Abstract
The compounds of the formula I <IMAGE> in which the substituents have the meaning given in Patent Claim 1, and their acid addition salts, have a long-lasting blocking effect on adrenergic beta -receptors. They can be used as medicines. They are obtained by acylation.

2. Verfahren nach Anspruch 1 zur Herstellung der Verbindungen der allgemeinen Formel 1, in denen Rl Phenyl bedeutet, und
R2 und R die in Anspruch 1 angegebene Bedeutung besitzen, dadurch gekennzeichnet, dass man Verbindungen der Formel II, in denen R2 und R3 die in Anspruch 1 angegebene Bedeutung besitzen, entsprechend acyliert.
3. Verfahren nach Anspruch 1 zur Herstellung von 4-(2 -Benzoyloxy-3-tert-butylamino-propoxy)-2-methylindol, da durch gekennzeichnet, dass man I-tert-Butylamino3 l-tert-Butylamino-3-(2-me- thylindol-4-yloxy)-2-propanol entsprechend acyliert.
4. Verfahren nach Anspruch 1 zur Herstellung der Verbindungen der Formel I in (S)-optisch aktiver Form, dadurch gekennzeichnet, dass man die Verbindungen der Formel II in (S)-optisch aktiver Form acyliert.
5. Verfahren nach Anspruch 1 zur Herstellung von der Verbindung des Anspruches 3 in (S)-optisch aktiver Form, dadurch gekennzeichnet, dass man das l-tert-Butylamino-3- -(2-methylindol-4-yloxy)-2-propanol in (S)-optisch aktiver Form acyliert.

Rl bedeutet vorzugsweise unsubstituiertes Phenyl. Falls Rl eine vorstehend definierte Phenoxyalkylgruppe bedeutet, stellt es vorzugsweise 1-(p-Chlorphenoxy)-1-methyläthyl dar.
Die Verbindungen der Formel I besitzen in ss-Stellung zum Stickstoffatom in der Seitenkette ein asymmetrisches Kohlenstoffatom und können daher in Form von optisch aktiven Verbindungen oder Racematen vorliegen. Bevorzugt sind Enantiomere mit (S)-Konfiguration an diesem Kohlenstoffatom.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel

