Dimethyl(N-cyanimido)carbonat hat sich als bevorzugtes Reagens bei der Synthese von Verbindungen, die Hystamin-Antagonisten an H2-Rezeptoren sind, z.B. Cimetidin, erwiesen. Aus J. Org. Chem. 1974, 39,1522 ist bekannt und darin beschrieben ein Verfahren zur Herstellung von Dimethyl(N-cyanimido)carbonat aus Dimethyl(N-cyanimido)dithiocar-bonat. Ferner ist bekannt und in der Literatur beschrieben die Synthese von Diethyl(N-cyanimido)carbonat mit Cyanamid im nicht wässerigen Medium bei 0 °C (Chem. Ber. 1967,100,2604). In der EP-Al-0014064 ist ein Verfahren zur Herstellung von (N-Cyanimido)carbonaten beschrieben, bei welchem die Umsetzung im System Wasser/mit Wasser nicht mischbares organisches Lösungsmittel, vorzugsweise Toluol, erfolgt. Die Umsetzung verläuft derart, dass das intermediäre disubstituierte Imidocarbonat aus Chlorcyan oder Brom-cyan und dem entsprechenden Alkohol in Gegenwart einer Base in situ gebildet wird. Vor der Vesetzung mit Cyanamid muss das Reaktionsgemisch mit conc. The reaction proceeds in such a way that the intermediate disubstituted imidocarbonate is formed in situ from cyanogen chloride or cyanogen bromide and the corresponding alcohol in the presence of a base. Before the addition of cyanamide, the reaction mixture must be treated with conc. HCl to be neutralized to pH 7.

Das erfmdungsmässige Verfahren ist in Patentanspruch 1 io definiert und beruht auf der Verwendung von Natriumcyanid als Ausgangsstoff. Vorzugsweise wird eine äquivalente Menge Natriumcyanid in Wasser gelöst und danach mit überschüssigem Natriumhydroxid und dem entsprechenden Alkohol, vorzugsweise Methanol, versetzt. In die gebildete, 15 auf 0 °C abgekühlte Lösung wird eine äquivalente Chlormenge mit einer solchen Geschwindigkeit eingeleitet, dass die Temperatur von 5 C nicht überschritten wird. Nach beendetem Einleiten von Chlor und Neutralisation des Reaktionsgemisches mit konzentrierter Salzsäure wird mit Me-20 thylenchlorid und danach mit einer wässerigen Cyanamid-Lösung und konzentrierter Salzsäure versetzt.The inventive method is defined in claim 1 io and is based on the use of sodium cyanide as a starting material. Die wässerige Phase wird zweimal mit Methylenchlorid extrahiert, die vereinigten Extrakte werden mit Na2S04 getrocknet und zur Trockne eingedampft.
