Aus der US-Patenischrift Nr. 3 940 400 ist bereits das 3-Methyl-4-phenyl-1,2,3,44efrahy & benzgl isochinolin als Anxiolytikum bekannt. Die Verbindungen der vorliegenden Erfindung sind gesättigt in Stellung 10,10a und unterscheiden sich ausserdem von der vorbekannten Verbindung sowohl durch die Substitution in Stellung 3, 4 und 5 wie auch durch ein neuartiges pharmakologisches Wirkungsmuster.
Man gelangt zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man a) zur Herstellung von Verbindungen der Formel Ia,

Sofern nicht anders angegeben, enthalten die in den Verbindungen der Formel I vorstehend definierten Alkyl-, Alkoxy- oder Alkylthiogruppen vorzugsweise 1 oder 2 Kohlenstoffatome und stellen insbesondere Methyl, Methoxy oder Methylthio dar. Sofern ein Substituent für vorstehend definiertes Halogen steht, stellt er vorzugsweise Chlor dar.
Der Substituent R1 steht vorzugsweise für Wasserstoff.
Ein sonstiger Substituent R1 ist vorzugsweise in 7- oder
8-Stellung des tricyclischen Ringgerüstes angeordnet.
Der Substituent R2 steht vorzugsweise für Wasserstoff oder bedeutet eine vorstehend definierte Alkylgruppe, insbesondere Methyl oder Isopropyl. Falls R2 eine vorstehend definierte Alkenyl- oder Alkinylgruppe bedeutet, enthält diese vorzugsweise 3-4 Kohlenstoffatome. Eine vorstehend definierte Hydroxylalkylgruppe R2 enthält vorzugsweise 2 oder 3 Kohlenstoffatome. Falls R2 eine vorstehend definierte Alkylcarbonylalkylgruppe darstellt, so enthält diese vorzugswei se einen Acetylrest und bedeutet insbesondere Acetonyl. Eine durch R2 symbolisierte vorstehend definierte Phenylalkylgruppe stellt vorzugsweise eine Benzylgruppe dar, welche gegebenenfalls durch ein vorstehend definiertes Halogen, insbesondere Chlor, substituiert sein kann.
Bevorzugt steht der Substituent R3 für Wasserstoff, niederes Alkyl oder eine vorzugsweise unsubstituierte Phenylgruppe.
Die Wasserabspaltung aus Verbindungen der Formel II gemäss Verfahren a) kann auf an sich zur Wasserabspaltung aus analogen Carbinolen bekannte Weise, beispielsweise durch Einwirkung geeigneter wasserabspaltender Mittel, gegebe nenfalls in Gegenwart eines unter den Reaktionsbedingungen inerten organischen Lösungsmittels, beispielsweise eines niederen Alkohols erfolgen.
Die Abspaltung des Restes R4 aus den Verbindungen der Formel III gemäss Verfahren b) kann auf an sich zur Ab spaltung von Aminoschutzgruppen aus heterocyclischen Aminen übliche Weise solvolytisch, insbesondere hydrolytisch erfolgen, beispielsweise nach zur Urethanspaltung bekannten Methoden.
Die Alkylierung der Verbindungen der Formel Ib gemäss Verfahrensvariante c) kann nach an sich zur Alkylie rung sekundärer Amine üblichen Methoden durchgeführt werden. Beispielsweise können die Verbindungen der Formel
Ib mit Verbindungen der Formel IV, R2lI-X IV worin R2lI obige Bedeutung besitzt und X für den Säurerest eines reaktionsfähigen Esters, vorzugsweise für Halogen oder einen organischen Sulfonsäurerest steht, auf an sich bekannte Weise umgesetzt werden.