worin R2 und R, obige Bedeutung besitzen, entsprechend acyliert und die so erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
Die Acylierung der Verbindungen der Formel II kann nach an sich zur Acylierung analoger sekundärer Alkohole bekannten Methoden, beispielsweise durch Umsetzung mit Säureanhydriden oder Halogeniden von Säuren der Formel RlCOOH, worin Rl obige Bedeutung besitzt, vorzugsweise deren Anhydriden erfolgen. Falls als Acylierungsmittel Säureanhydride verwendet werden, kann die Umsetzung beispielsweise bei Temperaturen zwischen ca. 0 u. 100 , vorzugsweise unter Verwendung eines Überschusses an Säureanhydrid erfolgen.
Es kann sich als zweckmässig erweisen, die Aminogruppe vorher zu protonisieren, beispielsweise durch Zusatz einer Säure, insbesondere der Säure RlCOOH, oder indem man die Verbindungen der Formel II in Form von Säureadditionssalzen, beispielsweise Hydrochloriden einsetzt. Bei Verwendung von Säurehalogeniden wird die Acylierung vorzugsweise bei Raumtemperatur oder leicht erhöhter Temperatur durchgeführt.
Bei der Acylierung bleibt die Konfiguration an dem asymmetrischen Kohlenstoffatom unverändert, so dass aus racemischen Ausgangsverbindungen racemische Endprodukte und aus optisch aktiven Ausgangsverbindungen optisch aktive Endprodukte erhalten werden.
Die erfindungsgemäss erhaltenen Verbindungen der Formel I können in Form der freien Basen oder ihrer Säure
additionssalze vorliegen. Die freien Basen können auf an sich bekannte Weise in ihre Säureadditionssalze überführt werden und umgekehrt. So können die erfindungsgemässen Verbindungen der Formel I z.B. mit anorganischen Säuren wie Chlorwasserstoff oder organischen Säuren wie Fumarsäure Säureadditionssalze bilden.
Die Ausgangsverbindungen sind bekannt oder können nach an sich bekannten Verfahren bzw. analog zu an sich bekannten Verfahren hergestellt werden.
Die Verbindungen der Formel I und ihre physiologisch verträglichen Säureadditionssalze zeichnen sich durch interessante pharmakologische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Insbesondere besitzen die Substanzen eine länger anhaltende und stärker ausgeprägte Blockerwirkung auf die adrenergischen ss-Rezeptoren als für Verbindungen dieses Strukturtyps erwartet werden kann, wie sich in Standard-Tests, beispielsweise durch Hemmung der positiv-inotropen Adrenalinwirkung am spontan schlagenden isolierten Meerschweinchenvorhof bei Badkonzentrationen um etwa 0,005 bis etwa 2,5 mg/kg und durch Hemmung der durch Isoproterenol bedingten Tachycardie und Blutdrucksenkung beim Infusionsversuch an der narkotisierten Katze bei Dosen von etwa 0,02 bis etwa 1 mg/kg zeigt. Aufgrund ihrer ss-Rezeptoren blockierenden Wirkung sind die Substanzen u.a. nützlich bei der Phophylaxe und Therapie von Coronarerkrankungen, insbesondere zur Behandlung von Angina pectoris, sowie der essentiellen Hypertonie.
Die Verbindungen weisen auch antiarrhythmische Wirkungen auf. Aufgrund dieser Wirkungen eignen sie sich auch zur Behandlung von Herzrhythmusstörungen.
Für obige Anwendungen beträgt die Tagesdosis etwa 1 bis etwa 200 mg, vorzugsweise etwa 1 bis 5 mg. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für orale Applikationen enthalten die Teildosen ca. 0,25 bis ca. l 100 mg der neuen Verbindungen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
Als Heilmittel können die Verbindungen der Formel I unh ihre physiologisch verträglichen Säureadditionssalze zusammen mit geeigneten pharmazeutischen Hilfsstoffen in zur enteralen, vorzugsweise oralen, oder parenteralen Applikation geeigneten festen oder flüssigen galenischen Zubereitungen, gewünschtenfalls mit vorzögerter Wirkstoffabgabe enthalten sein. Beispiele solcher Zubereitungen zur enteralen Applikation sind u.a. Pulver, Granulate, Tabletten, Kapseln, Dragees, Sirupe, Tropfen usw. Als Beispiele von Zubereitungen zur parenteralen Applikation seien Injektionslösungen oder injizierbare wässrige oder ölige Suspensionen genannt.
Die Herstellung der galenischen Zubereitungen kann nach an sich in der pharmazeutischen Technik üblichen Verfahren unter Verwendung üblicher fester oder flüssiger Träger- und Hilfsstoffe erfolgen.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern ,erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel I
4-62-Bel1zoyloxy-3-terf-blttylamillopropoxy)-2-methylil?d
26 g Benzoesäure werden in 50 ml Hexamethylphosphorsäuretriamid warm gelöst und mit 3,5 g 1-tert-Butylamino-3-(2-methylindol-4-yloxy)-2-propanol versetzt. Nach Abkühlen gibt man 3,0 g Benzoesäureanhydrid zu und rührt während 20 Stunden bei Raumtemperatur. Die klare gelbe Lösung wird auf Eis gegossen, 0,5 Liter Äther zugefügt und während 2 Stunden gerührt. Nach Alkalischstellen mit konzentriertem Ammoniak wird die Ätherphase abgetrennt, mit Weinsäure ausgeschüttelt, unter Eiskühlung mit Sodalösung alkalisch gestellt und mit Methylenchlorid extrahiert. Nach Abdampfen des Lösungsmittels wird der Rückstand mit 1 Mol Fumarsäure aus Methanol und Aceton kristallisiert (Smp. des Hydrogenfumarats der Titelverbindung 189-1910).
Analog zu Beispiel 1 erhält man durch Acylierung der entsprechenden Verbindungen der Formel II folgende Verbindungen der Formel 1:

3. The method according to claim 1 for the preparation of 4- (2-benzoyloxy-3-tert-butylamino-propoxy) -2-methylindole, characterized in that I-tert-butylamino3 l-tert-butylamino-3- (2nd -me- thylindol-4-yloxy) -2-propanol acylated accordingly.
4. The method according to claim 1 for the preparation of the compounds of formula I in (S) -optically active form, characterized in that the compounds of formula II in (S) -optically active form.
5. The method according to claim 1 for the preparation of the compound of claim 3 in (S) -optically active form, characterized in that the l-tert-butylamino-3- - (2-methylindol-4-yloxy) -2- propanol acylated in (S) -optically active form.
The invention relates to a process for the preparation of new tert-butylaminopropoxy indoles of the formula 1,

