PATENTANSPRÜCHE
1. Kutan applizierbare Zusammensetzung, welche 5 Chlor-4-(2-imidazolin-2-ylamino)-2, 1,3-benzothiadiazol als pharmazeutischen Wirkstoff enthält.
2. Kutan applizierbare Zusammensetzung gemäss Anspruch 1 in Form einer Mikroemulsion aus pharmazeutischem Wirkstoff, einem organischen Lösungsmittel, einem Emulgatorgemisch und Wasser.
3. Emulsion gemäss Anspruch 2, gekennzeichnet durch ein organisches Lösungsmittel a bestehend aus al) einem Ester aus einem aliphatischen (C3-18) Alkohol mit einer aliphatischen (Clo-u) Carbonsäure oder a2) einem Ester mit mindestens einer freien Hydroxylgruppe aus einem Poly-(2-7)-ethylenglykol-glycerol und einer aliphatischen (cm22) Carbonsäure oder a3) einem Ester aus einem aliphatischen (C12-22) Alkohol mit Milchsäure oder a4) einem gesättigten oder ein bis sechs Doppelbindungen enthaltenden (C24 40) Kohlenwasserstoff mit einer geraden (Cl2 32) Kohlenstoffkette,
welche mit 6 bis 16 Methylgruppen substituiert ist und ein Emulgatorgemisch b + c bestehend aus bl) einem aliphatischen (C12-20) Alkohol, welche über 32 "C flüssig ist oder b2) einem Monoester aus Ethylen- oder Propylenglykol mit einer aliphatischen (cm22) Carbonsäure oder b3) einem Ester beschrieben unter a2), sofern nicht bereits als organisches Lösungsmittel vorhanden oder b4) einem Ester beschrieben unter a3), sofern nicht bereits als organisches Lösungsmittel vorhanden und c) einem Monoäther aus einem Polyethylenglykol mit einem aliphatischen (C1218) Alkohol, wobei die Komponente c des Emulgatorgemisches einen HLB-Wert von 10-18 hat.

8. Verfahren zur Herstellung der Emulsion gemäss einem der Ansprüche 2 bis 7, dadurch gekennzeichnet, dass die Komponenten unter Rühren bei erhöhter Temperatur bis maximal 100 "C vermischt und danach abgekühlt werden.
Gegenstand der Erfindung ist eine kutan applizierbare Zusammensetzung, besonders eine Mikroemulsion von 5 Chlor4-(2-imidazdin-2-ylamino)-2, 1, 3-benzothiadiazol in einem organischen Lösungsmittel, einem Emulgatorgemisch und Wasser.
Der pharmakologische Wirkstoff ist ein Myotonolytikum mit antitremor- und antirigor-Wirkung und ist ein Muskelentspanner. Er ist in der englischen Patentschrift 1 429926 beschrieben worden.
Es hat sich nun gezeigt, dass der pharmakologische Aktivstoff seine systemische Wirkung entfalten kann, wenn er kutan, z. B. als Salbe, Lotion, Lösung oder Emulsion appliziert wird, besonders, wenn die Grösse der Aktivteilchen gering ist und die kutan applizierbare Zusammensetzung in Form einer Mikroemulsion vorliegt. Eine Mikroemulsion ist eine durchsichtige Emulsion hoher Stabilität. Ihre Teilchengrösse ist kleiner als 1500 AE (150 nm), was weniger als etwa V4 der Wellenlänge des sichtbaren Lichtes bedeutet, wodurch die Teilchen das Licht nicht streuen, das System transparent ist und in nur einer optisch wahrnehmbaren Phase existiert.
Durch geeigneter Wahl ihrer Komponenten lassen sich Mikroemulsionen mit höherer Viskosität herstellen, die man als Mikrogele bezeichnen kann und die, je nach Zusammensetzung bei Raumtemperatur viskoelastische Eigenschaften aufweisen können (swinging gels), bei einer Hauttemperatur von etwa 32 "C in den flüssigen Zustand übergehen und sich sehr gut und rasch in die Haut verreiben lassen.
