PATENTANSPRÜCHE
Verfahren zur Herstellung von zur Drogenproduktion verwendbaren Sporenkulturen eines Ergocristin produzieren den, auf Roggen virulenten Claviceps purpurea Stammes, da durch gekennzeichnet, dass man die Sporenkultur durch
Züchten des bei dem ungarischen Landesinstitut für Volkshygiene unter Nr. OKI 00163 deponierten Claviceps purpurea
Stammes herstellt.
Die Erfindung betrifft ein Verfahren zur Herstellung einer Sporenkultur eines neuen, Ergocristin produzierenden, auf Roggenpflanzen virulenten Claviceps purpurea Stammes.
Zur Herstellung von Mutterkornalkaloiden sind zur Zeit zwei verschiedene Wege bekannt, einerseits die klassischen Verfahren, in welchen die auf Roggenpflanzen gezüchteten und eingesammelten Drogen als Ausgangsstoff verwendet und die Alkaloide aus diesen durch Extraktion gewonnen werden, und andererseits die auf sterile Fermentation mit geeigneten Claviceps Stämmen gegründete Alkaloidproduktion. Es sind schon mehrere, grossbetrieblich ausführbare Verfahren zur Realisierung der letzteren Methode bekannt (vgl. z.B. die ungarischen Patentschriften Nr. 150 631, 151 724, 238 und 164 816), doch hat die Züchtung von Mutterkorn-Drogen seine Bedeutung nicht verloren; ein erheblicher Teil der Weltproduktion von Peptidalkaloiden ist auch heute noch auf die Verwendung von gezüchteten, an verschiedenen Alkaloid-Typen angereicherten Drogen basiert.
Das ist besonders der Fall in der Produktion von Ergocristin und Ergotamin. Die Seitenkette der Peptidalkaloide besteht aus Prolin, einer Gruppe von charakteristischen Aminosäuren und aus einer für das betreffende Alkaloid (innerhalb der erwähnten Gruppe) spezifisch charakteristischen Aminosäure. Die für die erwähnten beiden Alkaloide (Ergocristin und Ergotamin) charakteristische Aminosäure ist das Phenylalanin. Es ist aber äusserst schwierig, die Bildung bzw. die Überproduktion von Phenylalanin während der sich innerhalb von einigen Tagen abspielenden fermentativen Biosynthese zu sichern, da während der selben Zeit auch der biosynthetische Aufbau des Lysergsäure-Teils des Alkaloidmoleküls stattfindet und dazu aus der Synthese der aromatischen Aminosäuren erhebliche Mengen von Tryptophan erforderlich sind.
Im Laufe der Aufrechterhaltung des Stammes und der auf einander folgenden Umimpfungen kann daher der ursprünglich anwesende Anteil der Ergotamin und Ergocristin produzierenden Stämme durch die ausgleichende Regulation der Stoffwechselwege in die Richtung der Produktion von Ergocornin und dann von wasserlöslichen Alkaloiden verschoben werden. Auch dieser Umstand dürfte dazu beigetragen haben, dass gewisse Fermentationsverfahren später nicht mehr mit Erfolg reproduziert werden konnten (vgl. z.B. tschechoslovakische Patentschrift 104 613, DE-PS 1 215 637, GB-PS 1192 912). Da man bei den auf die Aufarbeitung von Drogen basierten Verfahren mit solchen Schwierigkeiten nicht rechnen muss, werden solche Verfahren auch weiterhin in grossem Massstab verwendet und es besteht auch weiterhin der Bedarf nach der technologischen Weiterentwicklung solcher Verfahren.
Die grundlegende Bedingung einer solchen Weiterentwicklung ist aber die Herstellung von Claviceps purpurea Stämmen mit entsprechend hoher Alkaloidproduktionsfähigkeit.
Verschiedene Verfahren zur Herstellung von Claviceps purpurea Stämmen mit guter Produktionsfähigkeit für Ergolin-Alkaloiden wurden schon in mehreren Patentschriften beschrieben; es wurden verschiedene Wege zur Gewinnung von Stämmen mit höherer Produktionsfähigkeit vorgeschlagen.
