worin X in Anspruch 1 definiert ist, mit einer äquimolaren Menge an Natriumhydrogencarbonat unter Kochen in Wasser umsetzt, wobei sich das entsprechende Natriumsalz bildet, dass man anschliessend diese Lösung eindampft und dann das erhaltene feste Natriumsalz mit Äthyl- oder Isopropylalkohol unter Kochen umsetzt.
Die Erfindung bezieht sich auf neue 4-Hydroxycumarin Alkoholkomplexe sowie auf ein Verfahren zu ihrer Herstellung.
Die genannten Alkoholkomplexe von 4-Hydroxycumarinderivaten weisen eine gerinnungshemmende Wirksamkeit auf.
Zur Zeit werden in der medizinischen Praxis solche 4-Hydroxycumarinderivate wie Warfarin, Pelentan (Äthylbiscumacetat) oder Acenocumarol angewandt.
Es ist auch ein Arzneimittel bekannt, in welchem als Wirkstoff 3-(1 -Phenyl-3-oxopentyl)-4-hydroxycumarin enthalten ist, in Kombination mit einem Füllstoff ist dieses Arzneimittel unter dem Namen Phepromaron bekannt. In einem anderen Arzneimittel wird als Wirkstoff 3-(1-p-Nitrophenyl- 3-oxopentyl)-4-hydroxycumarin eingesetzt (SU-PS 216 744) und in Kombination mit einem Füllstoff ist das letztere Arzneimittel unter dem Namen Nitrofarin bekannt.
Diese Antikoagulantien können zur Prophylaxe und Behandlung thromboembolischer Erkrankungen des kardiovaskulären Systems, bei ischämischer Herzkrankheit, bei einem Myokardinfarkt usw. angewandt werden. Die Arzneimittel Phepromaron und Nitrofarin unterscheiden sich vorteilhaft von den obenerwähnten Vertretern der bekannten Arzneimittel, indem sie eine mildere Wirkung, eine geringere Resistenz der Patienten und eine Herabsetzung des Schmerzsyndroms besitzen. Ein allgemeiner Nachteil aller oben aufgezählten Präparate ist die Unmöglichkeit, diese für Injektionen zu benutzen, weil sie in Lösungsmitteln, die in der medizinischen Praxis zu diesen Zielen allgemein angewandt werden, insbesondere Wasser, unlöslich sind.
Es ist jedoch bekannt, dass oft beim schweren Zustand des Patienten gerade die Verabreichung der Antikoagulantien durch Injektion erforderlich ist, und ausserdem ist bei Ulkuskrankheiten des Magens und des Magendarmkanals die perorale Verabreichung von Antikoagulantien nicht angezeigt. Zur Zeit wird in der ärztlichen Praxis für Injektionen ein Heparin Naturantikoagulans mit direkter Wirkung angewandt, das das Blutgerinnungsvermögen für einen kurzen Zeitraum - 2 bis 3 Stunden - herabsetzt; nach Ablauf dieser Zeit ist es jedoch notwendig, das Präparat wiederholt zu verabreichen.
Die Hydroxycumarinderivate haben im Unterschied zu Heparin eine anhaltende (4 bis 5 Tage) gerinnungshemmende Wirkung; daher wäre es vorteilhaft, diese in Form von Injektionen zu verabreichen. Bisher war es jedoch unmöglich, diese, wie oben gezeigt wurde, für Injektionen zu verwenden.
Es ist auch unmöglich, Natriumsalze der 4-Hydroxycumarine für Injektionen zu benutzen, weil diese Salze nicht stabil sind, und ihre Zusammensetzung verändert sich beim Aufbewahren in fester Form als auch in Form von wässrigen Lösungen und das Natriumsalz geht wieder in das entsprechende 4-Hydroxyoumarin über.
In der Literatur sind Angaben über den wasserlöslichen Isopropanolkomplex von Na-Warfarin (3-(1-Phenyl-3-oxo- butyl)-4-hydroxycumarin) mit Isopropylalkohol (US-PS 3 246 013, Kl. 260-343, 2, 1966) vorhanden, der als Arzneimittel mit gerinnungshemmender Wirkung verwendet werden kann. Dieser Isopropanolkomplex kann durch Umsetzung von Warfarin mit einem Natriumalkoholat (Methylat bis Butylat) in Isopropylalkohol erhalten werden, oder man kann Warfarin mit Alkali (Ätznatron) auch in Isopropylalkohol mit darauffolgender Abkühlung der gebildeten Lösung umsetzen, wobei Kristalle des Komplexes ausfallen.
Der Komplex enthält 5,7 Gew.% Isopropylalkohol, was einem Alkoholmolekül für drei Natriumwarfarinatmoleküle entspricht. Die Fähigkeit von Isopropylalkohol, einen Komplex mit Natriumwarfarinat zu bilden, muss als eine einzigartige Eigenschaft hervorgehoben werden, da andere Alkohole (CI-C4) und Lösungsmittel, wie z.B. Äthylacetat, Aceton oder
Dimethylformamid, mit Natriumwarfarinat kein kristallines wasserlösliches Produkt bilden.
Der erwähnte Komplex von Na-Warfarin und Isopropylalkohol kann in Form einer wässrigen Injektionslösung ver abreicht werden, jedoch ist dieser Komplex besonders giftig, was eine ständige ärztliche Kontrolle bei dessen Anwendung wegen der Gefahr einer hämorrhagischen Diatese erforderlich macht.
Der Erfindung wurde die Aufgabe zugrundegelegt, Alkoholkomplexe von 4-Hydroxycumarinderivaten zu entwikkeln, die bei einer gerinnungshemmenden Wirkung und einer geringen Toxizität stabile wässrige Lösungen ergeben und für Injektionen eingesetzt werden können.
Diese Aufgabe wird dadurch gelöst, dass Alkoholkomplexe von 4-Hydroxycumarinderivaten vorgeschlagen werden, die der folgenden allgemeinen Formel entsprechen

