, evaporated, slurried with 30 ml of ethanol, filtered, the filter residue was washed with ethanol and dried.