Zur Herstellung von Verbindungen der Formel Ic, worin R2ll für eine vorstehend definierte Alkanoylalkylgruppe, de ren Carbonylgruppe durch 2 Kohlenstoffatome von dem
Stickstoffatom getrennt ist, steht, kann die Alkylierung auch durch Addition entsprechender a,ss-ungesättigter Ketone auf an sich bekannte Weise erfolgen. Die Einführung eines Hy droxyalkylrestes, dessen Hydroxygruppe durch 2 Kohlenstoffatome von dem Stickstoffatom getrennt ist, kann durch Anlagerung entsprechender Epoxyde erfolgen.
Die erhaltenen Verbindungen der Formel I können in
Form der freien Basen oder ihrer Säureadditionssalze vorliegen. Die freien Basen können auf an sich bekannte Weise in ihre Säureadditionssalze überführt werden und umgekehrt. Zur Säureadditionssalzbildung können z.B. folgende Säuren verwendet werden: Chlorwasserstoff-, Schwefel-, Fumar-, Malein- oder Malonsäure.

Verbindungen der Formel III können beispielsweise erhalten werden, indem man in solchen Verbindungen der Formel I, worin R2 Methyl bedeutet, auf an sich bekannte Weise die Methylgruppe in einen solvolytisch abspaltbaren Rest R4 überführt, z.B. durch Umsetzung mit einem Halogenderivat eines Restes R4 wie z.B. Chlorameisensäureestern oder Bromcyan. Verbindungen der Formel III, worin R3 Wasserstoff und R4 eine Carbalkoxygruppe bedeutet, können auch durch Abspaltung von Wasser aus entsprechenden 5-Hydroxyderivaten analog Verfahren a) hergestellt werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I in freier Form oder in Form ihrer pharmakologisch verträglichen Säureadditionssalze zeichnen sich im Tierversuch durch vielfältige pharmakodynamische Eigenschaften aus und können daher als Heil mittel verwendet werden. So eignen sich die Verbindungen insbesondere als Antidepressiva.
Ausserdem besitzen die Verbindungen der Formel I analgetische Eigenschaften. Aufgrund ihrer analgetischen Wirksamkeit können die Verbindungen zur Behandlung von Schmerzen verschiedener Genese verwendet werden.
Ferner besitzen die Verbindungen der Formel I, in welchen R3 Wasserstoff oder eine Alkylgruppe bedeutet, antiaggressive Eigenschaften und eignen sich daher zur Behandlung von aggressiven Erregungszuständen, beispielsweise zur Dämpfung von aggressivem Verhalten von Psychopathen und Schwachsinnigen.
Schliesslich eignen sich die Verbindungen der Formel I, in welchen R3 eine Thienylgruppe oder eine gegebenenfalls substituierte Phenylgruppe bedeutet, als Blutplättchen-Aggregations-Hemmer, z.B. für die Prophylaxe und Therapie von Krankheitszuständen bei welchen thrombo-embolische Komplikationen eine wesentliche Rolle spielen.
Die Erfindung betrifft auch Heilmittel, die eine Verbindung der Formel I in freier Form oder in Form eines pharmakologisch verträglichen Säureadditionssalzes enthalten.
Diese Heilmittel können nach bekannten Methoden, unter Verwendung der üblichen Hilfs- und Trägerstoffe, hergestellt werden.
In den nachfolgenden Beispielen, welche die Erfindung näher erläutern, ihren Umfang aber in keiner Weise, einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1
1,2,3,4,10, iOa-Hexahydro-2-isopropyl-5-phenylbenz[g]- isochinolin [Verfahren a)]
5,9 g rohes trans- 1 ,2,3,4,4a, 1 0a-Hexahydro-2-isopropyl- -5-phenyl- 1 OH-benz[g] isochinolin-5-ol werden in 180 ml Isopropanol und 36 ml 7 N isopropanolischer Chlorwasserstofflösung gelöst, 2 Stunden zum Sieden erhitzt und eingeengt. Der Rückstand wird mit Wasser versetzt, mit konz.
Natronlauge alkalisch gestellt und mit Methylenchlorid extrahiert. Die Extrakte werden mit Wasser gewaschen, getrocknet, eingedampft und die zurückbleibende Titelverbindung wird in Isopropanol in ihr Fumarat übergeführt.