Diese Komplexe sind feste kristalline Stoffe. Bei X=Was
serstoffsind die Komplexe des Natriumsalzes von Phenyl-3-oxopentyl)-4-hydroxycumarin mit Äthyl- oder Isopropylalkohol kristalline Stoffe von weisser Farbe, die keinen deutlichen Schmelzpunkt haben und sich bei einer Temperatur von 100 bis 104"C in das Natriumsalz und den entsprechenden Alkohol zersetzen. Bei X=NO2 sind die Komplexe des Natriumsalzes von 3-(l-p-Nitrophenyl-3-oxopentyl)-4-hydroxycumarin mit Äthyl- oder Isopropylalkohol kristalline Stoffe von gelber Farbe, die keinen deutlichen Schmelzpunkt haben und sich bei einer Temperatur von 140 bis 145C in das Natriumsalz und den entsprechenden Alkohol zersetzen.
Das äquimolare Verhältnis der Natriumsalze mit Alkoholen in Komplexen wurde durch Titration der wässrigen Komplexlösungen mit 0,1 n-Salzsäure auf dem Hochfrequenztitrator sowie durch die Angaben der Elementaranalyse, durch Analyse der im deuterierten Wasser aufgenommenen Integralkurven der PMR-Spektren der Komplexe und durch Analyse der TG-Kurven bei der thermochemischen Zersetzung der Komplexe im Temperaturbereich von 30 bis 300"C bewiesen.
Die erfindungsgemässen Alkoholkomplexe der 4-Hydroxycumarinderivate sind in fester Form beim Aufbewahren an der Luft während anderthalb Jahren beständig, und beim Auflösen in destilliertem Wasser ergeben sie Lösungen, die 1 bis 2 Tage lang klar bleiben. Diese Eigenschaften der Komplexe ermöglichen, Natriumsalze der bekannten Antikoagulantien Phepromaron und Nitrofarin in Injektionsformen überzuführen.
Das erfindungsgemässe Verfahren (Verfahren I) zur Herstellung der erfindungsgemässen Verbindungen ist dadurch gekennzeichnet, dass man eine Verbindung der allgemeinen Formel

