Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung des neuen Antibioticums SL 3733.
Erfindungsgemäss gelangt man zu dem neuen Antibioticum SL 3733, indem man einen Stamm der Pilzgattung Coniothyrium sp. auf oder in einem Nährmedium züchtet und das Antibioticum aus der Fermentationsbrühe durch Extraktion und/ oder Adsorption isoliert und hierauf chromatographisch oder mittels Kristallisation reinigt.
Der neue erfindungsgemäss verwendete Stamm von Coniothyrium sp. wurde aus einer in Zentralafrika gefundenen Erdprobe isoliert und eine Probe davon beim United States Department of Agriculture (Northern Utilization Research and Development Division), Peoria, 111./USA unter der Nummer NRRL 5494 deponiert.
Der neue Stamm der Pilzgattung Coniothyrium wächst auf Malzextrakt-Agar bei 12 bis 33O; bei Temperaturen bis zu 40O erfolgt kein Wachstum, doch wird der Stamm nicht abgetötet.
Das Wachstumsoptimum liegt zwischen 27 und 30O, die Pyknidienbildung erfolgt im ganzen Temperaturbereich des Wachstums. Der Pilz wächst bei niederen Temperaturen (bis 210) regelmässig flächig, bei höheren Temperaturen rosettenartig mit unregelmässig berandeten Lappen, deren Ränder durch die dort dicht stehenden Pyknidien deutlich markiert sind.
In der Reinkultur bildet der Pilz in grosser Zahl kugelige, oft auch flächenförmige oder sehr unregelmässig ellipsoidische, 10S200 Ij grosse Pyknidien, die sich am Scheitel mit einer regelmässig rundlichen Mündung öffnen. Ihre Wand besteht aus 3-4 Lagen von plattenförmigen, relativ dünnwandigen, aussen braunen, innen farblosen, 8-15 F grossen Zellen, an die sich innen eine Schicht von isodiametrischen, zartwandigen, farblosen, 4-7 11 grossen Zellen anlagert. Aus diesen gehen die der ganzen inneren Wand entlang angeordneten, konidienbildenden Zellen (Phialiden) hervor; diese sind flaschenförmig, an der Basis 3-4 F breit und oben in eine mehr oder weniger ausgeprägte Spitze ausgezogen.
Die nacheinander sich entwikkelnden Konidien sind unregelmässig ellipsoidisch, braun, 5-8 x 3,54 F gross und glatt.
Der Stamm NRRL 5494 von Coniothyrium sp. entspricht in seiner Morphologie und Physiologie der Beschreibung in F.
Petrek und H. Sydow Die Phaeosporen Sphaeropsideen und die Gattung Macrophoma Rep. spec. nov. regn. veget 42, 1927, 327 ff.
Für das erfindungsgemässe Verfahren können auch Stämme venvendet werden, wie sie z. B. durch Selektion oder Mutation unter Einwirkung von Ultraviolett- oder Röntgenstrahlen oder durch Anwendung anderer Massnahmen, wie z. B. Behandlung von Laboratoriumskulturen mit geeigneten Chemikalien, aus dem neuen Stamm der Pilzgattung Coniothyrium gewonnen werden können.
Der neue Stamm NRRL 5494 lässt sich auf vielartigen Nährböden, die die üblichen Nährstoffe für Pilze enthalten, züchten. So verwendet dieser Stamm die für die kohlenstoffheterotrophen Organismen üblicherweise benutzten Nährstoffe, z. B. Glucose, Saccharose, Glycerin, Maltose, Lactose usw. als Kohlenstoffquelle, organische und anorganische stickstoffhaltige Verbindungen, wie Pepton, Hefe- oder Fleischextrakte, Ammoniumoxalat, Ammoniumnitrat, Aminosäuren usw. als Stickstoffquelle sowie die üblichen Mineralsalze und Spurenelemente.
Das neue Antibioticum SL 3733 kann man in der Weise herstellen, dass ein flüssiges Medium mit einer Konidien- und Mycelsuspension des Stammes NRRL 5494 beimpft und die Kultur 6-11 Tage, vorzugsweise 7 Tage, bei 240 und bei einem pH-Wert von 3,7-5,2, vorzugsweise bei 4,4, inkubiert wird.
