BESCHREIBUNG
Die Erfindung betrifft neue Dihydroapovincaminsäureester, wie sie in den Ansprüchen 1 bis 3 definiert sind, Verfahren zu ihrer Herstellung sowie die Verwendung der Verbindungen.
In der BE-PS 904619 sind neue Verfahren zur Herstellung von teilweise bekannten, teilweise neuen, optisch aktiven Eburnamenin-Derivaten der Formel Ia oder lb der vorliegenden Ansprüche 7 und 8 sowie deren Racemate und Säureadditionssalze beschrieben.
Jenes Verfahren ermöglicht es, die in Frage stehenden Verbindungen auf einfache Weise, in guter Ausbeute und sogar stereoselektiv herzustellen.
Die der vorliegenden Erfindung zugrunde liegende Aufgabe war nun die Schaffung neuer Ausgangsstoffe für die Durchführungjenes Verfahrens.
Erfindungsgemäss wird diese Aufgabe durch die in den Ansprüchen 4 bis 6 definierten Verfahren gelöst.
Die Verbindungen der Formeln IIIa und IIlb sind, wie erwähnt, neu. Sie sind nicht nur wertvolle Ausgangsstoffe für die Verfahren der genannten BE-PS, sondern können auch als Ausgangsstoffe für die Herstellung anderer pharmakologisch wirksamer Verbindungen dienen.
In der HU-PS 171163 sind zwar die racemischen trans 14,15-Dihydroeburnamenin-Derivate, welche aus den entsprechenden 3,4-Dehydroeburnamenin- Derivaten durch Reduktion hergestellt werden, beschrieben. Die Beschreibung enthält jedoch keinen Hinweis auf die Konfiguration der Gruppen am C-Atom in 14-Stellung. Aus der Natur der Reaktion folgt jedoch, dass nur das eine Racemat des tans-Derivates entstehen kann.
Die Herstellung der als Ausgangsstoffe dienenden Apovino aminsäureester der Formel In rist in der HU-Patentanmeldung 1753/81 beschrieben. Gemäss dieser Anmeldung werden die gewünschten Verbindungen durch Behandeln von Hydroxyimino octahydro-indolo(2,3-a)chinazolin-Derivaten mit konzentrierten Mineralsäuren oder mit aliphatischen oder aromatischen Sulfonsäuren in einem inerten organischen Lösungsmittel hergestellt.
In den Verbindungen der Formeln IIIa und IIlb können die Ringe C und D in cis- oder in trans-Verbindung stehen. Bei einer cis-Verbindung der Ringe stehen das 3-ständige Wasserstoffatom und die 14-ständige Gruppe Ri in cis-Stellung zueinander, während sie bei einer trans-Verbindung der Ringe trans-ständig zueinander sind. Das Wasserstoffatom im 14-Stellung und die Gruppe ROOC- können räumlich axial oder äquatorial zueinander stehen.
In den Formeln Ia, lb, IIIa, IIlb und IV können die Alkylgruppen R1 und R' gerade oder verzweigt sein. Insbesondere können sie Methyl-, Äthyl-, n-Propyl-, i-Propyl-, n-Butyl-, sec.-Butyl oder tert.-Butylgruppen sein; vorzugsweise sind sie Methyl- oder Äthylgruppen.
Die Erfindung wird im folgenden ausführlicher beschrieben.
Gemäss dem Verfahren nach Anspruch 4 wird ein ApoWlcamin- säureester der Formel IV katalytisch hydriert. Die katalytische Hydrierung wird vorzugsweise in Gegenwart von 5- bis lOprozen- tiger Palladiumaktivkohle in einem alkoholischen Lösungsmittelmedium, zum Beispiel methanolischen Lösungsmittelmedium, mit elementarem Wasserstoff oder einer anderen Wasserstoffquelle, vorzugsweise Ameisensäure, bei Temperaturen zwischen Raumtemperatur und 60 " C, vorzugsweise bei etwa 40 " C vorgenommen.
Der grosse Vorteil dieses Verfahrens besteht darin, dass nur ein einziges Epimer des Dehydroapovincaminsäure-Derivates entsteht, und zwar die sich unter kinetisch kontrollierten Bedingungen bildende, eine axiale Alkoxycarbonylgruppe enthaltende Verbindung der Formel IIIa.