worin X in Anspruch 1 definiert ist, mit einer äquimolaren Menge an Natriumhydrogencarbonat unter Kochen in Wasser umsetzt, wobei sich das entsprechende Natriumsalz bildet, dass man anschliessend diese Lösung eindampft und dann das erhaltene feste Natriumsalz mit Äthyl- oder Isopropylalkohol unter Kochen umsetzt.
Die Verwendung von Natriumhydrogencarbonat im erfindungsgemässen Verfahren ermöglicht, bei einem niedrigeren pH-Wert im Reaktionsmedium zu arbeiten und dadurch die Zerstörung des Cumarinringgerüstes zu vermindern, die bei der Umsetzung von Hydroxycumarinderivaten mit Ätznatron immer stattfindet (US-PS Nr. 3 246 013).
Die erfindungsgemässen Verbindungen können jedoch auch nach bekannten Verfahren hergestellt werden, z.B.
durch Umsetzung entsprechender 4-Hydroxycumarinderivate mit Natriumalkoholaten (C2-C3) in einem Alkoholmedium (C2-C3) (Verfahren 2). Die Natriumalkoholate können durch das Auflösen von metallischem Natrium im Medium eines entsprechenden Alkohols erhalten werden.
Die Untersuchung der gerinnungshemmenden Eigenschaften der erfindungsgemässen Alkoholkomplexe von 4-Hydroxycumarinderivaten wurde an Kaninchen durch intravenöse Einführung der Präparate durchgeführt. Die gerinnungshemmende Wirkung wurde durch die Verlängerung der Tromboplastinzeit nach Quick (siehe Tabelle 1) bestimmt. Die Untersuchungen zeigten eine höhere antikoagulierende Wirksamkeit der Komplexe bei deren intravenöser Einführung im Vergleich zur peroralen Einführung von Phepromaron, in welchem der Wirkstoff 3-(1-Pheny13- oxopentyl)-4-hydroxycumarin ist, und von Nitrofarin, in welchem der Wirkstoff 3-(1 -p-Nitrophenyl-3-oxopentyl)-4- hydroxycumarin ist.

Die erfindungsgemässen Alkoholkomplexe von 4-Hydroxycumarinderivaten, im Unterschied zu bekannten Antikoagulantien,
die peroral verabreicht werden, weisen eine schnellere Entwicklung der gerinnungshemmenden Wirkung auf und diese konnte schon 8 Stunden nach der Verabreichung des Komplexes nachgewiesen werden. Die Verbindungen weisen eine weniger anhaltende Wirkungsdauer - 3 bis 5 Tage bei den Präparaten, die peroral verabreicht werden - sowie eine beständigere gerinnungshemmende Wirkung auf. Das Wirkungsmaximum der Komplexe wird nach 24 Stunden, und bei Präparaten, die peroral verabreicht werden - nach 48 bis 72 Stunden - festgestellt.
Die an Mäusen bestimmten Angaben über die Toxizität (Das) (siehe Tabelle 2) weisen eine gewisse Erhöhung der Toxizität beim Übergang von Phepromaron, worin der Wirkstoff 3-(1 -Phenyl-3-oxopentyl)-4-hydroxycumarin ist, und Nitrofarin, worin der Wirkstoff 3-( 1 -p-Nitrophenyl-3-oxo- pentyl)-4-hydroxycumarin ist, bis zu den erfindungsgemässen Komplexen auf, die jedoch eine höhere Wirksamkeit besitzen und daher mit niedrigeren Dosen verabreicht werden können, was diese negative Wirkung ausgleicht.

