PATENTANSPRÜCHE
1. Biozide Aktivität zeigendes Mittel, dadurch gekennzeichnet, dass es einen durch Polypyrrolidon und Jod gebildeten Komplex enthält.
2. Mittel gemäss Patentanspruch 1, bei dem das Polypyrrolidon in Form von Verbandsmaterial vorliegt.
3. Mittel gemäss Patentanspruch 1, bei dem das Polypyrrolidon in Form von Nähmaterial vorliegt.

Diese Erfindung bezieht sich auf biozide Aktivität zeigende Mittel sowie auf deren Verwendung in topisch aufzubringenden pharmazeutischen Präparaten.
Der Komplex, welcher sich beim Mischen von Polyvinylpyrrolidon und Jod bildet, ist zuerst in der US-Patentschrift Nr. 2739922 von Herman A. Shelanski beschrieben worden.
Dieser Komplex stellt heute die bevorzugte Anwendung der günstigen bioziden Eigenschaften von Jod ohne die Nebenwirkungen des genannten Elementes dar. Speziell bevorzugt wird das Mittel in der Chirurgie, wo es zur Bedeckung der Haut vor der Inzision angewendet wird.
Polyvinylpyrrolidon ist ein wasserlösliches Polymer, und auch der Polyvinylpyrrolidon-Jod-Komplex zeigt gute Wasserlöslichkeit. Nun gibt es aber Anwendungen, bei denen diese Wasserlöslichkeit eine höchst ungewünschte Eigenschaft darstellt. Beispielsweise ist die Effizienz von topischen Anwendungen solch wasserlöslicher Antiseptika nur von kurzer Dauer, da sie schnell sowohl durch Regen wie auch durch Transpiration weggewaschen werden. In solchen Fällen wäre ein unlösliches Präparat, welches die oben beschriebenen gezähmten Jodeigenschaften aufweist, viel nützlicher und effizienter wegen seiner längeren Wirkungsdauer.
Diese Erfindung hat nun den Hauptzweck, einen wasserunlöslichen und daher beständigeren Jodkomplex zu schaffen, welcher ähnliche pharmakologische und medizinale Eigenschaften aufweist wie der wasserlösliche Polyvinylpyrrolidon-Jod-Komplex.
Ein weiterer Zweck dieser Erfindung ist es, einen wasserunlöslichen Jodkomplex zu schaffen, welcher elementares Jod in kleinen, aber wirkungsvollen Mengen, langsam und über eine lange Wirkungsdauer abgibt, wodurch einerseits die biozide Aktivität des Mittels verlängert wird und andererseits die schädlichen Nebenwirkungen, wie Hautreizungen usw., minimalisiert werden.
Es ist ein weiterer Zweck dieser Erfindung, neue Verbandsund Nähmaterialien für medizinale und chirurgische Zwecke zu schaffen, welche selbst die bakteriziden und fungiziden Wirkstoffe sind. Verbandsmaterialien dieser Art bestehen heute sowohl aus gewobenen wie auch aus nicht gewobenen Textilien.
Ein weiterer Zweck dieser Erfindung ist es, Langzeitsalben und -cremen mit antiseptischer Wirkung für topische Anwendungen zu schaffen.

Gefunden wurde, dass Polypyrrolidon, welches auch als Nylon-4 bekannt ist und ein Polymer der Struktur ist, mit elementarem Jod zu einem Komplex reagiert werden kann, welcher sehr ähnlich ist dem Polyvinylpyrrolidon-Jod-Komplex. Im Gegensatz zum Polyvinylpyrrolidon-Jod-Komplex aber ist der Nylon-4-Jod-Komplex in Wasser unlöslich.
Das erfindungsgemässe, biozide Aktivität zeigende Mittel ist im vorangehenden Patentanspruch 1 charakterisiert.