Diese kann nach dem Verfahren nach Anspruch 5, in an sich bekannter Weise, in einem alkoholischen Medium, zweckmässigerweise dem der Bedeutung von R' entsprechenden Alkohol, in Gegenwart eines in katalytischer Menge eingesetzten basischen Katalysators, vorzugsweise eines Alkalialkoholates wie Lithium-, Natrium- oder Kaliumalkoholat, quantitativ zu dem eine äquatoriale Alkoxycarbonylgruppe enthaltenden Dihydroapovincaminsäureester der Formel IIIb epimerisiert werden.
Die Verbindungen der Formeln IIIa und IIlb können isoliert und/oder gewünschtenfalls in ihre Säureadditionssalze überführt werden.
Bei der Herstellung der cis- oder der tans-Stereoisomeren von Verbindungen der Formeln IIIa und IIlb kann man von dem entsprechenden Stereoisomeren eines Apovincaminsäureesters der Formel IV ausgehen und diesen auf die für das Verfahren nach Anspruch 4 angegebene Weise katalytisch hydrieren. Bei der Reaktion entstehen gleichzeitig zwei cis- bzw. trans-Dihydroapovincaminsäureester, die hinsichtlich ihres C-Atoms in 14-Stellung einander epimer sind.
Die Epimeren können durch selektive fraktionierte Kristallisation voneinander getrennt werden. Die der Formel IIlb entsprechenden A -Epimeren kristallisieren gut aus dem der Gruppe R' entsprechenden Alkohol. Die aus der Mutterlauge durch Eindampfen isolierbaren, der Formel IIIa entsprechenden B Epimeren können gut gereinigt werden, zum Beispiel durch Umkristallisieren aus Diisopropyläther. Die B -epimeren Dihydroapovincaminsäureester können auf die für das Verfahren nach Anspruch 5 beschriebene Weise zu A -Epimeren epimerisiert werden.
Die nach den erfmdungsgemässen Verfahren erhaltenen Verbindungen der Formeln IIIa und IIIb können isoliert werden, indem man den Katalysator zum Beispiel durch Filtrieren aus dem Reaktionsgemisch entfernt, die erhaltene Lösung eindampft, den Rückstand mit einem nicht wassermischbaren Lösungsmittel, zum Beispiel Dichlormethan, Chloroform, Dichloräthan, Benzol oder Toluol, vermischt, gewünschtenfalls alkalisch stellt (vorzugsweise mit 5prozentiger Sodalösung), dann abtrennt, die Lösung mit Wasser wäscht, trocknet und schliesslich eindampft. Das als Eindampfrückstand erhaltene rohe Produkt kann gewünschtenfalls durch Umkristallisieren gereinigt werden.
Die erhaltenen Verbindungen der Formeln IIIa und IIIb können gewünschtenfalls in ihre Säureadditionssalze übergeführt werden. Die Salzbildung kann in einem inerten Lösungsmittel, zum Beispiel einem aliphatischen Alkohol mit 1 bis 6 Kohlenstoffatomen oder einem dipolar-aprotischen Lösungsmittel, wie Äther oder Aceton, vorgenommen werden, indem man die Verbindung in dem Lösungsmittel löst und die Lösung so lange mit der entsprechenden Säure bzw. deren mit demselben Lösungsmittel bereiteten Lösung versetzt, bis das Gemisch schwach sauer reagiert. Dann wird das ausgefallene Säureadditionssalz auf geeignete Weise, zum Beispiel durch Filtrieren, aus dem Reaktionsgemisch abgetrennt.
Die erfindungsgemässen Verfahren sowie zwei Möglichkeiten der Verwendung nach den Ansprüchen 7 und 8 werden an Hand der Beispiele näher erläutert.
Beispiel 1 (-)-3S, 1 6R, 145-14,1 14,15-Dihydro- 14-methoxycarbonyl-eburname- nin (IIIa)
47 g(+)-3S, 16R- l4-Methoxycarbonyl-eburnamenin werden in 380 ml Methanol suspendiert. Nach Spülen mit Stickstoffwird die Suspension mit der Suspension von 12 g 10prozentiger Palladiumaktivkohle in 50 ml Dimethylformamid versetzt. Bei Zimmertemperatur werden 25 ml Ameisensäure zugegeben, und das Gemisch wird unter Stickstoffatmosphäre bei 50 "C 90 Minuten lang gerührt. Nach Zusatz von 300 ml Wasser wird der Katalysator abffltriert und dann zweimal mit insgesamt 100 ml 50prozentigem wässrigem Methanol gewaschen.
