1. Verfahren zur Herstellung eines neuen Tetrahydropyridinderivats der Formel I

und deren Säureadditionssalze beschrieben. In der obigen Formel können die Variabeln verschiedene, definierte Bedeutungen haben. So kann Rl u.a. Wasserstoff oder Methyl, R2 und Rt, je Wasserstoff oder niedere Alkylgruppen, R3 und R u.a. Wasserstoff, eine niedere Alkyl- oder Alkoxygruppe oder Halogen und R3 überdies u.a. auch eine Cycloalkylgruppe bedeuten, während X und Y je ein Wasserstoffatom oder zusammen eine zusätzliche Bindung bedeuten.
Diese Verbindungen besitzen pharmakologische Eigenschaften, die für Antidepressiva charakteristisch sind, insbesondere hemmen sie an der Ratte die A-Form der Monoaminoxidase, die Aufnahme von Noradrenalin ins Herz und die Aufnahme von Serotonin in die Mitteihirnsynaptosomen und antagonisieren die Wirkung von Tetrabenazin.
Die vorliegende Erfindung betrifft nun ein Verfahren zur Herstellung einer von der obigen allgemeinen Formel Ia umfassten, aber in der genannten Offenlegungsschrift nicht beschriebenen Verbindung, die sich gegenüber den dort beschriebenen Verbindungen insbesondere durch eine besonders starke Monoaminoxidase-hemmende Wirkung auszeichnet.
Die erfindungsgemäss herstellbare neue Verbindung ist das 4-(7-Brom-5-methoxy-2-benzofuranyl)- 1 -methyl- 1,2,3,6-tetra- hydropyridin der Formel I

Ebenfalls Gegenstand der Erfindung ist die Herstellung der Additionssalze, insbesondere der pharmazeutisch annehmbaren Additionssalze der Verbindung der Formel I mit anorganischen und organischen Säuren.
Die Verbindung der Formel I und ihre Additionssalze mit anorganischen und organischen Säuren hemmen, wie aus den Resultaten der isotopischen Bestimmung der Enzymaktivität hervorgeht, an der Ratte und an weiteren Versuchstieren die Monoaminoxidase, insbesondere selektiv und reversibel deren A-Form, nach oraler oder subcutaner Verabreichung von Dosen ab 1,0 mg/kg. Nach Verabreichung von je 100 mg/kg Testsubstanz an Ratten per os, Töten der Tiere nach 2 Stunden, Herstellen von Hirnhomogenisaten und Leberhomogenisaten und Bestimmen der durch diese bewirkten, aber gehemmten Oxidation von isotopisch markiertem Serotonin als Substrat der A-Form der Monoaminoxidase in % der durch entsprechende Organhomogenisate von unbehandelten Ratten bewirkten, nicht gehemmten Oxidation des gleichen Substrats wurden für das Hydrochlorid der Verbindung der Formel I Werte von 4% bzw.
6% (Hirn- bzw. Leberhomogenisat) gefunden, während durch die Hydrochloride der in bezug auf Monoaminoxidase-Hemmung wirksamsten der im Hauptpatent beschriebenen Verbindungen, des 4-(5-Brom-2-benzofuranyl)-piperidins und des l-Methyl-4-(5-cyclohexyl-2-benzofuranyl3-piperidins, bei gleicher Dosierung eine weniger starke Verminderung der Oxidation des gleichen Substrats auf 9% bzw. 12% und 11% bzw. 12% bewirkt wurde.
Die neue Verbindung der Formel I zeigt auch eine, im Vergleich zur sehr starken MAO-A-Hemmung weniger ausgeprägte, Hemmung der Aufnahme von Noradrenalin ins Herz der Ratte bei oraler oder subcutaner Verabreichung von Dosen von 10 bis 100 mg/kg und hemmt auch die Aufnahme von Serotonin in die Mittelhirnsynaptosomen von Ratten. Ferner antagonisiert sie bei intraperitonealer Verabreichung an der Ratte in Dosen von 2 bis 40 mgXkg die hypotherme Wirkung von Reserpin. Zusammen mit einem günstigen therapeutischen Index charakterisieren die obengenannten Eigenschaften die Verbindung der Formel I und ihre pharmazeutisch annehmbaren Salze mit anorganischen und organischen Säuren als Antidepressiva, die z.B. oral oder parenteral zur Behandlung von Gemütsdepressionen verabreicht werden können.
