Somit haben die erfindungsgemässen Verbindungen eine ganz einzigartige chemische Struktur und sind nicht vergleichbar mit herkömmlichen Antidepressiva oder bekannten Verbindungen mit antidepressiver Wirkung.
Die vorliegende Erfindung bezieht sich auf neue tetracyclische Verbindungen der Formel:

In den obigen Definitionen umfasst der Ausdruck Alkyl mit 1 bis 3 Kohlenstoffatomen Methyl, Äthyl und Isopropyl. Der Ausdruck Halogen umfasst Fluor, Chlor und Brom.
Die tetracyclischen Verbindungen der Formel I vermögen Säureadditionssalze, z. B. Hydrochloride, Hydrobromide, Sulfate, Acetate, OxaIate, Citrate, Fumarate, Maleate, Lactate und Tartrate, sowie quaternäre Ammoniumsalze, z. B. Methochloride und Methojodide, zu bilden.
Die tetracyclischen Verbindungen der Formel I und ihre pharmazeutisch unbedenklichen Salze weisen pharmakologische Aktivitäten auf und sind brauchbar als Arzneimittel. Im allgemeinen beeinflussen sie die Funktionen des Zentralnervensystems, d. h. sie hemmen die depressive Wirkung auf das Zentralnervensystem, die durch Tetrabenazin und Reserpin hervorgerufen wird, und potenzieren auch die zentralen Wirkungen von Norepinephrin. Daher sind sie brauchbar als Antidepressiva.

Die tetracyclischen Verbindungen der Formel I und deren pharmazeutisch unbedenkliche Salze können in Form von herkömmlichen pharmazeutischen Präparaten parenteral oder oral verabreicht werden, wobei die Dosierung an die individuellen Anforderungen angepasst werden kann und 10 bis 300 mg pro menschlichen Körper bzw. 10 bis 300 mg pro 60 kg Körpergewicht und Tag betragen kann.
Zum Beispiel können sie in Form eines herkömmlichen festen pharmazeutischen Präparates, wie Tabletten oder Kapseln, oder in Form eines herkömmlichen flüssigen pharmazeutischen Präparates, wie Suspensionen, Emulsionen oder Lösungen, verabreicht werden.
Die tetracyclischen Verbindungen der Formel I werden erfindungsgemäss nach verschiedenen Verfahren hergestellt: (a) Tetracyclische Verbindungen der Formel:

Die Reduktion kann unter Verwendung eines Reduktionsmittels, das herkömmlicherweise für die Reduktion eines Amides zu einem Amin verwendet wird, ausgeführt werden. Eines der bevorzugtesten Reduktionsmittel ist ein Metallhydrid, wie Lithiumaluminiumhydrid, Natrium-bis (2-methoxy-äthoxy)-aluminiumhydrid oder Natriumdihydrodiäthylaluminat. Das Reduktionsmittel kann in einer mindestens äquimolaren Menge, bezogen auf das Amid der Formel II, verwendet werden. Wenn man Natriumborhydrid als Reduktionsmittel verwendet, wird das Vorhandensein eines Salzes, wie Aluminiumchlorid, bevorzugt. Gewünschtenfalls kann ein inertes Lösungsmittel, wie ein Äther, z. B. Diäthyläther, Tetrahydrofuran, Dioxan oder Äthylenglycoldimethyläther, ein aliphatischer Kohlenwasserstoff, z. B. n-Hexan, n-Heptan oder Cyclohexan, oder ein aromatischer Kohlenwasserstoff, z. B.

Die Oxydation kann unter Verwendung eines Oxydationsmittels, das herkömmlicherweise für die Oxydation eines Alkohols zu einem Keton verwendet wird, ausgeführt werden. Bevorzugte Oxydationsmittel sind Mangandioxyd, Chromsäure, Chromate usw. Die Oxydationsreaktion wird gewöhnlich in einem inerten Lösungsmittel, wie einem aliphatischen Kohlenwasserstoff, z. Die so hergestellten tetracyclischen Verbindungen der Formel I können nach herkömmlichen Verfahren in ihre Salze übergeführt werden, und auch die Überführung der Salze in die ursprünglichen freien Basen kann in herkömmlicher Weise erfolgen.
Verschiedene Ausgangsmaterialien für die obigen Verfahren sind neu und können nach dem folgenden Reaktionsschema hergestellt werden:

Somit können die tricyclischen Verbindungen der Formel IX mit Alkoxycarbonylcarbenen, die aus Alkyldiazoacetaten hergestellt sind, zu den Cyclopropancarbonsäurealkylestern der Formel X umgesetzt werden. Die Carbonsäureester der Formel X können zu den entsprechenden Carbonsäuren verseift werden, die dann ihre reaktionsfähigen Derivate, wie Säureanhydride oder Säurehalogenide, übergeführt und mit Aminen zu den tetracyclischen Amiden der Formel XI umgesetzt werden können. Ferner können die Amide der Formel XI, worin X5 -NCOR5- bedeutet, mit Metallhydridkomplexen, wie Lithiumaluminiumhydrid, bei niedrigen Temperaturen zu den Amiden der Formel XII umgesetzt werden, oder die Amide der Formel XI, worin X5 -CO bedeutet, können zu den Aminen der Formel IV reduziert werden.
Die Carbonsäureester der Formel X können aber auch zu den entsprechenden Alkoholen der Formel XIII reduziert und dann in reaktionsfähige Ester der Formel V übergeführt werden. Die Verbindungen der Formel XIII, worin X6 -CHOH- bedeutet, können zu den entsprechenden Ketonen oxydiert und dann in reaktionsfähige Ester der Formel V, worin X4 -CO- bedeutet, übergeführt werden.
Einige der tricyclischen Verbindungen der Formel IX und ihre Herstellung sind bekannt, und die anderen Verbindungen können aus diesen nach an sich bekannten Verfahren oder in ähnlicher Weise synthetisiert werden.
Zum Beispiel sind Dibenzo-[b,fl-oxepin (IX: X5 = o; yl = H) und sein 2-Chlorderivat (IX: X5 = o; yl = Cl) [V.

Chem. 12, 913 (1969)] usw. bekannt. 2-Fluordibenzo-[b,fl-oxepin (IX: X5 = O; Y1 = F), 2-Bromdibenzo-[b,f]-oxepin (IX: X5 = 0; = = Br), 2-Fluordibenzo-[b,fl-thiepin (IX: X5 = S; yl = F), 2-Chlordibenzo-[b,f]-thiepin (IX: X5 = S; yl = Cl) und 2-Bromdibenzo-[b,f]-thiepin (IX: X5 = S; yl = Br) können in der gleichen Weise hergestellt werden, wie in der Veröffentlichung von V. Seidlova et al. oder der Veröffentlichung von J. O. Jilek et al. beschrieben. 2-Fluordibenzo-[b,-tro- pon (IX: X5 = CO; yl = F) und 2-Bromdibenzo-[b,f]-tropon (IX: X5 = CO; yl = Br) können nach dem gleichen Verfahren hergestellt werden, wie in der Veröffentlichung von C.A.
Stone et al. beschrieben. 2-Brom-5H-dibenzo-[b,f]azepin kann nach dem gleichen Verfahren hergestellt werden, wie in der Veröffentlichung von R. S. Varma et al. beschrieben.
Ferner liefert die Methylierung von SH-Dibenzo-[b,f]- azepin nach einem an sich bekannten Verfahren 5-Methyl5H-dibenzo-[b,f]-azepin [IX: X5 = N(CH3); yl = H] vom Schmelzpunkt 130 bis 131 C. In gleicher Weise kann man durch Acylierung von 5H-Dibenzo-[b,I]-azepin mit einem Acetylierungsmittel bzw. einem Carbäthoxylierungsmittel 5 Acetyl-5H-dibenzo-[b,fj-azepin [IX: X5 = N(COCH3); Y1 = H] (Smp. 121 bis 122 C) und 5-Carbäthoxy-5H-dibenzo [b,f]-azepin [IX: X5 = N(COOC2H5); yl = Hj (Smp. 142 bis 144 C) erhalten.
Beispiele von Verbindungen, die durch Alkylierung oder Acylierung von 2-Fluor-5H-dibenzo-[b,f] azepin, 2-Chlor-5H-dibenzo-[b,fl-azepin oder 2-Brom-5H dibenzo4b,-azepin erhältlich sind, sind 2-Fluor-5-(CI¯3)-al- kyl-5H-dibenzo-[b,f]-azepin [IX: X5 = N(C13-Alkyl); yl = F], 2-Fluor-5-(C2-4)-alkanoyl-5H-dibenzo-[b,f]-azepin [IX: X5 = NCO(CI 3-Alkyl); yl = F], 2-Fluor-5-(C2 4)-carb- alkoxy-5H-dibenzo-[b,fl-azepin [IX: X5 = NCOO(C1-3-Al- kyl); Y1 = F], 2-Chlor-5-(C1-3)-alkyl-5H-dibenzo-[b,f]-aze- pin [IX: X5 = N(Cl 3-Alkyl); yl = Cl], 2-Chlor-5-(C2X)-al- kanoyl-5H-dibenzo-[b,f]-azepin fIX:

Die folgenden Beispiele erläutern die Erfindung.
Beispiel 1
Eine Lösung von 1,25 g Lithiumaluminiumhydrid in 20 ml Ather wurde unter Eiskühlung mit einer Lösung von 2,10 g 1-(N,N-Dimethylcarbamoyl)-3-chlor-1,1 a,6,1 Ob-tetra- hydrodibenzo-[b,f]-cyclopropa-[d]-oxepin in 20 ml Tetrahydrofuran versetzt, worauf das resultierende Gemisch 3 Stunden lang bei Raumtemperatur gerührt wurde. Das abgekühlte Reaktionsgemisch wurde mit Wasser versetzt, um überschüssiges Lithiumaluminiumhydrid zu zersetzen. Das Reaktionsgemisch wurde über Natriumsulfat getrocknet und filtriert. Das Filtrat wurde eingedampft und chromatographiert und ergab 1-(N,N-Dimethylaminomethyl)-3-chlor- 1,1 a,6, 1 0b-tetrahydrodibenzo-[b,f]-cyclopropa-[d]-oxepin als ölige Substanz; das Hydrochlorid hatte den Schmelzpunkt 243 bis 244 C.
Beispiel 2
Eine Lösung von 0,45 g Lithiumaluminiumhydrid in 10 ml Äther wurde unter Eiskühlung mit einer Lösung von 0,67 g 1 -(N-Methylcarbamoyl)-6-äthoxycarbonyl- 1,1 a,6, 1 0b-tetrahydrodibenzo-[b,f]-cyclopropa-[d]-azepin in 40 ml Tetrahydrofuran versetzt, worauf das resultierende Gemisch 6 Stunden lang bei Raumtemperatur gerührt und dann 6 Stunden lang zum Rückfluss erhitzt wurde. Das abgekühlte Reaktionsgemisch wurde mit Wasser versetzt, um überschüssiges Lithiumaluminiumhydrid zu zersetzen. Das Reaktionsgemisch wurde über Natriumsulfat getrocknet und filtriert. Das Filtrat wurde eingedampft und chromatographiert und ergab 1 -(N-Methylaminomethy1)-1 ,la,6,10b- tetrahydrodibenzo-[b,f]-cyclopropa-[d]-azepin als kristallines Material vom Schmelzpunkt 104 bis 107 "C.
Beispiel 3
Ein Gemisch aus 0,25 g l-(N,N-Dimethylaminomethyl) 6-hydroxy-1, la,6, 10b-tetrahydrodibenzo-[a,e]-cyclopropa- [c]-cycloheptan in 20 ml Chloroform und 2,7 g Mangandioxid wurde bei 5 C 90 Minuten lang und dann bei Raumtemperatur 4 Stunden lang gerührt. Nach Abfiltrieren von anorganischen Materialien wurde das Filtrat eingedampft und chromatographiert und ergab l-(N,N-Dimethylaminomethyl)- 1,1 a,6,1 Ob-tetrahydrodibenzo-[a,e]-cyclopropa-[c]- cycloheptan-6-on als kristallines Material vom Schmelzpunkt 78 bis 79 C.
Beispiel 4
Eine Lösung von 0,30 g Diisopropylamin in 5 ml Äthanol wurde bei Raumtemperatur mit einer Lösung von 0,35 g 1 -Chlormethyl-6-methyl- 1,1 a,6, 1 0b-tetrahydro- dibenzo-[b,f]-cyclopropa-[d]-azepin in 5 ml Äthanol versetzt, worauf das Gemisch 7 Stunden lang unter Rückfluss gerührt wurde. Das Reaktionsgemisch wurde eingedampft und in eine 10%ige wässrige Lösung von Natriumhydroxid gegossen. Der Chloroformextrakt wurde getrocknet und chromatographiert und ergab l-(N,N-Diisopropylaminomethyl)-6methyl-l,1 a,6,10b-tetrahydrodibenzo-[b,f]-cyclopropa-[d]- azepin als öliges Material. Das Hydrochlorid hatte den Schmelzpunkt 233 bis 235 C.
Beispiel 5
Eine Lösung von 0,25 g l-(N,N-Dimethylaminomethyl)- 1,1a,6,10b-tetrahydrodibenzo-[b,f]-cyclopropa-[d]-azepin in 10 ml Tetrahydrofuran wurde bei Raumtemperatur mit 60 mg Natriumamid versetzt, worauf das resultierende Gemisch 1 Stunde lang zum Rückfluss erhitzt wurde. Nach dem Abkühlen wurde das Gemisch unter Eiskühlung mit einer Lösung von 0,25 g Methyljodid in 1 ml Tetrahydrofuran versetzt, worauf das Gemisch 1 Stunde lang zum Rückfluss erhitzt wurde. Nach dem Abkühlen wurde Wasser zugesetzt, worauf die anorganischen Materialien abfiltriert wurden.

