Die Erfindung betrifft ein Verfahren zur Herstellung von Algen mit biologischer Wirkung.
Die Algen werden von der Menschheit seit Urzeiten zu Ernährungs- und Fütterungszwecken genutzt. So verzehren die fernöstlichen Völker in erster Linie Meeresalgen, neuerdings werden diese aber auch in getrockneter Form beziehungsweise als Tabletten in entwickelten Ländern verbraucht. Die Algen sind Träger sehr wertvoller Nährstoffe, ihre getrockneten Formen enthalten in hoher Konzentration die für ein gesundes Leben notwendigen Substanzen, wie beispielsweise Vitamine, Proteine, Protein-Mikroelement-Komplexe, Saccharide, mehrfach ungesättigte Fettsäuren usw.
In den letzten Jahren hat die Umweltverschmutzung Weltmassstab angenommen, so dass die Verunreinigung der Meere, Ozeane durch toxische Schwermetalle (Pb, Hg, Cd, Cu) beziehungsweise karzinogene Agenzien, wie beispielsweise polykondensierte zyklische aromatische Verbindungen, ein nicht zu vernachlässigendes Ausmass angenommen hat. Ein besonderes Problem ist dabei, dass die Meeresalgen diese Agenzien in ihren Organismen anhäufen. Für menschliche oder tierische Nahrungs-, Fütterungs-, kosmetische oder therapeutische Zwecke sind deshalb die aus dem Meer gewonnenen oder unter anderen natürlichen Bedingungen gezüchteten "ganzen Algen" nicht nutzbar, es kann nur die Fraktion genutzt werden, die von den giftigen Stoffen beziehungsweise Stoffen mit schädlicher biologischer Wirkung gereinigt wurde.
Einen solchen Reinigungsvorgang beschreibt beispielsweise Carames de Gouvea (Cosmetics and Toiletries, 95, 47/1980/). Das Reinigen ist jedoch ein Eingriff, der einen Teil der biologisch wirksamen Stoffe der Algen vernichtet, so verringert sich der biologische Wert der auf diese Weise hergestellten Algenfraktion bedeutend (Zajic: Properties and Products of Algae. Edition Planum, New York, 1970).
Aus den oben aufgeführten Gründen hat die Herstellung von umweltfreundlichen sterilen Algen an Bedeutung gewonnen. So sind zahlreiche Verfahren zur künstlichen Züchtung von Algen bekannt. Diese Verfahren können in offenen Becken unter Sonnenlicht oder unter geschlossenen, die Sterilität gewährleistenden Bedingungen, bei natürlicher oder künstlicher Beleuchtung oder ohne Beleuchtung durchgeführt werden.
Gemäss der japanischen Offenlegungsschrift Nr. 5 696 690 werden Algen in sterilisiertem Meerwasser, in aus verschiedenen Nährsalzen bestehender Lösung, unter Verwendung von Meeresalgenstämmen und mit Hilfe einer künstlichen Lichtquelle gezüchtet.
Die japanische Offenlegungsschrift Nr. 4 517 146 beschreibt die industrielle Züchtung von einzelligen grünen Chlorella-Algen, wonach die Algen in steriler Nährlösung unter Ausschluss von Licht und Kohlendioxyd, in süsswässrigem Medium gezüchtet werden.
Auch die französische Patentschrift Nr. 2 103 462 beschreibt die Züchtung von einzelligen grünen Süsswasseralgen, wonach die industrielle Algenzüchtung durch die Verwendung von die entsprechenden Nährsalze enthaltender Nährlösung, auf fotosynthetischem Wege verwirklicht wird.
Die bekanntgemachten ungarischen Patentanmeldungen Nr. 4613/84 und 4614/84 beschreiben die Algenzüchtung in Mineralwasser, Heilwasser, Thermalwasser natürlichen Ursprungs oder in bis zu einer Konzentration von 10<-><2> Mol/Liter mit Metallverbindungen angereicherten Gemischen dieser.
