REVENDICATIONS
1. Dérivés du pipéridylinodole et leurs sels d'addition avec les acides minéraux ou organiques, répondant à la formule générale

9. Composition pharmaceutique selon la revendication 8, caractérisée en ce que le principe actif est constitute par l'un des composes définis aux revendications 2 ou 3 ou par l'un de leurs sels d'addition avec les acides pharmaceutiquement acceptables.

La présente invention a pour objet de nouveaux dérivés du pipéridylindole et leurs sels d'addition avec les acides minéraux ou organiques, lesdits dérivés répondant à la formule générale:

Dans la formule I et dans ce qui suit, le substituant X peut metre, par exemple, en positions 4, 5 ou 6 de l'indole, le radical alcoxy renfermant de I à 3 atomes de carbone désigne, par exemple, un radical methoxy, éthoxy ou propoxy.
Les sels d'addition avec les acides minéraux ou organiques peuvent metre, par exemple, les sels forms avec les acides chlorhydrique, bromhydrique, iodydrique, nitrique, sulfurique, phosphorique, acétique, formique, benzoïque, maleïque, fumarique, succinique, tartrique, citrique, oxalique, glyoxylique, aspartique, alcanesulfoniques tel l'acide méthanesulfonique, et arylsulfoniques tel l'acide benzènesulfonique.
Parmi les nouveaux dérivés du pipéridylindole, objets de la présente invention, on peut citer notamment ceux répondant à la formule générale I ci-dessus, ainsi que leurs sels d'addition avec les acides minéraux ou organiques, formule I dans laquelle X représente un atome de chlore, lorsque Y et Z représentent un atome d'hydro gene ou forment ensemble une double liaison, ou X represent un atome d'hydrogène ou un radical méthoxy lorsque Y et Z forment ensemble une double liaison.
Parmi ces derniers dérivés, on retient plus particulièrement les dérivés du pipéridylindole répondant à la formule générale I, ainsi que leurs sels d'addition avec les acides minéraux ou organiques, formule I dans laquelle X represent un atome de chlore et Y et Z representent un atome d'hydrogène ou forment ensemble une double liaison.
Parmi les produits préférés de l'invention, on retient tout particulièrement les produits décrits dans les exemples.