Diese transparenten Gele sind wasserhaltig, nicht klebend und nicht fettend und, falls erforderlich, leicht abwaschbar.
Mikroemulsionen für kosmetische bzw. lokale Anwendung sind bekannt. Bei geeigneten Gewichtsverhältnissen der Komponenten sind Mikroemulsionen herstellbar aus einem pharmakologisch aktiven Wirkstoff mit lokaler oder systemischer Wirkung, Kohlenwasserstoffen unterschiedlicher Kettenlänge als organischen Lösungsmitteln, aus Poly-(9)oxyethylenlauryläther und Dekanol als Emulgatorgemisch (APV-Kongress 1980, Amsterdam).
Gegenstand der Erfindung sind besonders solche Emulsionen, gekennzeichnet durch ein organisches Lösungsmittel a, bestehend aus: al) einem Ester aus einem aliphatischen (C18) Alkohol mit einer aliphatischen (Cl0 22) Carbonsäure oder a2) einem Ester mit mindestens einer freien Hydroxylgruppe aus einem Poly-(2-7)-ethylenglykol-glycerol und einer aliphatischen (C6-22) Carbonsäure oder a3) einem Ester aus einem aliphatischen (C12 22) Alkohol mit Milchsäure oder a4) einem gesättigten oder ein bis sechs Doppelbindungen enthaltenden (C24 40) Kohlenwasserstoff mit einer geraden (Cl2¯32) Kohlenstoffkette,
welche mit 6 bis 16 Methylgruppen substituiert ist und ein Emulgatorgemisch b + c bestehend aus bl) einem aliphatischen (Cl2 20} Alkohol, welche über 32 "C flüssig ist oder b2) einem Monoester aus Ethylen- oder Propylenglykol mit einer aliphatischen (CS22) Carbonsäure oder b3) einem Ester beschrieben unter a2), sofern nicht bereits als organisches Lösungsmittel vorhanden oder b4) einem Ester beschrieben unter a3), sofern nicht bereits als organisches Lösungsmittel vorhanden und c) einem Monoäther aus einem Polyethylenglykol mit einem aliphatischen (Cl2 l8) Alkohol, wobei die Komponente c des Emulgatorgemisches einen HLB-Wert von 10-18 hat.
Der HLB-Wert (hydrophilic-lipophilic balance-Wert) ist ein Mass für den relativen Anteil von hydrophilen Gruppen des Emulgatormoleküls (vgl. Pharm. Act. Helv. 44, 9 (1969) und H.P. Fiedler, Lexikon der Hilfsstoffe für Pharmazie, Kosmetik und angrenzende Gebiete, Seite 263, Editio Cantor KG, 1971).
Wo von aliphatischen Carbonsäuren gesprochen wird, sind solche gemeint, welche geradkettig oder verzweigt und gesättigt oder ungesättigt sind. Falls sie ungesättigt sind, enthalten sie vorzugsweise ein oder zwei Doppelbindungen.
Wo von aliphatischen Alkoholen die Rede ist, sind einwertige Alkohole gemeint. Sie sind vorzugsweise primär oder sekundär, besonders primär, weiter sind sie geradkettig oder
verzweigt, vorzugsweise geradkettig und gesättigt oder ungesättigt, vorzugsweise gesättigt. Sind sie ungesättigt, enthalten sie vorzugsweise ein oder zwei Doppelbindungen.
Überraschend ist, dass die organischen Komponenten in Gegenwart einer für den gewünschten therapeutischen Effekt ausreichenden Wirkstoffkonzentration welche 15 Gew.-% übersteigen kann, eine Mikroemulsion bilden können.
Diese Mikroemulsionen sind stabil bei Temperaturen zwischen 15 und 40 "C, speziell zwischen 20 und 32 C, besonders wenn sie 610% an pharmakologischem Wirkstoff.

Die spezifischen Komponenten der Emulsion werden im Rahmen ihrer oben beschriebenen Aufzählung aufeinander abgestimmt, besonders die spezifischen Komponenten des zu verwendenden Emulgatorgemisches, mit dem, wie bekannt, die Teilchengrösse der Emulsion gesteuert werden kann.