Das Verfahren zur Herstellung der erfindungsgemässen Alkoholkomplexe von 4-Hydroxycumarinderivaten ist technologisch einfach und kann bevorzugt folgendermassen ausgeführt werden:

Zuerst werden 3-(1 -Phenyl-3-oxopentyl)-4-hydroxycu- marin- und 3-( 1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxycu- marinnatriumsalze hergestellt. Zu diesem Zweck werden äquimolare Mengen der genannten 4-Hydroxycumarine und Natriumhydrogencarbonats in einer geringen Menge destilliertem Wasser bei Siedetemperatur erwärmt und der ungelöste Teil wird nach dem Abkühlen abfiltriert. Die Lösung wird in einem Luftbad bis zum Erhalten eines dicken Sirups eingedampft und im Exsikkator mit P205 stehengelassen. Der trockene Rückstand wird zerrieben und mit Äthyl- oder Isopropylalkohol gekocht. Wenn sich dabei eine Suspension bildet, wird diese abf(ftriert und die Mutterlauge wird langsam abgekühlt.
Die beim Stehen aus der Lösung ausgefallenen Kristalle werden abfiltriert, auf dem Filter einer geringen Menge des entsprechenden Alkohols gewaschen und an der Luft oder im Trockenschrank bei einer Temperatur von 30 bis 40"C getrocknet.
Die erfindungsgemässen Alkoholkomplexe von 4-Hydroxycumarinderivaten können auch nach dem bekannten Verfahren unter Verwendung von Natriumalkoholaten erhalten werden. Dazu wird z.B. zuerst eine Suspension von Phenyl-3-oxopentyl)-4-hydroxycumarin oder 3-( 1 -p-Nitro- phenyl-3-oxopentyl)-4-hydroxycumarin in Äthyl- oder Isopropylalkohol bereitgestellt. In einem anderen Kolben wird ein entsprechendes Natriumalkoholat durch Auflösen von metallischem Natrium in Alkohol (Cl-C4) bereitgestellt. Das erhaltene Natriumalkoholat wird zur Suspension unter intensivem Rühren hinzugegossen, wobei die Umsetzung der Reagentien stattfindet. Die beschriebene Umsetzung kann auch unter Kochen der Reaktionsmasse durchgeführt werden, die durch Abgiessen der Suspension und der Natriumalkoholatlösung erhalten wird.
Nach der Umsetzung wird der ungelöste Teil, falls sich dieser bildet, abfiltriert und die Lösung wird abgekühlt. Es fallen Kristalle des Endproduktes aus, die abfiltriert, auf dem Filter mit einer geringen Menge des entsprechenden Alkohols gewaschen und bei einer Temperatur von 30 bis 40"C getrocknet werden.
Zum besseren Verständnis der vorliegenden Erfindung sowie des Verfahrens 2 werden folgende Beispiele angeführt.
Beispiel 1
Komplex des 3 -(1 -Phenyl-3-oxopentyl)-4-hydroxycuma- rinnatriumsalzes mit Äthylalkohol, Verfahren 1.
12,0 g 3-(1 -Phenyl-3-oxopentyl)-4-hydroxycumarin und 3,2 g Natriumhydrogencarbonat werden in 100 ml destilliertem Wasser 0,5 Stunden lang gekocht. Der ungelöste Teil wird schnell abfiltriert, die Mutterlauge wird in eine Porzellanschale gegeben, auf dem Luftbad unter ständigem Rühren bis zum Erhalten eines dicken Sirups eingedampft und dann in einem Exsikkator, der mit P2OS gefüllt ist, getrocknet.
Der feste Rückstand wird zerrieben und es werden 12,8 g 3 -( I -Phenyl-3 -oxopentyl)-4-hydroxycumarinnatriumsalz erhalten (quantitative Ausbeute).
12,8 g 3-( 1 -Phenyl-3-oxopentyl)-4-hydroxycumarinnatri umsalz werden in 40 bis 45 ml wasserfreiem Äthanol gelöst und 10 bis 15 min lang gekocht. Der ungelöste Teil wird abfiltriert und die heisse Lösung wird im Exsikkator über einem Trockenmittel (pos, CaClr u.a.) bis zur Kristallbildung stehengelassen. Die ausgeschiedenen Kristalle werden abfiltriert, mit 5 ml Äthanol gewaschen und getrocknet. Es werden 6,0 g (43,0%) eines weissen kristallinen Stoffes erhalten, der keinen deutlichen Schmelzpunkt hat. Bei einer Temperatur von 104"C wird der Komplex unter der Ausscheidung von Alkohol zersetzt. Der Alkohol ist nach den Angaben der termögravimetrischen Analyse in einer Menge von 11,95% (11,78% der Theorie) enthalten.