Nach dem in der sowjetischen Patentschrift 232 468 beschrie benen Verfahren wird ein durch bestimmte kolonienmorpho logische Eigenschaften und Sporenmasse gekennzeichneter
Stamm für die Produktion von Ergotoxin ausgewählt. In der sowjetischen Patentschrift 232467 ist ein auf künstlichem
Nährboden beinahe mit den obigen identische morphologi sche Eigenschaften zeigender, in 80% Ergocryptin produzie render Stamm beschrieben. In der ungarischen Patentschrift
154 035 ist eine schnell durchführbare und wirksame Methode beschrieben, nach welchem die zur Produktion von Ergotoxin fähige Kulturen auf Grund der Farbe der Kolonien gegenüber den Ergotamin produzierenden Individuen angereichert werden können.
Zur Herstellung von Stämmen mit hoher Alkaloidproduktion sind die folgenden wichtigeren Schritte erforderlich:
1. die Züchtung eines veredelten Claviceps purpurea Stammes;
2. die Herstellung von zur Infizierung von Roggenpflanzen geeigneten Sporen durch fermentative Vermehrung des Stammes,
3. die Konservierung dieser Sporen bis zu deren Verwendung.
Die Zielsetzung der vorliegenden Erfindung war die Herstellung eines besonders Ergocristin in höherer Ausbeute produzierenden Stammes, welcher besonders die folgenden vorteilhaften Eigenschaften zeigt: - der Stamm zeigt an künstlichen Nährböden, sowohl auf Agar-Oberfläche, als auch in submersen, belüfteten Kulturen gute Sporenbildung: - die Sporen sind genügend resistent und somit gut erhaltbar; - die Sporen sind lebensfähig und virulent so dass sie, wenn sie wieder in ein Nährbodenmedium kommen, in 98 bis 100% auskeimen und auf Pflanzen aufgebracht die Pflanze wirksam infizieren; - die Quantität der pro Flächeneinheit produzierten Droge ist hoch; - die Droge zeigt einen hohen Gehalt an Hauptalkaloiden und enthält neben diesen Hauptalkaloiden nur unbeträchtliche Mengen von Nebenalkaloiden.
Der neue Stamm wurde durch die kombinierte Anwendung der geschlechtlichen und der selektiven Veredelung auf die folgende Weise hergestellt:
Aus dem Ausgangsstamm, welcher in der eigenen Sammlung des ungarischen Heilpflanzen-Forschungsinstituts unter Nr. 661/1974 registriert wurde, wurden unter Berücksichtigung der morphologischen Gesichtspunkte gut entwickelte, äusserlich einwandfreie Sklerotien mit glatter Oberfläche ausgewählt. Diese Sklerotien wurden auf feuchten Sand gelegt und 6 Wochen lang zwischen 0 C und 5 C gehalten, dann abgespült und auf einen feucht gehaltenen sterilen Schwamm gebracht. Nach 12 Tagen begann die Entwicklung von Stromen; die erschienenen Stromen haben sich in 6 Tagen voll entwickelt.
Zur Gewinnung von Sporen wurden die Stromen derjenigen Sklerotien ausgewählt, welche nur einige (3 bis 4) Pilzköpfe getrieben haben; von diesen wurden solche Stromen entnommen, deren Durchmesser grösser als 4 mm war und an deren Oberfläche durch mikroskopische Untersuchung die grössten Ausbuchtungen von Peritetien gesichtet wurden.
Diese Stromen wurden dann über einem flüssigen Malz Nährboden, unter sterilen Bedingungen, durch Wärmeeinwirkung zur Sporulation gebracht. Aus der mikroskopisch kontrollierten, Ascosporen enthaltenden Flüssigkeit wurden dann auf festem Malz-Agar-Nährboden Ausbreitungen gemacht. Die sich aus je einer Spore ausbildenden Kolonien wurden auf Grund ihres Entwicklungstempo und ihrer Sporenbildungsfähigkeit selektiert. Die Sporen der ausgewählten Kolonien wurden nach ihrer Vermehrung isoliert und unter parasitische Bedingungen gebracht, d.h. es wurden die Ähren
von sich im Vorantese-Zustand befindenden Roggenpflanzen mit Hilfe von Impfnadeln mit diesen Sporen infiziert. Von den Roggenpflanzen wurden dann diejenigen Sklerotien ausgewählt, bei welchen die Sklerotienansätze innerhalb der kürzesten Zeit nach dem Erscheinen des Honigtaus erschienen sind. Die auf diese Weise eingesammelten Körner wurden durch Dünnschichtchromatographie analysiert. Die weitere Selektion erfolgte auf Grund des Ergocristingehalts und der Menge der anwesenden verunreinigten Nebenalkaloiden.