Dans des conditions préférées de mise en oeuvre des procédés cidessus décrits, on opere comme suit: a) la reduction du produit de formule III est effectuée au moyen de l'hydrogène en presence d'un catalyseur tel que l'oxyde de platine ou l'hydroxyde de palladium; b) la saponification du produit de formule IV est effectuée au moyen de l'hydroxyde de potassium, au sein d'un alcanol tel que le propanol, au reflux du mélange réactionnel;
c) I'acide fort en presence duquel on fait réagir le produit de formule II' avec le chlorhydrate de 4-pipéridone est constitué par un acide tel que l'acide phosphorique, et la reaction est effectuée à une temperature comprise entre la temperature ambiante et la tempéra- ture d'ébullition du mélange réactionnel.
Les composes de formule I présentent un caractere basique. On peut avantageusement préparer les sels d'addition de ces dérivés en faisant réagir en proportions sensiblement stoechiométriques un acide mineral ou organique avec lesdits dérivés.
Les composes de formule I possèdent de très interessantes propriétés pharmacologiques; ils sont doués notamment de remarquables propriétés antidépressives, antiparkinsonniennes et antiémétiques.
Ces propriétés justifient l'utilisation des nouveaux dérivés du pipéridylindole de formule I et de leurs sels d'addition avec les acides pharmaceutiquement acceptables, à titre de médicaments.
Parmi ces médicaments, on retient notamment ceux qui sont constitués par les nouveaux dérivés du pipéridylindole répondant à la formule I, dans laquelle X représente un atome de chlore, lorsque Y et Z representent un atome d'hydrogène ou forment ensemble une double liaison, ou X represent un atome d'hydrogène ou un radical methoxy lorsque Y et Z forment ensemble une double liaison, ainsi que les sels d'addition avec les acides pharmaceutiquement acceptables desdits dérivés.
Parmi ces derniers, on retient tout particulièrement ceux répon- dant à la formule I, dans laquelle X represent un atome de chlore et
Y et Z representent un atome d'hydrogène ou forment ensemble une double liaison, ainsi que leurs sels d'addition avec les acides pharmaceutiquement acceptables.
Parmi les médicaments préférés, on retient tout particulièrement ceux qui sont constitutes par les produits décrits dans les exemples.
Ces médicaments trouvent par exemple leur emploi dans le traitement des troubles psychiques, des troubles du comportement, des troubles caractériels, dans le traitement des etats akinétiques, et dyskinetiques ainsi que dans le traitement des vomissements et nausées de toutes origines. La dose usuelle, variable selon le produit utilise, le sujet trait et l'affection en cause peut metre, par exemple, de 5 à 500 mg/d, par voie orale chez l'homme.
Les nouveaux dérivés du pipéridylindole de formule I, ainsi que leurs sels d'addition avec les acides pharmaceutiquement acceptables, peuvent metre employs pour préparer des compositions pharmaceutiques renfermant, à titre de principe actif, I'un au moins desdits dérivés ou I'un au moins desdits sels.
Ainsi, à titre de médicaments, les dérivés répondant à la formule I et leurs sels d'addition avec les acides pharmaceutiquement acceptables peuvent metre incorporés dans des compositions pharmaceutiques destinies à la voie digestive ou parentérale. Ces compositions pharmaceutiques peuvent etre, par exemple, solides ou liquides et se presenter sous les formes pharmaceutiques couramment utilises en médicine humaine, comme par exemple les comprimés, simples ou dragéfiés, les gélules, les capsules, les granules, les suppositoires, les preparations injectables; elles sont prepares selon les méthodes usuelles.
Le ou les principes actifs peuvent y être incorporés à des excipients habituellement employs dans ces compositions pharmaceutiques tels que le talc, la gomme arabique, le lactose, I'amidon, le stéarate de magnesium, le beurre de cacao, les véhicules aqueux ou non, les corps gras d'origine animale ou végétale, les dérivés paraffiniques, les glycols, les divers agents amouillants, dispersants ou émulsifiants, les conservateurs.

On donne ci-après des exemples des procédés de préparation.
Exemple 1:
Chlorhydrate du 5-chloro 3-(4-pipéridyl) 1H-indole
Stade A: 3-(I-acétyl 1,4-dihydro 4-pyridyl) 5-chloro 1H-indole
A 120 cm3 de dioxanne et 11,2 cm de chlorure d'acétyle, refroidis par un bain d'eau glacée, on ajoute 27 cm de pyridine redistillée en maintenant la température intérieure entre 8 et 15 C.
On ajoute à la suspension ainsi obtenue 22 g de 5-chloro lH-indole dans 120 cm de dioxanne en maintenant la température entre 10 et 15 C, puis on agite 7h à température ambiante et à l'abri de la lumière.
On verse la suspension obtenue dans 500 cm3 d'eau, agite 5 min et ajoute de nouveau 500 cm3 d'eau.
On filtre, empâte avec40 cm3 d'acétonitrile, filtre, rince à l'acétonitrile et une fois à l'éther.
On obtient 13,5 g de 3-(1-acétyl 1,4-dihydro 4-pyridyl) 5-chloro IH-indole sous forme d'un solide de couleur jaune pâle (F. = 202 C).
Analyse pour C15H13ClN2O = 272,747:

Stade B: 3-(1-acétyl 4-pipéridyl) 5-chloro IH-indole
Dans 420 cm3 d'éthanol, on introduit 8,49 g de 3-(1-acétyl 1,4-dihydro 4-pyridyl) 5-chloro IH-indole et 850 mg d'oxyde de platine et absorbe l'hydrogène jusqu'à saturation, filtre, rince à l'éthanol, évapore à sec.
On obtient 9 g de produit brut que l'on reprend par 10 cm3 d'acétonitrile. Après 20 min d'agitation à température ambiante, on filtre et rince Åa l'acétonitrile. On obtient 6,99 g de 3-(1-acétyl 4-pipéridyl) 5-chloro IH-indole que l'on purifie par chaud et froid dans l'éthanol.
Après séchage, on recueille 4,78 g de 3-(1-acétyl 4-pipéridyl) 5-chloro IH-indole sous forme d'un solide incolore fondant à 201 C.
Analyse pour C15H17ClN2O = 276,779:
Calculi: C 65,1 H 6,19 Cl 12,81 N 10,12%
Trouvé: C 65,2 H 6,3 Cl 12,6 N 10,1 %
Stade C. 5-chloro 3-(4-pipéridyl) IH-indole et son chlorhydrate
On introduit 6,02 g de 3-(1-acétyl 4-pipéridyl) 5-chloro IH-indole et 6 g d'hydroxyde de potassium dans 50 cm de propanol. On porte et maintient au reflux pendant 4 h. Après refroidissement, on verse la solution obtenue dans 500 cm3 d'eau glacée, agite 45 min à température ambiante, filtre rince à l'eau et sèche sous vide à 50 C.
On obtient 5,02 g de 5-chloro 3-(4-pipéridyV 1H-indole(F. = 208 C).
Préparation du chlorhydrate
On met en suspension dans 120 cm3 d'acétate d'éthyle, 5,5 g de 5-chloro 3-(4-pipéridyl) 1H-indole préparés comme indiqué cidessus. On glace, agite et ajoute 10 cm d'acétate d'éthyle saturé d'acide chlorhydrique. On agite 15 min au bain de glace, filtre, rince à l'acétate d'ethyle puis à l'éther. On obtient 5,89 g de chlorhydrate brut que l'on purifie par chaud et froid dans l'éthanol. On rince à l'éthanol, à l'éther et sèche vide à température ambiante.
On recueille 2,86 g de chlorhydrate du 5-chloro 3-(4-pipéridyl)
IH-indole sous forme d'un solide incolore (F. = 260-262 C).
Analyse pour C13H15Cl2N2 = 271,198:

Trouvé: C57,3 H6,0 C125,8 N 10,2 %
Exemple 2:
Succinate neutre de 3-(1,2,3,6-tétrahydro 4-pyridyl) IH-indole
On dissout 10 g d'indole dans 200 cm3 d'acide acétique, chauffe à 95-100 C sous agitation et atmosphere d'azote. On ajoute 50 cm3 d'acide phosphorique aqueux N et 39,3 g de chlorhydrate de 4-pipéridone monohydraté. On chauffe à 100 C pendant I h, laisse refroidir, verse sur de la glace additionnée de 350 cm d'ammoniaque concentrée et extrait à l'acétate d'éthyle.
On lave à l'eau, à l'eau salée, sèche sur sulfate de magnesium et évapore à sec. On obtient 14,7 g de produit brut que l'on empâte sous azote avec 75 cm3 de methanol.
On filtre sous vide, rince au methanol et à l'éther. On obtient 1,42 g de 3-(1,2,3,6-tétrahydro-4-pyridyl) 1H-indole (F. = 185-186 C).
On évapore les liqueurs meres et purifie par chromatographie sur silice le produit brut obtenu en Burnt au mélange chloroforme/ méthanol/triéthylamine (6/3/1). On recueille 4,55 g de produit de Rf égal à 0,15 que l'on empâte dans l'éther. On obtient finalement 4,295 g de 3-(1,2,3,6-tétrahydro 4-pyridyl) IH-indole, soit au total 5,715 g avec le premier jet. On purifie le produit obtenu par chaud et froid dans l'isopropanol et obtient 3,56 g de 3-(1,2,3-tétrahydro 4-pyridyl) IH-indole (F = 190-191 C).
Préparation du succinate neutre
On dissout 3,8 g de 3-(1,2,3-tétrahydro 4-pyridyl) IH-indole dans 200 cm3 de methanol et ajoute 2,26 g d'acide succinique, On recueille le succinate et le redissout au reflux dans le methanol. On filtre à chaud, concentre et laisse cristalliser. Après avoir effectué une seconde purification dans les mimes conditions, on recueille 2,65 g de succinate de 3-(1,2,3-tétrahydro 4-pyridyl) 1H-indole (F. = 238-240 C).
Analyse pour C30H34N4O4 = 514,60:
Calculi: C 70,02 H 6,66 N 10,89%
Trouvé: C69,7 H6,6 N10,9 %
Exemple 3:
Succinate neutre de 5-méthoxy 3-(1,2,3-tétrahydro 4-pyridyl)
IH-indole
On dissout à 100 C 12,6 g de 5-méthoxy IH-indole dans 240 cm3 d'acide acétique, ajoute 44 g de chlorhydrate de 4-pipéridone monohydratée et maintient le chauffage 30 min à 100 C.
Après refroidissement, on verse de l'eau glacée additionnée de 400 cm d'ammoniaque concentrée, extrait à l'acétate d'éthyle et lave la phase organique à l'eau salée. On sèche sur sulfate de magnesium et évapore à sec. On obtient 20 g de produit brut que l'on purifie par chromatographie sur silice en éluant au mélange chloroforme/ méthanol/triéthylamine (7/2/1).
On obtient 5,26 g de 5-méthoxy 3-(1,2,3,6-tétrahydro 4-pyridyl) lH-indole sous forme d'une résine.
Préparation du succinate neutre
On dissout 5,26 g de 5-méthoxy 3-(1,2,3,6-tétrahydro 4-pyridyl) lH-indole dans 100 cm3 de méthanol, ajoute 1,22 g d'acide succinique dissous dans 10 cm de methanol, laisse cristalliser, filtre, rince au méthanol puis à l'éthanol.
On recueille 4,4 g de succinate neutre de 5-méthoxy 3-(1,2,3,6-tétrahydro 4-pyridyl) IH-indole sous forme de cristaux (F. = 255-258 C).
Analyse pour C32H38N4O6 = 574,683:

Exemple 4:
Succinate neutre 5-chloro 3-(1,2,3,6-tétrahydro 4-pyridyl)
IH-indole
On dissout 9,05 g de 5-chloro 1H-indole dans 180 cm3 d'acide acétique, chauffe à 90 C environ et introduit 47,5 g de chlorhydrate de 4-pipéridone monohydratée. On maintient le chauffage à 90-100 C pendant I h, laisse refroidir puis verse sur de l'eau glance additionnée de 300 cm d'ammoniaque concentrée.
On extrait à l'acétate d'éthyle, lave à l'eau salée, sèche sur sulfate de magnesium et évapore à sec. On obtient 12,816 g de produit brut que
I'on reprend avec le melange chloroforme/méthanol/triéthylamine (6/3/1). On filtre sous vide, recueille le filtrat et chromatographie ce dernier sur silice en éluant au mélange chloroforme/méthanol/ triethylamine (6/3/1).
Aprils evaporation de l'éluant, on obtient 5,973 g de 5-chloro 3-(1,2,3,6-tétrahydro 4-pyridyl) 1H-indole sous forme d'une résine jaune.
Préparation du succinate neutre
On dissout 5,973 g de Chloro 3-(1,2,3,6-tétrahydro 4-pyridyl) 1H-indole dans 50 cm de methanol, ajoute 3 g d'acide succinique, laisse cristalliser et glace pendant 30 min. On filtre sous vide, rince au methanol, seche et obtient 5,466 g de succinate neutre de 5-chloro 3-(1,2,3,6-tétrahydro 4-pyridyl) 1H-indole sous forme de cristaux jaunes (F. = 253-254 C).
Analyse pour C30H32Cl2N4O4 = 583,52:
Calculé: C 61,75 H 5,52 Cl 12,15 N 9,60%
Trouvé: C 61,5 H 5,6 Cl 12,2 N 9,4 %
Exemple 5:
Succinate neutre de 4-methoxy 3-(1,2,3,6-tétrahydro 4-pyridyl)
IH-indole
En opérant comme à l'exemple 3, mais au départ du 4-méthoxy lH-indole, on obtient le succinate neutre de 4-méthoxy 3-(1,2,3,6-tétrahydro 4-pyridyl) 1H-indole, sous forme de cristaux (F. = 160 C, puis 194-196"C).

