dessen Enantiomers. des racemischen Gemisches und von pharmakologisch anwendbaren Salzen derselben. Die neue Synthese ist einfacher und effektiver als die bekannten Verfahren.
In einer früheren Patentschrift (HU-PS 174 114 = DOS 2 518 160.0) von Chinoin wurden mehrere Synthesen zur Herstellung von y-Glutamyl-taurin beschrieben. Gemeinsam in diesen Synthesen ist die acylierende Verbindung des Taurins nach der Umwandlung in das aktive Derivat z. B. in Aktivester-Derivat. Diese Verbindung ist ein auf der Aminogruppe der Glutaminsäure geschütztes a-Halbester-Derivat der Glutaminsäure z. B. Benzyloxycarbonyl-glutaminsäureu-benzylester.
Der Gegenstand der vorliegenden Verbindung ist ein synthetisches Verfahren wodurch das y-Glutamyl-taurin im Vergleich mit den bekannten Verfahren einfacher, in kürzerer Zeit und mit weniger Chemikalien und mit besserer Ausbeute hergestellt werden kann.

entfernt und man erhält so die gewünsechte Verbindung der Formel (I).
Das erfindungsgemässe Verfahren wird so ausgeführt, dass man das Phtalyl-glutaminsäure-anhydrid der Formel (III) (F.E. King, D.A.A. Kidd, J. Chem. Soc. 1949, 3315) in einer Lösung mit Taurin der Formel (II) umsetzt. Als Lösungsmittel verwendet man bevorzugt ein organisches Lösungsmittel. Durch Zugabe einer Base als Säurebindemittel läuft die Reaktion besser ab. Das gebildete Phtalyl-yglutamyl-taurin der Formel (IV) wird mit einem für die Entfernung der Phtalylgruppe geeigneten Mittel, zweckmässig mit Hydrazin-hydrat umgesetzt (Chem. Ber. 83, 246), und man erhält y-Glutamyl-taurin (I). Das Produkt wird umkristallisiert und/oder durch Ionenaustausch-chromatographie gereinigt und erwünschtenfalls ins Salz übergeführt.
Untenstehend werden die Vorteile des neuen Verfahrens im Vergleich mit den bekannten Verfahren aufgezählt:
Aus Glutaminsäure erhält man durch eine kurze 4-stufige Synthese y-Glutamyl-taurin. Die früheren Verfahren waren 6-stufige oder noch mehr-stufige Synthesen.
Die aus Glutaminsäure ausgehende Synthese besteht
etwa aus halb so vielen Operationen und dauert etwa halb so viele Arbeitsstunden als das einfachste frühere Verfahren.
Man verwendet wesentlich weniger Chemikalien als das am meisten ökonomische frühere Verfahren.
Gemäss dem neuen synthetischen Verfahren erhält man aus Glutaminsäure das y-Glutamyl-taurin mit einer Totalausbeute von 32%. Diese Ausbeute ist im allgemeinen viel höher als bei den früheren Verfahren auf die Glutaminsäure bezogen.
Weitere Einzelheiten sind den folgenden Beispielen zu entnehmen ohne den Schutzumfang auf die Beispiele einzuschränken.
Beispiel 1
Zu 3,05 g (11 mM) Phtalyl-L-glutaminsäure-anhydrid gibt man 15 ml wasserfreies Acetonitril. Die gebildete Suspension wird auf 0 C abgekühlt und man tropft unter Rühren eine Lösung von 1,25 g (10 mM) Taurin in 2,56 ml (20 mM) Tetramethyl-guanidin und 5 ml Acetonitril dazu.
Die Lösung wird dann 30 Minuten bei 0 C und weitere 30 Minuten bei 20 C gerührt. Nach Beendigung des Rührens wird die Suspension klar. Zu dem Reaktionsgemisch gibt man 1,4 ml (20 mM) 70%-iges Hydrazin-hydrat und 10 ml Ethanol. Die Lösung wird über Nacht bei Raumtemperatur stehen gelassen und bei vermindertem Druck eingeengt. Der Rückstand wird in 50 ml Wasser aufgelöst und man gibt 1,2 ml Eisessig zu. Das ausgeschiedene Phtalyl-hydrazin wird durch Filtrieren oder Zentrifugieren entfernt. Der pH der reinen Lösung wird durch die Zugabe von Ameisensäure auf 2,5 eingestellt. Die Lösung wird auf eine vorher mit einem 1:4:45 Vol. Verhältnis Gemisch von Ameisensäure: Es sigsäure : Wasser (ca. pH = 2,1) equilibrierte 2,2 > < x 40 40 cm Do- wex 50W x 2 (l00-200mesh) Säule in dem H+-Zyclus, aufgetragen.
Mit der gleichen Ameisensäure-Essigsäure-Lösung wird eluiert. Am Anfang des Musterauftragens wird das Eluat schon gesammelt und man hört nach der Beendigung des Musterauftragens nach Auslaufen von 160 ml Eluat auf.
Die Lösung wird bei vermindertem Druck eingeengt. Der kristalline Rückstand wird in einem Exsiccator über Lauge getrocknet und aus einem Gemisch von Dimethylformamid und Wasser = 8:2 Vol. Verhältnis umkristallisiert. Die Mutterlauge wird eingeengt und der so erhaltene Rückstand wird auch umkristallisiert. Die zwei kristallinen Produkte werden vereinigt. Man erhält 1,85 g y-L-Glutamyltaurin (73%).
Schmelzpunkt: 222-223 C. [a]D25 = +20.3 (Wasser. c 3.6).
Papierelektrophoretische Mobilität bei pH = 6,5 auf Cysteinsäure bezogen: 0,73, bei pH = 7 0,53. Papierchromatographischer R°Wert: 0,19 (Butanol-Pyridin-Eisessig-Wasser = 15:10:3:12) Analyse auf die Formel C7H,4N206S (954.26) berechnet: C 33,07%, H 5.55%. N 11.0?oo
O 37,75%. S 17.61% gefunden: C 33.11%, H 5,710/0, N 10. 97%