Es ist überraschend und konnte nicht ohne weiteres vorausgesehen werden, dass das Nylon-4 mit dem Jod einen Komplex bilden soll. Der Pyrrolidon-Ring wird nämlich bei der Polymerisation zum Polypyrrolidon geöffnet, er liegt also in Nylon-4, das bekanntlich eine Polyamidstruktur hat, nicht mehr vor. Im Gegensatz dazu ist der Pyrrolidon-Ring in Polyvinylpyrrolidon intakt. Trotzdem wurde, wie gesagt,.
gefunden, dass das Polypyrrolidon mit Jod einen Komplex bildet, dessen Eigenschaften sehr nahe demjenigen des Polyvinylpyrrolidon-Jod-Komplexes sind. Der einzige, aber sehr wichtige Unterschied ist die Wasserunlöslichkeit des neuen Komplexes.
Das Polypyrrolidon, welches für die Herstellung des gesuchten Komplexes mit Jod verwendet wird, kann in einer beliebigen physikalischen und dimensionellen Form vorliegen. Beispiele solcher Formen sind Pulver, Brei, Fasern, extrudierte Stäbe oder Fäden, Textilien, Folien oder auch formgepresste oder gegossene Formstücke. Für die Herstellung von Salben oder Pasten wird feines Pulver verwendet, wobei das mittlere Molekulargewicht des Polymeren (d.h. der Wert von n in der obigen Formel) nicht wichtig ist. Zur Herstellung von Verband- und Nähmaterialien aus gewobenen Textilien ist es nötig, das Harz vorerst zu schmelzen und zu extrudieren, um so die Fasern und Fäden zu erhalten, mit denen die genannten Materialien hergestellt werden können.
Für diese Zwecke wird naturgemäss ein Polymer eingesetzt, welches höhere thermische Stabilität aufweist, und hier spielt demnach das mittlere Molekulargewicht des Polymeren eine wichtige Rolle. Es gilt als angenommen, dass dazu ein Molekulargewicht nötig ist, welches dem Material eine inhärente Viskosität von mindestens 2,0 dl pro Gramm erteilt, wenn die Messung in einer Lösung von 0,5 Gramm Material pro dl Lösung in Hexafluorisopropanol bei 25"C ausgeführt wird.
Vorteilhafterweise hat das Harz eine Viskosität von 3,0 bis 4,0 dl pro Gramm, wenn es in Spinnverfahren eingesetzt werden soll. Verfahren zur Herstellung von Polymeren auf Basis von 2-Pyrrolidon mit den genannten Viskositätscharakteristiken werden in USA-Patent Nr. 3 721 652 dargelegt. Im gleichen Patent wird die Bestimmungsmethode für die inhärente Viskosität beschrieben.
Am leichtesten lässt sich der Komplex von Nylon-4 mit Jod durch Eintauchen von Nylon-4-Körpern in eine Lösung erreichen, welche sowohl Kaliumjodid wie auch freies Jod enthält. Einebevorzugte Rezeptur ist die folgende: Kaliumjodid 3,3 g Jod 2,0 g Wasser 100 ml
Nach Eintauchen von weissen Polypyrrolidon-Materialien in eine solche Lösung werden die genannten Materialien beinahe sofort schokoladebraun, dunkelbraun und dann beinahe schwarz. Nach dem Trocknen der Materialien wird die
Farbe wieder heller. Die Tauchzeiten betragen von einigen wenigen Sekunden bis zu 2 oder 3 Minuten. Kurze Tauchzeiten ergeben goldgelbe Produkte nach der Trocknung, währenddem längere Tauchzeiten solche mit dunkelbrauner Farbe ergeben. Die Farbe des getrockneten Komplexproduktes ist in etwa ein Mass für die darin enthaltene Jodmenge. Übermässiger Gehalt an Jod wird übrigens das Polymer langsam abbauen und sollte daher vermieden werden. Vor allem gilt dies für diejenigen Fälle, wo ein solcher Abbau vermieden werden soll, wie in den Textilfasern.
Der Jodkomplex kann mittels anderer Methoden, wie z.B.