Die auf diese Weise durch geschlechtliche Vermehrung gewonnenen Sklerotien-Kulturen wurden dann einer weiteren Selektionsreihe unterworfen, in welcher aus den Sklerotienteilchen auf festem Nährboden Kolonien gezüchtet und mit diesen flüssige Nährböden eingeimpft wurden; aus den so hergestellten submersen Kulturen wurden dann in Feldversuchen die am besten sporierenden, lebensfähigsten und höchste Virulenz zeigenden Kulturen auf Grund der Menge und des durchschnittlichen Alkaloidgehalts der produzierten Droge selektiert.
Nach dieser Methode wurde eine Reihe von Selektionen in den auf einander folgenden Schritten von zwei - auch parasitisches Wachstum in sich schliessenden - Vermehrungszyklen durchgeführt, wodurch ein neuer, gegenüber den bisher bekannten Stämmen vorteilhaftere Eigenschaften zeigender Claviceps purpurea Stamm erzeugt werden konnte.
Der neue, bei dem ungarischen Landesinstitut für Volkshygienie H - 1966 Budapest,Gyáli ut 2-4 (OKI) am 2. Mai
1977 unter Nr. MNG 00163 deponierte Stamm ist besonders zur betrieblichen Herstellung von Impfsporen-Präparaten vorteilhaft geeignet, da er die Sporenproduktion der Fermentation erhöht; er zeigt aber auch in der Feldproduktion der Droge vorteilhafte Eigenschaften, erhöht die Menge der produzierten Droge und verbessert erheblich die Qualität der Droge.
In der nachstehenden Tabelle sind die Ergebnisse der mit dem neuen Stamm OKI/MNG 00163 und (zum Vergleich) mit dem Ausgangsstamm Nr. 661/1974 des ungarischen Heilpflanzen-Forschungsinstituts durchgeführten Fermentations- und Feldversuche zusammengefasst.

Die morphologische Charakterisierung des durch die er findungsgemässe Selektion erhaltenen neuen Stammes und des Ausgangsstammes:
Stamm OKI 00163: a) Malz-Agar: nach 28 Tagen weisse, 0,5 bis 0,6 cm grosse Kolonien mit kurzen Luftmyzelien; vom 12 Tag Konidienbildung; Grössenverteilung der Konidien: 1,3 llm bis 3,8 ,um, häufigste Grösse: 2,5 llm.
b) Mannit-Citrat-Agar: nach 28 tagen flache, cremefarbige, 1,0 bis 1,5 cm grosse, faltige Kolonien; Konidienbildung vom 15. Tag angefangen.
c) Durchschnittliche Grösse der Konidien (von 100 unausgewählten Körnern) 2,5 llm; Oberfläche glatt.
Ausgangsstamm 661/1974: a) Malz-Agar: nach 28 Tagen grauweisse, 0,8 bis 12,0 cm grosse Kolonien, mit ausgebreiteten Luftmyzelien; Konidienbildung vom 15 Tag; durchschnittliche Grösse der Konidien: 3,4 tim.
b) Mannit-Citrat-Agar: nach 28 Tagen weisse, 1,5 bis 2,0 cm grosse, filzartige, ausgebreitete Kolonien; Konidienbildung vom 20. Tag angefangen; durchschnittliche Grösse der Konidien: 4 gm.
c) Durchschnittliche Grösse der Sklerotien (von 100 unausgewählten Körnern) 18 mm; Oberfläche rissig.
Die Impfsporenproduktion der Fermentation wird durch die Anwendung des neuen Stammes auf das Zweifache erhöht; die Drogenproduktion wird um 50% erhöht und der Alkaloidgehalt der erhaltenen Droge ist beinahe um 100% höher. Durch den wesentlich höheren Alkaloidgehalt wird auch die Alkaloidausbeute-günstig beeinflusst; als weiterer Vorteil kann noch erwähnt werden, dass die Erhöhung des Alkaloidgehalts nicht von einer erhöhten Anhäufung der Fettstoffe begleitet wird, so dass die relative Menge der Fettstoffe in der Droge niedriger wird.
Das erfindungsgemässe Verfahren wird durch die nachstehenden Beispiele näher veranschaulicht.
Beispiels
In einem Glasfermentor mit 5 Liter Rauminhalt wird eine 24 Tage alte Malz-Agar-Kultur des Stammes OKI 00163 auf einen Nährboden mit folgender Zusammensetzung geimpft: Malzextrakt 2,0% Kartoffelpulver 1,0% Glucose 0,5% pH-Wert des Nährbodens: 6,0
Die Kultur wird bei 24 C, unter Rühren mit 440 U/min Belüftung mit 0,5 Liter Luft pro Liter Fermentationsflüssigkeit pro Minute 3 Tage langinkubiert.
