In der obigen Formel sowie in der folgenden Beschreibung und in den Ansprüchen werden die Octahydrobenzo[g]chinoline nur durch die Formel eines einzigen Stereoisomers wiedergegeben; die Erfindung umfasst jedoch sowohl die Verbindungen mit der Struktur der Formel I als auch ihre optischen Antipoden und die Gemische, besonders die Racemate, dieser Enantiomeren. Als optisch aktive Enantiomeren sind die (3R, 4aR, 10aR)-Benzochinolinderivate bevorzugt.
Die erfindungsgemässen Verbindungen der Formel I können in Form der freien Basen oder ihrer Säureadditionssalze vorliegen. Die freien Basen können auf an sich bekannte Weise in ihre Säureadditionssalze überführt werden und umgekehrt. So können die erfindungsgemässen Verbindungen der Formel I z.B. mit anorganischen Säuren wie Chlorwasserstoffsäure oder mit organischen Säuren wie Maleinsäure Säureadditonssalze bilden.
In den Verbindungen der Formel I sind die nachfolgenden Bedeutungen sowie deren Kombinationen bevorzugt.

Zu den erfindungsgemässen Verbindungen und ihren Säureadditionssalzen gelangt man beispielsweise, indem man

und gewünschtenfalls die erhaltenen Verbindungen der Formel I in ihre Säureadditionssalze überführt.
Das Verfahren a) ist eine klassische Alkylierung und kann z.B. durch Umsetzung mit einer Verbindung der Formel R1Z, worin Z für einen reaktionsfähigen Rest steht, erfolgen. Z bedeutet vorzugsweise Cl, Br, J oder den Säuerest einer organischen Sulfonsäure, z.B. Methansulfonyloxy oder p-Tosyloxy. Die Reaktion wird vorzugsweise in Gegenwart eines säurebindenden Mittels, z.B. Na2CO3 oder einer organischen Base, durchgeführt.
Die Ätherspaltung nach Verfahren b) kann in an sich bekannter Weise, z.B. mittels HBr oder BBr3 erfolgen.
Die Acylierung nach Verfahren c) erfolgt ebenfalls auf üblichem Wege, z.B. mit reaktiven Derivaten der oben erwähnten Säuren, vorzugsweise mit Säurehalogeniden oder Säureanhydriden.
Die Ausgangsverbindungen der Formel II können beispielsweise hergestellt werden, indem man
1) eine Verbindung der Formel III in einem basischen Milieu in eine Verbindung der Formel IV umlagert unter gleichzeitiger Hydrolyse der Ester- zu einer Carboxylgruppe:

Dieses Verfahren kannn nach an sich für die Herstellung von Ureiden aus Carbonsäuren und Carbodiimiden allgemein bekannten Verfahren durchgeführt werden. Vorzugsweise lässt man die Verbindungen der Formeln IV und V während mehreren Stunden in einem inerten Lösungsmittel, wie z.B. Tetrahydrofuran, unter Rückfluss miteinander reagieren. Gegebenenfalls kann eine organische Base, wie N-Äthyldiisopropylamin, zugegeben werden.
Bei dieser Reaktion entstehen sowohl Verbindungen der Forme VI, worin R2 für