Bei jeder dieser dargelegten Lösungen werden unter Anwendung steriler Bedingungen zu Ernährungs-, Fütterungs- oder therapeutischen Zwekken verwendbare Algen künstlich hergestellt. Gemäss diesen Verfahren werden die Algen unter Bedingungen gezüchtet, die gewährleisten, dass die Eigenschaften der entstehenden Algen denen der in der Natur spontan, unter von Umweltschäden freien Bedingungen entstehenden Algen entsprechen, beziehungsweise dass die Eigenschaften der hergestellten Algen in Abhängigkeit von der Art und Weise der künstlichen sterilen Züchtung von diesen nur in geringem Mass abweichen.
Die gemäss den bekannten Verfahren künstlich gezüchteten Algen enthalten zahlreiche chemische Elemente, wie zum Beispiel Selen, Zink, Silber usw., überhaupt nicht oder nur in Spuren.
Es ist weiterhin bekannt, dass das Selen vielseitige physiologische Funktionen ausübt. Zusammenfassende Artikel hierzu veröffentlichten Thressa und Mitarbeiter (Nutrition Review 35, 7 [1977]), Shamberger (J. of Env. Path. and Tox. 4, 305 [1980]) sowie Masukawa und Mitarbeiter (Experientia 39, 405 [1983]). Aus diesen Artikeln ist also bekannt, dass das Selen an sich ein blutdrucksenkender Stoff ist, die ischämischen, hypoxischen und infarktiösen Zustände des Herzens bessert, und die ceroide Lipofuscinose des Zentralnervensystems ver hindert. Ebenfalls wirksam ist es bei Periodontitis, und ausserdem verringert es mit Sicherheit die Möglichkeit der Entstehung von Krebserkrankungen, weiterhin wird es auch noch als mutagener Inhibitor registriert.
Der Mangel an Selen verursacht z.B. die folgenden Veränderungen beziehungsweise Erkrankungen: Lebernekrose, Muskelnekrose, Destruktion der Erythrocytenmembran, Bindegewebsläsion, als EKG-Veränderung S-T-Elevation, Kwashiorkor-Syndrom, Sclerosis multiplex.
Die günstige Wirkung des Selens beruht hauptsächlich darauf, dass es das Enzym Glutathion-Peroxydase aktiviert. Das Selen ist ein unentbehrlicher Bestandteil der prostetischen Gruppe des Enzyms Glutathion-Peroxydase, das der wichtigste endogene Hemmer der schädlichen Peroxydationsvorgänge ist. Das Selen ist also eine der wichtigsten, essentiellen Substanzen für das Leben, das sich im Organismus nicht anreichert. Daher muss ständig für seinen Nachschub gesorgt werden. Die Aufnahme des Selens in den Organismus erfolgte bisher ausschliesslich über anorganische Verbindungen (SeO2, NaSeO3 usw.).
Ziel der Erfindung war die Herstellung einer Alge mit besonderen biologischen Eigenschaften, die fähig ist, die biologische Wirkung des Selens aufgrund seines erhöhten Selengehalt zu verwirklichen.
Die Erfindung beruht auf der Erkenntnis, dass die Algen in selenhaltiger Nährlösung unter bestimmten Bedingungen nicht nur am Leben erhalten werden können, d.h. nicht kaputtgehen, sondern dass sie in einer das Selen in hoher Konzentration enthaltenden Nährlösung auch wachsen, d.h. dass sie züchtbar sind. Diese Erkenntnis ist überraschend, weil nach dem Stand der Technik zu erwarten war, dass die Algen von dem für sie giftigen Selen zugrunde gehen.
Die Erfindung beruht weiterhin auf der Erkenntnis, dass die Algen bei Züchtung in selenhaltiger Nährlösung unter bestimmten Bedingungen während ihres Wachstums das ansonsten für sie giftige Selen in ihren eigenen Organismus einbauen.
Daher ist Gegenstand der Erfindung ein Verfahren zur Herstellung von Algen mit verbesserter biologischer Wirkung, wobei man den ausgewählten Algenstamm unter sterilen Bedingungen in einer aus Süsswasser und Nährsalzen zusammengestellten Nährlösung, in Gegenwart von Licht und Kohlendioxyd auf fotosynthetischem Wege, oder ohne Licht auf einem auch Kohlenstoff-, Wasserstoff-, Sauerstoff- und Stickstoffquellen enthaltenden Nährboden züchtet und dann die erhaltenen Algen isoliert. Im Sinne der Erfindung wird so verfahren, dass man der Nährlösung in einer Konzentration zwischen 10<-><7> Mol/Liter und 2.10<-><3> Mol/Liter anorganische und/oder organische Selenverbindung(en) zusetzt und diese Nährlösung mit einer Algen-Reinkultur, die in der Lage ist, das Selen gut zu inkorporieren, impft.