Among these latter derivatives, the derivatives of piperidylindole corresponding to general formula I are more particularly retained, as well as their addition salts with mineral or organic acids, formula I in which X represents a chlorine atom and Y and Z represent a hydrogen atom or together form a double bond.

The compounds of formula I have a basic character. It is advantageous to prepare the addition salts of these derivatives by reacting in substantially stoichiometric proportions a mineral or organic acid with said derivatives.
The compounds of formula I have very interesting pharmacological properties; they are endowed in particular with remarkable antidepressant, antiparkinsonnian and antiemetic properties.
These properties justify the use of the new piperidylindole derivatives of formula I and their addition salts with pharmaceutically acceptable acids, as medicaments.
Among these drugs, we notably retain those which consist of the new piperidylindole derivatives corresponding to formula I, in which X represents a chlorine atom, when Y and Z represent a hydrogen atom or together form a double bond, or X represents a hydrogen atom or a methoxy radical when Y and Z together form a double bond, as well as the addition salts with pharmaceutically acceptable acids of said derivatives.
Among the latter, there are very particularly those corresponding to formula I, in which X represents a chlorine atom and
Y and Z represent a hydrogen atom or together form a double bond, as well as their addition salts with pharmaceutically acceptable acids.
Among the preferred medicaments, there are particularly retained those which consist of the products described in the examples.
These drugs find, for example, their use in the treatment of mental disorders, behavioral disorders, character disorders, in the treatment of akinetic and dyskinetic states as well as in the treatment of vomiting and nausea of all origins. The usual dose, which varies according to the product used, the subject treated and the condition in question, can be, for example, from 5 to 500 mg / d, orally in humans.
The new piperidylindole derivatives of formula I, as well as their addition salts with pharmaceutically acceptable acids, can be used to prepare pharmaceutical compositions containing, as active principle, at least one of said derivatives or one at less of said salts.
Thus, as medicaments, the derivatives corresponding to formula I and their addition salts with pharmaceutically acceptable acids can be incorporated into pharmaceutical compositions intended for the digestive or parenteral route. These pharmaceutical compositions can be, for example, solid or liquid and are presented in the pharmaceutical forms commonly used in human medicine, such as for example tablets, simple or dragéfiés, capsules, capsules, granules, suppositories, injectable preparations ; they are prepared according to the usual methods.
The active ingredient (s) can be incorporated therein into excipients usually used in these pharmaceutical compositions such as talc, gum arabic, lactose, starch, magnesium stearate, cocoa butter, aqueous vehicles or not, fatty substances of animal or vegetable origin, paraffinic derivatives, glycols, various wetting agents, dispersants or emulsifiers, preservatives.