Diese beiden Verbindungen können auf üblichem Wege, z.B. durch selektive Kristallisation oder durch Chromatographie, voneinander getrennt werden.
3) aus einer Verbindung der Formel VI die OCH3-Gruppe in Stellung 1 abspaltet zur Herstellung einer Verbindung der Formel II.
Die Abspaltung kann vorzugsweise durch Reduktion, z.B. mit Zink und Essigsäure, vorgenommen werden.
Die Verbindungen der Formel III sind bekannt oder können auf an sich bekannte Weise hergestellt werden.
Die Verbindungen der Formel II sind neu und sind ein Teil der Erfindung.
Die optisch aktiven Enantiomeren der Verbindungen der Formel I können auf üblichem Wege, z.B. ausgehend von einem optisch aktiven Zwischenprodukt, beispielsweise einer optisch aktiven Verbindung der Formel III, hergestellt werden.
Racemate können z.B. durch Chromatographie über optisch aktive Verbindungen enthaltende Träger in ihre optischen Enantiomere getrennt werden.
In den nachfolgenden Beispielen werden alle Temperaturen in Celsiusgraden angegeben und sind unkorrigiert.
In den Beispielen werden die Racemate mit dem Namen einer der beiden Enantiomeren (z.B. 3 beta , 4a alpha , 10a beta ) gefolgt durch (Racemat) gekennzeichnet.
Beispiel 1:
1-Äthyl-3-(3-dimethylaminopropyl)-3-(1-allyl-6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin-3 beta -carbonyl)-harnstoff (Racemat)
900 mg 1-Äthyl-3-(3-dimethylaminopropyl)-3-(6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin-3 beta -carbonyl)-harnstoff (Racemat) werden in 9 ml Dimethylformamid gelöst, mit 340 mg Na2CO3 und 0,18 ml Allylbromid versetzt und 6 Stunden bei Raumtemperatur gerührt. Hiernach wird das Reaktionsgemisch auf Wasser gegossen und mit Essigester extrahiert.
Nach Trocknen und Eindampfen erhält man die Titelverbindung als braunes Harz. Dieses wird in wenig CH2Cl2 gelöst und über basischem Kieselgel mit CH2Cl2 + 0,5% CH3OH + 5% Ammoniaklösung (25%ig), flash chromatographiert. Das Dihydrochlorid der erhaltenen Titelverbindung schmilzt bei 185 DEG unter Aufschäumen.
Das Ausgangsprodukt kann wie folgt hergestellt werden:
a) 1-Methoxy-3 beta -carboxy-6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin (Racemat)
2,0 g 1-Methoxy-3 alpha -methoxycarbonyl-6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin (Racemat, 40 ml Äthanol, 5 ml Wasser und 2,0 g KOH werden zusammengegeben und über Nacht unter Rückfluss gehalten. Am Morgen wird eingeengt, auf 2N HCl gegossen und die ausgefallenen weissen Festkörper abgenutscht.

