Tous les composés préparés conformément au procédé selon l'invention possèdent au moins l'une des propriétés pharmacologiques suivantes: anti-inflammatoire, antiulcéreuse, analgésique, antipyrétique, antihistaminique et dépresseur du système nerveux central (SNC), ainsi qu'une faible toxicité, et plusieurs d'entre eux possèdent plusieurs de ces propriétés. C'est pourquoi il est à remarquer que certains des composés entrant dans le cadre de la présente invention sont intéressants comme nouveaux agents analgésiques, anti-inflammatoires et comme dépresseurs du SNC.

On peut faire réagir les substances de départ représentées par la formule générale (1) avec ces réactifs de formule générale (2), (4) ou (5).
Ces réactions ont lieu de préférence dans un solvant organique, par exemple toluène, xylène, benzène, tétrahydrofuranne, dioxanne, diméthylformamide, diglyme, chloroforme ou alcool.
Les réactions illustrées par les schémas I et II s'effectuent de préférence en présence d'un composé métallique, en particulier amidure de sodium ou de potassium, hydrure ou éthylate de sodium, d'une base organique, notamment pyridine ou trialkylamine, ou d'une base minérale, en particulier hydroxyde ou carbonate alcalin.
Les composés métalliques mentionnés en premier sont les plus efficaces pour l'obtention des composés de l'invention avec les rendements les plus élevés. La température souhaitable n'est pas critique et peut être la température ambiante ou une température plus élevée.
Dans le cas du schéma réactionnel III, on peut opérer en présence d'un solvant organique, notamment chloroforme, benzène, toluène ou xylène, en tube scellé à une température comprise entre 60 et 200" C, ou à reflux.
Dans le cas des schémas I à III, le mélange réactionnel peut être concentré sous pression réduite et le résidu obtenu peut être purifié par recristallisation à partir d'un solvant organique, en particulier éther, éther de pétrole, chloroforme ou méthanol, ou bien par chromatographie sur colonne, pour donner des cristaux purs des produits cherchés.
Les composés de départ de formule générale (1) peuvent être préparés par réaction de l'urée avec un dérivé d'anilino-2 hydroxyméthyl-3 pyridine.
Les composés obtenus peuvent être transformés en leurs sels d'addition avec différents acides organiques ou minéraux. On peut ainsi obtenir, par exemple, des chlorhydrates, sulfates, phosphates, acétates, benzoates, lactates, succinates, citrates, tartrates, fumarates, malonates et maléates. Ces sels, dont la formation sert à améliorer la solubilité et la stabilité des composés selon l'invention, entrent naturellement dans le cadre de celle-ci.

Pour de nombreux composés préparés conformément à l'in-
vention, on a étudié la toxicité aiguë, afin de s'assurer de leur
innocuité,
ainsi que leurs effets analgésique, anti-inflammatoire, et
de dépression du SNC, pour montrer leurs excellentes activités à ces
égards. Les résultats de chaque examen sont rassemblés dans le
tableau 2. Chaque essai est conduit de la manière suivante.
1. Toxicité aiguë
Le composé à essayer, mis en suspension dans une solution saline de gomme adragante à 0,5%, est administré par voie intrapéritonéale ou orale à des souris mâles de souche dd (16-24 g). La dose létale est déterminée à partir du nombre d'animaux morts 72 h après l'administration.
2. Activité anti-inflammatoire
On administre oralement à un groupe de 5 rats mâles de souche
Wistar (100-150 g) chaque composé essayé, mis en suspension dans une solution saline de gomme adragante à 0,5%. Au bout de 30 mn, on injecte dans la patte arrière, par voie sous-cutanée, une suspen sion dans l'eau à 0,5 à 1% de carraghénine. Au bout de 3 h, on mesure le volume de l'oedème provoqué par la carraghénine, et l'on détermine le pourcentage d'inhibition par comparaison avec les résultats obtenus chez les animaux témoins. Ces pourcentages sont représentés par les notations suivantes:

