Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Hydroxyamine, die wirksame ss-Rezeptor blokkierende Verbindungen darstellen. Diese Verbindungen blokkieren bei Verabreichung an Säugetiere, insbesondere Menschen, die ss-Rezeptoren des Herzens und sind daher wertvolle Heilmittel zur Behandlung der Symptome und Merkmale von cardiovaskulären Erkrankungen.

Keine der genannten meta-substituierten Verbindungen zeigt jedoch irgendwelche a-Rezeptoraktivität. Die ortho-substituierten Verbindungen der genannten britischen Patentschrift zeigen Nebenaktivitäten in Kombination mit ihrerssl Rezeptorblockierungsselektivität, die nicht gewünscht sind.
Die neuen erfindungsgemäss erhältlichen Verbindungen haben wertvolle pharmakologische Eigenschaften, so blockie

B. die cardialenss-Rezeptoren, was durch die Bestimmung des Antagonismus der Tachycardie nach intravenöser Injektion von 0,5,zm/kg von d/l-Isoproterenolsulfat an einer anästhesierten Katze nach intravenöser Injektion von 0,002 bis 2 mg/kg gezeigt werden kann. Die erfindungsgemäss erhältlichen Verbindungen blockieren auch die vaskulären p- Rezeptoren, was durch Bestimmung des Antagonismus der
Vasodilatation nach intravenöser Injektion von 0,5,ug/kg von d/l-lsoproterenolsulfat an einer anästhesierten Katze durch intravenöse Verabreichung von 0,002 bis 2 mg/kg oder mehr gezeigt wird. Die erfindungsgemäss erhältlichen Verbindungen haben ausserdem stimulierende Eigenschaften auf ss-Rezepto ren, d. h. sie zeigen eine spezifische Wirksamkeit (intrinsic ac tivity).
Diese Eigenschaft ist hinsichtlich der vaskulären ss-Rezeptoren, die Dilatation der peripheren Blutgefässe bewirken, besonders ausgeprägt'
Die neuen erfindungsgemäss erhältlichen Verbindungen können zur Behandlung von Arrhythmien, Angina pectoris und Hypertonie verwendet werden. Die periphere Vasodilata tion ist besonders wertvoll für die beiden zuletzt erwähnten
Indikationen. Man kann die Verbindungen ausserdem als Zwi schenprodukte zur Herstellung anderer wertvoller Verbindun gen mit pharmakologischen Eigenschaften verwenden.

Salzbildende Säuren können zur Herstellung von therapeutisch zulässigen Salzen der erfindungsgemäss erhältlichen Ver bindungen verwendet werden, beispielsweise Halogenwasserstoffsäuren, Schwefel-, Phosphor-, Salpeter- oder Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, beispielsweise Ameisen-, Essig-, Propion-, Bernstein-, Glykol-, Milch-, Äpfel-, Wein-, Citronen-, Ascorbin-, Malein-, Hydroxymalein-, Brenztrauben-, Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl-, p-Aminosalicyl-, Embon-, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon-, Äthylensulfon-, Halogenbenzolsulfon-, Toluolsulfon-, Naphthylsulfon- oder Sulfanilsäure, Methionin, Tryptophan, Lysin oder Arginin.
Die erfindungsgemäss erhältlichen Verbindungen können oral oder parenteral für akute und chronische Behandlung der oben erwähnten cardiovaskulären Erkrankungen verwendet werden. Die biologischen Wirkungen der neuen, erfindungsgemäss erhältlichen Verbindungen wurden geprüft, und die verschiedenen, mit den genannten Verbindungen ausge führten Tests sind weiter unten angegeben und erläutert.
Diese Umsetzung kann in üblicher Weise ausgeführt werden, vorzugsweise in Gegenwart eines basischen Kondensationsmittels und/oder eines Überschusses an Amin. Geeignete basische Kondensationsmittel sind z. B. Alkalialkoholate, vorzugsweise Natrium- oder Kalumalkoholat, oder auch Kalicarbonate, wie Natrium- oder Kaliumcarbonat.
Je nach den Verfahrensbedingungen und den verwendeten Ausgangsmaterialien wird das Endprodukt entweder in freier Form oder in Form seines Säureadditionssalzes erhalten, was in den Schutzumfang der Erfindung eingeschlossen ist. So können beispielsweise basische, neutrale oder gemischte Salze erhalten werden sowie Hemiamino, Sesqui- oder Polyhydrate.
