Die basischen Verbindungen der Formel I, welche sich nötigenfalls in an sich bekannter Weise aus erhaltenen Säureadditionssalzen freisetzen lassen, sind bei Raumtemperatur feste, gegebenenfalls kristalline oder ölige Verbindungen, die durch Umsetzen mit geeigneten anorganischen oder organischen Säuren in ihre Säureadditionssalze übergeführt werden können. Hierfür sind als anorganische Säuren beispielsweise Halogenwasserstoffsäuren und als organische Säuren beispielsweise Essigsäure, Maleinsäure usw. geeignet.
Die erfindungsgemäss hergestellten Verbindungen der Formel I zeichnen sich durch pharmakodynamische Eigenschaften aus. Insbesondere besitzen die Verbindungen der Formel I im Tierversuch eine Antirigorwirksamkeit.
Die Antirigorwirksamkeit der Verbindungen der Formel I zeigt sich bei Ratten in einer Dosis zwischen 0,001 und 0,1 mg/kg nach i.v.-Verabreichung.
Zur Feststellung der Antirigorwirksamkeit der Verbindungen der Formel I wird folgender Test verwendet:
Ratten werden jeweils 7,5 mg/kg Thalamonal i. p. injiziert, worauf diese Tiere einen Rigor entwickeln, der mit Hilfe eines Elektromyographen gemessen werden kann. Es wird die Dosis an Wirksubstanz festgestellt, die i. v. injiziert werden muss, um den Rigor der Ratten zu hemmen.
Aufgrund der obigen'Wirkung sind die Verbindungen der Formel I angezeigt zur Behandlung des Rigors.
Für die obige Anwendung hängt die zu verabreichende Dosis von der verwendeten Verbindung und der Verabreichungsart sowie der Behandlungsart ab. Im allgemeinen erhält man zufriedenstellende Resultate, wie oben angegeben, bei p. o. oder parenteraler Verabreichung von Verbindungen der Formel I in einer Dosis von 0,01 bis 50 mg/kg Tierkörpergewicht. Bei grösseren Säugetieren ist eine täglich zu verabreichende Menge zwischen 10 und 500 mg angezeigt. Diese täglich zu verabreichende Menge kann auch in kleineren Dosen 1-5mal täglich oder in Retardform verabreicht werden.
Eine Einheitsdosis, beispielsweise eine zur oralen Verabreichung geeignete Tablette, kann zwischen 10 und 60 mg des Wirkstoffes zusammen mit geeigneten, pharmazeutisch indifferenten Hilfsstoffen, wie Lactose, Maisstärke, Talk, Magnesiumstearat usw., enthalten.
Die Verbindungen der Formel I können ferner als Myotonolytika und Muskelrelaxantien verwendet werden, wie dies beispielsweise unter Verwendung des von Teschendorf et al.
[Arch. Exp. Pharmacol. 266 (1970), 467-468] beschriebenen Tests festgestellt wurde. Die für diese Anwendung zu verabreichende Dosis hängt von der verwendeten Verbindung, der Verabreichungsart sowie der Behandlungsart ab. Zufriedenstellende Resultate erhält man bei Verabreichung von Verbin dungen der Formel I in einer täglichen Dosis zwischen 0,0002 und 1 mg/kg Tierkörpergewicht, zweckmässigerweise verab reicht in mehreren Teilmengen 2-4mal täglich oder in Retard form.
Bei grösseren Säugetieren soll die tägliche Dosis von Ver bindungen der Formel I zwischen 0,01 und 10 mg, z.B. zwi schen 0,1 und 6 mg, vorzugsweise zwischen 0,15 und 3 mg, betragen. Eine Einheitsdosis, beispielsweise eine zur oralen
Verabreichung geeignete Tablette, kann zwischen 0,025 und 5 5 mg des Wirkstoffes zusammen mit geeigneten pharmazeu- tisch indifferenten Hilfsstoffen enthalten.
Die Verbindungen der Formel I können ebenfalls in Form von deren Säureadditionssalzen verabreicht werden, die den gleichen Grad an Aktivität besitzen wie die freien Basen.
Die Verabreichung von Verbindungen der Formel I bzw.
deren Salzen kann entweder oral in Form von Tabletten, Gra nulaten, Kapseln oder Dragees oder parenteral in Form von
Injektionslösungen erfolgen.
Vom pharmakodynamischen Standpunkt sind diejenigen 1Verbindungen der Formel I bevorzugt, worin Rt sich in Stel lung 1 befindet, R2 und R3 für Wasserstoff, Halogen oder
Alkyl mit 1-4 Kohlenstoffatomen stehen, und R4 eine Gruppe der Formel

Eine beispielsweise Tablettenzusammensetzung besteht aus 1 mg 1-Methyl-7-(2-thiazolin-2-yl-amino)-indazol, 70 mg Lactose, 5 mg Maisstärke, 5 mg Talk und 0,1 mg Magnesiumstearat. Die auf an sich bekannte Weise hergestellte Tablette ist mit einer Doppelkerbe versehen.
In den nachfolgenden Beispielen sind die Temperaturen in Grad Celsius angegeben, die Raumtemperatur beträgt zwischen 20 und 30 C, falls nicht anders gesagt wird. Die Chloroformlösungen werden entweder mit Natriumsulfat (wasserfrei) oder Sikkon getrocknet.
Beispiel 1
1 -Methyl-7-(2-thiazolin-2-yl-amino) -indazol
9,1 g N-(2-Hydroxyäthyl)-N'-(1-methyl-indazolyl-7)-thio- harnstoff werden in 25 ml konzentrierter Salzsäure gelöst und 30 Minuten auf dem Dampfbad erhitzt. Die Lösung wird mit 75 ml heissem Wasser verdünnt, mit etwas Aktivkohle behandelt und filtriert. Das farblose Filtrat wird mit konzentriertem Ammoniak basisch gestellt. Der Niederschlag wird abfiltriert.
Nach dem Umkristallisieren aus Wasser erhält man 1-Methyl7-(2-thiazolin-2-ylamino)-indazol vom Smp. 1350C.
Den in diesem Beispiel verwendeten N-(2-Hydroxyäthyl) N1-(1-methyl-indazolyl-7)-thioharnstoff erhält man wie folgt:
In ein Gemisch von 2 ml Äthanolamin und 100 ml Äther werden 3 g 1-Methyl-7-isothiocyan-indazol unter Rühren eingetragen. Es bildet sich spontan ein weisser Niederschlag.
Nach 2 Stunden wird dieser abfiltriert und reichlich mit Äther gewaschen. Der so erhaltene N-(2-Hydroxyäthyl)-N'-(1- methyl-indazolyl-7)-thioharnstoff vom Smp. 207-2090 wird ohne weitere Reinigung in obenstehende Reaktion eingesetzt.
Analog Beispiel 1 und unter Verwendung entsprechender Ausgangsstoffe in ungefähr äquivalenten Anteilen gelangt man zu folgenden Verbindungen der Formel I, wobei X die Position des 2-Thiazolin-2-yl-amino-Restes bedeutet: Beispiel R1 R2 R3 R4 Smp. C
