In-vitro-Kulturen tierischer und menschlicher primärer oder kontinuierlicher (transformierter) Zellinien repräsentieren biologisch höchst empfindliche Systeme. Die Zellen müssen für ihre optimale Proliferation neben Temperaturkonstanz, Serumzusatz und aufwendigen Nährlösungen vor allem um den physiologischen pH-Wert (pH 7,2-7,4) mit nichttoxischen Substanzen gut abgepuffert werden.
Während für einige wenige kontinuierliche Zellinien synthetische Puffersubstanzen wie Piperazinsulfonsäure-Derivate sich in allerdings nur niedrigen Konzentrationen (20-50 mM) als nichttoxisch erwiesen haben, ist man hingegen für die Kultivierung der meisten Zellinien auf das CO2/ NaHCO3-Puffersystem angewiesen. Dies hat den Nachteil, dass man in offenen Gefässen respektive Systemen arbeiten muss, um den Gasaustausch zu gewährleisten. Kontaminationen durch Bakterien, Mycoplasmen, Hefe und Pilze lassen sich nur durch den Einsatz bestimmter Verbindungen (Antibiotikas, Fungizide) auf ein Minimum reduzieren, wobei diese Verbindungen das Wachstum tierischer oder menschlicher Zellen beeinträchtigen. Antikontaminationsmittel sind vor allem dann nicht erwünscht, wenn auf menschlichen primären Zellinien Humanvaccine hergestellt werden müssen.
Diese Sterilitätsprobleme respektive der Einsatz von Antibiotikas lassen sich vollständig eliminieren, wenn es möglich ist, primäre Humanzellen in geschlossenen Systemen zu kultivieren, was dann den Einsatz nichttoxischer Puffer bedingt..
Es hat sich gezeigt, dass als nichttoxische Puffer hierfür niedere Peptide oder Tris-hydroxymethyl-methan substituierte niedere Peptide sehr geeignet sind. Niedere Peptide besitzen eine gute Pufferkapazität in einem weiten pH-Bereich (in Abhängigkeit der Aminosäuren-Zusammensetzung), kommen jedoch als Puffersubstanzen praktisch nicht in Frage, da ihre Löslichkeiten in protischen Lösungsmitteln zu gering sind und oft durch biologische Systeme metabolisiert, das heisst aufgebraucht werden.
Beide Nachteile lassen sich durch geeignete Substituenten an den Peptiden eliminieren. AIs Substituent besitzt hierfür der Tris-hydroxymethyl-methan-Rest die erforderlichen molekularen Eigenschaften, niedere Peptide in eine gut wasserlösliche Form zu überführen. Dies erfolgt durch die Umsetzung von Tris-hydroxymethyl-aminomethan mit N-2halogenacetylierten Aminosäuren respektive N-2-halogenacetylierten Di-, Tri- und Tetrapeptiden in wässriger Lösung.
Bei dieser Kondensationsreaktion wird die Aminosäure respektive das niedere Peptid am N-Terminus um einen Glycinrest verlängert, welcher seinerseits Tris-hydroxymethylmethan substituiert ist. Hierdurch lassen sich Tris-hydroxy- methyl-methan substituierte Peptide ganz allgemein, wie folgt, synthetisieren:

2. Synthese nach Anspruch 1, dadurch gekennzeichnet, dass die Aminosäure eine a-l- oder a-d-Aminosäure darstellt.
3. Synthese nach Anspruch 1, dadurch gekennzeichnet, dass das Di-, Tri- oder Tetrapeptid aus ss-, y- oder 6-Amino- säuren oder deren Gemische besteht.
4. Synthese nach Anspruch 1, dadurch gekennzeichnet, dass die Di-, Tri- und Tetrapeptide nach Anspruch 3 saure und basische Aminosäuren, zum Beispiel Aminodicarbonsäuren und Di-aminocarbonsäuren im Molekül enthalten.
5. Synthese nach Anspruch 1-4, dadurch gekennzeichnet, dass man die Alkalisalze der Aminosäuren respektive Di-, Tri- und Tetrapeptide verwendet.
6. Verwendung der nach Anspruch 1 und 2 erhaltenen Peptide als untoxische Puffer für sämtliche biologischen Systeme, wobei für die Aminosäure alle natürlichen l-Amino- säuren stehen.
7. Verwendung nach Anspruch 6, dadurch gekennzeichnet, dass man Tris-Gly-Ala als untoxische Puffersubstanz für kontinuierliche und primäre Zellinien einsetzt.
8. Verwendung nach Anspruch 6, dadurch gekennzeich net, dass man Tris-Gly-Met als untoxische Puffersubstanz für kontinuierliche und primäre Zellinien einsetzt.

Zu einer gesättigten, wässrigen Lösung von 1 Mol N-2-Jacetyl-l-tyrosin (Li-Salz) gibt man 1,05 Mol Tris-hydroxymethyl-aminomethan und erwärmt das Gemisch zum Siedepunkt unter Rückfluss und Lichtausschluss für 15 Minuten.
Nach Zusatz von Äthanol bis zum Trübungspunkt in der Hitze lässt man das Produkt (Tris-Gly-Tyr) unter Abkühlen auskristallisieren .
Beispiel 3
Zu einer gesättigten, wässrigen Lösung von 0,1 Mol N-2 Br-acetyl-l-gly-l-arg-d-asp-l-threo (K-Salz) gibt man 0,1 Mol Tris-hydroxymethyl-aminomethan und erwärmt das Gemisch zum Siedepunkt unter Rückfluss und Lichtausschluss für 45 Minuten. Nach Zusatz von Isopropanol bis zum Trübungspunkt in der Hitze lässt man das Produkt (Tris-Gly-Gly-arg-ser-threo) unter Abkühlen auskristallisieren.
Beispiel 4
Zu einer gesättigten, wässrigen Lösung von 0,05 Mol N2-chloracetyl-y-aminobuttersäure (K-Salz) gibt man 0,05 Mol Tris-hydroxymethyl-aminomethan und erwärmt das Gemisch zum Siedepunkt unter Rückfluss für fünf Stunden.
Nach Zusatz von Propanol bis zum Trübungspunkt in der Hitze lässt man das Produkt (Tris-Gly-y-butyr) unter Abkühlen auskristallisieren.

In vitro cultures of animal and human primary or continuous (transformed) cell lines represent biologically highly sensitive systems. For their optimal proliferation, the cells must be well buffered with non-toxic substances in addition to constant temperature, serum addition and complex nutrient solutions, especially around the physiological pH (pH 7.2-7.4).
While for a few continuous cell lines synthetic buffer substances such as piperazine sulfonic acid derivatives have proven to be non-toxic in only low concentrations (20-50 mM), the CO2 / NaHCO3 buffer system is required for the cultivation of most cell lines. This has the disadvantage that you have to work in open vessels or systems to ensure gas exchange.