Die Säureadditionssalze der neuen Verbindungen können in an sich bekannter Weise in die freien Verbindungen übergeführt werden, wobei basische Mittel, wie Alkalien, oder Ionenaustauscher verwendet werden. Anderseits können die erhaltenen freien Basen mit organischen oder anorganischen Säuren Salze bilden. Bei der Herstellung von Säureadditionssalzen werden vorzugsweise solche Säuren verwendet, die geeignete therapeutisch zulässige Salze bilden.
Solche Säuren sind beispielsweise Halogenwasserstoffsäuren, Schwefel-, Phosphor-, Salpeter- oder Perchlorsäure, aliphatische, alicyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie beispielsweise Ameisen-, Essig-, Propion-, Bemstein-, Glykol-, Milch-, Äpfel-, Wein-, Citronen-, Ascorbin-, Malein-, Hydroxymalein-, Bernstein-, Phenylessig-, Benzoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxybenzoe-, Salicyl-, p-Aminosalicyl- oder Embonsäure, Methansulfon-, Äthansulfon-, Hydroxyäthansulfon- oder Äthylensulfonsäuren, Halogenbenzolsulfon-, Toluolsulfon- oder Naphthylsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin oder Arginin.
Diese oder andere Salze der neuen Verbindungen, beispielsweise die Picrate, können dazu dienen, die erhaltenen freien Basen zu reinigen, indem die Basen in Salze übergeführt werden, indem diese abgetrennt werden und indem aus den Salzen die Basen wieder in Freiheit gesetzt werden. Wegen der engen Beziehung zwischen den neuen Verbindungen in freier Form und in Form ihrer Salze versteht es sich aus dem Voranstehenden und dem Folgenden, dass - falls möglich - auch die entsprechenden Salze verwendet werden können, wenn die freien Verbindungen genannt werden.
Je nach der Wahl der Ausgangsmaterialien und dem Verfahren können die neuen Verbindungen als optische Antipoden oder Racemate vorliegen oder, falls sie mindestens zwei asymmetrische Kohlenstoffatome enthalten, können sie in Form eines isomeren Gemisches (Racematgemisch) vorhanden sein.
Die erhaltenen isomeren Gemische (Racematmischungen) können je nach physikalisch-chemischen Unterschieden der Komponenten in die beiden stereoisomeren (diastereomeren) reinen Racemate getrennt werden, beispielsweise mittels Chromatographie und/oder durch fraktionierte Kristallisation.
Die erhaltenen Racemate können nach an sich bekannten Methoden aufgetrennt werden, z. B. durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mittels Mikroorganismen oder durch Umsetzung mit optisch aktiven Säuren, wobei Salze der Verbindungen gebildet werden, die dann beispielsweise aufgrund ihrer unterschiedlichen Löslichkeit in die diastereomeren Formen getrennt werden, aus denen die Antipoden dann durch geeignete Mittel in Freiheit gesetzt werden.
Geeignete optisch aktive Säuren sind beispielsweise die L- und D-Formen der Weinsäure, Di-o-tolylweinsäure, Äpfelsäure, Mandelsäure, Kampfersulfonsäure oder Chinasäure. Vorzugsweise wird der aktivere Teil der beiden Antipoden isoliert.
Bei der klinischen Anwendung können die erfindungsgemäss erhältlichen Verbindungen normalerweise oral, rektal oder durch injektion in Form eines pharmazeutischen Präparats, das eine wirksame Komponente entweder als freie Base oder als pharmazeutisch zulässiges, nicht-toxisches Säureadditionssalz, z. B. das Hydrochlorid, Lactat, Acetat, Sulfamat oder dergleichen, in Kombination mit einem pharmazeutischen Trägerstoff enthält, verabreicht werden.
Die Erwähnung der neuen erfindungsgemäss erhältlichen Verbindungen bezieht sich entweder auf die freie Aminbase oder auf die Säureadditionssalze der freien Base, auch wenn die Verbindungen allgemein oder speziell beschrieben sind, vorausgesetzt, dass der Zusammenhang, in dem solche Ausdrücke gebraucht werden, z. B. in den Beispielen, mit der breiten Bedeutung nicht übereinstimmt. Der Trägerstoff kann ein festes, halbfestes oder flüssiges Verdünnungsmittel oder eine Kapsel sein. Üblicherweise liegt die Menge an wirksamer Verbindung zwischen 0,1 bis 95 Gew. % des Präparats, vorzugsweise zwischen 0,5 und 20 Gew. % bei Präparaten zur Injektion und zwischen 2 und 50 Gew. % bei Präparaten für orale Verabreichung.
