Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Chinoxalin-di-N-oxid-Derivate für die Bekämpfung pathogener Mikroorganismen und zur Förderung des Wachstums von Tieren sowie die neuen Verbindungen enthaltende Mittel und die Verwendung dieser Verbindungen.

Die Verbindungen der Formel I lassen sich nach bekannten Methoden herstellen (vgl. Deutsche Offenlegungsschrift Nr. 1 670 935).
Die Verbindungen der Formel I werden erfindungsgemäss wie folgt hergestellt:

Die Umsetzung wird in für die Reaktionspartner inerten organischen Lösungsmitteln wie Alkoholen, Acetonitril, Dimethylformamid, Tetrahydrofuran, Dioxan, Benzol, Toluol oder Methylcellosolve (Äthylenglykolmonomethyläther), vorzugsweise in Methanol durchgeführt, wobei es vorteilhaft ist, wenn das eingesetzte Reaktionsmedium in möglichst wasserfreiem Zustand ist.
Die Reaktion erfolgt bei Temperaturen zwischen 0 und 70"C, vorzugsweise bei 30 bis 50"C, und wird in Gegenwart von Basen durchgeführt. Als solche kommen Amine, vorzugsweise primäre Amine sowie Ammoniak in Frage.
Erfindungsgemäss werden Verbindungen der Formel I auch hergestellt, indem man das in der Vorstufe durch Reaktion des Aminonitrils mit dem Diketen als Ausgangsstoff erhaltene Acetessigsäureamid der Formel III in situ mit dem Benzofuroxan der Formel II zu der gexvünschten Verbindung der Formel I umsetzt.
Als weitere mögliche Herstellungsmethode für die Verbindungen der Formel I steht folgende aus der britischen Patentschrift Nr. 1 308 370 bekannte Synthese zur Verfügung:

Die für die erfindungsgemässe Herstellung der Verbindungen der Formel I Verwendung findenden Ausgangsstoffe der Formel III sind teils neu, teils bekannt. Sie können nach an sich bekannten Methoden aus Aminen und Diketen (vgl.
Houben-Weyl 7/4 S. 234 und 8 S. 658) gewonnen werden:

Die Herstellung der Ausgangsverbindungen der Formel III wird bei Temperaturen zwischen - 150 und 30"C, vorzugsweise zwischen - 10" und 0 C durchgeführt. Die Umsetzung erfolgt in inerten organischen Lösungsmitteln. Als solche kommen Alkohole wie Äthanol oder Isopropanol, vorzugsweise Methanol in Betracht. Es ist vorteilhaft, die Reaktion in wasserfreiem Medium durchzuführen.

Den erfindungsgemäss hergestellten Verbindungen der Formel I ähnliche Verbindungen sind aus der Deutschen Offenlegungsschrift Nr. 1 670 935, der Deutschen Offenlegungsschrift Nr. 1 620 114 und der britischen Patentschrift Nr. 1 223 720 bereits bekannt.
Die erfindungsgemäss hergestellten Verbindungen der Formel I sind den bekannten Verbindungen ähnlicher Struktur bei der Bekämpfung pathogener Mikroorganismen weit überlegen und zeichnen sich ihnen gegenüber besonders durch stärkere therapeutische Wirksamkeit aus. Die neuen Wirkstoffe eignen sich insbesondere für den Einsatz im veterinärmedizinischen Bereich. So können sie beispielsweise mit gutem Erfolg gegen durch E. coli hervorgerufene Luftwegerkrankungen beim Geflügel sowie gegen Infektionen des Intestinaltraktes, wie Schweinediarrhoe, und des Urogenitalsystems eingesetzt werden.
Ferner besitzen die Verbindungen der Formel I gute Eigenschaften zur Förderung des Wachstums von Tieren.
In ihren biologischen Eigenschaften ragen durch besondere Aktivität folgende Verbindungen hervor: 1 ,4-Dioxido-3-methyl-chinoxalin(2)-N-(2'-cyanoäthyl)- -carbonsäureamid
1 ,4-Dioxido-3-methyl-chinoxalin(2)-N-(cyanomethyl) -carbonsäureamid
1 ,4-Dioxido-3 -methyl-chinoxalin(2)-N-(1'-cyanoisopro- pyl)-carbonsäureamid
1 ,4-Dioxido-3-methyl-chinoxalin(2)-N-(3 > -cyanopropyl)- -carbonsäureamid
1 ,4-Dioxido-3-methyl-chinoxalin(2)-N-(4'-cyanobutyl)- -carbonsäureamid.
Die Wirkstoffe der Formel I können entsprechend der beabsichtigten Anwendung als solche der oder in Kombination mit inerten Trägerstoffen bzw. Verdünnungsmitteln in Form von Lösungen, Emulsionen, Suspensionen, Pulvern, Tabletten, Bolussen und Kapseln peroral, abomasal oder via Injektion den Tieren direkt und zwar als Einzeldosis wie auch wiederholt verabreicht werden. Die Wirkstoffe bzw.
sie enthaltende Gemische können auch dem Futter oder den Tränken direkt zugesetzt werden oder sind in sogenannten Premixes (Futtervormischungen) enthalten. Die Konzentration der Wirkstoffe bezogen auf das Gesamtfutter bzw.
-tränke beträgt vorzugsweise jeweils 1 bis 500 ppm.
Geeignete Vormischungen bestehen z.B. aus einer Mischung des Wirkstoffs mit Kaolin, Kalk, Aluminiumoxid, gemahlene Muschelschalen, Bolus alba, Aerosol, Stärke oder Lactose. Zur Herstellung einer Futtermischung wird die nötige Menge Vormischung (Premix) mit der entsprechenden Menge eines handelsüblichen Standardfutters gründlich vermischt.
Die überraschenderweise ausgezeichnete therapeutische Wirksamkeit der Verbindungen der Formel I konnte sowohl in vitro als auch insbesondere am Tier bei akuten bakteriel len Infektionen nach oraler sowie subcutaner Applikation festgestellt werden. Das Wirkungsspektrum der Verbindun gen umfasst grampositive und gramnegative Bakterien.
Wie der nachfolgenden Zusammenstellung von Ergebnis sen aus Vergleichsversuchen mit bekannten Verbindungen zu entnehmen ist, lässt sich die Überlegenheit der erfin dungsgemäss hergestellten Verbindungen signifikant nach weisen.
Bei Versuchen an der weissen Maus werden die intra peritoneal infizierten Tiere subcutan bzw. peroral folgender massen behandelt:

Nachweis der wachstums fördern den Wirksamkeit
Eine Gruppe von 8 Jungschweinen (je 4 e und g) wurden mit einem Normalfutter enthaltend 17,6% Rohprotein und 3,4% Rohfaser unter Zusatz von 50 ppm 1,4 Dioxido -3 -methyl-chinoxalin(2)-N-(2'-cyanoäthyl)-carbonsäureamid (Beispiel 1) 28 Tage lang gefüttert. Das durchschnittliche Anfangsgewicht der einzelnen Versuchstiere betrug zu Versuchsbeginn ca. 9 kg. Nach Versuchsende wurden die Tiere einzeln gewogen und der Futterverbrauch gruppenweise bestimmt. Zur Kontrolle wurde ein Parallelversuch gleicher Versuchsanordnung unter Verwendung des gleichen Futters ohne Wirksubstanz durchgeführt.

Beispiel 1 l,4-Diondo-3-methyl-chinoxalin-2-N-(-cyanäthyl)-carbon- säureamid
In eine Lösung von 23,8 g N-P-Cyanäthyl-azetessigsäure- amid in 80 ml Methanol trägt man 19,2 g Benzofuroxan portionsweise ein und leitet dann 2 Stunden über KOH getrockneten Ammoniak ein, wobei man anfangs durch gelegentliches Kühlen, die Temperatur der rasch dunkel gefärbten Lösung unter 450C hält. Nach dem Abklingen der Reaktionswärmebildung hält man die Temperatur der Reaktionsmischung auf 40-450C und rührt nach beendeter Ammoniakeinleitung bei gleicher Temperatur noch 10-12 Stunden nach.
Das Reaktionsprodukt scheidet sich in beigefarbenen Kristallen ab. Man rührt, nutscht ab und wäscht mit kaltem Methanol aus. Das erhaltene, reine kristalline 1,4-Dioxido-3 -methyl-chinoxalin-2-N-(ss-cyanäthyl)-carbonsäureamid schmilzt bei 198-1990C. Es lässt sich aus Dimethylformamid/Äthanol umkristallisieren.
Beispiel 2
1,4-Dioxid o-3-methyl-chinoxalin-2-N-(ss-cyanäthyl)-carbon- säureamid
In eine Lösung von 80 ml Methanol und 9,9 g Aminopropionitril tropft man unter Rühren bei -10 bis 0 C 11,8 g frisch destilliertes Diketen zu, lässt auf Raumtemperatur kommen und rührt weitere 2 Stunden bei 350C. In die nun vorliegende Lösung von N-ss-Cyanäthyl-acetessigsäureamid trägt man 19,2 g Benzofuroxan portionsweise ein und leitet dann 8 Stunden über KOH getrockneten Ammoniak ein, wobei man anfangs durch gelegentliches Kühlen die Temperatur der rasch dunkel gefärbten Lösung unter 450C hält.
Nach dem Abklingen der Reaktionswärmebildung hält man die Temperatur der Reaktionsmischung auf 40-45 C und rührt nach beendeter Ammoniakeinleitung bei gleicher Temperatur noch 10-12 Stunden nach. Das Reaktionsprodukt scheidet sich in beigefarbenen Kristallen ab. Man kühlt, nutscht ab und wäscht mit kaltem Methanol aus. Das erhaltene, reine kristalline 1 ,4-Dioxido-3-methyl-chinoxalin-2-N- -(ss-cyanäthyl)-carbonsäureamid schmilzt bei 198-199 C. Es lässt sich aus Dimethylformamid/Äthanol umkristallisieren.
Beispiel 3
A cetessigsäure-N-cyanmethylamid
In eine Lösung von 400 ml Methanol und 56 g frisch destilliertem Aminoacetonitril tropft man unter Rühren bei - 10 bis 0 C 92 g ebenfalls frisch destilliertes Diketen zu, lässt auf Raumtemperatur kommen und rührt noch 2 Stunden bei 35"C im Wasserbad nach. Am Rotationsverdampfer destilliert man das Lösungsmittel ab und schlemmt den festen Rückstand in kaltem Äther auf. Nutscht man ab, so erhält man das farblose kristalline Acetessigsäure-N-cyanmethylamid in reiner Form mit dem Schmelzpunkt von 63-65 C.
Die folgenden Verbindungen wurden analog dem in den Beispielen 1 und 2 beschriebenen Verfahren hergestellt:

Anschliessend mahlt man zur gewünschten Teilchengrösse von z.B. 5-10 Mikron. Diese erfindungsgemässen Futterzusatzmittel können mit 5800 Gew.-Teilen Standardfutter vermischt oder zu 6000 Gew.-Teilen Fertigtränken verarbeitet werden.
Ausserdem können diese erfindungsgemässen Futterzusatzmittel zu 6000 Gew.-Teilen Standardfutter pelletiert werden (Futterpellets).

The reaction is carried out in organic solvents which are inert to the reactants, such as alcohols, acetonitrile, dimethylformamide, tetrahydrofuran, dioxane, benzene, toluene or methyl cellosolve (ethylene glycol monomethyl ether), preferably in methanol, it being advantageous if the reaction medium used is as anhydrous as possible.
The reaction takes place at temperatures between 0 and 70 "C, preferably at 30 to 50" C, and is carried out in the presence of bases. Such amines, preferably primary amines and ammonia are suitable.
According to the invention, compounds of the formula I are also prepared by reacting the acetoacetic acid amide of the formula III obtained as a starting material in the precursor by reaction of the aminonitrile with the diketene in situ with the benzofuroxane of the formula II to give the desired compound of the formula I.
The following synthesis known from British Patent No. 1 308 370 is available as a further possible production method for the compounds of the formula I:

The starting materials of the formula III which are used for the preparation of the compounds of the formula I according to the invention are partly new and partly known. They can be prepared from amines and diketene (cf.
Houben-Weyl 7/4 p. 234 and 8 p. 658):

The preparation of the starting compounds of formula III is carried out at temperatures between - 150 and 30 "C, preferably between - 10" and 0 C. The reaction takes place in inert organic solvents. Alcohols such as ethanol or isopropanol, preferably methanol, are suitable as such. It is advantageous to carry out the reaction in an anhydrous medium.

The recovery of the benzofuroxane of the formula II is described in Organic Syntheses IV page 74.
Compounds similar to the compounds of formula I prepared according to the invention are already known from German Offenlegungsschrift No. 1,670,935, German Offenlegungsschrift No. 1,620,114 and British Patent No. 1,223,720.
The compounds of the formula I prepared according to the invention are far superior to the known compounds of similar structure in combating pathogenic microorganisms and are distinguished in particular by their greater therapeutic activity. The new active ingredients are particularly suitable for use in the veterinary field. For example, they can be used with good success against airway diseases in poultry caused by E. coli and against infections of the intestinal tract, such as swine diarrhea, and the urogenital system.
Furthermore, the compounds of formula I have good properties for promoting the growth of animals.
The following compounds stand out in their biological properties through special activity: 1,4-dioxido-3-methyl-quinoxaline (2) -N- (2'-cyanoethyl) - -carboxamide