Smp.: 199-200 .
Das Ausgangsmaterial wird wie folgt hergestellt: a) 47,5 g 3-Benzyl-4-oxopiperidin-1 -carbonsäureäthyl- ester werden mit 15,6 g Acetoncyanhydrin versetzt und nach Zugabe einer Spatelspitze mit Methanol angefeuchtetem Kaliumcarbonat wird das Reaktionsgemisch 5-8 Stunden bei Raumtemperatur weitergerührt. Nach 5-10 Stunden Stehen bei Raumtemperatur wird das breiige Reaktionsgemisch mit 20 ml Äther und 20 ml Petroläther versetzt, das feste Produkt abgesaugt, nachgewaschen und bei 50 getrocknet.
Smp. des erhaltenen 3-Benzyl-4-cyan-4-hydroxypip eridin- 1 -carbonsäureäthylesters: 120-121 (aus Äther/Petroläther).
b) Eine Lösung von 16,0 g des vorstehenden Produktes in 20 ml wasserfreiem Pyridin und 14 ml wasserfreiem Benzol wird bei 5O mit 6,5 ml Thionylchlorid tropfenweise versetzt. Das Reaktionsgemisch wird 5 Stunden bei 0-5" und 2 Stunden bei Raumtemperatur stehen gelassen, auf 100 ml Eiswasser gegossen, die organische Phase wird abgetrennt und mit Wasser neutral gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Das als öl erhaltene Gemisch von 5-Benzyl-4-cyan-1,2,3,6-tetrahydropyridin-1-carbonsäureäthylester und 3-Benzyl-4cyan- 1 ,2,5,6-tetrahydropyridin- 1- -carbonsäureäthylester wird roh weiterverarbeitet.
c) Eine Lösung von 20,0 g des vorstehendes Gemisches in 300 ml Essigsäureäthylester wird in Gegenwart von 2,0 g 10%dem Palladium/Kohle bei 30 atü Wasserstoffdruck und Normaltemperatur hydriert. Nach Aufnahme der theoretischen Menge Wasserstoff wird der Katalysator abfiltriert und das Lösungsmittel unter vermindertem Druck abgedampft. Das als öl erhaltene 3-Benzyl-4-cyanpiperidin- 1-car- bonsäureäthylester (cis-trans-Isomerengemisch) wird roh weiterverarbeitet.
d) Zur Lösung von 20,0 g des vorstehenden Produktes in 400 ml Aceton und 30 ml 2 N Natriumcarbonatlösung werden 99,7 ml 40%iger Wasserstoffperoxidlösung zugegeben und das Ganze 2 Tage bei Raumtemperatur gerührt, dann das Aceton abgedampft, der Rückstand mit Wasser verdünnt und mit Methylenchlorid extrahiert. Die organischen Lösungen werden nach dem Waschen mit Natriumcarbonatlösung und mit Wasser über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Äther fraktioniert kristallisiert, wobei zuerst der trans-3-Benzyl-4-carbamoylpi peridin-1-carbonsäureäthylester vom'Smp. 155-156 und dann das cis-Isomere vom Smp. 136-138 auskristallisiert.
e) 20,0 g des vorstehenden Produktes werden mit 120 ml konz. Salzsäure und 240 ml Wasser 4 Stunden zum Sieden erhitzt, auf Raumtemperatur abgekühlt und mit Methylenchlorid extrahiert. Die Extrakte werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingedampft und die zurückbleibende 1-Äthoxycarbonyl-3-benzyl-4-pipe- ridincarbonsäure (cis-trans-Isomerengemisch) wird aus Aceton/Hexan umkristallisiert. Smp.: 143-148 .