Activité analgésique
Chaque composé essayé, mis en suspension dans une solution saline de gomme adragante à 0,5%, est administré oralement à des souris mâles de souche dd (18-20 g). Au bout de 1 h, on injecte par voie intrapéritonéale une solution d'acide acétique à 0s6% à raison de 0,1 ml/lOg. Le syndrome de crispation est observé pendant 10 mn au bout de 30 mn après l'injection: on calcule par la méthode de
Litchfield-Wilcoxon la dose analgésique active à 50% (DA50) et ses limites de fiabilité à 95%.
4. Activité dépressive sur le SNC
Chaque composé essayé, mis en suspension dans une solution saline de gomme adragante à 0,5%, est injecté par voie intrapéritonéale à des souris mâles de souche dd (16-24 g). La disparition du réflexe de sursaut est étudiée en milieu insonorisé. La dose nécessaire pour obtenir cette disparition est indiquée par les notations suivantes:

La présente invention est illustrée par les exemples suivants.
Exemple I
On ajoute à une solution de 3 g de (m-bromophényl)-l oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine dans 100 ml de diméthylformamide sec, 0,6 g d'hydrure de sodium à 50%, et l'on agite le mélange pendant 30 mn. On additionne ensuite le tout de 3,5 g de bromure de propargyle et on laisse réagir pendant 1 h à la température ambiante. Lorsque la réaction est achevée, on chasse le solvant du mélange réactionnel par distillation sous pression réduite. Le résidu ainsi obtenu est dilué avec de l'eau pour libérer un produit huileux, que l'on extrait ensuite à l'éther.
La solution éthérée est déshydratée sur du sulfate de magnésium, et séparation et purification sont réalisées par passage de cette solution sur une colonne d'alumine. L'éluat ainsi obtenu donne par concentration 2,9 g de (m-bromophényl)-l propargyle-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, sous forme de prismes incolores, fondant à 118-119 C.

Exemple 2
A une solution de 0,7 g de (m-trifluorométhylphényl)-l oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine dans 10 ml de diméthylformamide, est ajouté 0,13 g d'hydrure de sodium. On additionne d'un mélange de 5,1 g d'iodure de méthyle et 5 ml de diméthylformamide, goutte à goutte sous refroidissement, et le mélange résultant est immédiatement chauffé pendant 1 V2 h dans un bain d'huile maintenu à 100" C. Lorsque la réaction est terminée, le solvant est chassé par distillation sous pression réduite, abandonnant un résidu qui est additionné d'eau glacée. On extrait à l'éther, lave et déshydrate.
La séparation et la purification sont réalisées par passage de la solution sur une colonne d'alumine, et l'éluat obtenu, par concentration, donne 0,6 g de (m-trifluorométhylphényl)- 1 méthyl-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, sous forme de prismes incolores, fondant à 122-124" C.

Exemple 3
1,2 g d'hydrure de sodium à 50% est ajouté à une solution de 4,5 g de phényle oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine dans 50 ml de diméthylformamide sec, le mélange est agité pendant 30 mn à température ambiante, puis chauffé à une température de 90-100" C. On l'additionne graduellement de 1,28 g de p-toluènesulfonate d'isopropyle et on laisse réagir le tout pendant
1 h. Lorsque la réaction est achevée, le solvant est éliminé du mélange par distillation sous pression réduite. Le résidu obtenu est dilué avec de l'eau pour donner un précipité brut qui est recueilli par filtration et ensuite dissous dans l'éther éthylique.
La séparation et la purification sont réalisées par passage de la solution sur une colonne d'alumine, et L'élut par concentration donne 4,1 g de phényl-l isopropyl-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]pyrimidine, sous forme de prismes incolores fondant à 164-165" C.

Trouvé: C 71,75 H 6,41 N 15,72.
Exemple 4
Un mélange de 0,5 g de (m-trifluorométhylphényl)-1 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, 0,8 g de carbonate de diméthyle et 10 ml de diméthylformamide est mis à réagir à 160-170"C en tube scellé pendant 20 h. Lorsque la réaction est terminée, le solvant est chassé du mélange par distillation pour laisser un résidu que l'on additionne d'eau. Ce résidu est extrait à l'éther, puis déshydraté. La solution éthérée est envoyée sur une colonne d'alumine, et ensuite éluée à l'éther. Après évaporation de l'éther, l'éluat donne 0,4 g de (m-trifluorométhylphényl)-1 méthyl-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, sous forme de prismes incolores, fondant à 122-124" C.