Das neue Tetrahydropyridinderivat der Formel I und seine Säureadditionssalze werden erfindungsgemäss hergestellt, indem man in an sich bekannter Weise eine Verbindung der allgemeinen Formel II

in welcher ZE ein einwertiges Anion der das Normaläquivalent eines mehrwertigen Anions bedeutet, partiell reduziert, und gewünschtenfalls die erhaltene Verbindung der Formel I in ein Additionssalz mit einer anorganischen oder organischen Säure überführt.
Die partielle Reduktion von Verbindungen der allgemeinen Formel II wird vorzugsweise mit Hilfe von Natriumoder Kaliumborhydrid in organisch-wässerigem Medium durchgeführt, indem man beispielsweise zur vorgelegten Lösung des Ausgangsstoffes der allgemeinen Formel II in einem organischen, mit Wasser mischbaren Lösungsmittel, z.B. in einem niederen Alkanol wie Methanol oder Äthanol oder deren Gemischen mit Wasser, allmählich eine wässerige Lösung von Natriumborhydrid zugefügt und das Reaktionsgemisch anschliessend noch einige Zeit weiterreagieren lässt, wobei eine Reaktionstemperatur zwischen ca. 5 und 60"C, vorzugsweise Raumtemperatur bis 35"C, eingehalten wird.
Die Ausgangsstoffe für das obengenannte Verfahren lassen sich in mehreren Stufen aus dem 4-(7!Brom-5-methoxy-2- -benzofuranyl)-pyridin herstellen. Letztere Verbindung kann ihrerseits beispielsweise hergestellt werden, indem man 3 -Brom-5-methoxysalicylald'ehyd mit einen 4-(Halogenmethyl) -pyridin, insbesondere 4-(Chlormethyl)- oder 4-(Brommethyl) -pyridin, in Gegenwart eines säurebindenden Mittels, wie z.B. Kaliumcarbonat, sowie gegebenenfalls Natrium- oder Kaliumjodid unter Erhitzen in einem organischen Lösungsmittel wie Dimethylformamid umsetzt, wobei zusätzlich zur Ätherbildung zum intermediären 2-[(4-Pyridyl)-methoxy]-3 - zbrom-5-methoxybenzaldehyd auch unter Wasseraustritt die Bildung des Benzofuranringes erfolgt.
Aus dem 4-(7-Brom-5-methoxy-2-benzofuranyl)-pyridin erhält man in an sich bekannter Weise durch Quaternierung mit reaktionsfähigen Estern des Methanols, z.B. den weiter unten genannten, die Verbindungen der allgemeinen Formel tI. Die Quaternierung kann in üblicher Weise in einem inerten organischen Lösungsmittel, z.B. in Methanol, Äthylmethylketon, Äthyl'acetat, Tetrahydrofuran oder Dioxan bei Raumtemperatur oder mässig erhöhten Temperaturen bis ca.
100" durchgeführt werden.
Als reaktionsfähige Ester des Methanols für die vorgenannte Quaternierung eignen sich beispielsweise Halogenwasserstoffsäureester, insbesondere das Bromid oder das Jodid, weiter niedere Alkansuifonsäureester und Arensulfonsäureester wie der Methansulfonsäureester, bzw. der Benzolsulfonsäure- und der p-Toluolsulfonsäureester, sowie auch Ester von anderen starken Säuren, z.B. Schwefelsäureester, d.h. Dimethylsulfat.
Die nach dem erfindungsgemässen Verfahren erhaltene Verbindung der Formel I kann gewünschtenfalls in üblicher Weise in ihre Additionssalze mit anorganischen oder organischen Säuren übergeführt werden. Beispielsweise versetzt man ihre Lösung in einem organischen Lösungsmittel mit der als Salzkomponente gewünschten Säure. Vorzugsweise wählt man für die Umsetzung organische Lösungsmittel, in denen das entstehende Salz schwer löslich ist, damit es durch Filtration abgetrennt werden kann. Solche Lösungsmittel sind z.B.
Äthylacetat, Methanol, Äther, Aceton, Äthylmethylketon, Aceton-Äther, Aceton-Äthanol, Methanol-Äther oder Äthanol-Äther.
Zur Salzbildung mit der Verbindung der Formel I können z.B. die Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, 2-Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Bersteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure verwendet werden.