Die Algen-Reinkultur wird derart gewonnen, dass man den ausgewählten Algenstamm auf einen anorganische und gegebenenfalls auch organische Nährstoffe enthaltenden flüssigen Nährboden impft, dann mit N-Methyl-N min -nitro-N-nitrosoguanidin behandelt, danach den Stamm auf einem auch Selen enthaltenden Nährboden züchtet, dann die Individuen des auf diese Weise durch Mutation gewonnenen Stammes isoliert werden, die nach der Mutation bei der Züchtung das Selen gut inkorporieren und deren Wuchskraft mindestens so stark wie die des wilden Stammes ist. Nach Einimpfen mit der so gewonnenen Reinkultur - nach Ablauf der Züchtungszeit - werden die Algen isoliert, gewünschtenfalls bis zu einem beliebigen Feuchtigkeitsgehalt getrocknet und auf an sich bekannte Weise durch Ultraschall oder Mahlen aufgeschlossen.
Im Sinne der Erfindung wird aus vorzugsweise die zum Inkorporieren des Selens fähige Reinkultur vorzugsweise aus einzelligen Grün- beziehungsweise Blaualgen, beispielsweise aus Chlorella sp., Scenedesmus sp., Spirulina sp. derart hergestellt, dass man nach der Behandlung mit N-Methyl-N min -nitro-N-nitrosoguanidin die Zellen sorgfältig auswäscht und auf einer nach halbierender Verdünnung hergestellten festen Nährbodenreihe verteilt, die durch Selenverbindungen in einer Konzentration von 2.10<-><3> Mol/Liter bis zu 10<-><7> Mol/Liter ergänzt wird. Von den sich hier entwickelnden, schnell wachsenden, das Selen gut inkorporierenden Stämmen werden die vorteilhaftesten herausgesucht und auf flüssigem Nährboden weitergezüchtet, und man unterzieht die so aufrechterhaltene Algen-Reinkultur industrieller Herstellung.
Der wie oben behandelte Algenstamm baut also während seines Wachstums das Selen in seinen eigenen Organismus ein, und zwar im Vergleich zum ursprünglichen Algenstamm durchschnittlich in einer 10<4>-fachen Konzentration.
Zur industriellen Züchtung von Algenstämmen, die das Selen gut inkorporieren können, werden mit destilliertem Wasser bereitete Lösungen verwendet, die neben den gebräuchlichen bekannten Nährstoffen auch durch anorganische und/oder organische Selenverbindungen ergänzt werden. Die so hergestellte und sterilisierte Nährlösung wird mit dem auf die beschriebene Weise gewonnenen, das Selen gut inkorporierenden Algenstamm geimpft. Die Algen bauen während ihres Wachstums das Selen - zusammen mit wichtigen Spurenelementen - in ihren eigenen Organismus stufenweise, ohne die toxischen, unerwünschten, früher beschriebenen verschmutzenden Agenzien, ein.
Die so hergestellten Algen werden aus der Nährlösung isoliert, dann auf bekannte Weise unter milden Bedingungen, optimal bei 65 DEG C, aber höchstens bei 80 DEG C, vorsichtig getrocknet. Die getrockneten Algen werden vorteilhaft durch Mahlen aufgeschlossen (Feinheit des Mahlens: 1 mu m). Eine andere Möglichkeit ist, das erhaltene nasse Algenkonzentrat mittels Ultraschall aufzuschliessen, dann die aufgeschlossene Zellmasse unter den obigen Bedingungen zu trocknen.