f) Ein Gemisch von 20,0 g des vorstehenden Produktes und 120 g Polyphosphorsäure wird 1 Stunde bei 80" erhitzt, abgekühlt, auf Eiswasser gegossen und mehrmals mit Benzol extrahiert. Die organischen Lösungen werden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingedampft Der zurückbleibende trans-1,2,3,4,4a,10a-Hehaxydro-5-oxo- - 1 OH-benz [gl isochinolin-2-carbonsäureäthylester wird aus Äther kristallisiert Smp. 90-920.
g) Ein Gemisch von 15,0 g des vorstehenden Produktes und 95 ml Essigsäure und 36,5 ml 48%iger Bromwasserstoffsäure wird 3,5 Stunden zum Sieden erhitzt, abgekühlt und mit konz. Natronlauge alkalisch gestellt. Das als öl ausgeschiedene Produkt wird mit Methylenchlorid extrahiert, die organischen Lösungen werden mit Wasser gewaschen, über Kaliumcarbonat getrocknet und eingedampft. Das zurückbleibende trans-1,2,3,4,4a, 10a-Hexahydro-10H-benz[gJ iso- chinolin-5-on wird aus Aceton umkristallisiert. Smp. des Hydrochlorids: Zers. ab 290".
h) Ein Gemisch von 18,0 g des vorstehenden Produktes, 20,0 g Natriumcarbonat und 0,2 g Natriumjodid in 180 ml Dimethylformamid wird auf 500 vorgeheizt. Zu dieser Suspension werden 16,5 g Isopropylbromid in 50 ml Dimethylformamid innert 2 Stunden zugetropft, das Reaktionsgemisch wird 3,5 Stunden bei gleicher Temperatur weitergerührt, abgekühlt und auf Eiswasser gegossen. Die organische Substanz wird mit Äther extrahiert, die Extrakte werden mit Wasser gewaschen, über Kaliumcarbonat getrocknet und eingedampft. Das zurückbleibende trans-1,2,3,4, 4a,10a-Hexahydro-2-isopropyl-lOH-benz[g] isochinolin-5-on wird roh weiterverarbeitet.
i) Aus 0,77 g Lithium und 7,2 g Brombenzol wird in 50 ml wasserfreiem Äther Phenyllithium hergestellt. Nachdem das Lithium gelöst ist, wird bei 10-15 eine Lösung von 4,8 g des vorstehenden Produktes in 70 ml wasserfreiem Äther zugetropft. Das Reaktionsgemisch wird nun 1 Stunde bei Raumtemperatur und 18 Stunden am Rückfluss gekocht und gerührt, unter starkem Kühlen bei ca. 0-10 mit Wasser versetzt und die organische Phase wird abgetrennt.
Sie wird mit Wasser gewaschen, über Natriumsulfat getrock net und eingedampft. Das zurückbleibende trans-1,2,3,4,4a,- 1 Oa-Hexahydro-2-isopropyl-5-phenyl- 1 OH-benz [g] isochinolin -5-ol wird roh weiterverarbeitet.
Analog Beispiel 1 können auch die folgenden Verbindungen der Formel Ia durch Wasserabspaltung aus den entsprechenden Verbindungen der Formel II erhalten werden:

Beispiel 2 1,2,3,4,10,10a-Hexahydrobenz[g]isochinolin [Verfahren b)]
Ein Gemisch von 10,0 g 1,2,3,4,10,1 0a-Hexahydrobenz- Lg]isochinolin-2-carbonsäureäthylester in 100 ml Eisessig und 40 ml 48%iger Bromwasserstoffsäure wird 3-4 Stunden zum Sieden erhitzt, eingeengt und unter Kühlen mit konz.
Natronlauge alkalisch gestellt. Das Gemisch wird mit Methylenchlorid extrahiert, die Extrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet, eingedampft und die als öliger Rückstand verbleibende Titelverbindung wird in das Hydrochlorid in Äthanol übergeführt. Smp.: Zers. ab 250".
