Description
The invention relates to a process for the preparation of new benzoic acid derivatives of the formula I as defined in claim 1.
The new compounds of formula I are characterized by a blood sugar-lowering effect.
It is e.g. from CH-PS 486 456, it is known that sulfone amide derivatives, such as sulfonylureas, sulfonylsemicarbazides, sulfonamidopyrimidines, and some other sulfonamide heterocycles and sulfonyl urethanes on the non-pretreated test animal have a hypoglycemic effect. Some of these compounds have also found widespread use in human therapy for the treatment of diabetes mellitus. It has now surprisingly been found that substituted benzoic acids, their salts and esters also bring about a reduction in the blood sugar level.
In addition to the ammonium salt, the salts in particular are the alkali metal salts and the alkaline earth metal salts and salts of other metals. In view of the pharmaceutical use of the products, however, the metals are limited to those that are physiologically compatible.

In principle, all alcohols are suitable for a possible esterification of the carboxyl group. The lower monohydric alcohols, such as methanol, ethanol or propanol, and polyhydric alcohols, for. As glycol, or alcohols with other functional groups, such as ethanolamine or glycoether.
For the group X 'in formula I, primarily mononuclear aromatic or heteroaromatic ring systems, in particular the unsubstituted or mono- to polysubstituted phenyl radical, the pyridine radical or the thiophene radical, are suitable. However, polynuclear aromatic or heteroaromatic ring systems are also equally suitable, including those which are partially hydrogenated, such as, for example, benzofuran, dihydrobenzofuran, chroman, chromene or homochroman and quinoline.
Z 'primarily means hydrogen, but substitution of the central phenyl nucleus by one or more substituents is not excluded.
It has been found that those substituents X 'and Z' which have up to 6 carbon atoms show the best activity. Halogen in the above sense is primarily chlorine and bromine, but especially in the case where X 'is a phenyl radical, also fluorine. The corresponding iodine compounds are usually also effective, but their use is less recommended.
A preferred position as a bridge member Y 'is taken by hydrocarbon bridges with 2 carbon atoms, which in turn can be substituted - also by alkyl groups. Accordingly, the group -CH2-CH2- and the group -CH (CH3) -CH2- are particularly preferred. The other straight-chain or branched hydrocarbon bridges with up to 3 carbon atoms are less preferred.
The compounds of formula I are prepared using the method defined in claim 1.
The starting compounds of formula II can be prepared by using an amine of formula III
durch Umsetzung mit einer den Rest X'-CO- abgebenden Verbindung acyliert oder indem man in eine Verbindung der Formel IV by reaction with a compound which donates the radical X'-CO or by converting into a compound of the formula IV

Compounds of the formula IV in which X 'represents a phenyl radical which is substituted by an amino group can be obtained from the corresponding nitro compounds by reduction.
The compounds obtainable according to the invention have a hypoglycemic effect. This can be determined by feeding the compounds as such, their salts or esters in doses of 10 to 400 mg, preferably about 100 mg / kg, to normal-fed rabbits and the blood sugar over a longer period of time according to the known hawthorn method. Jensen or with an au-tonalyzer.
The compounds obtainable according to the invention are preferably used for the production of orally administrable preparations with a blood sugar-lowering activity for the treatment of diabetes mellitus. They can be applied as such or, if appropriate, in the form of their salts or esters or in the presence of substances which lead to salt formation. For example, alkaline agents such as alkali or alkaline earth metal hydroxides, carbonates or bicarbonates can be used for salt formation.
Suitable medical preparations are preferably tablets which, in addition to the process products, contain the customary excipients and auxiliaries, such as talc, starch, lactose, tragacanth or magnesium stearate.
A preparation that contains the compounds described as an active ingredient, for. B. a tablet or a powder with or without additives is suitably placed in a suitably dosed form. The dose to be selected is one that is adapted to the effectiveness of the active ingredient used and the desired effect. The dosage per unit is expediently about 0.1 to 2 g, preferably 0.5 to 1 g, but dosage units above or below it can also be used, which may have to be divided or multiplied before application.
The benzoic acid derivatives obtainable according to the invention can be used both for the treatment of diabetes mellitus alone and in combination with other oral antidiabetic agents. As such, not only blood-sugar-lowering sulfonylureas come into consideration, but also compounds of different chemical structures, such as biguanides, in particular phenylethyl biguanide or dimethyl biguanide.