Mit der auf obige Weise erhaltenen Inoculum-Kultur werden dann 100 Liter Nährboden der folgenden Zusammensetzung eingeimpft: Mannit 8,0% Citronensäure 0,7% Maisquellwasser (auf Trockensubstanz berechnet) 0,1% Kaliumdihydrogenphosphat 0,025% Magnesiumsulfat 0,025%
Die Fermentation wird in einem mit Rührwerk ausgerüsteten, säurefesten Fermentor mit 160 Liter Rauminhalt durchgeführt.
Die Inkubation wird bei 24 C, unter Rühren mit 230 U/min und Belüftung mit 1,0 Liter Luft pro Liter Fermenta tionsflüssigkeit pro Minute, bei pH 4,5 bis 5,0, sechs Tage lang fortgesetzt. Der pH-Wert der Fermentationsflüssigkeit wird durch die Zugabe von Ammoniumhydroxyd zwischen den angegebenen Grenzen gehalten.
Am Ende der Fermentation zeigt die Fermentationsflüssigkeit eine Sporenzahl von 4,2:108/ml.
Die erhaltene Biomasse wird abgetrennt, mit 30 Liter 10%aber Saccharoselösung verdünnt, in Flaschen abgefüllt und unter Tielkühlung aufbewahrt.
Das auf obige Weise erhaltene Impfsporen-Präparat zeigt 99% Keimfähigkeit. Dieses Impfsporen-Präparat wird dann auf eine Konzentration von 30 000 Sporen/mm3 verdünnt und in diesem Zustand zum Infizieren von Roggenpflanzen vor der Ahrenbildung verwendet. Aus dem Ernteertrag werden auf diese Weise 250 kg Droge pro ha gewonnen; der Gehalt der Droge an fetten Olen beträgt 20%.
Ergocristingehalt: 6,2 mg/g Ergotamingehalt: 0,1 mg/g Ergometringehalt: 0,2 mg/g.
Beispiel 2
Der Stamm OKI 00163 wird auf Malz-Agar bei 24 C 21Tage inkubiert; die erhaltenen Sporen werden mit steriler physiologischer Kochsalzlösung von der Oberfläche der Kultur abgewaschen. Mit dieser sporenhaltigen Kochsalzlösung werden dann die im Phytotron gezüchteten Roggenpflanzen infiziert; die Sporen werden mit Hilfe einer Injektionsspritze, durch 5 Stiche je Ähre in den Pflanzenkörper eingebracht.
Zum Vergleichen wurde die gleiche Prozedur mit dem Ausgangsstamm 661/1974 in gleicher Weise durchgeführt. Mit den beiden Stämmen wurden je 100 Ahren infiziert.

Beispiel 3
Eine auf Malz-Agar gezüchtete, 28 Tage alte Kultur des Stammes OKI 00163 wurde mit steriler Kochsalzlösung abgewaschen und die Sporenzahl der Waschflüssigkeit auf 30 000 Sporen/mm3 eingestellt. Mit dieser Flüssigkeit wurden auf 30 m2 grosse Versuchsparzellen Roggenpflanzen in vier Parallelversuchen infiziert. Zum Vergleich wurden unter identischen Umständen Parallelversuche mit einer auf gleiche Weise vorbereiteten Kultur des Ausgangsstammes 661/1974 durchgeführt.
Die Infizierung wurde mit der Hand, mit Hilfe von zierplatten durchgeführt; die reifen Sklerotien wurden ebenfalls mit der Hand, verlustfrei eingesammelt. The invention relates to a method for producing a spore culture of a new, Ergocristin-producing Claviceps purpurea strain, which is virulent on rye plants.
Two different ways of producing ergot alkaloids are currently known, on the one hand the classic methods in which the drugs grown and collected on rye plants are used as the starting material and the alkaloids are obtained from them by extraction, and on the other hand the method based on sterile fermentation with suitable Claviceps strains Alkaloid production. Several large-scale procedures for realizing the latter method are already known (see, for example, Hungarian Patent Nos. 150 631, 151 724, 238 and 164 816), but breeding ergot drugs has not lost its importance; A significant part of the world production of peptide alkaloids is still based today on the use of bred drugs enriched with different types of alkaloid.