Bei der Herstellung von pharmazeutischen Präparaten für orale Verabreichung kann die erfindungsgemäss erhältliche Verbindung mit einem festen, pulverförmigen Trägerstoff vermischt werden, beispielsweise mit Lactose, Saccharose, Sorbit, Mannit, Stärke, beispielsweise Kartoffelstärke, Maisstärke oder Amylopectin, Cellulosederivaten oder Gelatine, oder auch zusammen mit einem Gleitmittel, wie Magnesiumstearat, Calciumstearat, Polyäthylenglykolwachsen oder dergleichen; die Masse kann dann zu Tabletten verpresst werden.
Falls Dragees hergestellt werden sollen, können die wie oben hergestellten Kerne mit konzentrierten Lösungen von Zucker überzogen werden. Diese Lösungen können auch beispielsweise Gummi arabicum, Gelatine, Talk, Titandioxyd oder dergleichen enthalten. Darüber hinaus können die Dragees mit einem in einem leicht flüchtigen organischen Lösungsmittel oder Gemisch von Lösungsmitteln gelösten Lack überzogen werden. Diesen Überzügen kann ein Farbstoff zugesetzt werden, um die leichte Unterscheidung zwischen Tabletten mit verschiedenen wirksamen Verbindungen oder mit unterschiedlichen Mengen an wirksamer Verbindung zu ermöglichen.
Bei der Herstellung von Weichgelatinekapseln (perlförmige, geschlossene Kapseln), die aus Gelatine und beispielsweise Glycerin bestehen, oder bei der Herstellung von ähnlichen geschlossenen Kapseln kann die wirksame Verbindung mit einem pflanzlichen Ö1 vermischt werden. Hartgelatinekapseln können Körnchen der wirksamen Verbindung in Kombination mit einem festen, pulverförmigen Trägerstoff enthalten, bei spielsweise mit Lactose, Saccharose, Sorbit, Mannit, Stärke (z. B. Kartoffelstärke, Maisstärke oder Amylopectin), Cellulo sederivaten oder Gelatine.
Einheitsdosierungsformen für rektale Verabreichung können in Form von Suppositorien hergestellt werden, die die wirksame Substanz im Gemisch mit einer neutralen Fettbasis enthalten, oder sie können in Form von Gelatinerektalkapseln hergestellt werden, die die wirksame Substanz im Gemisch mit einem pflanzlichen Öl oder Paraffinöl enthalten.
Flüssige Präparate für orale Verabreichung können in Form von Sirupi oder Suspensionen vorliegen, z. B. Lösungen, die etwa 0,2 bis etwa 20 Gew. % an wirksamer Substanz enthalten, wobei der Rest aus Zucker und einem Gemisch von Äthanol, Wasser, Glycerin und Propylenglykol besteht. Falls gewünscht, können solche flüssigen Präparate Farbstoffe, Aromastoffe, Saccharin und/oder Carboxymethylcellulose als Verdickungsmittel enthalten
Lösungen für parenterale Verabreichung durch Injektion können in Form einer wässrigen Lösung eines wasserlöslichen pharmazeutisch zulässigen Salzes der wirksamen Verbindung, vorzugsweise in einer Konzentration von etwa 0,5 bis 10,0 Gew. %, hergestellt werden. Diese Lösungen können auch Stabilisierungsmittel und/oder Puffersubstanzen enthalten und können in Ampullen mit verschiedener Dosierung erhalten werden.
Im folgenden wird an Hand eines Beispiels eine bevorzugte Ausführungsform des erfindungsgemässen Verfahrens näher erläutert. Die Temperaturangaben erfolgen in " C.
Beispiel
10 g 1,2-Epoxy-3-o-allylphenoxypropan wurden in 100 ml Äthanol gelöst. Die Lösung wurde mit gasförmigem Ammoniak gesättigt. Diese Reaktionsmischung wurde nun in einem Autoklav über siedendem Wasser 4 Stunden lang erwärmt.