Behandlung von Nylon-4-Stücken in Joddampf oder Mischen von pulverförmigen Nylonharz mit festen Jodkristallen unter Rühren, gebildet werden. Andererseits kann das Harz auch in einer Kugelmühle vorerst mit Jodkristallen vermischt und anschliessend vermahlen werden. Erhalten wird so ein pulverförmiger Komplexstoff, welcher sehr gut geeignet ist als Trockensubstanz in Rezepturen für Salben und Cremen. Weitere Ingredienzien, wie Mittel gegen das Zusammenbacken oder weitere therapeutische Wirkstoffe, können der Mischung zugegeben werden. Für die fertige Rezeptur kann die geeignete Fettbasis, wie beispielsweise Lanolin, Petrolatum, Kaliumstearat und viele andere, verwendet werden.
Das Jod wird im Komplex ziemlich stabil zurückgehalten.
Wenn beispielsweise Baumwolle (ähnliche hydrophile Fasern) in eine den obigen Mischungen entsprechende Jodlösung getaucht wird, färbt sich das Material in ähnlicher Weise dunkelbraun. Das Jod wird jedoch in der Baumwolle nur absorptiv zurückgehalten. Dies zeigt sich beim Auswaschen von derart präparierter Baumwolle mit Wasser. In einer Minute ungefähr ist alles Jod herausgewaschen. Der dunkelbraune Komplex aus Nylon-4 und Jod hingegen wird nicht so leicht mittels Wasser ausgewaschen. Kleine Mengen von Jod treten natürlich aus, wobei die Austrittsgeschwindigkeit u.a. von der Temperatur abhängig ist. Bei Raumtemperatur ist diese Austrittsgeschwindigkeit sehr klein. Bei ungefähr 1 000C hingegen geht die Abgabe von Jod aus dem Komplex sehr rasch von sich.
Falls also Nylon-4-Jod-Komplex Material in siedendes Wasser gegeben wird, wird das Jod in der Zeit von einigen Minuten vollständig aus dem Komplex abgegeben. Ähnlich ergeht es Komplexmaterial, das in einen Ofen bei 1 000C gegeben wird. Das Jod sublimiert in kurzer Zeit aus dem Komplex und hinterlässt das farblose Nylon-4
Material. Proben des genannten Komplexmaterials können bei Raumtemperatur einige Monate lang ohne sichtbaren Jodverlust aufbewahrt werden. Bei 40"C ist nach 24 Stunden ebenfalls noch kein messbarer Verlust von Jod festzustellen.
Das Komplexmaterial ist zudem ziemlich pH-unabhängig. In Lösungen von pH 5 bis pH 10 zeigt sich praktisch kein Jodverlust.
Um gute therapeutische Wirkung zu erreichen, muss die Jodabgabe bei Körpertemperatur die richtige Grösse haben.
Aus verschiedenen Gründen müssen Verbände alle 24 Stunden gewechselt werden, in dieser Zeit sollte aber eben ein Verband aus dem erfindungsgemässen Komplexmaterial wirksam sein. Wie weiter oben schon bemerkt, ist eine sehr wichtige Eigenschaft des Nylon-4-Jod-Komplexes gegenüber dem Polyvinylpvrrolidon-Jod-Komplex die Tatsache, dass Bandagen oder ähnliche Verbandstoffe durch Transpiration nicht inaktiv werden, da das Material aus dem Nylon-4-Jod Komplex nicht mitgelöst wird.
Nähmaterialien aus dem erfindungsgemässen Mittel können wie folgt dargestellt werden: das Polypyrrolidonharz wird schmelzextrudiert, bei Temperaturen von ungefähr 290"C und unter hohem Druck. Beim Extrudieren muss darauf geachtet werden, dass das Harz auf mindestens 0,03 Restfeuchtigkeit getrocknet wird. Während des Extrudierens muss das flüssige Kunstharz unter trockenem Stickstoff gehalten werden, um so jede weitere Aufnahme von Feuchtigkeit während des Extrudierens zu verunmöglichen.
Die flüssige Masse wird durch die Düse ausgepresst, wobei diese eine einlochige, oder, vorteilhafterweise, eine mehrlochige Düse ist. Die Dimensionen und Anzahl Löcher sind durch das gewünschte Produkt und dessen gesuchten Eigenschaften gegeben. Nach dem Austritt aus der Düse werden die Fäden durch das oben beschriebene Jodbad geführt, wobei auf die zu bestimmende Kontaktzeit zu achten ist.