Von der mit der Waschflüssigkeit vereinigten Lösung werden unter vermindertem Druck 300 ml abdestilliert, und der Rückstand wird unter intensivem Rühren mit wässrigem Ammoniak auf pH 9 gestellt. Die ausgefallene Substanz wird abfiltnert, zuerst mit Wasser, dann mit destilliertem Wasser neutral gewaschen und schliesslich getrocknet.
Das erhaltene Rohprodukt (47 g) wird in 100 ml Methanol aufgekocht und dann bei 0 "C abffltriert. Man erhält 42,5 g (89,9 Prozent) der kristallinen Tltelverbindung, die bei 217-218 "C schmilzt.

Beispiel 2 ( + )-3 S, 16R, 14R- 14,1 5-Dihydro- 14-methoxycarbonyl-eburnamenin (IIlb)
1 g der gemäss Beispiel 1 erhaltenen Substanz wird in der Lösung von 0,1 g metallischem Natrium in 50 ml wasserfreiem Methanol gelöst und die Lösung zwei Stunden lang am Rückfluss gekocht. Dann werden im Vakuum 25 ml Methanol abdestilliert und 0,5 ml Essigsäure zugesetzt. Nach Eindampfen zur Trockene wird der Rückstand in einem Gemisch aus 50 ml Chloroform und
15 ml Wasser aufgenommen und unter Rühren mit konzentriertem wässrigem Ammoniak auf pH 9 gestellt. Die organische Phase wird abgetrennt, über Magnesiumsulfat getrocknet und nach dem Filtrieren eingedampft. Das erhaltene blassgelbe Öl wird in 4 ml Diisopropyläther aufgekocht.
Die entstandenen Kristalle werden bei 0 C abfiltriert und mit 0,5 ml kaltem Diisopropyläther gewaschen. Man erhält 0,8 g (80 Prozent) der Trtelver- bindung, die bei 113-115 Cschmilzt.

Beispiel 3 (-)-3S,16R, 14R-14, 15-Dihydo-14-hydroxymethyl-eburnamenin (Ia)
36,5 g des gemäss Beispiel 1 erhaltenen Produktes werden in 700 ml wasserfreiem Tetrahydrofuran gelöst. Die Lösung wird unter Stickstoffatmosphäre auf -30 "C gekühlt und mit 6 g Lithiumaluminiumhydrid versetzt. Man lässt die Temperatur innerhalb von 15 Minuten auf 0 C ansteigen, innerhalb weiterer Minuten dann auf 20 "C. Bei dieser Temperatur wird das Reaktionsgemisch noch eine Stunde lang gerührt und dann tropfenweise zuerst mit 10 ml Äthanol, anschliessend mit 10 ml Wasser versetzt. Der ausgefallene Niederschlag wird bei Raumtemperatur abffltriert und durch dreimaliges Suspendieren in insgesamt 300 ml kochendem Chloroform gewaschen.
Das erste, tetrahydrofuranhaltige Filtrat wird im Vakuum bis zur öligen Konsistenz eingedampft. Dann werden die vereinigten Waschflüssigkeiten zugegeben, und das Gemisch wird mit 50 ml gesättigter Kochsalzlösung extrahiert. Die organische Phase wird über Magnesiumsulfat getrocknet, filtriert und im Vakuum zur Trockene eingedampft.
Der Rückstand wird in 60 ml Äthanol aufgekocht, bei 0 " C fil- triert, und die Kristalle werden mit 10 ml kaltem Äthanol gewaschen. Man erhält 30,8 g (92 Prozent) der Trtelverbindung, die bei 208-209 0C schmilzt [ai2D0 = -132,1" (c = 1, Chloroform)

Beispiel 4 ( + )-3S,16R314S- 14,15-Dihydro- 14-hydroxymethyl-eburnamenin (lb)
Die Lösung von 0,4 g (+)-3S,16R,14R-14, 15-Dihydro-
14-methoxycarbonyl-eburnamenin (erhalten nach Beispiel 2) in
15 ml wasserfreiem Tetrahydrofuran wird auf -30 " C gekühlt und unter Stickstoffatmosphäre mit 0,1 g Lithiumaluminiumhydrid versetzt. Man lässt die Temperatur des Gemisches auf 20 "C ansteigen und rührt bei dieser Temperatur eine Stunde lang. Nach Zusatz von 1 ml Methanol und 0,5 ml Wasser wird der gebildete Niederschlag abfiltriert und zweimal mit insgesamt 40 ml kochendem Chloroform gewaschen. Das tetrahydrofuranhaltige Filtrat wird im Vakuum eingedampft, und zu dem erhaltenen öligen
Rückstand wird das zum Waschen benutzte Chloroform gegeben.