Beispiel 2

12,0 g 3-( 1 -Phenyl-3 -oxopentyl)-4-hydroxycumarin und 3,2 g Natriumhydrogencarbonat werden in 100 ml destilliertem Wasser 0,5 Stunden lang gekocht. Der ungelöste Teil wird schnell abfiltriert, die Mutterlauge wird in eine Porzellanschale gegeben, auf dem Luftbad unter ständigem Rühren bis zum Erhalten eines dicken Sirups eingedampft und nachher in einem Exsikkator, der ein Trockenmittel enthält, bis zur vollen Trocknung untergebracht. Der feste Rückstand wird zerrieben; es werden 12,8 g 3-(l-Phenyl-3-oxopentyl)-4-hydroxycumarinnatriumsalz erhalten.
12,8 g 3-( 1 - Phenyl-3-oxopentyl)-4-hydroxycumarinnat- riumsalz werden in 50 bis 55 ml wasserfreiem Isopropylalkohol gelöst und 10 bis 15 min lang gekocht. Der ungelöste Teil wird abfiltriert und man lässt die heisse Lösung im Exsikkator über einem Trockenmittel bis zur Kristallbildung stehen. Die ausgefallenen Kristalle werden abfiltriert, mit 15 ml Isopropylalkohol gewaschen und getrocknet. Es werden 11 g (86%) eines weissen kristallinen Stoffes erhalten, der keinen deutlichen Schmelzpunkt hat. Bei einer Temperatur von 100 C wird der Komplex unter Ausscheidung von Alkohol zersetzt. Der Alkohol ist nach den Angaben der termögravimetrischen Analyse in einer Menge von 14,67% (14,83% der Theorie) im Komplex enthalten.

Komplex des 3-( 1 -Phenyl-3-oxopentyl)-4-hydroxycuma- rinnatriumsalzes mit Isopropylalkohol, Verfahren 2.
32,2 g 3-(1-Phenyl-3-oxopentyl)-4-hydroxycumarin werden in 150 bis 200 ml wasserfreien Isopropylalkohols suspendiert. Zur erhaltenen Suspension wird unter intensivem Rühren die Lösung von Natriumisopropylat in Isopropylalkohol zugegeben, die durch das Auflösen von 2,3 g metallischem Natrium in 50 bis 60 ml Isopropylalkohol erhalten wurde. Die Reaktionslösung wird bis zum Sieden erwärmt, der ungelöste Teil wird abfiltriert und man lässt die Mutterlauge bis zur Kristallbildung im Exsikkator über einem Trockenmittel stehen. Die ausgefallenen Kristalle werden abfiltriert, auf dem Filter mit 20 ml Isopropylalkohol gewaschen und an der Luft oder im Trockenschrank bei einer Temperatur von 30 bis 40"C getrocknet. Es werden 20 g (72%) eines weissen kristallinen Stoffes erhalten, der keinen deutlichen Schmelzpunkt hat.
Bei einer Temperatur von 100"C wird der Komplex unter Ausscheidung von Alkohol zerstört.
Der Alkohol ist nach den Angaben der termögravimetrischen Analyse im Komplex in einer Menge von 14,9% (14,83% der Theorie) enthalten.