b) 1-Äthyl-3-(3-dimethylaminopropyl)-3-(1-methoxy-6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin-3 beta -carbonyl)-harnstoff (Racemat)
500 mg des Endproduktes der Stufe a), 323 mg N-Äthyl-N min -(3 min -dimethylaminopropyl)-carbodiimid, 0,36 ml N-Äthyldiisopropylamin und 10 ml Tetrahydrofuran werden zusammengegeben und über Nacht unter Argon gekocht. Am Morgen wird eingedampft. Der farblose harzige Rückstand wird in CH2Cl2 gelöst und 2 mal mit 2N NaOH ausgeschüttelt. Hiernach wird die CH2Cl2-Lösung 2 mal mit 2n HCl ausgeschüttelt, die saure Phase mit NaOH conc. alkalisch gestellt und mit CH2Cl2 extrahiert. Die organische Phase wird getrocknet und eingedampft.
Als Rückstand erhält man ein farbloses Harz, das in wenig CH2Cl2 gelöst und mit CH2Cl2 + 4% CH3OH flashchromatographiert wird, wobei als zweites Produkt die Titelverbindung und als erstes Produkt die Isomere 3-Äthyl-1-(3-dimethylaminopropyl)-Verbindung erhalten wird.
c) 1-Äthyl-3-(3-dimethylaminopropyl)-3-(6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin-3 beta -carbonyl)-harnstoff (Racemat)
1,64 g des Endproduktes der Stufe b) werden in 20 ml Essigsäure und 10 ml Wasser suspendiert und unter Rühren bei Raumtemperatur portionenweise mit 5,8 g Zinkstaub versetzt, wobei eine exotherme Reaktion zu beobachten ist. Nach beendeter Zugabe lässt man noch 6 Stunden bei Raumtemperatur nachrühren.
Anschliessend wird filtriert und mit Wasser und Essigester nachgewaschen. Das Filtrat wird eingeengt, mit NaHCO3-Lösung versetzt und mit Essigester extrahiert. Die organische Phase wird 1 mal mit Sole gewaschen, getrocknet und eingeengt. Man erhält die Titelverbindung.
Das optisch aktive (3R,4aR,10aR)-Enantiomer der Titelverbindung des Beispiels 1 kann z.B. ausgehend von (-)-(3S,4aR,10aR)-1,2,3,4,4a,5,10,10a-Octahydro-1,6-dimethoxy-3-carboxy-benzo[g]chinolin hergestellt werden.
Beispiel 2:
1-Äthyl-3-(3-dimethylaminopropyl)-3-(1-propyl-6-methoxy-1,2,3,4,4a alpha ,5,10,10a beta -octahydrobenzo[g]chinolin-3 beta -carbonyl)-harnstoff (Racemat)
Analog zu Beispiel 1 wird die Titelverbindung hergestellt.
Smp. des Dihydrochloris (Schaum) ca. 140 DEG unter Zersetzung.
Die Verbindungen der Formel I sowie die pharmazeutisch akzeptablen Säureadditionssalze dieser Verbindungen weisen im Tierversuch interessante pharmakodynamische Eigenschaften auf. Sie können daher als Arzneimittel verwendet werden.
Insbesondere besitzen die erfindungsgemässen Verbindungen der Formel I und ihre Salze eine dopaminerge (Dopaminrezeptoren stimulierende) und prolaktin (PRL)-sekretionshemmende Wirkung. Diese Wirkung kann z.B. durch die Hemmung der basalen Prolaktinsekretion bei männlichen Ratten nach der Methode E. Flückiger et al. (Experientia 34, 1330, 1978) nachgewiesen werden. In diesem Test zeigen sich die Verbindungen aktiv bei Dosen zwischen 0,005 und 0,1 mg/kg s.c. Peroral verabreicht bei Dosen zwischen 0,03 und 0,5 mg/kg zeigen die Verbindungen ausserdem eine länger anhaltende Wirkung als die üblichen PRL-sekretionshemmer, z.B. als Bromokriptin.
Die Verbindung des Beispiels 1 ist das bevorzugte Racemat. Das 3R,4aR,10aR-Enantiomer davon ist das bevorzugte Isomer.
Aufgrund ihrer PRL-sekretionshemmenden Wirkung sind die Verbindungen der Formel I und ihre pharmazeutisch akzeptablen Säureadditionssalze wertvoll als PRL-Sekretionshemmer, z.B. bei der Behandlung von Erkrankungen, worin eine Senkung des PRL-Spiegels erwünscht ist, z.B. bei der Behandlung der Galaktorrhoe, bei der Behandlung von PRL-abhängigen menstruellen Störungen, z.B. der Oligo-und Amenorrhoe, zur Verhinderung der Laktation, sowie bei der Behandlung von hyperprolaktinämischem Hypogonadismus bei Mann und Frau und bei der Behandlung von Prolaktinomen.
Aufgrund ihrer dopaminergen Wirkung sind die Verbindung auch verwendbar bei der Behandlung des Morbus Parkinson.
Die Prolaktinsekretionshemmung ist bevorzugte Indikation.
Für die oben genannten Anwendungen variiert die zu verwendende Dosis selbstverständlich je nach verwendeter Substanz, Art der Administration und der gewünschten Behandlung. Im allgemeinen liegt eine geeignete Einzeldosis im Bereich von ca. 0,5 bis ca. 10 mg, einmal pro Tag bis zu einmal in 5 Tagen verabreicht. Einzeldosen können ca. 0,5 bis 50 mg enthalten.
Die Verbindungen der Formel I können, als freie Base oder als pharmazeutisch akzeptable Säureadditionssalze, enteral (z.B. oral, beispielsweise als Tabletten oder Kapseln) oder parenteral (z.B. als Injektionslösungen oder Suspensionen) verabreicht werden.
Die Erfindung betrifft auch pharmazeutische Präparate, die eine Verbindung der Formel I oder ein pharmazeutisch akzeptables Säureadditionssalz davon enthalten. Diese Präparate, beispielsweise eine Lösung oder eine Tablette, können nach bekannten Methoden, unter Verwendung der üblichen Hilfs- und Trägerstoffe, hergestellt werden.
Die Erfindung betrifft weiter die Verbindungen der Formel I oder ihre pharmazeutisch akzeptablen Säureadditionssalze als Arzneimittel, besonders als Prolaktin-Hemmer/Dopaminagonisten, d.h. sie eignen sich zur Behandlung der oben erwähnten Indikationen.

Racemates can e.g. be separated into their optical enantiomers by chromatography using optically active compounds.
In the following examples, all temperatures are given in degrees Celsius and are uncorrected.
In the examples, the racemates are identified by the name of one of the two enantiomers (e.g. 3 beta, 4a alpha, 10a beta) followed by (racemate).
Example 1:

900 mg 1-ethyl-3- (3-dimethylaminopropyl) -3- (6-methoxy-1,2,3,4,4a alpha, 5,10,10a beta-octahydrobenzo [g] quinoline-3 beta-carbonyl) urea (racemate) are dissolved in 9 ml of dimethylformamide, 340 mg of Na2CO3 and 0.18 ml of allyl bromide are added and the mixture is stirred at room temperature for 6 hours. The reaction mixture is then poured onto water and extracted with ethyl acetate.
After drying and evaporation, the title compound is obtained as a brown resin. This is dissolved in a little CH2Cl2 and flash chromatographed over basic silica gel with CH2Cl2 + 0.5% CH3OH + 5% ammonia solution (25%). The dihydrochloride of the title compound obtained melts at 185 ° with foaming.