Die Züchtung kann unter aeroben Bedingungen in einer Oberflächenkultur oder submers unter Schütteln oder in Fermentern mit Begasung durch Luft oder Sauerstoff unter Rühren erfolgen. Sobald eine maximale Menge an Antibioticum produziert ist, wird filtriert und das Antibioticum durch extraktive und/oder adsorptive Arbeitsmethoden auf an sich bekannte Weise aus der Kulturlösung gewonnen.
Eine Methode, die sich als vorzugsweise geeignet erwiesen hat, ist die Extraktion der Kulturbrühe mit Essigester; jedoch können auch andere organische Lösungsmittel, wie z. B. Benzol, Chloroform, Methylenchlorid oder Propylacetat verwendet werden. Der nach Abdestillieren des Lösungsmittels verbleibende Extraktionsrückstand wird zur Isolierung des neuen Antibioticums auf chromatographischem Wege an adsorbierenden Mitteln, wie beispielsweise Kieselgel oder Magnesiumsilikat, gereinigt und gegebenenfalls anschliessend z. B. aus Methylenchlorid kristallisiert.
Das neue Antibioticum SL 3733 kann wie folgt charakterisiert werden: Hellgelbe, kristalline Substanz der Summenformel C20H1007(MG= 362,3) Smp. 237-2390C; [ai20 = -637" (c = 0,5 in Chloroform) UV-Spektrum: Maxima bei 215,5 nm (log e = 4,46) (Fig. 1) 268,5 nm (log e = 3,88)
346,5 nm (log e = 3,64) IR-Spektrum u.a. Banden bei 3050, 1682, 1668, 1635, in Methylenchlorid: 1592, 1470, 1316, 1212, (Fig. 2) 1020, 945, 830 cm-t.
Das Antibioticum SL 3733 ist wirksam gegen pathogene Pilze, insbesondere gegen die Mykosen erzeugenden Arten Trichophyton tonsurans, Mikrosporum canis und Allescheria boydii.
Ausserdem zeichnet es sich durch interessante pharmakologische Eigenschaften aus und kann daher als Heilmittel Verwendung finden. Es zeigt cytostatische Wirkung, wie in vitro an einem aus einem Maustumor herrührenden Zellstamm (P815-Mastocytom) festgestellt werden konnte. Eine 50%ige Hemmung der Zellvermehrung wurde bei einem Substanzgehalt von 0,13 mg/l beobachtet.
Zudem zeichnet sich das Antibioticum durch ausgeprägte immunosuppressive Eigenschaften aus, wie sich in verschiedenen Tests erwies: a) Im target cell destruction Test konnte bei einem Gehalt von etwa 10-8-5 bis 10-6.5 g/ml eine ausgeprägte Hemmung der Zellyse gezeigt werden.
b) Im Chemotaxistest konnte eine 100%ige Hemmung der Wanderung von Kaninchen-Leukozyten bei einem Gehalt von etwa 10-7 bis 10-6,5 g/ml festgestellt werden.
c) Im LHG-Test nach Jerne konnte an der Maus eine leichte Hemmung der plaque forming cells in einem Dosenbereich von etwa 1,5 bis 3 mg/kg gezeigt werden.
d) In Hauttransplantationsversuchen an der Maus zeigte sich bei Verabreichung von 1,5 mg/kg i. p. die Überlebenszeit des Transpiantates bei H-2-Kompatibilität (Spender DBA/2 Maus, Empfänger Balb/c-Maus) hochsignifikant verlängert (P < 0,005).
Das genannte Antibioticum ist aufgrund dieser Wirkung zur Unterdrückung unerwünschter Immunreaktionen geeignet.
Für die genannten Anwendungen wird die Dosis davon abhängen, welche Verabreichungsform und Behandlung gewünscht wird. Befriedigende Resultate werden jedoch erreicht mit einer Dosis von etwa 0,1 bis 5 mg/kg Körpergewicht des Testtieres, die nötigenfalls in 2-3 Anteilen verabreicht werden können. Für grössere Säugetiere liegt die Tagesdosis bei etwa 10-300 mg; für orale Applikationen enthalten die Teildosen etwa 5 bis 100 mg der wirksamen Substanz neben festen oder flüssigen Trägersubstanzen.
Das Antibioticum SL 3733 kann als Arzneimittel allein oder in entsprechenden Arzneiformen für orale, enterale oder parenterale Verabreichung verwendet werden.