Examples of the preparation methods are given below.
Example 1:
5-chloro 3- (4-piperidyl) 1H-indole hydrochloride
Stage A: 3- (I-acetyl 1,4-dihydro 4-pyridyl) 5-chloro 1H-indole
To 120 cm3 of dioxane and 11.2 cm of acetyl chloride, cooled by an ice-water bath, 27 cm of redistilled pyridine are added while maintaining the internal temperature between 8 and 15 C.
22 g of 5-chloro 1H-indole in 120 cm of dioxane are added to the suspension thus obtained, keeping the temperature between 10 and 15 ° C., then the mixture is stirred for 7 hours at room temperature and protected from light.
The suspension obtained is poured into 500 cm3 of water, stirred for 5 min and again added 500 cm3 of water.
Filter, paste with 40 cm 3 of acetonitrile, filter, rinse with acetonitrile and once with ether.
13.5 g of 3- (1-acetyl 1,4-dihydro 4-pyridyl) 5-chloro IH-indole are obtained in the form of a pale yellow solid (F. = 202 C).

Stage B: 3- (1-acetyl 4-piperidyl) 5-chloro IH-indole
In 420 cm3 of ethanol, 8.49 g of 3- (1-acetyl 1,4-dihydro 4-pyridyl) 5-chloro IH-indole and 850 mg of platinum oxide are introduced and the hydrogen is absorbed up to at saturation, filter, rinse with ethanol, evaporate to dryness.
9 g of crude product are obtained which is taken up in 10 cm 3 of acetonitrile. After 20 min of stirring at room temperature, the mixture is filtered and rinsed with acetonitrile. 6.99 g of 3- (1-acetyl 4-piperidyl) 5-chloro IH-indole are obtained which are purified by hot and cold in ethanol.
After drying, 4.78 g of 3- (1-acetyl 4-piperidyl) 5-chloro IH-indole are collected in the form of a colorless solid melting at 201 C.

Stage C. 5-chloro 3- (4-piperidyl) IH-indole and its hydrochloride
6.02 g of 3- (1-acetyl 4-piperidyl) 5-chloro IH-indole and 6 g of potassium hydroxide in 50 cm of propanol are introduced. It is brought to and maintained at reflux for 4 h. After cooling, the solution obtained is poured into 500 cm3 of ice water, stirred 45 min at room temperature, filter rinsed with water and dried under vacuum at 50 C.
5.02 g of 5-chloro 3- (4-piperidyV 1H-indole (F = 208 C) are obtained.
Preparation of the hydrochloride
5.5 g of 5-chloro 3- (4-piperidyl) 1H-indole, prepared as indicated above, are suspended in 120 cm3 of ethyl acetate. Ice is stirred and add 10 cm of ethyl acetate saturated with hydrochloric acid. The mixture is stirred for 15 min in an ice bath, filter, rinsed with ethyl acetate and then with ether. 5.89 g of crude hydrochloride are obtained which is purified by hot and cold in ethanol. Rinsed with ethanol, ether and vacuum dried at room temperature.
2.86 g of 5-chloro 3- (4-piperidyl) hydrochloride are collected
IH-indole in the form of a colorless solid (F. = 260-262 C).

Example 2:
3- (1,2,3,6-tetrahydro 4-pyridyl) IH-indole neutral succinate
10 g of indole are dissolved in 200 cm3 of acetic acid, heated to 95-100 C with stirring and nitrogen atmosphere. 50 cm3 of aqueous phosphoric acid N and 39.3 g of 4-piperidone hydrochloride monohydrate are added. The mixture is heated at 100 ° C. for 1 hour, allowed to cool, poured onto ice with 350 cm of concentrated ammonia added and extracted with ethyl acetate.
Wash with water, with salt water, dry over magnesium sulphate and evaporate to dryness. 14.7 g of crude product are obtained which is impasted under nitrogen with 75 cm 3 of methanol.
Filtered under vacuum, rinsed with methanol and ether. 1.42 g of 3- (1,2,3,6-tetrahydro-4-pyridyl) 1H-indole are obtained (F. = 185-186 C).
The mother liquors are evaporated and the crude product obtained in Burnt is purified by chromatography on silica using a chloroform / methanol / triethylamine mixture (6/3/1). 4.55 g of Rf product, equal to 0.15, is collected which is pasted in ether. 4.295 g of 3- (1,2,3,6-tetrahydro 4-pyridyl) IH-indole are finally obtained, that is to say 5.715 g with the first spray. The product obtained is purified by hot and cold in isopropanol and 3.56 g of 3- (1,2,3-tetrahydro 4-pyridyl) IH-indole (F = 190-191 C) are obtained.
Preparation of neutral succinate
3.8 g of 3- (1,2,3-tetrahydro 4-pyridyl) IH-indole are dissolved in 200 cm 3 of methanol and 2.26 g of succinic acid are added. The succinate is collected and redissolved at reflux in methanol. Filtered while hot, concentrated and allowed to crystallize. After having carried out a second purification under the same conditions, 2.65 g of 3- (1,2,3-tetrahydro 4-pyridyl) 1H-indole succinate (F. = 238-240 C) are collected.