Das erhaltene Algenpulver, dessen Selengehalt 250-4000 mu g/g beträgt, kann vorteilhaft an sich direkt konsumiert beziehungsweise verbraucht werden, beziehungsweise kann es in Lebensmitteln, Futter, Kosmetika oder zusammen mit therapeutisch wirksamen, biologisch aktiven Substanzen und/oder diesen zugesetzt, in im Handel befindlichen Ausführungsformen dieser Produkte, wie vorzugsweise in kapsulierter, tablettierter oder anderer Form, Verwendung finden.
Die Erfindung hat die folgenden wichtigen Vorteile:
a) Die Algen können in einer einfachen Vorrichtung, mit einem leicht verwirklichbaren Verfahren, billig gezüchtet werden.
b) Nach der Züchtung ist keine Reinigung oder Aufarbeitung in mehreren Schritten erforderlich.
c) Die Algen werden unter sterilen Bedingungen, durch Ausschalten von Umweltverschmutzungen gezüchtet, so dass die gewonnenen Algen zum Verbrauch durch den Menschen eindeutig geeignet sind.
d) Das Verfahren ermöglicht die Herstellung von Algen mit hohem Selengehalt und besserer biologischer und physiologischer Wirkung als die der bisher bekannten Algen.
e) Die mit dem erfindungsgemässen Verfahren gezüchteten Algen können auf vielen Gebieten, so in der Ernährung, in der kosmetischen Industrie, in der Therapie und auch auf anderen Gebieten, vorteilhaft angewendet werden.
Besonders vorteilhaft können die mit dem erfindungsgemässen Verfahren gezüchteten Algen in der Therapie verwendet werden, da das Selen die prostetische Gruppe des in den physiologischen Vorgängen eine wichtige Rolle spielenden Enzyms Glutathion-Peroxydase aktiviert. Da das Selen nicht im Organismus angereichert wird, ermöglicht die erfindungsgemässe Alge auch den Selennachschub. Dadurch eröffnet sich eine gute Möglichkeit zum Behandeln aller Gesundheitsschäden, die unter anderem auf Selenmangel zurückzuführen sind.
Das erfindungsgemässe Verfahren wird in den folgenden Beispielen näher erläutert, ohne dass dabei der Schutzumfang auf diese Beispiele eingeschränkt wird.
Beispiel 1
Auf flüssigem Bold-Nährboden, der 100-500 mu g/ml N-Methyl-N min -nitro-N-nitrosoguanidin enthält, wird die Kultur Scenedesmus obtisiusculus in einen 250 ml-Kolben bei 25-27 DEG C in einer geschüttelten Kultur 30 Minuten behandelt. Nach der Behandlung werden die Zellen sorgfältig ausgewaschen, dann auf einer mit Agar verhärteten Bold-Nährboden-Reihe verteilt. Die Nährboden-Reihe enthält Selen in einer Menge, die sich von 3,125 mu g/ml auf eine Grössenordnung von 400 mu g/ml erhöht. Die aus den auf dem Nährboden wachsenden, überlebenden Zellen entstehenden Kolonien werden isoliert, und in Laborausmassen werden die ausgewählten Zellinien in Bold-Nährlösung, die mindestens 20 mu g/ml Selen enthält, vermehrt.
Es werden die Kolonien vermehrt, die das Selen am besten inkorporieren können und in einer mindestens 20 mu g/ml Selen enthaltenden Nährlösung die gleiche Wuchskraft aufweisen wie der wilde Kontrollstamm.
Die isolierten und mit Wasser sorgfältig gewaschenen Zellen werden bei 65 DEG C getrocknet und dann mit Ultraschall aufgeschlossen. Der Selengehalt des Algenpulvers wird mit Hilfe der Atomabsorptions-Methode bestimmt.

Beispiel 2
In ein Algenzüchtungsglas mit einem Fassungsver mögen von 10 Liter werden 8 Liter Knop-Pringsheim-Nährlösung gefüllt, der 40 mg Natriumselenit zugegeben werden. Die so erhaltene Nährlösung wird bei 121 DEG C 30 Minuten lang bei einem Überdruck von 1 bar sterilisiert. Danach wird die sterile Lösung gekühlt und mit der das Selen gut inkorporierenden Algen-Reinkultur Scenedesmus obtisiusculus geimpft. Durch die Nährlösung wird bei 25 DEG C sterile, 5 Vol.-% Kohlendioxyd enthaltende Luft geleitet, und das System wird durch eine Leuchtstoffröhre (Leuchtkraft: 4000 lux, Wellenlänge: 440-520 und 640-700 mu m) beleuchtet. Nach einem 14-tägigen Züchtungszyklus werden die Algen von der Nährlösung isoliert und mit Wasser gewaschen. Die so erhaltene Algenmasse wird mit Ultraschall aufgeschlossen, dann vorsichtig bei einer Temperatur unter 65 DEG C getrocknet.