Die Lösung wird mit 10 ml gesättigter Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und nach dem Filtrieren im Vakuum-eingedampft. Der erhaltene ölige Rückstand wird in 2 ml Diisopropyläther aufgekocht, bei 0 "C abfiltriert und die kristalline Substanz getrocknet. Man erhält 0,31 g (84,5 Prozent) der Titelverbindung, die bei 165-167 "C schmilzt.

Beispiel 5 (+ )-3R,16S,14S- 14,15-Dihydro-14-äthoxyzarbonyl-eburnamenin (IIIa)
8 g (-)-3R,16S-l4-Äthoxycarbonyl-eburnamenin werden in
70 ml Äthanol suspendiert. Nach Spülen mit Stickstoffwird die mit 8 ml Dimethylformamid bereitete Suspension von 2 g l0pro- zentiger Palladiumaktivkohle zugegeben. Nach Zusatz von 4 ml
Ameisensäure wird das Gemisch bei 40 "C 3 Stunden lang gerührt. Bei der gleichen Temperatur wird der Katalysator abfil triert. Die auf dem Katalysator abgeschiedene Substanz wird zweimal mit insgesamt 200 ml Waschflüssigkeit (15 Prozent Was ser enthaltender Äthylalkohol mit einer Temperatur von 60
70 "C) gewaschen.
Das Filtrat wird im Vakuum auf ein Drittel sei nes Volumens eingedampft. Der Rückstand wird mit 70 ml
Dichlormethan und 100 ml Wasser versetzt und das Gemisch unter energischem Rühren mit wässrigem Ammoniak auf pH 9 alkalisch gestellt. Die Phasen werden voneinander getrennt; die wässrige Phase wird mit 20 ml Dichlormethan extrahiert. Die ver einigten organischen Phasen werden über Magnesiumsulfat getrocknet und dann eingedampft. Der Eindampfrückstand wird mit 10 ml Äthanol aufgekocht und bei 0 "C filtriert. Man erhält
7 g (87,5 Prozent) der Tltelverbindung, die bei 172-173 "C schmilzt.
[o]2D0 = + 175,5 (c = 1, Chloroform) Beispiel 6 (-)-3R, 16S, l4R- 14,1 5-Dihydro- 14-äthoxycarbonyl-eburnameni- n-hydrochlorid (IIIb.HCI)
1,5 g des gemäss Beispiel 5 erhaltenen Produktes werden in 20 ml wasserfreiem Äthanol gelöst und mit 0,1 g Kaliumtert.-butylat versetzt. Das Gemisch wird am Rückiluss 2 Stunden lang gekocht und dann mit Essigsäure auf pH 6 angesäuert. Die Lösung wird zur Trockene eingedampft, der Rückstand in 30 ml Dichlormethan gelöst und die Lösung mit 10 ml 2prozentiger Natriumhydrogencarbonatlösung extrahiert. Die organische Phase wird über Magnesiumsulfat getrocknet und im Vakuum eingedampft. Der ölige Rückstand wird in 3 ml Äthanol gelöst und mit äthanolischer Salzsäure auf pH 2-3 gestellt.
Das Gemisch wird auf 0 "C gekühlt, das ausgefallene Hydrochlorid abfiltriert und mit wenig kaltem Äthanol gewaschen. Man erhält 1,0 g der Titelverbindung, die bei 139-241 "C schmilzt.
[a]D - -80,4"
Beispiel 7 (+ )-3R, 165,145-14,1 14,15-Dihydro- 14-methoxycarbonyl-eburname- nin (IIIa)
Man arbeitet auf die im Beispiel 1 beschriebene Weise, verwendet jedoch als Ausgangsverbindung (-)-3R, 16S- l4-Methoxycarbonyl-eburnamenin. Man erhält 43,2 g (91,5 Prozent) der Tltelverbindung, die bei 219-221 "C schmilzt.
[ai20 = 181,5 (c = 1, Chloroform) Beispiel 8 3S,16S-14,1 14,15-Dihydro- 14-äthoxyvarbonyl-eburnamenin A - und B -Epimer
50 g (+ )-3S, 16S- 14-Äthoxycarbonyl-eburnamenin werden in 375 ml Äthanol suspendiert. Zu der Suspension gibt man die mit 40 ml Dimethylformamid bereitete Suspension von 10 g 10pro- zentiger Palladiumaktivkohle. Unter Rühren werden 225 ml Ameisensäure zu dem Gemisch gegeben, und dieses wird bei 40 " C 4 Stunden lang gerührt. Bei dieser Temperatur wird der Katalysator abfiltriert und zweimal mit insgesamt 150 ml 50prozentigem, auf 60 " C erwärmtem Äthanol gewaschen.