This is particularly the case in the production of ergocristine and ergotamine. The side chain of the peptide alkaloids consists of proline, a group of characteristic amino acids and an amino acid specific for the alkaloid in question (within the group mentioned). The amino acid characteristic of the two alkaloids mentioned (ergocristine and ergotamine) is phenylalanine. However, it is extremely difficult to ensure the formation or overproduction of phenylalanine during the fermentative biosynthesis that takes place within a few days, since the biosynthetic structure of the lysergic acid part of the alkaloid molecule takes place during the same time, and also from the synthesis of the aromatic ones Amino acids require substantial amounts of tryptophan.
In the course of maintaining the strain and the subsequent vaccinations, the originally present proportion of the strains producing ergotamine and ergocristine can be shifted in the direction of the production of ergocornin and then of water-soluble alkaloids through the balancing regulation of the metabolic pathways. This fact may also have contributed to the fact that certain fermentation processes could not be reproduced successfully later (see e.g. Czechoslovakian patent specification 104 613, DE-PS 1 215 637, GB-PS 1192 912). Since such difficulties are not to be expected in the processes based on the processing of drugs, such processes continue to be used on a large scale and there is still a need for the technological development of such processes.
However, the basic condition of such a further development is the production of Claviceps purpurea strains with a correspondingly high alkaloid production capacity.
Various processes for the production of Claviceps purpurea strains with good production ability for ergoline alkaloids have already been described in several patents; Various ways of obtaining strains with higher productivity have been proposed.

154 035 describes a method which can be carried out quickly and is effective, according to which the cultures capable of producing ergotoxin can be enriched against the individuals producing ergotamine on the basis of the color of the colonies.
The following major steps are required to produce high alkaloid production strains:
1. the cultivation of a grafted Claviceps purpurea strain;
2. the production of spores suitable for infecting rye plants by fermentative propagation of the strain,
3. the preservation of these spores until their use.
The aim of the present invention was to produce a strain which produces ergocristin in particular and which has the following advantageous properties: the strain shows good spore formation on artificial nutrient media, both on the agar surface and in submerged, aerated cultures: the spores are sufficiently resistant and therefore easy to maintain; - The spores are viable and virulent so that when they come back into a nutrient medium they germinate in 98 to 100% and effectively infect the plant when applied to plants; - the quantity of drug produced per unit area is high; - The drug shows a high content of main alkaloids and contains only insignificant amounts of secondary alkaloids in addition to these main alkaloids.
The new strain was created through the combined application of sexual and selective finishing in the following way:
From the original strain, which was registered in the Hungarian Medicinal Plant Research Institute's own collection under No. 661/1974, well developed, externally perfect sclerotia with a smooth surface were selected taking into account the morphological aspects. These sclerotia were placed on damp sand and held between 0 C and 5 C for 6 weeks, then rinsed and placed on a sterile sponge kept moist. The development of currents began after 12 days; the currents that appeared appeared fully in 6 days.
The currents of those sclerotia which drove only a few (3 to 4) mushroom heads were selected to obtain spores; From these currents were taken whose diameter was greater than 4 mm and on the surface of which the largest bulges of peritetia were seen by microscopic examination.
These streams were then sporulated over a liquid malt culture medium under sterile conditions. Spreads were then made from the microscopically controlled liquid containing ascospores on solid malt agar culture medium. The colonies that each formed from a spore were selected on the basis of their speed of development and their ability to form spores. The spores of the selected colonies were isolated after growing and placed under parasitic conditions, i.e. it was the ears of corn
infected with these spores by rye plants in the pre-condition with the help of vaccination needles. From the rye plants, those sclerotia were selected in which the sclerotic approaches appeared within the shortest time after the appearance of the honey dew. The grains collected in this way were analyzed by thin layer chromatography. The further selection was based on the ergocrist content and the amount of contaminated secondary alkaloids present.
The sclerotic cultures obtained in this way through sexual reproduction were then subjected to a further selection series in which colonies were grown from the sclerotic particles on solid nutrient media and inoculated with liquid nutrient media; From the submerged cultures thus produced, the best sporing, most viable and most virulent cultures were selected on the basis of the quantity and the average alkaloid content of the drug produced.
According to this method, a series of selections were carried out in the successive steps of two - also parasitic growth in closed - propagation cycles, whereby a new Claviceps purpurea strain exhibiting properties which were more advantageous than the previously known strains could be produced.

