Die erfindungsgemässen Umsetzungen werden zweckmässig in äquimolaren Mengen der jeweiligen Ausgangsstoffe durchgeführt. Bei flüchtigen Reaktionspartnern empfiehlt sich jedoch die Anwendung eines Überschusses. Die Umsetzungen werden vorteilhaft in einem Lösungs- oder Verteilungsmittel durchgeführt, jedoch sind bestimmte Reaktionen auch ohne Lösungs- oder Verteilungsmittel durchführbar, wie nachstehend ausgeführt wird.
Als Lösungs- oder Verteilungsmittel kommen beispielsweise in Frage:
Bei Verfahren a) aromatische. gegebenenfalls halogenierte Kohlenwasserstoffe, wie Benzol, Toluol, Xylol, Chlorbenzol, Dichlorbenzol, Trichlorbenzol, chlorierte, aliphatische Kohlenwasserstoffe, wie Methylenchlorid, Chloroform, aliphatische Äther. wie Di-isopropyläther, Äthylenglycoldi methyläther, -diiithyliither. Di.ithylenglycol-dimethyläther, Tetrahydrofuran Dioxan.
Besonders vorteilhaft ist es die zur Umsetzung verwendeten Carbonsäureamide oder Lactame der Formel 111 im iibcrsrhuss zu verwenden. Der Überschuss kann gegebenenfalls bei der Aufarbeitung des Reaktionsansatzes wiedergewonnen werden.
Bei Verfahren b) Alkohole, wie Methanol, Äthanol, Propanol. Butanol, Methoxyäthanol, Äthoxyiithanol oder besonders vorteilhaft die zur Umsetzung verwendeten Acetale der Carbonsiiureamide oder der Lactame der Formel 1V im Überschuss.
Die Umsetzungen nach Verfahren a) werden vorteilhafterweise in Gegenwart eines Kondensationsmittels durchgeführt. Als Kondensationsmittel kommen vorzugsweise anorganis he und organische Säurehalogenide in Frage, beispielsweise Thionylchlorid. Phosphortrichlorid, Phosphor pentachlorid, Phosphoroxychlorid, Chlorsulfonsäure, Phosgen, Oxalylchlorid, Chlorameisensiiurealkylester, Benzoylchlorid, Benzolsulfosäurechlorid. 4-Toluolsulfosäurechlorid.
Werden bei der Umsetzung nach Verfahren a) Carbon s:iurethioamide bzw. Thiolactame eingesetzt, empfiehlt sich die Mitverwendung eines schwefelbindenden Mittels. Als schwefelbindende Mittel kommen beispielsweise Schwerme talloxide. wie Quecksilberoxid und Bleioxid in Frage.
Die Reaktionskomponenten nach Verfahren a) werden zweckmiissig in iiquimolaren Mengen zur Umsetzung ge bracht. Die drei letztgenannten Komponenten ,insbesonde- re die Carbons':iureamide und Thioamide, Lactame und Thiolactame können mit Vorteil auch im Überschuss angewendet werden.
Die Reaktionstemperaturen liegen bei beiden Verfahrensvarianten im allgemeinen zwischen 0 und l()0 C, vorzugsweise zwischen 25 und 80"C.
Die Reaktionszeiten betragen je nach Verfahrensvariante und je nach Temperaturbereich wenige Minuten bis zu einigen Stunden.
Die Reaktionsprodukte nach Verfahren a) fallen in Form ihrer Salze an. Sie können als solche isoliert werden, oder gegebenenfalls durch Alkalischstellen der wässrigen Lösungen in die freien Basen übergeführt werden.
Zum Alkalischstellen benutzt man üblicherweise starke Basen, wie Ammoniak, Soda, Pottasche, Ätznatron, Ätzkali oder ihre wässrigen Lösungen. Die freigesetzten Basen können wiederum mit physiologisch verträglichen Säuren in Salze überführt werden.
Als physiologisch verträgliche Säuren kommen beispielsweise in Frage Halogenwasserstoffsäuren, insbesondere Salzsäure, ferner Schwefelsäure, Phosphorsäure, Essigsäure, Propionsäure, Milchsäure, Weinsäure.
Erforderlichenfalls kann eine Reinigung der Verfahrenserzeugnisse durch Umkristallisation aus einem geeigneten Lösungsmittel oder Lösungsmittelgemisch erfolgen.