The tetracyclic compounds of formula I are capable of acid addition salts, e.g. B. hydrochlorides, hydrobromides, sulfates, acetates, oxalates, citrates, fumarates, maleates, lactates and tartrates, and quaternary ammonium salts, e.g. B. methochlorides and methoiodides.
The tetracyclic compounds of formula I and their pharmaceutically acceptable salts have pharmacological activities and are useful as medicaments. In general, they affect the functions of the central nervous system, i.e. H. they inhibit the depressive effects on the central nervous system caused by tetrabenazine and reserpine and also potentiate the central effects of norepinephrine. Therefore, they are useful as antidepressants.

example 1
A solution of 1.25 g of lithium aluminum hydride in 20 ml of ether was cooled with ice with a solution of 2.10 g of 1- (N, N-dimethylcarbamoyl) -3-chloro-1.1 a, 6.1 Ob-tetra-hydrodibenzo - [b, f] -cyclopropa- [d] -oxepin in 20 ml of tetrahydrofuran was added and the resulting mixture was stirred for 3 hours at room temperature. Water was added to the cooled reaction mixture to decompose excess lithium aluminum hydride. The reaction mixture was dried over sodium sulfate and filtered. The filtrate was evaporated and chromatographed to give 1- (N, N-dimethylaminomethyl) -3-chloro-1,1 a, 6, 10b-tetrahydrodibenzo- [b, f] -cyclopropa- [d] -oxepine as an oily substance ; the hydrochloride had a melting point of 243 to 244 C.
Example 2
A solution of 0.45 g of lithium aluminum hydride in 10 ml of ether was cooled with ice with a solution of 0.67 g of 1 - (N-methylcarbamoyl) -6-ethoxycarbonyl-1,1 a, 6, 1 0b-tetrahydrodibenzo- [b, f] -cyclopropa- [d] -azepine in 40 ml of tetrahydrofuran was added and the resulting mixture was stirred at room temperature for 6 hours and then heated to reflux for 6 hours. Water was added to the cooled reaction mixture to decompose excess lithium aluminum hydride. The reaction mixture was dried over sodium sulfate and filtered. The filtrate was evaporated and chromatographed and gave 1 - (N-methylaminomethy1) -1, la, 6,10b-tetrahydrodibenzo- [b, f] -cyclopropa- [d] -azepine as crystalline material with melting point 104 to 107 "C.
Example 3
A mixture of 0.25 g l- (N, N-dimethylaminomethyl) 6-hydroxy-1, la, 6, 10b-tetrahydrodibenzo- [a, e] -cyclopropa- [c] -cycloheptane in 20 ml chloroform and 2, 7 g of manganese dioxide was stirred at 5 C for 90 minutes and then at room temperature for 4 hours. Example 4
A solution of 0.30 g of diisopropylamine in 5 ml of ethanol was treated at room temperature with a solution of 0.35 g of 1-chloromethyl-6-methyl-1,1 a, 6, 1 0b-tetrahydro-dibenzo- [b, f] -cyclopropa- [d] -azepine in 5 ml of ethanol was added and the mixture was stirred under reflux for 7 hours. The reaction mixture was evaporated and poured into a 10% aqueous solution of sodium hydroxide. The chloroform extract was dried and chromatographed to give l- (N, N-diisopropylaminomethyl) -6methyl-1, 1 a, 6,10b-tetrahydrodibenzo- [b, f] -cyclopropa- [d] - azepine as an oily material. The hydrochloride had a melting point of 233 to 235 C.
Example 5
A solution of 0.25 g of l- (N, N-dimethylaminomethyl) - 1,1a, 6,10b-tetrahydrodibenzo- [b, f] -cyclopropa- [d] -azepine in 10 ml of tetrahydrofuran was mixed with 60 mg at room temperature Sodium amide was added and the resulting mixture was heated to reflux for 1 hour.