Beispiel 3
Komplex des 3-( 1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxy- cumarinnatriumsalzes mit Äthylalkohol, Verfahren 1.
11,3 g 3-(1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxycumarin und 2,6 Natriumhydrogencarbonat werden in 100 ml destilliertem Wasser 0,5 Stunden lang gekocht. Der ungelöste Teil wird schnell abfiltriert, die Mutterlauge wird in eine Porzellanschale gegeben und auf dem Luftbad unter ständigem Rühren bis zum Erhalt eines dicken Sirups eingedampft, den man dann in einer Exsikkator mit einem Trockenmittel bis zur vollen Trocknung gibt. Der feste Rückstand wird zerrieben und es werden 12 g 3-(1-p-Nitrophenyl-3-oxopentyl)- 4-hydroxycumarinnatriumsalz erhalten.
12 g 3-(1-p-Nitrophenyl-3-oxopentyl)-4-hydroxycumarinnatriumsalz werden in 40 bis 45 ml wasserfreiem Äthanol gelöst und 5 bis 10 min lang gekocht. Der ungelöste Teil wird abfiltriert und man lässt die heisse Lösung im Exsikkator mit einem Trockenmittel bis zur Kristallbildung stehen. Die niedergeschlagenen Kristalle werden abfiltriert, auf dem Filter mit 5 ml Äthanol gewaschen und getrocknet. Es werden
11 g (78%) eines gelben kristallinen Stoffes erhalten, der keinen deutlichen Schmelzpunkt hat. Bei einer Temperatur von 140"C wird der Komplex unter Ausscheidung von Alkohol zerstört, der nach den Angaben der termögravimetrischen Analyse in einer Menge von 10,66% (10,56% der Theorie) im Komplex enthalten ist.

Komplex des 3-(1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxy- cumarinnatriumsalzes mit Äthylalkohol, Verfahren 2.
36,8 g 3 -( I -p-Nitrophenyl-3-oxopentyl)-4-hydroxycumarin werden in 150 ml wasserfreiem Äthylalkohol suspendiert und zur erhaltenen Suspension gibt man unter intensivem Rühren die Lösung von Natriumäthylat im Äthylalkohol zu, die durch Auflösen von 2,3 g metallischem Natrium in 50 ml Äthylalkohol erhalten wurde. Nach 2 bis 3 min wird der ungelöste Teil abfiltriert und man lässt die Mutterlauge bis zur Kristallbildung im Exsikkator über einem Trockenmittel stehen. Komplex des 3-(1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxy- cumarinnatriumsalzes mit Isopropylalkohol, Verfahren 1.
11,3 g 3-(1-p-Nitrophenyl-3-oxopentyl)-4-hydroxycumarin und 2,6 g Natriumhydrogencarbonat werden in 10 ml destilliertem Wasser 0,5 Stunden lang gekocht. Der ungelöste Teil wird schnell abfiltriert und die Mutterlauge wird in eine Porzellanschale gegeben und auf dem Luftbad unter ständigem Rühren bis zum Erhalten eines dicken Sirups eingedampft.
Dann trocknet man die Mutterlauge im Exsikkator mit einem Trockenmittel. Der feste Rückstand wird zerrieben, und man erhält 12 g 3-( 1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxycuma- rinnatriumsalz.
12 g 3-(1 -p-Nitrophenyl-3-oxopentyl)-4-hydroxycumarinnatriumsalz werden in 50 ml wasserfreiem Isopropylalkohol gelöst und 5 min lang gekocht. Der ungelöste Teil wird abfiltriert und man lässt die heisse Lösung im Exsikkator mit einem Trockenmittel bis zur Kristallbildung stehen. Die ausgefallenen Kristalle werden abfiltriert, auf dem Filter mit 5 ml Isopropylalkohol gewaschen und getrocknet. Es werden 11,5 g (85%) eines gelben kristallinen Stoffes erhalten. Nach den Angaben der termögravimetrischen Analyse wird der Komplex bei einer Temperatur von 145C unter Ausscheidung des Alkohols zerstört, der im Komplex in einer Menge von 13,10% (13,35% der Theorie) enthalten ist.