The strain deposited in 1977 under number MNG 00163 is particularly suitable for the production of vaccine spore preparations because it increases the spore production of the fermentation; but it also shows beneficial properties in the field production of the drug, increases the amount of drug produced and significantly improves the quality of the drug.
The table below summarizes the results of the fermentation and field tests carried out with the new strain OKI / MNG 00163 and (for comparison) with the starting strain no. 661/1974 of the Hungarian Medicinal Plant Research Institute.

The morphological characterization of the new strain and the starting strain obtained by the selection according to the invention:
Strain OKI 00163: a) malt agar: after 28 days white, 0.5 to 0.6 cm large colonies with short aerial mycelia; from the 12th day conidia formation; Size distribution of the conidia: 1.3 llm to 3.8 µm, most common size: 2.5 llm.
b) Mannitol citrate agar: after 28 days flat, cream-colored, 1.0 to 1.5 cm large, wrinkled colonies; Conidia formation started on day 15.
c) Average conidia size (out of 100 unselected grains) 2.5 llm; Smooth surface.
Starting strain 661/1974: a) Malt agar: after 28 days gray-white, 0.8 to 12.0 cm large colonies, with spreading aerial mycelia; Conidia formation from the 15th day; average size of the conidia: 3.4 tim.
b) Mannitol citrate agar: after 28 days, white, 1.5 to 2.0 cm large, felt-like, expanded colonies; Conidia formation started on day 20; average size of the conidia: 4 gm.
c) Average size of sclerotia (out of 100 unselected grains) 18 mm; Cracked surface.
The vaccine spore production of the fermentation is increased twice by the application of the new strain; drug production is increased by 50% and the alkaloid content of the received drug is almost 100% higher. The alkaloid yield is also favorably influenced by the significantly higher alkaloid content; as a further advantage it can be mentioned that the increase in the alkaloid content is not accompanied by an increased accumulation of the fatty substances, so that the relative amount of the fatty substances in the drug is lower.
The process according to the invention is illustrated in more detail by the examples below.
Example
In a glass fermentor with a volume of 5 liters, a 24-day-old malt agar culture of the OKI 00163 strain is inoculated onto a nutrient medium with the following composition: malt extract 2.0% potato powder 1.0% glucose 0.5% pH value of the nutrient medium: 6.0
The culture is incubated for 3 days at 24 ° C. with stirring with 440 rpm aeration with 0.5 liters of air per liter of fermentation liquid per minute.
The inoculum culture obtained in the above manner is then used to inoculate 100 liters of medium of the following composition: mannitol 8.0% citric acid 0.7% corn steep liquor (calculated on dry matter) 0.1% potassium dihydrogen phosphate 0.025% magnesium sulfate 0.025%
The fermentation is carried out in an acid-proof fermenter with a volume of 160 liters equipped with a stirrer.
The incubation is continued at 24 C, with stirring at 230 rpm and aeration with 1.0 liter of air per liter of fermentation liquid per minute, at pH 4.5 to 5.0, for six days. The pH of the fermentation liquid is kept between the specified limits by the addition of ammonium hydroxide.
At the end of the fermentation, the fermentation liquid shows a spore count of 4.2: 108 / ml.
The biomass obtained is separated off, diluted with 30 liters of 10% but sucrose solution, bottled and stored under cooling.
The vaccine spore preparation obtained in the above manner shows 99% germination capacity. This inoculation spore preparation is then diluted to a concentration of 30,000 spores / mm 3 and is used in this state to infect rye plants before ear formation. In this way, 250 kg of drug per hectare are obtained from the harvest; the content of the drug in fatty oils is 20%.

Example 2
The OKI 00163 strain is incubated on malt agar at 24 C 21 days; the spores obtained are washed off the surface of the culture with sterile physiological saline. The rye plants grown in the phytotron are then infected with this spore-containing saline solution; the spores are injected into the plant body using 5 syringes per ear.
For comparison, the same procedure was carried out in the same way with the parent strain 661/1974. 100 ears were infected with each of the two strains.

Example 3
A 28 day old culture of the OKI 00163 strain grown on malt agar was washed off with sterile saline solution and the spore number of the washing liquid was adjusted to 30,000 spores / mm 3. With this liquid, rye plants were infected in four parallel experiments on 30 m2 test plots. For comparison, parallel tests were carried out under identical circumstances with a culture of the parent strain 661/1974 prepared in the same way.
The infection was carried out by hand using decorative plates; the mature sclerotia were also collected by hand, without loss.