Rl is preferably unsubstituted phenyl. If R 1 is a phenoxyalkyl group as defined above, it is preferably 1- (p-chlorophenoxy) -1-methylethyl.
The compounds of the formula I have an asymmetric carbon atom in the ss-position to the nitrogen atom in the side chain and can therefore be in the form of optically active compounds or racemates. Enantiomers with (S) configuration on this carbon atom are preferred.
According to the invention, the new compounds of the formula I and their acid addition salts are obtained by using compounds of the formula

wherein R2 and R, as defined above, are acylated accordingly and, if desired, the compounds of the formula I thus obtained are converted into their acid addition salts.
The acylation of the compounds of the formula II can be carried out by methods known per se for the acylation of secondary alcohols, for example by reaction with acid anhydrides or halides of acids of the formula RlCOOH, in which Rl has the above meaning, preferably their anhydrides. If acid anhydrides are used as the acylating agent, the reaction can be carried out, for example, at temperatures between about 0 and. 100, preferably using an excess of acid anhydride.
It may prove expedient to protonize the amino group beforehand, for example by adding an acid, in particular the acid RlCOOH, or by using the compounds of the formula II in the form of acid addition salts, for example hydrochlorides. When using acid halides, the acylation is preferably carried out at room temperature or slightly elevated temperature.
During the acylation, the configuration on the asymmetric carbon atom remains unchanged, so that racemic end products are obtained from racemic starting compounds and optically active end products are obtained from optically active starting compounds.
The compounds of the formula I obtained according to the invention can be in the form of the free bases or their acid
addition salts are present. The free bases can be converted into their acid addition salts in a manner known per se and vice versa. Thus the compounds of formula I according to the invention can e.g. form acid addition salts with inorganic acids such as hydrogen chloride or organic acids such as fumaric acid.
The starting compounds are known or can be prepared by processes known per se or analogously to processes known per se.
The compounds of the formula I and their physiologically tolerated acid addition salts are distinguished by interesting pharmacological properties and can therefore be used as medicines.
In particular, the substances have a longer-lasting and more pronounced blocker effect on the adrenergic ss receptors than can be expected for compounds of this structure type, as can be seen in standard tests, for example by inhibiting the positive inotropic adrenaline effect on the spontaneously beating isolated guinea pig atrium at bath concentrations by about 0.005 to about 2.5 mg / kg and by inhibiting isoproterenol-related tachycardia and lowering blood pressure when trying to infuse the anesthetized cat at doses of about 0.02 to about 1 mg / kg. Due to their ss-receptor blocking effect, the substances are among others useful in the prevention and therapy of coronary diseases, especially for the treatment of angina pectoris, as well as essential hypertension.
The compounds also have antiarrhythmic effects. Because of these effects, they are also suitable for the treatment of cardiac arrhythmias.
For the above applications, the daily dose is about 1 to about 200 mg, preferably about 1 to 5 mg. If necessary, this dose can be administered in 2 to 4 portions or as a slow-release form. For oral applications, the partial doses contain approx. 0.25 to approx. 100 mg of the new compounds in addition to solid or liquid carriers or diluents.
The compounds of the formula I and their physiologically tolerable acid addition salts, together with suitable pharmaceutical auxiliaries, can be present as medicinal products in solid or liquid pharmaceutical preparations suitable for enteral, preferably oral or parenteral administration, if desired with a delayed release of active ingredient. Examples of such preparations for enteral application include Powders, granules, tablets, capsules, dragees, syrups, drops, etc. Injection solutions or injectable aqueous or oily suspensions may be mentioned as examples of preparations for parenteral administration.
The galenical preparations can be prepared by conventional methods in pharmaceutical technology using conventional solid or liquid carriers and auxiliaries.
In the following examples, which explain the invention in more detail, all temperatures are given in degrees Celsius.
Example I

26 g of benzoic acid are dissolved warm in 50 ml of hexamethylphosphoric triamide and 3.5 g of 1-tert-butylamino-3- (2-methylindol-4-yloxy) -2-propanol are added. After cooling, 3.0 g of benzoic anhydride are added and the mixture is stirred at room temperature for 20 hours. The clear yellow solution is poured onto ice, 0.5 liters of ether are added and the mixture is stirred for 2 hours. After making alkaline with concentrated ammonia, the ether phase is separated off, shaken out with tartaric acid, made alkaline with ice cooling with sodium carbonate solution and extracted with methylene chloride. After evaporation of the solvent, the residue is crystallized from methanol and acetone with 1 mol of fumaric acid (mp. Of the hydrogen fumarate of the title compound 189-1910).