The invention relates to new 4-hydroxycoumarin alcohol complexes and to a process for their preparation.
The alcohol complexes of 4-hydroxycoumarin derivatives mentioned have an anticoagulant activity.
4-Hydroxycoumarin derivatives such as warfarin, pelentan (ethyl biscum acetate) or acenocoumarol are currently used in medical practice.
A medicament is also known, in which 3- (1-phenyl-3-oxopentyl) -4-hydroxycoumarin is contained as active ingredient. In combination with a filler, this medicament is known under the name phepromarone. In another drug, 3- (1-p-nitrophenyl-3-oxopentyl) -4-hydroxycoumarin is used as active ingredient (SU-PS 216 744) and in combination with a filler, the latter drug is known under the name of nitrofarin.
These anticoagulants can be used to prevent and treat thromboembolic disorders of the cardiovascular system, ischemic heart disease, myocardial infarction, etc. The medicinal products phepromaron and nitrofarin advantageously differ from the above-mentioned representatives of the known medicinal products in that they have a milder effect, less resistance to the patient and a reduction in the pain syndrome. A general disadvantage of all the preparations listed above is the impossibility of using them for injections because they are insoluble in solvents which are generally used in medical practice for these purposes, in particular water.
However, it is known that the administration of the anticoagulants by injection is often required in the severe condition of the patient, and in addition, peroral administration of anticoagulants is not indicated in gastric and gastrointestinal tract ulcer diseases. At present, a heparin natural anticoagulant with a direct effect is used in the medical practice for injections, which reduces the blood clotting ability for a short period of time - 2 to 3 hours; after this time, however, it is necessary to administer the preparation repeatedly.
In contrast to heparin, the hydroxycoumarin derivatives have a persistent (4 to 5 days) anticoagulant effect; therefore it would be advantageous to administer them in the form of injections. So far, however, it has been impossible to use them for injections as shown above.
It is also impossible to use sodium salts of 4-hydroxycoumarins for injections because these salts are not stable and their composition changes when stored in solid form as well as in aqueous solutions and the sodium salt goes back into the corresponding 4-hydroxyyoumarin about.
In the literature, information about the water-soluble isopropanol complex of sodium warfarin (3- (1-phenyl-3-oxobutyl) -4-hydroxycoumarin) with isopropyl alcohol (US Pat. No. 3,246,013, cl. 260-343, 2, 1966), which can be used as a medicine with an anticoagulant effect. This isopropanol complex can be obtained by reacting warfarin with a sodium alcoholate (methylate to butylate) in isopropyl alcohol, or warfarin with alkali (caustic soda) can also be converted into isopropyl alcohol with subsequent cooling of the solution formed, crystals of the complex precipitating out.
The complex contains 5.7% by weight of isopropyl alcohol, which corresponds to one alcohol molecule for three sodium warfarinate molecules. The ability of isopropyl alcohol to complex with sodium warfarinate has to be emphasized as a unique property since other alcohols (CI-C4) and solvents such as e.g. Ethyl acetate, acetone or
Dimethylformamide, do not form a crystalline water-soluble product with sodium waxarinate.
The above-mentioned complex of sodium warfarin and isopropyl alcohol can be administered in the form of an aqueous solution for injection, but this complex is particularly toxic, which necessitates constant medical monitoring when it is used because of the risk of hemorrhagic diate tea.
The invention was based on the object of developing alcohol complexes of 4-hydroxycoumarin derivatives which, with an anticoagulant action and low toxicity, give stable aqueous solutions and can be used for injections.

The equimolar ratio of the sodium salts with alcohols in complexes was determined by titration of the aqueous complex solutions with 0.1 N hydrochloric acid on the high-frequency titrator as well as by the data from the elementary analysis, by analysis of the integral curves of the PMR spectra of the complexes recorded in deuterated water and by analysis of the TG curves proved in the thermochemical decomposition of the complexes in the temperature range from 30 to 300 "C.
The alcohol complexes of the 4-hydroxycoumarin derivatives according to the invention are stable in solid form when stored in the air for one and a half years, and when dissolved in distilled water they give solutions which remain clear for 1 to 2 days. These properties of the complexes make it possible to convert sodium salts of the known anticoagulants phepromaron and nitrofarin into injection forms.