Das Ausgangsmaterial kann wie folgt hergestellt werden: a) 20,0 g trans-1,2,3,4,4a,10a-Hexahydro-5-oxo-lOH- -benz[g] isochinolin-2-carbonsäureäthylester (Herstellung siehe Beispiel 1 Stunfen a-f) werden in 110 ml 95%igem Äthanol gelöst und auf 40 vorgeheizt. Bei dieser Temperatur wird eine Lösung von 7,0 g Natriumborhydrid in 32,5 ml Wasser und 2 ml konz. Natronlauge zugetropft, die Reaktionslösung wird 2 Stunden beig leicher Temperatur gerührt, 30 Minuten zum Sieden erhitzt, abgekühlt und zur Trockne eingedampft. Der als Rückstand verbleibende trans-1,2,3,4,4a,10a-Hexa- hydro-5-hydroxy- 1 0H-benz[gJ isochinolin-2-carbonsäure- äthylester wird ohne weitere Reinigung für die nächste Stufe verwendet.
b) 15,0 g des vorstehenden Produktes werden in 200 ml 5 H isopropanolischer Chlorwasserstofflösung aufgenommen, unter Rühren 15 Stunden auf 40-500 erhitzt und eingedampft. Der Rückstand wird in Wasser und Methylenchlorid verteilt, die organische Lösung abgetrennt, mit Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft.
Der als Rückstand verbleibende 1,2,3,4,10,10a-Hexahydro benz[g]isochinolin-2-carbonsäureäthylester wird roh für die letzte Stufe eingesetzt.
Beispiel 3 1,2,3,4,10,1 0a.Hexahydro-2-isopropyl-5-phenylbenz[g]- isochinolin [Verfahren c)
Ein Gemisch von 22,0 g 1,2,3,4,10,10a-Hexahydro-5 .phenylbenz[gjisochinolin, 20,0 g Natriumcarbonat und 0,2 g Natriumjodid in 180 ml Dimethylformamid wird auf 50 vorgeheizt. Zu dieser Suspension werden 16,5 g Isopropylbromid in 50 ml Dimethylformamid innert 2 Stunden zugetropft, das Reaktionsgemisch wird 3,5 Stunden bei gleicher Temperatur weitergerührt, abgekühlt und auf Eiswasser gegossen. Die organische Substanz wird mit Benzol extrahiert, die Extrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet, eingedampft und die als Rückstand verbleibende Titelverbindung wird in Isopropanol in ihr Fumarat übergeführt. Smp.: 199-2000.
Analog Beispiel 3 können auch die Verbindungen der Beispiele 1p bis 1N durch Alkylierung der entsprechenden Verbindungen der Formel Ib erhalten werden.

The compounds of the formula I in free form or in the form of their pharmacologically acceptable acid addition salts are distinguished by a wide range of pharmacodynamic properties in animal experiments and can therefore be used as medicinal agents. The compounds are particularly suitable as antidepressants.
In addition, the compounds of formula I have analgesic properties. Because of their analgesic activity, the compounds can be used to treat pain of various origins.

The invention also relates to medicaments which contain a compound of formula I in free form or in the form of a pharmacologically acceptable acid addition salt.
These remedies can be produced by known methods using the usual auxiliaries and carriers.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all the temperatures are given in degrees Celsius.
example 1
1,2,3,4,10, iOa-hexahydro-2-isopropyl-5-phenylbenz [g] - isoquinoline [process a)]
5.9 g of crude trans-1, 2,3,4,4a, 1,0a-hexahydro-2-isopropyl--5-phenyl-1 OH-benz [g] isoquinolin-5-ol are dissolved in 180 ml of isopropanol and 36 ml 7 N isopropanolic hydrogen chloride solution dissolved, heated to boiling for 2 hours and concentrated. The residue is mixed with water, with conc.
Sodium hydroxide solution made alkaline and extracted with methylene chloride. The extracts are washed with water, dried, evaporated and the remaining title compound is converted into its fumarate in isopropanol.
M.p .: 199-200.