Exemple 5
A une solution de 2,4 g de (p-fluorophényl)-1 oxo-2 tétrahydro1,2,3,4 pyrido-[2,3d]-pyrimidine dans 15 ml de diméthylformamide, on ajoute 0,5 g d'hydrure de sodium à 50% environ, et on laisse reposer le mélange jusqu'à ce que cesse le dégagement d'hydrogène.
On y ajoute alors 5,9 g d'oxalate de diméthyle, et on fait chauffer ce mélange à 170"C en tube scellé pendant 12 h. Lorsque la réaction est achevée, on élimine le solvant du mélange réactionnel par distillation sous pression réduite. Le résidu ainsi obtenu est dilué avec de l'eau, extrait avec de l'éther et déshydraté. Cette solution éthérée par concentration abandonne 1,6 g de (p-fiuorophényl)-1 méthyl-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, sous forme d'aiguilles incolores, fondant à 212-213"C.

Exemple 6
On ajoute 0,6 g d'hydrure de sodium à une solution de 2,6 g de (m-chlorophényl)-1 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimi- dine dans 20 ml de diméthylformamide, et l'on agite ce mélange pendant 15 mn. Ensuite, on l'additionne de 3,4 g de fluorosulfate de méthyle, et l'on continue l'agitation à température ambiante pendant 1 V2 h. Lorsque la réaction est terminée, le solvant est chassé du mélange par distillation sous pression réduite. Le résidu ainsi obtenu est dilué avec de l'eau, extrait à l'éther et déshydraté, la solution éthérée est placée sur une colonne d'alumine, puis éluée à l'éther.
Par évaporation de l'éther de l'élut on obtient le résidu qui, par recristallisation à partir d'un mélange d'éther et d'éther de pétrole, fournit 2,1 g de (m-chlorophényl)-t méthyl-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, sous forme de prismes incolores, fondant à 139-140" C.

Exemple 7
A une solution de 2,2 g de phényl-1 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine dans 20 ml de diméthylformamide, on ajoute 0,5 g d'hydrure de sodium à 50% environ, et on laisse ce mélange reposer à température ambiante pendant 30 mn. Puis on y ajoute goutte à goutte 4,2 g de phosphate de triméthyle et on chauffe le tout en tube scellé pendant 7h à 170-180" C. Lorsque la réaction est achevée, le solvant est éliminé du mélange par distillation sous pression réduite. Le résidu ainsi obtenu est dilué avec de l'eau, extrait à éther et déshydraté.
L'éther est éliminé par évaporation et le résidu est recristallisé à partir d'un mélange d'éther et d'éther de pétrole pour donner 1,7 g de phényl-1 méthyl-3 oxo-2 tétra hydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, sous forme de prismes incolores, fondant à 172-173" C.

Exemple 8
Un mélange de 2,9 g de (m-trifluorométhylphényl)-l oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]-pyrimidine, 25 ml de diméthylformamide et 4,4 g d'acétal diéthylique de diméthylformamide, est mis à réagir à 145-150" C pendant 5 h. Lorsque la réaction est achevée, le solvant est chassé du mélange par distillation sous pression réduite. Le résidu ainsi obtenu est dilué avec de l'eau, extrait à l'éther et déshydraté. La solution éthérée est placée sur une colonne d'alumine et ensuite éluée à l'éther.
L'éluat est concentré par évaporation et le résidu obtenu est alors recristallisé à partir d'un mélange d'éther et d'éther de pétrole et donne 0,6 g de (m-trifluorométhylphényl)-l éthyl-3 oxo-2 tétrahydro-1,2,3,4 pyrido-[2,3d]- pyrimidine, sous forme de prismes incolores, fondant à 155-157" C.

For many compounds prepared in accordance with the
vention, the acute toxicity was studied to ensure their
harmlessness,
as well as their analgesic, anti-inflammatory, and
CNS depression, to show their excellent activities to these
regards. The results of each examination are compiled in the
Table 2. Each test is carried out as follows.
1. Acute toxicity
The test compound, suspended in 0.5% tragacanth saline, is administered intraperitoneally or orally to male dd strain mice (16-24 g). The lethal dose is determined from the number of animals that died 72 hours after administration.
2. Anti-inflammatory activity
A group of 5 strain male rats was orally administered
Wistar (100-150 g) each test compound, suspended in 0.5% tragacanth saline. After 30 min, a suspension in water containing 0.5 to 1% carrageenan is injected subcutaneously into the hind paw. After 3 hours, the volume of the edema caused by carrageenan is measured, and the percentage inhibition is determined by comparison with the results obtained in the control animals. These percentages are represented by the following notations:

Analgesic activity
Each test compound, suspended in 0.5% tragacanth saline, was administered orally to male dd strain mice (18-20 g). After 1 h, a solution of 0s6% acetic acid is injected intraperitoneally at a rate of 0.1 ml / 10 g. The crispation syndrome is observed for 10 minutes after 30 minutes after the injection: it is calculated by the method of
Litchfield-Wilcoxon the 50% active analgesic dose (DA50) and its 95% reliability limits.
4. CNS depressive activity
Each test compound, suspended in 0.5% tragacanth saline, is injected intraperitoneally into male mice of the dd strain (16-24 g). The disappearance of the startle reflex is studied in a soundproof environment. The dose necessary to obtain this disappearance is indicated by the following notations:

The present invention is illustrated by the following examples.
Example I
To a solution of 3 g of (m-bromophenyl) -1 oxo-2 tetrahydro-1,2,3,4 pyrido- [2,3d] -pyrimidine in 100 ml of dry dimethylformamide is added, 0.6 g of 50% sodium hydride, and the mixture is stirred for 30 min. 3.5 g of propargyl bromide are then added and the mixture is left to react for 1 hour at room temperature. When the reaction is complete, the solvent is removed from the reaction mixture by distillation under reduced pressure. The residue thus obtained is diluted with water to release an oily product, which is then extracted with ether.
The ethereal solution is dried over magnesium sulphate, and separation and purification are carried out by passing this solution through an alumina column. The eluate thus obtained gives, by concentration, 2.9 g of (m-bromophenyl) -1 propargyl-3 oxo-2 tetrahydro-1,2,3,4 pyrido- [2,3d] -pyrimidine, in the form of colorless prisms. , melting at 118-119 C.

Example 2
To a solution of 0.7 g of (m-trifluoromethylphenyl) -l oxo-2 tetrahydro-1,2,3,4 pyrido- [2,3d] -pyrimidine in 10 ml of dimethylformamide, is added 0.13 g of sodium hydride. A mixture of 5.1 g of methyl iodide and 5 ml of dimethylformamide is added dropwise with cooling, and the resulting mixture is immediately heated for 1.2 h in an oil bath maintained at 100 ° C. When the reaction is complete, the solvent is distilled off under reduced pressure, leaving a residue which is added with ice-water, extracted with ether, washed and dried.

Example 3
1.2 g of 50% sodium hydride is added to a solution of 4.5 g of phenyl oxo-2 tetrahydro-1,2,3,4 pyrido- [2,3d] -pyrimidine in 50 ml of dimethylformamide dry, the mixture is stirred for 30 min at room temperature, then heated to a temperature of 90-100 ° C. To this is gradually added 1.28 g of isopropyl p-toluenesulphonate and the whole is left to react for
1 hr. When the reaction is complete, the solvent is removed from the mixture by distillation under reduced pressure. The residue obtained is diluted with water to give a crude precipitate which is collected by filtration and then dissolved in ethyl ether.
The separation and purification are carried out by passing the solution through an alumina column, and the elut by concentration gives 4.1 g of phenyl-1 isopropyl-3 oxo-2 tetrahydro-1,2,3,4 pyrido - [2,3d] pyrimidine, in the form of colorless prisms melting at 164-165 "C.

Example 4
A mixture of 0.5 g of (m-trifluoromethylphenyl) -1 oxo-2 tetrahydro-1,2,3,4 pyrido- [2,3d] -pyrimidine, 0.8 g of dimethyl carbonate and 10 ml of dimethylformamide is reacted at 160-170 ° C in a sealed tube for 20 h. To a solution of 2.4 g of (p-fluorophenyl) -1 oxo-2 tetrahydro1,2,3,4 pyrido- [2,3d] -pyrimidine in 15 ml of dimethylformamide is added 0.5 g of hydride about 50% sodium, and the mixture is allowed to stand until the evolution of hydrogen ceases.

0.6 g of sodium hydride is added to a solution of 2.6 g of (m-chlorophenyl) -1 oxo-2 tetrahydro-1,2,3,4 pyrido- [2,3d] -pyrimidine in 20 ml of dimethylformamide, and this mixture is stirred for 15 min. Then 3.4 g of methyl fluorosulfate is added thereto, and stirring is continued at room temperature for 1.2 h.