Die neuen Verbindungen der Formel I sind gut verträglich und eignen sich zur Bekämpfung von Protozoenerkrankungen beim Mensch und Tier, wie sie z.B. durch Infektionen mit Trichomonas vaginalis und Entamoeba histolytica hervorgerufen werden. Die neuen Verbindungen der Formel I wirken hier bereits in geringeren Dosen als Metronidazol. Ausserdem wirken sie gegen Ilelminthen und gegen Ektoparasiten wie Zecken.
Die neuen Verbindungen können oral oder lokal angewendet werden. Die orale Anwendung kann in pharmazeutisch üblichen Zubereitungen, z.B. in Form von Tabletten oder Kapseln. die pro Tagesdosis etwa 10 bis 750 mg des Wirkstoffes in Mischung mit einem gebräuchlichen Trägerstoff und/oder Konstituens enthalten, erfolgen. Für die lokale Anwendung können z.B. Gelees, Crems, Salben oder Suppositorien verwendet werden.

Beispiel I 1.1) l-Methyl-2-(4-dimethylamino-methylenimino-phenyl- -oxymethyl)-5-nitro-imidazol
248 g (1 Mol) l-Methyl-2-(4-aminophenyl-oxymethyl)-5- -nitro-imidazol werden in 1250 ml Dimethylformamid gelöst. Zu dieser Mischung werden unter Rühren bei 30-40"C 154 g (1 Mol) Phosphoroxichlorid zugetropft. Danach wird noch 2 Stunden bei 25"C weiter gerührt. Das Endprodukt wird als Salz durch Zugabe von 2,5 1 Methylenchlorid unter Rühren ausgefällt, abgesaugt, mit Methylenchlorid gewaschen, trockengesaugt. in Wasser gelöst, mit wässrigen, konz.
Ammoniak alkalisch gemacht und die freie Base mit Methylenchlorid mehrfach ausgeschüttelt. Die vereinigten Extrakte werden mit Natriumsulfat getrocknet, eingedampft und der Rückstand aus Alkohol unter Kohlenzusatz umkristallisiert. Man erhiilt so 252 g (83 ^'s der Theorie) l-Me thyl-2-(4-dimethylamino-methylenimino-phenyl.oxymethyl) -5-nitro-imidazol in Form von gelben Kristallen mit einem Fp. 1450C.
Aus der freien Base kann auf übliche Weise mit molaren Mengen alkoholischer Salzsiiure das Hydrochlorid mit Fp. 187"C in Form von schwach gclblichen Kristallen dargestellt werden.
Das als Ausgangsstoff verwendete l-Mcthyl-2-(4-amino- phenyloxymethyl)-5-nitro-imidazol (orangerote Kristalle, Fp. 152"C) kann durch Verseifung von l-Methyl-2-(4-acet- aminophenyl-oxymethyl)-5-nitro-imidazol mittels 40% der Schwefelsäure (2 Stunden, 80-90"C) in 85% Ausbeute dargestellt werden.
Das zur Darstellung des Ausgangsstoffes verwendete 1 -Methyl-2-(4-acetaminophenyl-oxymethyl)-5-nitro-imidazol (blassgelbe Kristalle, Fp. 163"C) kann durch Umsetzung von molaren Mengen 4-Acetaminophenol mit 1-Methyl-2 -chlormethyl-5-nitro-imidazol in Dimethylformamid (1 Stunde 30-40"C) in Gegenwart von Kaliumcarbonat in 95 % Ausbeute dargestellt werden.
Die Darstellung von 1-Methyl-2-chlormethyl-5-nitro- -imidazol ist in DOS 1 595 929 beschrieben; sie erfolgt durch Umsetzen der l-Methyl-2-hydroxymethylverbindung mit Thionylchlorid. An Stelle von l-Methyl-2-chlormethyl-5- -nitro-imidazol kann auch l-Methyl-2-benzoyloxymethyl-5- -nitro-imidazol oder l-Methyl-2-(4-nitrobenzoyloxymethyl)-5- -nitro-imidazol verwendet werden, die aus der l-Methyl-2- -hydroxymethylverbindung mit Benzoylchlorid bzw. 4-Nitrobenzoylchlorid erhalten werden.

24.8 g (0.1 mol) of 1-methyl-2- (4-aminophenyloxymethyl) -5-nitro-imidazole are suspended in 250 ml of pyridine, 25 g (excess) of dimethylformamide diethyl acetal are added and the reaction mixture is 3 hours heated under reflux. The reaction medium pyridine is then distilled off under reduced pressure, the residue is recrystallized from alcohol with the addition of coal. This gives 24.0 g (79% of theory) of 1-methyl-2- (4-dimethylamino-methyleniminophenyloxymethyl) -5-nitro-imidazole, mp 145 C.