Das erhaltene Filtrat wird unter vermindertem Druck auf 200 ml eingedampft und dann mit 300 ml Dichlormethan sowie 200 ml Wasser versetzt. Unter Rühren wird das Gemisch mit konzentriertem wässrigem Ammoniak auf pH 9 alkalisch gestellt. Die Phasen werden voneinander getrennt, und die wässrige Phase wird mit 50 ml Dichlormethan extrahiert. Die vereinigten organischen Phasen werden zur Trockene eingedampft.
a) A -Epimer: ( + )-3S,16S,14R- 14,15-Dihydro- 14-äthoxycarbonyl-eburnamenin (IIlb)
Der Eindampfrückstand wird mit 100 ml Äthanol versetzt und bei 15 "C 2 Stunden lang gerührt. Die ausgeschiedenen Kri stalle werden abffltriert und zweimal mit 5 ml kaltem Äthanol gewaschen. Man erhält 21,5 g Titelverbindung, die bei 151 154 0C schmilzt.
[a]D = + 111,1 (C = 1, Chloroform) b) B -Epimer: (-)-3S,16S,14S-14,1 14,15-Dihydro- 14-äthoxycarbonyl-eburnamenin (IIIa)
Zu der wie unter a) erhaltenen Mutterlauge werden bei Raum temperatur unter Rühren 150 ml destilliertes Wasser gegeben. Die
Lösung nimmt eine milchige Färbung an, und bald beginnen sich
Kristalle abzuscheiden. Das Gemisch wird bei -5 "C 2 Stunden lang gerührt. Die Kristalle werden abfiltriert und zweimal mit
5 ml 50prozentigem, auf 0 " C gekühltem Äthanol gewaschen.
Man erhält 26,2 g Rohprodukt, das bei 88-90 "C schmilzt.
[a]D = -s9,9" (c = 1, Chloroform)
Das Rohprodukt wird aus 50 ml Diisopropyläther umkristalli siert. Auf diese Weise erhält man 20,4 g der Tltelverbindung, die bei 91-92 "C schmilzt.
[a]D = -102,8" (c = 1, Chloroform)
Beispiel 9 3R, 1 6R- 14,1 5-Dihydro- 14-methoxycarbonyl-eburnamenin A und B -Epimer
4 g (-)-3S,16S-14-Äthoxyzarbonyl-eburnamenin werden in
35 ml Methanol suspendiert. Zu dieser Suspension werden die mit 5 ml Dimethylformamid bereitete Suspension von 1,2 g 10prozentiger Palladiumaktivkohle sowie unter Rühren 3 ml
Ameisensäure gegeben. Das Gemisch wird bei 50 "C 90 Minuten lang gerührt. Dann wird der Katalysator durch Filtrieren entfernt und zweimal mit insgesamt 20 ml 50prozentigem, auf 40 " C erwärmtem Methanol gewaschen.
Die methanolische Lösung wird unter vermindertem Druck auf 20 ml eingedampft, mit 25 ml
Chloroform und 20 ml Wasser versetzt und unter Rühren mit konzentriertem wässrigem Ammoniak auf pH 9 alkalisch gestellt.
Die Phasen werden voneinander getrennt, und die wässrige Phase wird mit 20 ml Chloroform extrahiert. Die vereinigten Chloroformphasen werden getrocknet, filtriert und zur Trockene eingedampft.
a) A -Epimer: (-)-3R,16R,14R-14, 15-Dihydro- 14-methoxycarbonyl-eburnamenin (IIIb)
Der erhaltene Eindampfrückstand wird in 6,5 ml Methanol aufgekocht. Aus dem auf 10 abgekühlten Gemisch werden die Knstalle abfiltriert und mit wenig kaltem Methanol gewaschen.
Man erhält 1,2 g der Titelverbindung, die bei 157-159 "C schmilzt.

DESCRIPTION
The invention relates to new dihydroapovincaminic acid esters as defined in claims 1 to 3, processes for their preparation and the use of the compounds.
BE-PS 904619 describes new processes for the preparation of partly known, partly new, optically active eburnamenin derivatives of the formula Ia or 1b of the present claims 7 and 8 and their racemates and acid addition salts.