Der Selengehalt des Algenpulvers beträgt 1200 mu g/g.
Beispiel 3
In einen Algenzüchtungsfermentor von 10 Liter Fassungsvermögen werden 8 Liter Wasser gefüllt, in dem 8,0 g NaNO2, 0,8 g MgSO4.7H2O, 0,8 g K2HPO4, 2,5 ml Arnon-Spurenelementlösung, weiterhin 5 g Traubenzucker, 0,1 g Cystein und 0,1 g Methionin gelöst werden. Die Nährlösung wird durch 50 mg Natriumselenit ergänzt, dann durch einen sterilen Filter geleitet, und unter streng eingehaltenen sterilen Bedingungen mit der das Selen gut inkorporierenden Algen-Reinkultur Scenedesmus obtisiusculus geimpft. Nach einer 4-tägigen Züchtung bei 25-28 DEG C im Dunkeln werden die Algen von der Nährlösung isoliert, mit Wasser gewaschen, dann wird die erhaltene Algenmasse mit Ultraschall aufgeschlossen und schliesslich bei höchstens 65 DEG C getrocknet. Der Selengehalt des gewonnenen Algen pulvers beträgt 1380 mu g/g Algenpulver.
Beispiel 4
Es wird in allem wie in Beispiel 2 verfahren, mit dem Unterschied, dass die Nährlösung anstatt Wärmebehandlung durch Hindurchleiten durch einen sterilen Bakterienfilter vom Typ G-5 sterilisiert wird. Der Selengehalt des erhaltenen Algenpulvers beträgt 1300 mu g/g.
Beispiel 5
Es wird in allem wie in Beispiel 2 oder 3 verfahren, mit dem Unterschied, dass anstatt dem Stamm Scenedesmus obtisiusculus der das Selen gut inkorporierende Algenstamm Chlorella vulgaris verwendet wird, der einer Mutationsbehandlung gemäss Beispiel 1 unterzogen wurde. In ein Algenzüchtungsglas mit einem Fassungsvermögen von 10 Liter werden 8 Liter steril filtrierter Bold-Nährboden gefüllt, dem 5.10<-><3> Mol Na2SeO3 zugesetzt werden. Die Nährlösung wird mit der wie in Beispiel 1 beschrieben hergestellten, Selen gut inkorporierenden Reinkultur Chlorella fusca geimpft, und die Algen werden 14 Tage lang auf die in Beispiel 2 beschriebene Weise gezüchtet. Dann wird die Algenmasse filtriert, sorgfältig mit Wasser gewaschen und bei einer Temperatur unter 80 DEG C getrocknet. Der Selengehalt des gewonnenen Algenpulvers beträgt 2700 mg/g.
Beispiel 11
Der auf die in Beispiel 1 beschriebene Weise gezüchtete, Selen gut inkorporierende Algenstamm Scenedesmus obliquus wird in ein Züchtungsgefäss, das 8 Liter steril filtrierte Bold-Nährlösung und 10<-><7> Mol Na2SeO3 enthält, eingeimpft. Im weiteren wird wie in Beispiel 10 verfahren. Der Selengehalt des gewonnenen Algenpulvers beträgt 300 mu g/g.
Beispiel 12
Es wird in allem wie in Beispiel 2 verfahren, mit dem Unterschied, dass die isolierten Algen bei 65 DEG C getrocknet und dann durch Mahlen auf 1 mu m aufgeschlossen werden.
The invention relates to a method for producing algae with a biological effect.