b) 1-ethyl-3- (3-dimethylaminopropyl) -3- (1-methoxy-6-methoxy-1,2,3,4,4a alpha, 5,10,10a beta-octahydrobenzo [g] quinoline-3 beta-carbonyl) urea (racemate)
500 mg of the end product of stage a), 323 mg of N-ethyl-N min - (3 min -dimethylaminopropyl) carbodiimide, 0.36 ml of N-ethyldiisopropylamine and 10 ml of tetrahydrofuran are combined and boiled overnight under argon. The residue obtained is a colorless resin, which is dissolved in a little CH2Cl2 and flash chromatographed with CH2Cl2 + 4% CH3OH, the second product being the title compound and the first product being the isomers 3-ethyl-1- (3-dimethylaminopropyl) compound .
c) 1-ethyl-3- (3-dimethylaminopropyl) -3- (6-methoxy-1,2,3,4,4a alpha, 5,10,10a beta-octahydrobenzo [g] quinoline-3 beta-carbonyl) -urea (racemate)
1.64 g of the end product of stage b) are suspended in 20 ml of acetic acid and 10 ml of water, and 5.8 g of zinc dust are added in portions with stirring at room temperature, an exothermic reaction being observed. It is then filtered and washed with water and ethyl acetate. The filtrate is concentrated, mixed with NaHCO3 solution and extracted with ethyl acetate. The organic phase is washed once with brine, dried and concentrated. The title compound is obtained.
The optically active (3R, 4aR, 10aR) enantiomer of the title compound of Example 1 can e.g. starting from (-) - (3S, 4aR, 10aR) -1,2,3,4,4a, 5,10,10a-octahydro-1,6-dimethoxy-3-carboxy-benzo [g] quinoline.
Example 2:

The compounds of the formula I and the pharmaceutically acceptable acid addition salts of these compounds have interesting pharmacodynamic properties in animal experiments. They can therefore be used as medicines.
In particular, the compounds of the formula I according to the invention and their salts have a dopaminergic (stimulating dopamine receptors) and prolactin (PRL) secretion-inhibiting action. This effect can e.g. by inhibiting basal prolactin secretion in male rats according to the E. Flückiger et al. (Experientia 34, 1330, 1978). In this test, the compounds are shown to be active at doses between 0.005 and 0.1 mg / kg s.c. Administered orally at doses between 0.03 and 0.5 mg / kg, the compounds also show a longer-lasting effect than the usual PRL secretion inhibitors, e.g. as bromocriptine.
The compound of Example 1 is the preferred racemate. The 3R, 4aR, 10aR enantiomer thereof is the preferred isomer.
Because of their PRL secretion-inhibiting activity, the compounds of formula I and their pharmaceutically acceptable acid addition salts are valuable as PRL secretion inhibitors, e.g. in the treatment of diseases in which a lowering of the PRL level is desired, e.g. in the treatment of galactorrhea, in the treatment of PRL-dependent menstrual disorders, e.g. Oligo and amenorrhea, to prevent lactation, as well as in the treatment of hyperprolactinemic hypogonadism in men and women and in the treatment of prolactinomas.
Because of their dopaminergic effects, the compound can also be used in the treatment of Parkinson's disease.
Inhibition of prolactin secretion is the preferred indication.
For the above-mentioned applications, the dose to be used naturally varies depending on the substance used, the type of administration and the desired treatment. In general, a suitable single dose is in the range of about 0.5 to about 10 mg, administered once a day up to once in 5 days. Single doses can contain approximately 0.5 to 50 mg.
The compounds of formula I, as free base or as pharmaceutically acceptable acid addition salts, can be administered enterally (e.g. orally, e.g. as tablets or capsules) or parenterally (e.g. as injection solutions or suspensions).
The invention also relates to pharmaceutical preparations which contain a compound of formula I or a pharmaceutically acceptable acid addition salt thereof. These preparations, for example a solution or a tablet, can be prepared by known methods using the customary auxiliaries and carriers.
The invention further relates to the compounds of formula I or their pharmaceutically acceptable acid addition salts as medicaments, especially as prolactin inhibitors / dopamine agonists, i.e. they are suitable for the treatment of the above-mentioned indications.