Example 3:
Neutral 5-methoxy 3- (1,2,3-tetrahydro 4-pyridyl) succinate
IH-indole
12.6 g of 5-methoxy IH-indole are dissolved at 100 ° C. in 240 cm 3 of acetic acid, 44 g of 4-piperidone hydrochloride monohydrate are added and heating is maintained at 100 ° C. for 30 min.
After cooling, ice water is added with 400 cm of concentrated ammonia, extracted with ethyl acetate and the organic phase is washed with salt water. It is dried over magnesium sulphate and evaporated to dryness. 20 g of crude product are obtained, which product is purified by chromatography on silica, eluting with a chloroform / methanol / triethylamine mixture (7/2/1).
5.26 g of 5-methoxy 3- (1,2,3,6-tetrahydro 4-pyridyl) 1H-indole are obtained in the form of a resin.
Preparation of neutral succinate
5.26 g of 5-methoxy 3- (1,2,3,6-tetrahydro 4-pyridyl) 1H-indole are dissolved in 100 cm 3 of methanol, 1.22 g of succinic acid dissolved in 10 cm of methanol are added , leaves to crystallize, filters, rinses with methanol and then with ethanol.
4.4 g of neutral 5-methoxy 3- (1,2,3,6-tetrahydro 4-pyridyl) IH-indole succinate are collected in the form of crystals (F. = 255-258 C).

Example 4:
Neutral succinate 5-chloro 3- (1,2,3,6-tetrahydro 4-pyridyl)
IH-indole
9.05 g of 5H-1H-indole are dissolved in 180 cm3 of acetic acid, the mixture is heated to approximately 90 ° C. and 47.5 g of 4-piperidone hydrochloride monohydrate are introduced. The heating is maintained at 90-100 ° C. for 1 h, allowed to cool and then poured into water sprayed with 300 cm of concentrated ammonia.
It is extracted with ethyl acetate, washed with salt water, dried over magnesium sulphate and evaporated to dryness. 12.816 g of crude product are obtained,
We resume with the chloroform / methanol / triethylamine mixture (6/3/1). Filtered under vacuum, the filtrate is collected and chromatographed on silica, eluting with a chloroform / methanol / triethylamine mixture (6/3/1).
After evaporation of the eluent, 5.973 g of 5-chloro 3- (1,2,3,6-tetrahydro 4-pyridyl) 1H-indole are obtained in the form of a yellow resin.
Preparation of neutral succinate
5.973 g of Chloro 3- (1,2,3,6-tetrahydro 4-pyridyl) 1H-indole are dissolved in 50 cm of methanol, 3 g of succinic acid are added, allowed to crystallize and ice for 30 min. Filtered under vacuum, rinsed with methanol, dried and obtained 5.466 g of neutral succinate of 5-chloro 3- (1,2,3,6-tetrahydro 4-pyridyl) 1H-indole in the form of yellow crystals (F. = 253 -254 C).

By operating as in Example 3, but starting from 4-methoxy 1H-indole, the neutral succinate of 4-methoxy 3- (1,2,3,6-tetrahydro 4-pyridyl) 1H-indole is obtained, under form of crystals (F. = 160 C, then 194-196 "C).