The algae have been used by mankind since time immemorial for nutritional and feeding purposes. For example, the Far Eastern peoples primarily consume seaweed, but recently they have also been used in dried form or as tablets in developed countries. The algae are carriers of very valuable nutrients, their dried forms contain in high concentration the substances necessary for a healthy life, such as vitamins, proteins, protein-microelement complexes, saccharides, polyunsaturated fatty acids etc.
In recent years, pollution has taken on a global scale, so that pollution of the seas, oceans by toxic heavy metals (Pb, Hg, Cd, Cu) or carcinogenic agents, such as polycondensed cyclic aromatic compounds, has reached a level that should not be neglected. A particular problem is that marine algae accumulate these agents in their organisms. The "whole algae" extracted from the sea or grown under other natural conditions cannot therefore be used for human or animal food, feeding, cosmetic or therapeutic purposes, only the fraction that contains the toxic substances or substances can be used harmful biological effects.
Such a cleaning process is described, for example, by Carames de Gouvea (Cosmetics and Toiletries, 95, 47/1980 /). These procedures can be performed in open pools under sunlight or in closed conditions that ensure sterility, with natural or artificial lighting or without lighting.
According to Japanese Patent Application Laid-Open No. 5 696 690, algae are grown in sterilized sea water, in a solution consisting of various nutrient salts, using sea algae strains and using an artificial light source.
Japanese Patent Application Laid-Open No. 4,517,146 describes the industrial cultivation of single-celled green chlorella algae, according to which the algae are grown in sterile nutrient solution with the exclusion of light and carbon dioxide, in a fresh aqueous medium.
French Patent No. 2 103 462 also describes the cultivation of unicellular green freshwater algae, according to which industrial algae cultivation is carried out by the use of nutrient solution containing the corresponding nutrient salts, in a photosynthetic manner.
The published Hungarian patent applications No. 4613/84 and 4614/84 describe the algae cultivation in mineral water, medicinal water, thermal water of natural origin or in mixtures of these enriched with metal compounds up to a concentration of 10 <-> 2 mol / liter.
In each of these solutions, algae that can be used for nutritional, feeding or therapeutic purposes are produced artificially using sterile conditions. According to these methods, the algae are grown under conditions which ensure that the properties of the algae formed correspond to those of the algae spontaneously in nature under conditions free from environmental damage, or that the properties of the algae produced depend on the nature of the Artificial sterile breeding deviate only slightly from these.
The algae artificially grown according to the known methods contain numerous chemical elements, such as selenium, zinc, silver, etc., not at all or only in traces.
It is also known that selenium has a variety of physiological functions. Thressa et al. (Nutrition Review 35, 7 [1977]), Shamberger (J. of Env. Path. And Tox. 4, 305 [1980]) and Masukawa et al. (Experientia 39, 405 [1983]) published summary articles on this. . It is therefore known from these articles that selenium itself is an antihypertensive substance, which improves ischemic, hypoxic and infarctial states of the heart, and prevents ceroid lipofuscinosis of the central nervous system. It is also effective for periodontitis, and it also reduces the possibility of developing cancer, and it is also registered as a mutagenic inhibitor.
The lack of selenium causes e.g. the following changes or diseases: liver necrosis, muscle necrosis, destruction of the erythrocyte membrane, connective tissue lesion, as an ECG change S-T elevation, Kwashiorkor syndrome, sclerosis multiplex.
The beneficial effect of selenium is mainly due to the fact that it activates the enzyme glutathione peroxidase. Selenium is an indispensable component of the prostatic group of the enzyme glutathione peroxidase, which is the most important endogenous inhibitor of the harmful peroxidation processes. So selenium is one of the most important, essential substances for life that does not accumulate in the organism. Therefore, it has to be constantly replenished. So far, selenium has been absorbed into the organism exclusively via inorganic compounds (SeO2, NaSeO3, etc.).
The aim of the invention was to produce an algae with special biological properties, which is able to achieve the biological effect of selenium due to its increased selenium content.
The invention is based on the knowledge that the algae in selenium-containing nutrient solution can not only be kept alive under certain conditions, i.e. not break, but that they also grow in a nutrient solution containing the selenium in high concentration, i.e. that they are breedable. This finding is surprising because, according to the state of the art, it was to be expected that the algae would perish from the selenium which was toxic to them.
