Das Verfahren kann analog zu bekannten Methoden erfolgen. Der Rest Z steht in den Verbindungen der Formel II beispielsweise .für Chlor oder Brom, oder für einen aliphatischen oder aromatischen Sulfonyloxyrest, vorzugsweise den Mesyloxy- oder den p-Tosyloxyrest. Man geht beispielsweise so vor, dass man Verbindungen der Formel II mit einem Cyanogruppen-Donor, beispielsweise einem Alkalimetallcyanid wie Natrium- oder Kaliumcyanid, umsetzt.
Man arbeitet mit Vorteil unter Erwärmen, z.B. auf 50 bis 100"C 1 ,6-Dimethyl-8o-acetonitril-ergolin kann z.B. in freier Form als Base, oder in Form seiner Additionssalze mit Säuren vorliegen. Aus der freien Base lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Die Ausgangsprodukte sind bekannt oder nach an sich bekannten Methoden herstellbar.
1 ,6-Dimethyl-8a-acetonitril-ergolin in freier Form oder in Form von physiologisch verträglichen Additionssalzen mit Säuren zeichnet sich durch interessante pharmakodynamische Eigenschaften aus. Es kann als Heilmittel verwendet werden.
So zeichnet sich die erfindungsgemässe Verbindung durch eine ausgeprägte prolactinsekretionshemmende Wirkung aus. Sie ist daher geeignet zur Prophylaxe oder Behandlung von prolactinabhängigen Funktionsstörungen, wie z.B. physiologischer Lactation und Galactorrhoe, von Prostatahypertrophie, von Prostata- und Mammacarcionomen; des weiteren kann sie bei Nephropathien und chronischen Nierenentzündungen, zur Regulierung des Wasser- und Elektrolythaushaltes, bei Oedemen, zur Behandlung von essentiellen und renalen Hypertonieformen sowie bei akuten Zuständen wie vor Migräneattacken und beim praemenstruellen Syndrom Anwendung finden.
Die erfindungsgemässe Verbindung besitzt ebenfalls dopaminerge Eigenschaften und kann bei der Behandlung des Morbus Parkinson und depressiven Zuständen Anwendung finden.
Die Prolactinsekretionshemmung konnte an Ratten deutlich gemacht werden. Bei lactierenden Ratten, die zeitweilig von ihren Jungen getrennt waren, kommt es bei erneuter Kontaktaufnahme zu einer massiven Prolactinausschüttung, die z.B. als Depletion des hypophysären Prolactingehaltes erfasst werden kann. Dieses Phänomen [beschrieben
von Grosvenor und Mena in J. Endocr. 52, 11 (1972)lergibt eine reproduzierbare Versuchsanordnung, um die Prolactinsekretionshemmung pharmakologisch zu erfassen. Es zeigte sich, dass 1 bis 5 mg/kg s.c. der erfindungsgemässen Verbindung nicht nur signifikant die durch sensorische Reize ausgelöste Prolactinfreisetzung hemmen, sondern gleichzeitig zu einem leichten Anstieg des Prolactingehaltes führen, wodurch deutlich gemacht wird, dass die Verbindungen die physiologisch stimulierte Prolactinsekretion zu hemmen vermögen.
Auch an männlichen Ratten konnte eine Wirkung auf den Prolactinspiegel festgestellt werden; die erfindungsgemässe Verbindung hemmt die tonische Prolactinsekretion mit ED5o Werten zwischen 0,005 und 1,0 mg/kg s.c. (durch Prolactinbestimmungen der Rattenseren mittels Radio-immunoassay ).
Für oben genannte Anwendung variiert die zu verwendende Dosis selbstverständlich je nach verwendeter Substanz, Art der Administration und der gewünschten Behandlung. Im allgemeinen werden aber befriedigende Resultate mit Dosen von ungefähr 0,005 bis 1,0 mg/kg Körpergewicht erreicht: die Administration kann mit einer Dosis täglich vorgenommen werden oder nötigenfalls in mehreren beispielsweise 7 bis 4 Teildosen erfolgen. Für grössere Säugetiere liegt die Tagesdosis im Bereich von etwa 0,5 bis 50 mg der Sub stand geeignete Dosierungsformen für z.B. orale Anwendungen enthalten im allgemeinen ungefähr 0,1 bis 100 mg wirksame Substanz neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Die Erfindung betrifft auch Heilmittel, die die erfindungsgemässe Verbindung in freier Form oder in Form ihrer physiologisch verträglichen Additionssalze mit Säuren enthalten.
Diese Heilmittel, beispielsweise eine Lösung oder eine Tablette, können nach bekannten Methoden, unter Verwendung der üblichen Hilfs- und Trägerstoffe, hergestellt werden.
Die Erfindung ist im experimentellen Teil erläutert.
Die Temperaturangaben erfolgen in Celsiusgraden.
Beispiel 1 ,6-Dimethyl-8a-acetonitril-ergolin
2,89 g (10 mMol) 8x-Chlormethyl-l,6-dimethyl-ergolin wurden in 20 ml Dimethylsulfoxid gelöst, mit einer Lösung von 1,95 g Kaliumcyanid in 10 ml Wasser versetzt und das Reaktionsgemisch 5 Stunden bei 110 gerührt. Darauf wurde zwischen Methylenchlorid und 2 N Ammoniak verteilt, die organischen Phasen mit Natriumsulfat getrocknet und eingeengt und der Rückstand an 100 g Kieselgel mittels Methylenchlorid/Methanol (95:5) chromatographiert und die reines 1 ,6-Dimethyl-8o-acetonitril-ergolin enthaltenden Fraktionen eingedampft und als Hydrogentartrat aus Aceton-Petroläther kristallisiert.

The inhibition of prolactin secretion could be demonstrated in rats.