Die neuen Wirkstoffe werden peroral, rektal oder parenteral verabreicht. Die Dosierung hängt von der Applikationsweise, der Spezies, dem Alter und von dem individuellen Zustand ab. Die täglichen Dosen der freien Basen oder von pharmazeutisch annehmbaren Säureadditionssalzen der freien Basen bewegen sich zwischen 0,01 mg/kg und 1,0 mg/kg für Warmblüter. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen können in üblicher Weise hergestellt werden und enthalten vorzugsweise 0,5 bis 10 mg eines erfindungsgemässen Wirkstoffes.
Das nachfolgende Beispiel erläutert die Herstellung der neuen Verbindung der Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, soll jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
Zu einer Lösung von 31,2 g (0,07 Mol) 4-(7-Brom-5 -methoxy-2-benzofuranyl) -1 -methyl-pyridiniumjodid in 200 ml Methanol wird unter Rühren und Aussenkühlung eine Lösung von 10,7 g Natriumborhydrid in 60 ml Wasser so zugetropft, dass die Reaktionstemperatur nicht über 35 steigt.
Anschliessend wird die Lösung 20 Stunden bei Raumtemperatur gerührt. Hierauf wird das Methanol im Vakuum abgedampft, die zurückbleibende wässerige Phase zweimal mit je 500 ml Chloroform extrahiert, die Chloroformlösung über Natriumsulfat getrocknet, filtriert und eingedampft.
Der Rückstand wird aus Methanol-Wasser kristallisiert, wobei man das 4-(7-Brom-5-methoxy-2-benzofuranyl)- 1 -methyl- -1,2,3,6-tetrahydropyridin vom Smp. 73-77" erhält. Das daraus mit einer Lösung von Chlorwasserstoff in Äther-Tetrahydrofuran bereitete Hydrochlorid schmilzt nach Umkristallisation aus Äthanol bei 247-2500.
Der Ausgangsstoff kann auf folgende Weise hergestellt werden: a) 75,5 g (0,327 Mol) 3-Brom-5-methoxysalicylaldehyd, 53,6 g (0,327 Mol) 4-(Chlormethyl)-pyridin-hydrochlorid, 194 g Kaliumcarbonat und 15 g Kaliumjodid werden in 320 ml Dimethylformamid unter Stickstoff 20 Stunden auf 1500 unter Rühren erhitzt. Hierauf wird das Reaktionsgemisch auf ca. 50" gekühlt und bei dieser Temperatur genutscht. Der Filterrückstand wird mit 200 ml qimethylform- amid auf 100" erwärmt und abgenutscht, und der neue Fil terrückstand mit Dimethylformamid nachgewaschen. Die vereinigten Filtrate werden im Vakuum eingedampft, der Rückstand wird anschliessend durch Erhitzen während 2 Stunden im Hochvakuum auf 80" von flüchtigen Anteilen befreit.
Der Rückstand wird in wenig Methylenchlorid gelöst und an 800 g Aluminiumoxid (Aktivität II, Neutral) chromatographiert. Die erste mit 2 Liter Chloroform eluierte Fraktion ist das 4-(7-Brom-5-methoxy-2-benzofuranyl)-pyridin, das nach Umkristallisation aus Äthylacetat bei 149 bis 152" schmilzt.
b) 23,4 g (0,767 Mol) 4-(7-Brom-5-methoxy-2-benzofuranyl)-pyridin werden in 470 ml Äthylmethylketon gelöst und mit 11,5 ml Methyljodid 15 Stunden bei ca. 50 gerührt.
Hierauf wird die Lösung auf - 60 abgekühlt und das ausgeschiedene Salz abgenutscht. Das Filtergut wird mit Äther Petroläther nachgewaschen. Das so erhaltene 4-(7-Brom-5 -methoxy-2-benzofuranyl)-1-methyl-pyridinium-jodid schmilzt bei 260 - 265 und kann direkt weiterverarbeitet werden.

The new active ingredients are administered orally, rectally or parenterally. The dosage depends on the mode of administration, the species, the age and the individual condition. The residue is dissolved in a little methylene chloride and chromatographed on 800 g of aluminum oxide (activity II, neutral). The first fraction eluted with 2 liters of chloroform is 4- (7-bromo-5-methoxy-2-benzofuranyl) pyridine, which, after recrystallization from ethyl acetate, melts at 149 to 152 ".