The invention is further based on the knowledge that the algae, when grown in selenium-containing nutrient solution under certain conditions, incorporate the otherwise poisonous selenium into their own organism during their growth.
The invention therefore relates to a process for the production of algae with an improved biological effect, the selected algae strain being sterilized in a nutrient solution composed of fresh water and nutrient salts, in the presence of light and carbon dioxide by photosynthesis, or without light on a carbon as well -, Hydrogen, oxygen and nitrogen sources containing growing medium and then isolated the algae obtained. In the sense of the invention, the procedure is such that inorganic and / or organic selenium compound (s) and this nutrient solution are added to the nutrient solution in a concentration between 10 <-> 7 mol / liter and 2.10 <-> 3 mol / liter vaccinated with a pure algae culture that is able to incorporate the selenium well.
The pure algae culture is obtained in such a way that the selected algae strain is inoculated onto a liquid nutrient medium containing inorganic and possibly also organic nutrients, then treated with N-methyl-N min -nitro-N-nitrosoguanidine, and then the strain is treated with a selenium-containing one Culture medium, then the individuals of the strain obtained in this way by mutation are isolated, who incorporate the selenium well after the mutation in the culture and whose vigor is at least as strong as that of the wild strain. After inoculation with the pure culture obtained in this way - after the end of the breeding period - the algae are isolated, if desired dried to any moisture content and broken down in a manner known per se by ultrasound or grinding.
For the purposes of the invention, the pure culture capable of incorporating the selenium is preferably made from single-celled green or blue algae, for example from Chlorella sp., Scenedesmus sp., Spirulina sp. prepared in such a way that, after treatment with N-methyl-N min -nitro-N-nitrosoguanidine, the cells are carefully washed and distributed on a solid nutrient medium row prepared after halving the dilution, which is produced by selenium compounds in a concentration of 2.10 <-> <3> Mol / liter up to 10 <-> <7> mol / liter is added. The most advantageous of the developing, fast growing, selenium-incorporating strains are selected and cultivated on liquid nutrient medium, and the pure algae culture thus maintained is subjected to industrial production.
The algae strain treated as above thus incorporates the selenium into its own organism as it grows, and in average 10 × 4 times the concentration compared to the original algae strain.
For the industrial cultivation of algae strains which can incorporate the selenium well, solutions prepared with distilled water are used which, in addition to the customary known nutrients, are supplemented by inorganic and / or organic selenium compounds. The nutrient solution thus produced and sterilized is inoculated with the algae strain obtained in the manner described and well incorporating the selenium. During their growth, the algae gradually incorporate the selenium - together with important trace elements - into their own organism without the toxic, undesirable, polluting agents described earlier.
The algae thus produced are isolated from the nutrient solution, then carefully dried in a known manner under mild conditions, optimally at 65 ° C., but at most at 80 ° C. The dried algae are advantageously broken down by grinding (fineness of grinding: 1 μm). Another possibility is to digest the wet algae concentrate obtained by means of ultrasound, then to dry the digested cell mass under the above conditions.
The algae powder obtained, the selenium content of which is 250-4000 mu g / g, can advantageously be consumed or consumed directly per se, or it can be added to foods, feed, cosmetics or together with therapeutically active, biologically active substances and / or these Commercially available embodiments of these products, such as preferably in encapsulated, tableted or other form, are used.
The invention has the following important advantages:
a) The algae can be grown cheaply in a simple device, with an easily implementable process.
b) After breeding, no cleaning or processing in several steps is required.
c) The algae are grown under sterile conditions by switching off environmental pollution, so that the algae obtained are clearly suitable for human consumption.
d) The method enables the production of algae with a high selenium content and a better biological and physiological effect than that of the previously known algae.
e) The algae cultivated with the method according to the invention can be used advantageously in many fields, for example in nutrition, in the cosmetic industry, in therapy and also in other fields.
The algae grown with the method according to the invention can be used particularly advantageously in therapy since the selenium activates the prosthetic group of the enzyme glutathione peroxidase, which plays an important role in the physiological processes. Since the selenium is not accumulated in the organism, the algae according to the invention also make it possible to replenish selenium. This opens up a good opportunity to treat all health problems that are due, among other things, to selenium deficiency.