This process enables the compounds in question to be prepared in a simple manner, in good yield and even stereoselectively.
The object underlying the present invention was now to create new starting materials for carrying out that process.
According to the invention, this object is achieved by the method defined in claims 4 to 6.
As mentioned, the compounds of the formulas IIIa and IIlb are new. They are not only valuable starting materials for the processes of the BE-PS mentioned, but can also serve as starting materials for the production of other pharmacologically active compounds.
The HU-PS 171163 describes the racemic trans 14,15-dihydroeburnamenin derivatives which are prepared from the corresponding 3,4-dehydroeburnamenin derivatives by reduction. However, the description contains no reference to the configuration of the groups on the C atom in the 14-position. However, it follows from the nature of the reaction that only one racemate of the tans derivative can arise.
The preparation of the apovino amino acid esters of the formula In serving as starting materials is described in HU patent application 1753/81. According to this application, the desired compounds are prepared by treating hydroxyimino octahydro-indolo (2,3-a) quinazoline derivatives with concentrated mineral acids or with aliphatic or aromatic sulfonic acids in an inert organic solvent.
In the compounds of the formulas IIIa and IIlb, the rings C and D can be in cis or in trans compound. In the case of a cis connection of the rings, the 3-position hydrogen atom and the 14-position group Ri are in the cis position to one another, while in the case of a trans connection of the rings they are trans-to one another. The hydrogen atom in the 14-position and the group ROOC- can be spatially axially or equatorially to one another.
In the formulas Ia, Ib, IIIa, IIlb and IV, the alkyl groups R1 and R 'can be straight or branched. In particular, they can be methyl, ethyl, n-propyl, i-propyl, n-butyl, sec-butyl or tert-butyl groups; preferably they are methyl or ethyl groups.
The invention is described in more detail below.
According to the method of claim 4, an ApoWlcamin- esters of formula IV is catalytically hydrogenated. The catalytic hydrogenation is preferably carried out in the presence of 5 to 10 percent palladium activated carbon in an alcoholic solvent medium, for example methanolic solvent medium, with elemental hydrogen or another hydrogen source, preferably formic acid, at temperatures between room temperature and 60 ° C., preferably at about 40 ° C. The great advantage of this process is that only a single epimer of the dehydroapovincaminic acid derivative is formed, namely the compound of the formula IIIa which forms under kinetically controlled conditions and contains an axial alkoxycarbonyl group.

In the preparation of the cis or tans stereoisomers of compounds of the formulas IIIa and IIlb, one can start from the corresponding stereoisomer of an apovincamic acid ester of the formula IV and catalytically hydrogenate it in the manner specified for the process according to claim 4. During the reaction, two cis or trans-dihydroapovincamic acid esters are formed at the same time, which are epimeric with respect to one another in terms of their C atom in the 14-position.
The epimers can be separated from one another by selective fractional crystallization. The A epimers corresponding to formula IIlb crystallize well from the alcohol corresponding to group R '. The B epimers which can be isolated from the mother liquor by evaporation and correspond to the formula IIIa can be readily purified, for example by recrystallization from diisopropyl ether. The B epimer dihydroapovincamic acid esters can be epimerized to A epimers in the manner described for the process according to claim 5.
The compounds of the formulas IIIa and IIIb obtained by the processes according to the invention can be isolated by removing the catalyst from the reaction mixture, for example by filtration, evaporating the solution obtained, and the residue with a water-immiscible solvent, for example dichloromethane, chloroform, dichloroethane, Benzene or toluene, mixed, if necessary alkaline (preferably with 5 percent sodium carbonate solution), then separated, the solution washed with water, dried and finally evaporated. The crude product obtained as the evaporation residue can, if desired, be purified by recrystallization.
If desired, the compounds of the formulas IIIa and IIIb obtained can be converted into their acid addition salts. Example 2 (+) -3 S, 16R, 14R- 14,1 5-dihydro- 14-methoxycarbonyl-eburnamenin (IIlb)
1 g of the substance obtained according to Example 1 is dissolved in the solution of 0.1 g of metallic sodium in 50 ml of anhydrous methanol and the solution is refluxed for two hours. Example 3 (-) - 3S, 16R, 14R-14, 15-dihydo-14-hydroxymethyl-eburnamenin (Ia)
36.5 g of the product obtained in Example 1 are dissolved in 700 ml of anhydrous tetrahydrofuran.