Das Emulgatorgemisch besteht aus zwei Komponententypen b) und c), wovon c) den Emulgator und b) den Co Emulgator darstellt. Eine genaue Grenze zwischen beiden ist nicht zu ziehen. Ohne den Emulgator ist keine Emulsion herstellbar und ohne den Co-Emulgator ist ihre Stabilität nicht gewährleistet. Vorzugsweise wird ein Emulgatorgemisch aus bl) und c) verwendet.
Damit eine Mikroemulsion entstehen kann, wird das Emulgatorgemisch auf die anderen Komponenten, wie das organische Lösungsmittel und besonders auf den pharmakologisch aktiven Wirkstoff abgestimmt.
Geeignete Co-Emulgatoren aus der Gruppe bl) sind Dodecanol, Tetradecanol, Oleylalkohol, 2-Hexyldecanol und 2 Oktyldodecanol, aus der Gruppe b2) Propylenglykolmonolaurat, Propylenglykolmonomyristat, aus der Gruppe b3) Polyethylenglycol-7-glycerylcocoat und aus der Gruppe b4) Lauryllactat und Myristyllactat. Besonders das Dodecanol und das Tetradecanol aus der Gruppe bl) sind geeignet.
Die geeigneten Emulgatoren sind zu beschreiben als solche der Gruppe c) mit einem HLB-Wert von 10-18, besonders als solche mit einem HLB-Wert von 12-15. Besonders der handelsübliche Polyoxyethylen-10-oleyl-äther aus der Gruppe cl) ist geeignet, der einen HLB-Wert von 12,4 hat.
Als organisches Lösungsmittel sind aus der Gruppe al) Isopropyllaurat und Isopropylmyristat, Hexyllaurat, Decyllaurat und Laurylmyristat geeignet und aus der Gruppe a4), 2,6,10,15,1 9,23-Hexamethyl-2,6, 10,14,1 8,22-tetracosanhe- xan (C30Hso) (Squalen) und Squalan, das als Perhydrosqualen (C30Hso) bezeichnet werden kann.
Emulsionen mit einem organischen Lösungsmittel al) und einem Emulgatorgemisch aus bl) und c) werden bevorzugt, besonders die Kombination aus Isopropyllaurat oder -myristat, Dodecanol oder Tetradecanol und Polyoxyethy len- 1 0-oleyläther. Sie werden hergestellt, indem die Komponenten eventuell unter Zusatz eines pH-Regulators, unter Rühren bei erhöhter Temperatur bis maximal 100 0C vermischt und danach abgekühlt werden.
Der pharmakologisch aktive Wirkstoff, wenn lokal appliziert, dringt in die Haut und wird in der Hornschicht angereichert, wodurch eine Depotbildung stattfindet. Von dort aus kann er, bei lokaler Aktivität heilend auf die Haut, und bei systemischer Aktivität, heilend auf die inneren Teile des Körpers einwirken.
Eine Mikroemulsion ist daher ausgezeichnet als Depotform für Wirkstoffe mit systemischer Aktivität geeignet, was nach der Methode beschrieben von H. Schaefer in Adv.
Pharmacol. Ther. [Proc. Int. Congr. Pharmacol. 7th] 9, 223-235 (1978) ed. by Y. Cohen; Pergamon, Oxford 1979, in folgender Weise an isolierter menschlicher Haut in vitro getestet werden kann:
Mikroemulsionen oder Mikrogele mit radioaktiv markiertem Aktivstoff werden in einer Menge von 5-7, 5 mg/ cm2 aufisolierte Stücke von menschlicher Abdominalhaut aufgetragen. Nach Ablauf von 16 Stunden bei 32 "C wird die Verteilung der Radioaktivität in Hornschicht, Epidermis, oberem Corium, unterem Corium, Subcutis, sowie im Diffusionkammerwasser gemessen, woraus sich die mittlere Penetrationsrate, d. h. diejenige Menge, welche die Hornschicht pro Stunde passiert hat, berechnen lässt.
Derjenige Anteil, der die Hornschicht passiert hat, ist für die Wirkung relevant. Dieser Anteil würde in vivo über das Kapillarsystem in den Kreislauf und damit in die inneren Teile des Körpers abtransportiert werden.
