EMI2.1

in which R1 is hydrogen, halogen (preferably chlorine), a lower alkyl or alkoxy group or a hydroxyl group, R2 and R3 are identical or different and represent lower alkyl groups with at least 3 C atoms together, R4 is hydrogen, an organic group or a cation a pharmaceutically acceptable base and Z is either an oxygen or a sulfur atom, with the limitation, however, that when R4 is an organic group meaning lower alkyl, Z must be a sulfur atom.
The process according to the invention for the preparation of these compounds consists in that a diphenylmethane derivative of the formula
EMI1.5
condenses with a compound of formula
EMI1.6
where X and X 'are two under condensation conditions, with the exception of a group which is split off in an oxygen or sulfur atom present in the same, and the optionally free carboxyl group is converted into a salt if desired. This procedure is easiest if one starts from a 4'-hydroxy- or 4-mercapto-diphenylmethane which is appropriately substituted in the 4'-position and this with an a-halogen fatty acid ester, most suitably an a-bromo or also a - Chloro fatty acid ester.
Similar phenoxycarboxylic acid derivatives, including those with a p-chlorobenzyl substituent in the p-position of the phenyl group, are known, for example, from GB-PS 860 303 and
DE-OS 23 56 655 known; it is also known that such
Compounds are able to lower the cholesterol and lipid content of the blood. It has now been shown that diphenylmethane derivatives obtainable according to the invention, in particular the methyl, ethyl and pyridylmethyl esters and the salts with pharmacologically acceptable metal ions (e.g. with alkali or alkaline earth metals, aluminum and the like) surprisingly increase such properties Have mass.
Example 1 2-Methyl-2- [4- (4'-chlorobenzyl) phenyl mercaptol butyric acid (Sgd 37874)
EMI2.1
3.1 g of sodium hydride (60% suspension in oil) are suspended in 60 ml of dimethylformamide (DMF). A solution of 11.8 g of 4- (4'-chlorobenzyl) thiophenol in 12 ml of DMF is then slowly added dropwise, with stirring, as the mixture warms. After continued stirring for one hour, a solution of 15.7 g of 2-bromo-2-methylbutyric acid ethyl ester in 16 ml of DMF is added dropwise and the mixture is stirred at 70 ° C. for a further 2 hours. The solvent is then distilled off under reduced pressure. The residue is treated with 1N NaOlI and the insoluble ester is extracted with ether.
Evaporation of the ethereal solution gives a crude yield of 17.7 g (97.5%). The 2-methyl-2 [4- (4'-chlorobenzyl) phenylmercapto) 1-butyric acid ethyl ester thus obtained as an intermediate is boiled under reflux for 1 hour in the uncleaned state for saponification with 25 ml of methanolic KOH solution (25%) . Then the solvent is distilled off and the residue is dissolved in water and treated with activated carbon. After filtration, it is acidified with dilute hydrochloric acid and extracted with ether. The evaporated residue gives 13.7 g of crude product (80%), from which the pure acid with mp. 88-89 C is obtained after recrystallization from gasoline.