1,4-dioxido-3-methyl-quinoxaline (2) -N- (4'-cyanobutyl) - carboxamide.
Depending on the intended use, the active compounds of the formula I, as such, or in combination with inert carriers or diluents in the form of solutions, emulsions, suspensions, powders, tablets, boluses and capsules, orally, abomasally or via injection directly to the animals, specifically as Single dose as well as administered repeatedly. The active ingredients or
mixtures containing them can also be added directly to the feed or the drinkers or are contained in so-called premixes (feed premixes). The concentration of the active ingredients in relation to the total feed or
potions is preferably 1 to 500 ppm each.
Suitable premixes are e.g. from a mixture of the active ingredient with kaolin, lime, aluminum oxide, ground mussel shells, bolus alba, aerosol, starch or lactose. To produce a feed mix, the necessary amount of premix is thoroughly mixed with the corresponding amount of a standard commercial feed.
The surprisingly excellent therapeutic activity of the compounds of the formula I was found both in vitro and, in particular, in animals in acute bacterial infections after oral and subcutaneous application. The spectrum of activity of the compounds includes gram-positive and gram-negative bacteria.
As can be seen in the following compilation of results from comparative experiments with known compounds, the superiority of the compounds produced according to the invention can be demonstrated significantly.
In experiments on the white mouse, the intra-peritoneally infected animals were treated subcutaneously or orally as follows:

Proof of growth will promote effectiveness
A group of 8 young pigs (4 e and g each) were fed a normal feed containing 17.6% crude protein and 3.4% crude fiber with the addition of 50 ppm 1.4 dioxido -3-methyl-quinoxaline (2) -N- ( 2'-cyanoethyl) carboxamide (Example 1) fed for 28 days. The average initial weight of the individual test animals was approximately 9 kg at the start of the test. After the end of the experiment, the animals were weighed individually and the feed consumption was determined in groups. As a control, a parallel test of the same test arrangement was carried out using the same feed without active substance.

Example 1 1, 4-Diondo-3-methyl-quinoxaline-2-N - (- cyanoethyl) -carbon- acid amide
In a solution of 23.8 g of NP-cyanoethyl-acetoacetic acid amide in 80 ml of methanol, 19.2 g of benzofuroxane are added in portions and then ammonia dried over KOH is passed in for 2 hours, initially by cooling occasionally, the temperature of the rapidly keeps dark colored solution below 450C. After the reaction heat has subsided, the temperature of the reaction mixture is kept at 40-450C and, after the introduction of ammonia has ended, the mixture is stirred at the same temperature for a further 10-12 hours.
The reaction product separates out in beige crystals. The mixture is stirred, suction filtered and washed with cold methanol. The pure crystalline 1,4-dioxido-3-methyl-quinoxalin-2-N- (ss-cyanoethyl) -carboxamide obtained melts at 198-1990C. It can be recrystallized from dimethylformamide / ethanol.
Example 2
1,4-Dioxide o-3-methyl-quinoxaline-2-N- (ss-cyanoethyl) -carbon- acid amide
11.8 g of freshly distilled diketene are added dropwise to a solution of 80 ml of methanol and 9.9 g of aminopropionitrile with stirring at -10 to 0 ° C., the mixture is allowed to come to room temperature and stirred at 350 ° C. for a further 2 hours. In the solution of N-ss-cyanoethyl acetoacetic acid amide present, 19.2 g of benzofuroxane are added in portions and then ammonia dried over KOH is passed in for 8 hours, the temperature of the rapidly dark colored solution initially being kept below 450 ° C. by occasional cooling.
After the heat of reaction has subsided, the temperature of the reaction mixture is kept at 40-45 ° C. and stirring is continued for 10-12 hours at the same temperature after the introduction of ammonia has ended. The reaction product separates out in beige crystals. It is cooled, filtered off and washed with cold methanol. The pure crystalline 1,4-dioxido-3-methyl-quinoxaline-2-N- (ss-cyanoethyl) carboxamide obtained melts at 198-199 C. It can be recrystallized from dimethylformamide / ethanol.
Example 3
Acetoacetic acid-N-cyanomethylamide
92 g of likewise freshly distilled diketene are added dropwise to a solution of 400 ml of methanol and 56 g of freshly distilled aminoacetonitrile with stirring at -10 ° C. to 0 ° C., the mixture is allowed to come to room temperature and stirred for a further 2 hours at 35 ° C. in a water bath. On a rotary evaporator The solvent is distilled off and the solid residue is suspended in cold ether. If the residue is filtered off, the colorless crystalline acetoacetic acid-N-cyanomethylamide is obtained in pure form with a melting point of 63-65 C.

The compound of formula I is either admixed directly to the excipients or in suitable solvents, e.g. in chloroform, dissolved on the carriers.