Die Penetrationsrate kann z. B. 1,2 bis 3.10-8 Mol pro cm2 Haut pro Stunde betragen. Für eine systemische Wirkung wird eine mittlere Penetrationsrate von etwa I x 108 Mol. cm-2 Stunde - 1 angestrebt.
Die Penetrationsrate ist nicht nur von den Emulsionskomponenten sondern auch von verschiedenen anderen Faktoren abhängig, z.B. vom pH-Wert der Emulsion, vom Verteilungscoefficient des Wirkstoffes zwischen Mikroemulsion und Hornschicht und von der Konzentration des Wirkstoffes in der Emulsion.
Der pH-Wert der Emulsion muss auf den pK-Wert des Wirkstoffes abgestimmt werden, z. B. mit Milchsäure, sodass der Wirkstoff zumindestens teilweise als freie, lipophile Base in Gleichgewicht vorliegt, wodurch die Penetrationsrate positiv beeinflusst wird.
Da jedoch immer nur ein Bruchteil der aufgetragenen Menge perkutan resorbiert wird und die applizierbare Menge begrenzt ist, sollte ein systemisch wirkender Aktivstoff in einer möglichst niedrigen Dosierung therapeutisch wirksam sein, d. h. seine per orale notwendige Dosis sollte maximal 10 mg pro Tag betragen.

The invention relates to a cutaneously applicable composition, especially a microemulsion of 5 chloro4- (2-imidazdin-2-ylamino) -2, 1, 3-benzothiadiazole in an organic solvent, an emulsifier mixture and water.
The pharmacological active ingredient is a myotonolytic agent with antitremor and antirigor effects and is a muscle relaxer. It has been described in the English patent 1 429926.
It has now been shown that the pharmacological active ingredient can exert its systemic effect if it is cutaneous, e.g. B. is applied as an ointment, lotion, solution or emulsion, especially if the size of the active particles is small and the composition which can be applied cutaneously is in the form of a microemulsion. A microemulsion is a transparent, high stability emulsion. Their particle size is less than 1500 AU (150 nm), which means less than about V4 of the wavelength of visible light, which means that the particles do not scatter the light, the system is transparent and exists in only one optically perceptible phase.
By suitable choice of their components, microemulsions with a higher viscosity can be produced, which can be called microgels and which, depending on their composition, can have viscoelastic properties at room temperature (swinging gels), change to the liquid state at a skin temperature of around 32 "C and can be rubbed very well and quickly into the skin.
These transparent gels are water-containing, non-sticky and non-greasy and, if necessary, easily washable.
Microemulsions for cosmetic or local use are known. With suitable weight ratios of the components, microemulsions can be produced from a pharmacologically active ingredient with local or systemic action, hydrocarbons of different chain lengths as organic solvents, from poly (9) oxyethylene lauryl ether and decanol as an emulsifier mixture (APV Congress 1980, Amsterdam).
The invention particularly relates to such emulsions, characterized by an organic solvent a, consisting of: al) an ester of an aliphatic (C18) alcohol with an aliphatic (Cl0 22) carboxylic acid or a2) an ester with at least one free hydroxyl group from a poly - (2-7) ethylene glycol glycerol and an aliphatic (C6-22) carboxylic acid or a3) an ester of an aliphatic (C12 22) alcohol with lactic acid or a4) a saturated or containing one to six double bonds (C24 40) hydrocarbon with a straight (Cl2¯32) carbon chain,

The HLB value (hydrophilic-lipophilic balance value) is a measure of the relative proportion of hydrophilic groups in the emulsifier molecule (cf. Pharm. Act. Helv. 44, 9 (1969) and HP Fiedler, Lexicon of auxiliaries for pharmacy, cosmetics and adjacent areas, page 263, Editio Cantor KG, 1971).
Wherever aliphatic carboxylic acids are mentioned, those are meant which are straight-chain or branched and saturated or unsaturated. If they are unsaturated, they preferably contain one or two double bonds.