Anschliessend wurde daraus das Lösungsmittel abgedampft und der Rückstand in Äthylacetat aufgenommen. In diese Lösung wurde gasförmiges HC1 eingeführt. Dadurch fiel das Hydrochlorid des 1-Amino-3-o-allylphenoxy-propanol-2 aus. Der Feststoff wurde abfiltriert und in 60 ml Äthanol aufgelöst. Zu dieser Lösung wurden nun 20 mg 2-(4-Hydroxyphenyl)-1,1dimethyläthyliodid und 15 g KzCO3 gegeben. In einem Autoklav wurde diese Reaktionslösung 2 Stunden lang auf 1200 C gehalten. Anschliessend wurde das Lösungsmittel abfiltriert.
Zum Rückstand wurde Wasser gegeben und die Lösung wurde mittels Ammoniak alkalisch gemacht. Die wässrige Phase wurde nun mittels Äthylacetat extrahiert, Die Äthylacetatphase wurde über Magnesiumsulfat getrocknet und dann durch Einleiten von gasförmigem HC1 das Hydrochlorid des Produktes ausgefällt.
Die erfindungsgemäss erhältlichen, ss-Rezeptor blockieren den Mittel wurden hinsichtlich ihrer biologischen Eigenschaf ten geprüft. Sämtliche Verbindungen wurden an anästhetisier ten Katzen (männlich und weiblich mit einem Gewicht von 2,5 bis 3,5 kg), die etwa 16 Stunden vor den Versuchen mit Re serpin (5 mg/kg Körpergewicht, intramuskulär verabreicht) vorbehandelt worden waren, getestet. Die Tiere wurden mit Reserpin vorbehandelt, um die endogene sympathetische Steuerung des Herzschlags und den vaskulären glatten Muskeltonus zu eliminieren. Die Katzen wurden mit Pentobarbital (30 mg/kg Körpergewicht, intraperitoneal verabreicht) anästhetisiert und mit Zimmerluft künstlich beatmet. Es wurde eine bilaterale Vagotomie im Nacken durchgeführt.
Der Blutdruck wurde mit einer Kanüle in der Carotisarterie gemessen und der Herzschlag wurde mit einem Cardiotachometer, ausgelöst durch das Elektrocardiogramm, registriert (ECG). Die eigent liche beta-mimetische Wirksamkeit auf das Herz wurde durch einen erhöhten Herzschlag nach der Verabreichung des Mittels angezeigt. Die Testverbindungen wurden intravenös in loga rithmisch erhöhten Dosen verabreicht. Die erhaltenen Werte wurden als Dosis-Wirkung-Kurven aufgezeichnet, aus denen die Wirksamkeitswerte (EDso) (affinity values) bestimmt wur den. Am Ende jeden Versuchs wurden hohe Dosen an Isopre nalin gegeben, um den Höchstwert der Herzschlagwirkung zu erhalten.
Die Verbindungen wurden auch an Hunden, die bei Bewusstsein waren, getestet. Beagle's wurden dressiert, ruhig zu liegen und sich 2 Minuten lang durch Auflegen der Vorderpfo ten auf einen Tisch in eine aufrechte Stellung zu heben. Der arterielle Blutdruck wurde über einen Blutdruckübermittler, der in Höhe des Herzens befestigt war, registriert. Der Herzschlag wurde von den EKG ermittelt. Alle Hunde wurden mit Methylscopolamin vorbehandelt, um Vaguseinflüsse auszuschalten. Die Aufzeichnungen erfolgten vor und 15 und 75 Minuten nach der Verabreichung der Testverbindung, zuerst, indem die Hunde zwei Minuten auf dem Rücken lagen und zwei Minuten lang in aufrechter Stellung. Die Testverbindungen wurden in zweistündigen Intervallen in steigenden Dosen verabreicht.
Die Versuche beweisen, dass die untersuchten Verbindungen wirksame ss-Rezeptor-Antagonisten sind, mit oder ohne wirklichess-mimetische Wirksamkeit. Die Verbindungen senken ausserdem den Blutdruck bei Hunden, die bei Bewusstsein sind, deutlich mehr als Propranolol und Metoprolol. Die ausgesprochene blutdrucksenkende Wirkung der erfindungsgemäss erhältlichen Verbindungen an Hunden, die bei Bewusstsein sind, beruht auf einer vasodilatatorischen Wirkung in Kombination mit Herz-ss-Rezeptor-Blockierung.