Diese Kontaktzeit kann wie oben bei den Feststoffmischungen zwischen einigen Sekunden bis zu einigen Minuten dauern. Das Material wird darauf kurz mit Wasser gewaschen und luftgetrocknet.
Aus solchen Fasern können nun vorerst Fäden und daraus die gewünschten Gewebe hergestellt werden.
Andererseits können auch zuerst aus unbehandeltem Nylon-4 Gewebe hergestellt werden, welche anschliessend als Ganzes in einem Jodbad mit Jod beladen werden.
Der beschriebene Nylon-4-Jod-Komplex kann aber auch in vielen anderen Anwendungen eingesetzt werden. Beispielsweise könen Packungen innen teilweise mit solchem Komplex ausgelegt werden; sie dienen dann darin als Biozidbarriere, um den Inhalt der Packung steril zu halten. Ebenso können solche Komplexmaterialien an anderen Orten eingesetzt werden, wo dauernd eine minime Jodkonzentration zur Herstellung einer sterilen Atmosphäre gewünscht wird. Wie aus dem Gesagten folgt, gibt es sehr verschiedene Anwendungen des erfindungsgemässen Materials, die hier nicht aufgeführt sind.
Die Erfindung wird in den Ansprüchen definiert.

This invention relates to agents showing biocidal activity and to their use in topical pharmaceutical preparations.
The complex that forms when polyvinylpyrrolidone and iodine are mixed was first described in U.S. Patent No. 2739922 to Herman A. Shelanski.
This complex today represents the preferred application of the favorable biocidal properties of iodine without the side effects of the element mentioned. The agent is particularly preferred in surgery, where it is used to cover the skin before the incision.
Polyvinylpyrrolidone is a water-soluble polymer, and the polyvinylpyrrolidone-iodine complex also shows good water solubility. Now there are applications in which this water solubility is a highly undesirable property. For example, the efficiency of topical applications of such water-soluble antiseptics is short-lived since they are quickly washed away by both rain and perspiration. In such cases, an insoluble preparation that has the tamed iodine properties described above would be much more useful and efficient because of its longer duration of action.
The main purpose of this invention is to create a water-insoluble and therefore more stable iodine complex which has similar pharmacological and medical properties to the water-soluble polyvinylpyrrolidone-iodine complex.
Another purpose of this invention is to create a water-insoluble iodine complex which releases elemental iodine in small but effective amounts, slowly and over a long period of action, which on the one hand prolongs the biocidal activity of the agent and on the other hand the harmful side effects such as skin irritation, etc ., be minimized.
It is another purpose of this invention to provide new medical and surgical dressings and sutures which are themselves the bactericidal and fungicidal agents. Dressing materials of this type today consist of both woven and non-woven textiles.
Another purpose of this invention is to provide long-term antiseptic ointments and creams for topical applications.

It has been found that polypyrrolidone, also known as nylon-4 and a polymer of the structure, can be reacted with elemental iodine to form a complex that is very similar to the polyvinylpyrrolidone-iodine complex. In contrast to the polyvinylpyrrolidone-iodine complex, the nylon-4-iodine complex is insoluble in water.
The agent according to the invention exhibiting biocidal activity is characterized in the preceding patent claim 1.
It is surprising and could not have been foreseen that the nylon-4 should complex with the iodine. The pyrrolidone ring is opened during the polymerization to form polypyrrolidone, so it is no longer present in nylon-4, which is known to have a polyamide structure. In contrast, the pyrrolidone ring in polyvinylpyrrolidone is intact. Nevertheless, as I said,
found that the polypyrrolidone forms a complex with iodine, the properties of which are very close to those of the polyvinylpyrrolidone-iodine complex. The only but very important difference is the water insolubility of the new complex.
The polypyrrolidone, which is used for the preparation of the complex sought with iodine, can be in any physical and dimensional form. Examples of such shapes are powders, slurry, fibers, extruded rods or threads, textiles, foils or also compression molded or cast pieces. Fine powder is used in the manufacture of ointments or pastes, and the average molecular weight of the polymer (i.e. the value of n in the formula above) is not important. For the production of bandages and sewing materials from woven textiles, it is necessary to melt and extrude the resin first in order to obtain the fibers and threads with which the materials mentioned can be produced.