Beispiel 2
3,05 g (11 mM) Phtalyl-D-glutaminsäure-anhydrid werden mit 1,25 g (10 mM) Taurin gemäss Beispiel 1 umgesetzt.
Man erhält 1,80 gy-D-Glutamyl-taurin (710/0). Schmelzpunkt: 220-222 C.
[am25 = 20,4- (Wasser, c 5,1).
Beispiel 3
3,05 g (11 mM) Phtalyl-DL-glutaminsäure-anhydrid werden gemäss Beispiel 1 mit 1,25 g (10 mM) Taurin umgesetzt. Man erhält 1,88 g y-DL-Glutamyl-taurin (74%).
Schmelzpunkt: 209-212 C.

The process according to the invention is carried out by reacting the phthalyl-glutamic acid anhydride of the formula (III) (F.E. King, D.A.A. Kidd, J. Chem. Soc. 1949, 3315) in a solution with taurine of the formula (II). An organic solvent is preferably used as the solvent. The reaction proceeds better by adding a base as an acid binder. The phthalyl-yglutamyl-taurine of formula (IV) formed is reacted with an agent suitable for removing the phthalyl group, expediently with hydrazine hydrate (Chem. Ber. 83, 246), and y-glutamyl-taurine (I) is obtained. . The product is recrystallized and / or purified by ion exchange chromatography and, if desired, converted into the salt.
The advantages of the new method compared to the known methods are listed below:
A short 4-step synthesis gives y-glutamyl-taurine from glutamic acid. The earlier processes were 6-step or even more step syntheses.
The synthesis based on glutamic acid consists

example 1
15 ml of anhydrous acetonitrile are added to 3.05 g (11 mM) of phthalyl-L-glutamic acid anhydride. The suspension formed is cooled to 0 ° C. and a solution of 1.25 g (10 mM) taurine in 2.56 ml (20 mM) tetramethylguanidine and 5 ml acetonitrile is added dropwise with stirring.