Wherever aliphatic alcohols are mentioned, monohydric alcohols are meant. They are preferably primary or secondary, particularly primary, furthermore they are straight-chain or
branched, preferably straight-chain and saturated or unsaturated, preferably saturated. If they are unsaturated, they preferably contain one or two double bonds.
It is surprising that the organic components can form a microemulsion in the presence of an active substance concentration which is sufficient for the desired therapeutic effect and which can exceed 15% by weight.
These microemulsions are stable at temperatures between 15 and 40 "C, especially between 20 and 32 C, especially if they contain 610% of pharmacological agent.

The emulsifier mixture consists of two component types b) and c), of which c) represents the emulsifier and b) the co-emulsifier. A precise boundary between the two cannot be drawn. No emulsion can be produced without the emulsifier and its stability cannot be guaranteed without the co-emulsifier. An emulsifier mixture of bl) and c) is preferably used.
In order for a microemulsion to form, the emulsifier mixture is matched to the other components, such as the organic solvent, and in particular to the pharmacologically active ingredient.

Dodecanol and tetradecanol from group b1) are particularly suitable.
The suitable emulsifiers can be described as those of group c) with an HLB value of 10-18, especially as those with an HLB value of 12-15. The commercially available polyoxyethylene-10-oleyl ether from group cl), which has an HLB value of 12.4, is particularly suitable.
Suitable organic solvents from the group al) are isopropyl laurate and isopropyl myristate, hexyl laurate, decyl laurate and lauryl myristate and from group a4), 2,6,10,15,1 9,23-hexamethyl-2,6, 10,14,1 8,22-tetracosane hexane (C30Hso) (squalene) and squalane, which can be referred to as perhydrosqualene (C30Hso).
Emulsions with an organic solvent al) and an emulsifier mixture from bl) and c) are preferred, especially the combination of isopropyl laurate or myristate, dodecanol or tetradecanol and polyoxyethylene 1-oleyl ether. They are produced by mixing the components, possibly with the addition of a pH regulator, with stirring at an elevated temperature up to a maximum of 100 ° C., and then cooling them.
The pharmacologically active ingredient, when applied locally, penetrates the skin and accumulates in the horny layer, which leads to depot formation. From there it can have a healing effect on the skin in the case of local activity and a healing effect on the inner parts of the body in the case of systemic activity.
A microemulsion is therefore excellently suitable as a depot form for active substances with systemic activity, which is described by the method described by H. Schaefer in Adv.
Pharmacol. Ther. [Proc. Int. Congr. Pharmacol. 7th] 9, 223-235 (1978) ed. By Y. Cohen; Pergamon, Oxford 1979, can be tested in vitro on isolated human skin in the following ways:
Microemulsions or microgels with radioactively labeled active substance are applied in an amount of 5-7.5 mg / cm 2 on isolated pieces of human abdominal skin. After 16 hours at 32 "C, the distribution of radioactivity in the horny layer, epidermis, upper corium, lower corium, subcutis, and in the diffusion chamber water is measured, from which the average penetration rate, ie the amount that has passed the horny layer per hour, can be calculated.
The portion that has passed the horny layer is relevant for the effect. This part would be transported in vivo via the capillary system into the circulation and thus into the inner parts of the body.
The penetration rate can e.g. B. 1.2 to 3.10-8 moles per cm2 of skin per hour. For a systemic effect, an average penetration rate of about I x 108 mol. Cm-2 hour - 1 is aimed for.
The penetration rate depends not only on the emulsion components but also on various other factors, e.g. on the pH of the emulsion, on the distribution coefficient of the active ingredient between the microemulsion and horny layer and on the concentration of the active ingredient in the emulsion.
The pH of the emulsion must be matched to the pK of the active ingredient, e.g. B. with lactic acid, so that the active ingredient is at least partially in balance as a free, lipophilic base, whereby the penetration rate is positively influenced.
However, since only a fraction of the applied amount is absorbed percutaneously and the amount that can be applied is limited, a systemically active substance should be therapeutically effective in the lowest possible dosage, i.e. H. his orally necessary dose should be a maximum of 10 mg per day.