A polymer which has higher thermal stability is naturally used for these purposes, and the average molecular weight of the polymer therefore plays an important role here. It is assumed that this requires a molecular weight which gives the material an inherent viscosity of at least 2.0 dl per gram if the measurement is carried out in a solution of 0.5 gram of material per dl solution in hexafluoroisopropanol at 25 ° C. becomes.
The resin advantageously has a viscosity of 3.0 to 4.0 dl per gram if it is to be used in spinning processes. Processes for the production of polymers based on 2-pyrrolidone with the viscosity characteristics mentioned are set out in US Pat. No. 3,721,652. The easiest way to achieve the complex of nylon-4 with iodine is by immersing nylon-4 bodies in a solution that contains both potassium iodide and free iodine. A preferred recipe is the following: potassium iodide 3.3 g iodine 2.0 g water 100 ml

Short dipping times result in golden yellow products after drying, while longer dipping times result in products with a dark brown color. The color of the dried complex product is roughly a measure of the amount of iodine it contains. Incidentally, excessive iodine content will slowly degrade the polymer and should therefore be avoided. This is especially true for those cases where such degradation should be avoided, such as in textile fibers.
The iodine complex can be prepared using other methods, e.g.
Treatment of nylon 4 pieces in iodine vapor or mixing powdered nylon resin with solid iodine crystals with stirring. On the other hand, the resin can also be mixed with iodine crystals in a ball mill and then ground. This gives a powdery complex substance which is very suitable as a dry substance in formulations for ointments and creams. Other ingredients, such as anti-caking agents or other therapeutic agents, can be added to the mixture. The appropriate fat base, such as lanolin, petrolatum, potassium stearate and many others, can be used for the finished formulation.
The iodine is retained fairly stable in the complex.
For example, if cotton (similar hydrophilic fibers) is immersed in an iodine solution corresponding to the above mixtures, the material will turn dark brown in a similar manner. However, the iodine is only retained in the cotton by absorption. This can be seen when washing such prepared cotton with water. In about a minute, all of the iodine will be washed out. The dark brown complex of nylon 4 and iodine, on the other hand, is not easily washed out with water. Of course, small amounts of iodine leak out, and the rate of leakage, depends on the temperature. This exit speed is very low at room temperature. At about 1000C, however, iodine is released from the complex very quickly.
So if nylon 4-iodine complex material is added to boiling water, the iodine will be completely released from the complex within a few minutes. The same applies to complex material that is placed in an oven at 1000C. The iodine sublimes from the complex in a short time and leaves the colorless nylon-4
Material. Samples of the complex material mentioned can be stored at room temperature for a few months without any visible loss of iodine. At 40 "C there is still no measurable loss of iodine after 24 hours.
The complex material is also fairly pH-independent. There is practically no loss of iodine in solutions from pH 5 to pH 10.
In order to achieve good therapeutic effects, the iodine release at body temperature must be of the correct size.
For various reasons, dressings have to be changed every 24 hours, but during this time a dressing made of the complex material according to the invention should be effective. As already noted above, a very important property of the nylon 4-iodine complex compared to the polyvinylprrolidone-iodine complex is the fact that bandages or similar dressings do not become inactive due to perspiration, since the material consists of the nylon 4-iodine Complex is not resolved.
Sewing materials from the agent according to the invention can be represented as follows: the polypyrrolidone resin is melt extruded, at temperatures of approximately 290 ° C. and under high pressure. Such fibers can now be used to produce threads and the desired fabrics.
On the other hand, it can also be made from untreated nylon-4 fabric, which is then loaded with iodine as a whole in an iodine bath.
The nylon-4-iodine complex described can also be used in many other applications. For example, packs can be partially covered with such a complex on the inside; they then serve as a biocide barrier to keep the contents of the package sterile. Likewise, such complex materials can be used in other places where a minimum iodine concentration is constantly required to create a sterile atmosphere. As follows from what has been said, there are very different applications of the material according to the invention which are not listed here.