In dem folgenden Beispiel erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 10 Liter einer Nährlösung, die pro Liter
100 g Saccharose
4 g Glykokoll 1 g KH2PO4
1 MgS04 7Hz0
1 g Hefeextrakt und entmineralisiertes Wasser enthält, werden mit 1 Liter einer Vorkultur des Stammes NRRL 5494 der Pilzgattung Coniothyrium angeimpft und in einem Glasfermenter unter Rühren (Blattrührer, 100-400 Umdrehungen/Minute, steigernd) und Belüftung (1,0 Liter Luft/Minute/Liter Nährlösung) 7 Tage bei 240 inkubiert.
Die Kulturbrühe (pH 4,4) wird dreimal mit der gleichen Menge Essigester extrahiert und die vereinigten, mit Wasser gewaschenen Extrakte zur Trockne eingedampft. Den Rückstand chromatographiert man an Kieselgel unter Verwendung von Chloroform und anschliessend einem Gemisch von Chloroform/0,5 % Methanol. Die tiefgelben Eluatfraktionen werden vereinigt und eingedampft. Das erhaltene Rohprodukt wird aus Methylenchlorid kristallisiert, wobei das Antibioticum SL 3733 in Form hellgelber Kristalle anfällt, Smp. 237-239 [a]i, = -637" (c = 0,5 in Chloroform).
Die als Ausgangsmaterial verwendete Vorkultur wird wie folgt erhalten:
Die zur Beimpfung verwendete Konidien- und Mycelsuspension wird aus einer Kultur des ursprünglich isolierten Stammes NRRL 5494 hergestellt, welche 11 Tage bei 270 auf einem Agarmedium, das pro Liter 20 g Glucose 15 gAgar

2 g Malzextrakt
2 g Hefeextrakt 2 g KH2PO4
2 g MgSO4 - 7H20 und entmineralisiertes Wasser enthält, gezüchtet wird. Die Konidien und das Mycel dieser Kultur werden gesammelt und in physiologischer Kochsalzlösung homogenisiert.
Mit dieser Suspension wird 1 Liter einer Nährlösung der folgenden Zusammensetzung 100 g Saccharose
10 g Proflo
3 g Ammoniumoxalat 100 ml Knop-Lösung 10-fach (10 g Ca(NO3)2 - 4H20;
2,5 g MgSO4 - 7H20; 2,5 g KH2PO4; 1,25 g KCI und entmineralisiertes Wasser ad 1 Liter)
2 ml Fe/Zn-Lösung (8,34 g FeSO4 7H20; 3,44 g ZnSO'7HO; zu 7z0; 5 ml H2SO4 [95-97%ig] und entmineralisiertes Wasser ad 1 Liter) in einem 2 Liter-Erlenmeyerkolben angeimpft und auf einer Rundschüttelmaschine (180 Umdrehungen/Minute) bei 240 und pH 4,2-4,7 während 3 Tagen inkubiert. Diese Fermentationslösung dient als Impfmaterial für den 10 Liter-Glasfermenter.

The new antibiotic SL 3733 can be prepared in such a way that a liquid medium is inoculated with a conidia and mycelium suspension of the strain NRRL 5494 and the culture is 6-11 days, preferably 7 days, at 240 and a pH of 3, 7-5.2, preferably at 4.4.
The cultivation can take place under aerobic conditions in a surface culture or submerged with shaking or in fermenters with aeration by air or oxygen with stirring. As soon as a maximum amount of antibiotic has been produced, it is filtered and the antibiotic is obtained from the culture solution in a manner known per se by extractive and / or adsorptive working methods.
One method that has proven to be particularly suitable is extraction of the culture broth with ethyl acetate; however, other organic solvents, such as. B. benzene, chloroform, methylene chloride or propyl acetate can be used. d) In skin transplantation experiments on mice, administration of 1.5 mg / kg i. p. the survival time of the transpiantate with H-2 compatibility (donor DBA / 2 mouse, recipient Balb / c mouse) is increased significantly (P <0.005).
Because of this effect, the antibiotic mentioned is suitable for suppressing unwanted immune reactions.
For the applications mentioned, the dose will depend on the form of administration and treatment desired. However, satisfactory results are achieved with a dose of about 0.1 to 5 mg / kg body weight of the test animal, which if necessary can be administered in 2-3 portions. For larger mammals, the daily dose is around 10-300 mg; For oral applications, the partial doses contain about 5 to 100 mg of the active substance in addition to solid or liquid carrier substances.