The use of sodium bicarbonate in the process according to the invention makes it possible to work at a lower pH in the reaction medium and thereby to reduce the destruction of the coumarin ring structure, which always takes place in the reaction of hydroxycoumarin derivatives with caustic soda (US Pat. No. 3,246,013).
However, the compounds according to the invention can also be prepared by known processes, e.g.
by reacting corresponding 4-hydroxycoumarin derivatives with sodium alcoholates (C2-C3) in an alcohol medium (C2-C3) (method 2). The sodium alcoholates can be obtained by dissolving metallic sodium in the medium of an appropriate alcohol.
The investigation of the anticoagulant properties of the alcohol complexes of 4-hydroxycoumarin derivatives according to the invention was carried out on rabbits by intravenous introduction of the preparations. The anticoagulant effect was determined by extending the tromboplastin time according to Quick (see Table 1). The investigations showed a higher anticoagulant activity of the complexes when they were introduced intravenously compared to the oral introduction of phepromaron, in which the active ingredient is 3- (1-pheny13-oxopentyl) -4-hydroxycoumarin, and of nitrofarin, in which the active ingredient 3- Is (1-p-nitrophenyl-3-oxopentyl) -4-hydroxycoumarin.

The alcohol complexes of 4-hydroxycoumarin derivatives according to the invention, in contrast to known anticoagulants,
which are administered orally show a faster development of the anticoagulant effect and this could already be detected 8 hours after the administration of the complex. The compounds have a less persistent duration of action - 3 to 5 days for the preparations that are administered orally - and a more stable anticoagulant effect. The maximum effect of the complexes is determined after 24 hours, and for preparations that are administered orally - after 48 to 72 hours.
The toxicity data (Das) determined in mice (see Table 2) show a certain increase in toxicity during the transition from phepromaron, in which the active ingredient is 3- (1-phenyl-3-oxopentyl) -4-hydroxycoumarin, and nitrofarin, in which the active ingredient is 3- (1-p-nitrophenyl-3-oxopentyl) -4-hydroxycoumarin, up to the complexes according to the invention which, however, are more effective and can therefore be administered at lower doses, which has this negative effect compensates.

The process for preparing the alcohol complexes of 4-hydroxycoumarin derivatives according to the invention is technologically simple and can preferably be carried out as follows:

First, 3- (1-phenyl-3-oxopentyl) -4-hydroxycu- marine and 3- (1-p-nitrophenyl-3-oxopentyl) -4-hydroxycu- marine sodium salts are prepared. For this purpose, equimolar amounts of the 4-hydroxycoumarins and sodium bicarbonate mentioned are heated in a small amount of distilled water at boiling temperature and the undissolved part is filtered off after cooling. The solution is evaporated in an air bath until a thick syrup is obtained and left in the desiccator with P205. The dry residue is ground up and boiled with ethyl or isopropyl alcohol. If a suspension forms, it is filtered off and the mother liquor is slowly cooled.
The crystals which precipitate out of solution on standing are filtered off, washed on the filter with a small amount of the corresponding alcohol and dried in air or in a drying cabinet at a temperature of 30 to 40 ° C.
The alcohol complexes of 4-hydroxycoumarin derivatives according to the invention can also be obtained by the known method using sodium alcoholates. For this, e.g. First a suspension of phenyl-3-oxopentyl) -4-hydroxycoumarin or 3- (1-p-nitro-phenyl-3-oxopentyl) -4-hydroxycoumarin in ethyl or isopropyl alcohol is provided. The reaction described can also be carried out by boiling the reaction mass which is obtained by pouring off the suspension and the sodium alcoholate solution.
After the reaction, the undissolved part, if it forms, is filtered off and the solution is cooled. Crystals of the end product precipitate, which are filtered off, washed on the filter with a small amount of the corresponding alcohol and dried at a temperature of 30 to 40 ° C.

12.8 g of 3- (1-phenyl-3-oxopentyl) -4-hydroxycoumarin sodium salt are dissolved in 40 to 45 ml of anhydrous ethanol and boiled for 10 to 15 minutes. The undissolved part is filtered off and the hot solution is left in the desiccator over a drying agent (pos, CaClr etc.) until the crystals form. The crystals which have separated out are filtered off, washed with 5 ml of ethanol and dried. 6.0 g (43.0%) of a white crystalline substance are obtained which have no clear melting point. The complex is decomposed with the excretion of alcohol at a temperature of 104 ° C. According to the term gravimetric analysis, the alcohol is contained in an amount of 11.95% (11.78% of theory).