The starting material is prepared as follows: a) 47.5 g of 3-benzyl-4-oxopiperidine-1-carboxylic acid ethyl ester are mixed with 15.6 g of acetone cyanohydrin and, after adding a spatula tip with potassium carbonate moistened with methanol, the reaction mixture is 5-8 hours stirred further at room temperature. After standing for 5-10 hours at room temperature, the mushy reaction mixture is mixed with 20 ml of ether and 20 ml of petroleum ether, the solid product is suctioned off, washed and dried at 50.
Mp. Of the 3-benzyl-4-cyan-4-hydroxypip eridin-1-carboxylic acid ethyl ester obtained: 120-121 (from ether / petroleum ether).
b) A solution of 16.0 g of the above product in 20 ml of anhydrous pyridine and 14 ml of anhydrous benzene is added dropwise at 50 with 6.5 ml of thionyl chloride. The reaction mixture is left to stand at 0-5 "for 5 hours and at room temperature for 2 hours, poured onto 100 ml of ice water, the organic phase is separated off and washed neutral with water, dried over magnesium sulfate and evaporated. The mixture of 5-benzyl obtained as an oil -4-cyan-1,2,3,6-tetrahydropyridine-1-carboxylic acid ethyl ester and 3-benzyl-4cyan-1, 2,5,6-tetrahydropyridine-1-carboxylic acid ethyl ester is further processed raw.
c) A solution of 20.0 g of the above mixture in 300 ml of ethyl acetate is hydrogenated in the presence of 2.0 g of 10% palladium / carbon at 30 atm hydrogen pressure and normal temperature. After the theoretical amount of hydrogen has been taken up, the catalyst is filtered off and the solvent is evaporated off under reduced pressure. The 3-benzyl-4-cyanpiperidine-1-carboxylic acid ethyl ester (cis-trans isomer mixture) obtained as an oil is further processed raw.
d) 99.7 ml of 40% hydrogen peroxide solution are added to the solution of 20.0 g of the above product in 400 ml of acetone and 30 ml of 2N sodium carbonate solution and the whole is stirred at room temperature for 2 days, then the acetone is evaporated off, and the residue is dissolved in water diluted and extracted with methylene chloride. After washing with sodium carbonate solution and with water, the organic solutions are dried over sodium sulfate and evaporated. The residue is fractionally crystallized from ether, the trans-3-benzyl-4-carbamoylpi peridine-1-carboxylic acid ethyl ester from 'Smp. 155-156 and then the cis isomer of mp 136-138 crystallized.
e) 20.0 g of the above product are concentrated with 120 ml. Hydrochloric acid and 240 ml of water heated to boiling for 4 hours, cooled to room temperature and extracted with methylene chloride. The extracts are washed with water, dried over magnesium sulfate and evaporated, and the remaining 1-ethoxycarbonyl-3-benzyl-4-piperidincarboxylic acid (cis-trans isomer mixture) is recrystallized from acetone / hexane. M.p .: 143-148.
f) A mixture of 20.0 g of the above product and 120 g of polyphosphoric acid is heated at 80 "for 1 hour, cooled, poured onto ice water and extracted several times with benzene. The organic solutions are washed with water, dried over magnesium sulfate and evaporated. The remaining trans-1,2,3,4,4a, 10a-hehaxydro-5-oxo - 1 OH-benz [gl isoquinoline-2-carboxylic acid ethyl ester is crystallized from ether, mp 90-920.
g) A mixture of 15.0 g of the above product and 95 ml of acetic acid and 36.5 ml of 48% hydrobromic acid is heated to boiling for 3.5 hours, cooled and concentrated with conc. Sodium hydroxide solution made alkaline. The product which is excreted as an oil is extracted with methylene chloride, the organic solutions are washed with water, dried over potassium carbonate and evaporated. The remaining trans-1,2,3,4,4a, 10a-hexahydro-10H-benz [gJ isoquinolin-5-one is recrystallized from acetone. Mp of hydrochloride: decomp. from 290 ".
h) A mixture of 18.0 g of the above product, 20.0 g of sodium carbonate and 0.2 g of sodium iodide in 180 ml of dimethylformamide is preheated to 500. 16.5 g of isopropyl bromide in 50 ml of dimethylformamide are added dropwise to this suspension within 2 hours, the reaction mixture is stirred for a further 3.5 hours at the same temperature, cooled and poured onto ice water. The organic substance is extracted with ether, the extracts are washed with water, dried over potassium carbonate and evaporated. The remaining trans-1,2,3,4,4a, 10a-hexahydro-2-isopropyl-lOH-benz [g] isoquinolin-5-one is further processed raw.
i) Phenyllithium is produced from 0.77 g of lithium and 7.2 g of bromobenzene in 50 ml of anhydrous ether. After the lithium is dissolved, a solution of 4.8 g of the above product in 70 ml of anhydrous ether is added dropwise at 10-15. The reaction mixture is then boiled and stirred at reflux for 1 hour at room temperature and for 18 hours, water is added with vigorous cooling at about 0-10 and the organic phase is separated off.
It is washed with water, dried over sodium sulfate and evaporated. The remaining trans-1,2,3,4,4a, - 1 Oa-hexahydro-2-isopropyl-5-phenyl-1 OH-benz [g] isoquinoline -5-ol is further processed raw.

Example 2 1,2,3,4,10,10a-Hexahydrobenz [g] isoquinoline [Process b)]
A mixture of 10.0 g 1,2,3,4,10,1 0a-hexahydrobenz-Lg] isoquinoline-2-carboxylic acid ethyl ester in 100 ml glacial acetic acid and 40 ml 48% hydrobromic acid is heated to boiling for 3-4 hours, concentrated and under cooling with conc.
Sodium hydroxide solution made alkaline. The mixture is extracted with methylene chloride, the extracts are washed with water, dried over sodium sulfate, evaporated and the title compound remaining as an oily residue is converted into the hydrochloride in ethanol. M.p .: dec. from 250 ".
The starting material can be prepared as follows: a) 20.0 g of trans-1,2,3,4,4a, 10a-hexahydro-5-oxo-lOH- -benz [g] isoquinoline-2-carboxylic acid ethyl ester (preparation see example 1 step af) are dissolved in 110 ml of 95% ethanol and preheated to 40. At this temperature, a solution of 7.0 g of sodium borohydride in 32.5 ml of water and 2 ml of conc. Sodium hydroxide solution added dropwise, the reaction solution is stirred for 2 hours at a lower temperature, heated to boiling for 30 minutes, cooled and evaporated to dryness. The residue is distributed in chloroform and water, the organic solution is separated off, washed with water, dried over sodium sulfate and evaporated.
The remaining trans-1,2,3,4,4a, 10a-hexa- hydro-5-hydroxy-1 0H-benz [gJ isoquinoline-2-carboxylic acid - ethyl ester is used for the next step without further purification.
b) 15.0 g of the above product are taken up in 200 ml of 5 H isopropanolic hydrogen chloride solution, heated to 40-500 with stirring for 15 hours and evaporated. The residue is distributed in water and methylene chloride, the organic solution is separated off, washed with water, dried over sodium sulfate and evaporated.
The 1,2,3,4,10,10a-hexahydro benz [g] isoquinoline-2-carboxylic acid ethyl ester remaining as residue is used crude for the last stage.
Example 3 1,2,3,4,10,1 0a.Hexahydro-2-isopropyl-5-phenylbenz [g] - isoquinoline [Process c)
A mixture of 22.0 g of 1,2,3,4,10,10a-hexahydro-5-phenylbenz [gjisoquinoline, 20.0 g of sodium carbonate and 0.2 g of sodium iodide in 180 ml of dimethylformamide is preheated to 50. 16.5 g of isopropyl bromide in 50 ml of dimethylformamide are added dropwise to this suspension within 2 hours, the reaction mixture is stirred for a further 3.5 hours at the same temperature, cooled and poured onto ice water.
