Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Thiophosphorsäureester mit anthelmintischer Wirkung.

in den Substituenten R1 bis R4 vorkommenden Alkylgruppen können geradkettig oder verzweigt sein.
Thiophosphorsäureester der Formel I, in denen unabhänging voneinander R1 Alkoxy mit 1 oder Z Kohlenstoffatomen, R3 bzw. R4 Alkyl mit 1 bis 5 Kohlenstoffatomen und R5 A1- kyl oder Alkenyl mit 1 bis 5 Kohlenstoffatomen bedeuten, sind als bevorzugt hinsichtlich ihrer anthelmintischen Wirksamkeit anzusehen.
Die neuen Thiophosphorsäureester der Formel I werden erfindungsgemäss hergestellt, indem man ein O,S-Dialkylthio phosphorsäurechlorid bzw. ein O,S-Dialkyldithiophosphorsäurechlorid der Formel II

Die Umsetzung wird vorteilhaft in einem inerten organischen Lösungsmittel, wie z. B. Acetonitril, durchgeführt. Es ist weiterhin vorteilhaft, die Umsetzung in Gegenwart von Kupferpulver als Katalysator durchzuführen. Als säurebindende Mittel sind Alkali- und Erdalkalimetall-hydroxide und-carbonate, wie Natriumhydroxid, Natriumcarbonat, sowie tert.
Amine wie Triäthylamin oder Pyridin geeignet. Die Reaktionstemperaturen liegen zwischen 20 und 80" C, vorzugsweise zwischen 50 bis 60 C. Das Reaktionsprodukt wird durch Abdampfen des Lösungsmittels und Extraktion des Rückstandes mit einem geeigneten Lösungsmittel, vorzugsweise Methylenchlorid, isoliert und aus der erhaltenen Lösung durch Abdampfen des zur Extraktion verwendeten Lösungsmittels gewonnen. Die Herstellung der neuen Thiophosphorsäureester der Formel I wird durch die folgenden Beispiele näher erläutert.
Beispiel 1
In eine Mischung von 45,6 g 6-Brom-3-t-butylphenol, 24 g Soda, 0,5 g Kupferpulver und 200 ml Acetonitril werden bei 60 C unter ständigem Rühren während 15 Minuten 40,5 g O-Äthyl-S-propylthiophosphorsäurechlorid eingetropft. Nach beendigter Zugabe wird 4 Stunden bei 50 bis 60 C nachgerührt. Nach dem Erkalten werden die entstandenen Salze abgesaugt, das Lösungsmittel im Vakuum abgedampft und der Rückstand mit 200 ml Methylenchlorid aufgenommen. Die erhaltene Lösung wird zweimal mit je 60 ml 1n Natronlauge ausgewaschen. Nach dem Abdampfen des Lösungsmittels im Vakuum erhält man 60,8 g O-Äthyl-O-(2-brom-5-t-butyl- phenyl)-S-n-propyl-thiophosphat als farbloses Öl mit einem Brechungsindex von nD24 1,5285 (Verbindung Nr. 1).

Die Verbindungen der Formel I sind insbesondere wirksam gegen Nematoden, wie Ascariden, Trichostrongyliden, Ancylostomatiden, Strongyliden und Cestoden, wie Anoplocephaliden und Taenioiden.
Die anthelmintische Wirkung der Verbindungen der Formel I macht sie zur Behandlung von Wurmkrankheiten bei Haus- und Nutztieren geeignet. Die Verbindungen zeigern in sehr niedrigen Dosierungen eine ausgezeichnete Wirksamkeit und zeichnen sich darüber hinaus durch niedere Toxizität und gute Verträglichkeit aus.
Mittel, die Verbindungen der Formel I als aktive Komponente enthalten, können zur Bekämpfung parasitärer Helminthen bei Haus- und Nutztieren wie Rindern, Schafen, Ziegen, Pferden, Schweinen, Katzen, Hunden und Geflügel verwendet werden. Dabei können sie den Tieren sowohl als Einzeldosis wie auch wiederholt verabreicht werden, wobei die einzelnen Gaben je nach Tierart vorzugsweise zwischen 5 und 1000 mg Wirkstoff pro kg Körpergewicht betragen. Durch eine protrahierte Verabreichung erzielt man in manchen Fällen eine bessere Wirkung oder man kann mit geringen Gesamtdosen auskommen. Die Wirkstoffe der Formel I bzw. die sie enthaltenen Gemische können auch dem Futter oder den Tränken zugesetzt werden. Das Fertigfutter enthält die Substanzen der Formel I vorzugsweise in einer Konzentration von etwa 0,05 bis 1 Gew. %.
Die Mittel können in Form von Lösungen, Emulsionen, Suspensionen (Drenchs), Pulvern, Tabletten, Bolussen und Kapseln peroral oder abomasal den Tieren verabreicht werden.
Zur Bereitung dieser Applikationsformen dienen zum Beispiel übliche feste Trägerstoffe wie Kaolin, Talkum, Bentonit, Kochsalz, Calciumphosphat, Baumwollsaatmehl oder mit den Wirkstoffen nicht reagierende Flüssigkeiten wie Öle und andere für den tierischen Organismus unschädliche Lösungsoder Verdünnungsmittel. Soweit die physikalischen und toxikologischen Eigenschaften von Lösungen oder Emulsionen dies zulassen, können die Wirkstoffe den Tieren auch z. B.
subcutan injiziert werden.
Soll Wasser als Verdünnungsmittel für Applikationsformen wie z. B. Drenchs oder Emulsionen verwendet werden oder -sollen die Mittel in Form von Futterkonzentraten verabreicht werden, so ist durch eine geeignete Schutzformulierung (z. B.
Umhüllungsgranulate), durch Einbettung in hydrophobe Materialien (z. B. Paraffin, Silikonöle oder -wachse) oder durch Aufziehen auf inerte feste Trägerstoffe wie Kaolin, Talkum, Bentonit, Kieselgur, Bolus alba dafür zu sorgen, dass keine vorzeitige Zersetzung eintritt.
Liegen die anthelmintischen Mittel in Form von Futterkonzentraten vor, so dienen als Trägerstoffe z. B. Leistungsfutter, Futtergetreide oder Proteinkonzentrate. Solche Futter können ausser den Wirkstoffen noch Zusatzstoffe, Vitamine, Antibiotika, Chemotherapeutika oder andere Pestizide, vor nehmlich B akteriostatika, Fungistatika, Coccidiostatika oder auch Hormonpräparate, Stoffe mit anaboler Wirkung oder andere das Wachstum begünstigende, die Fleischqualität von Schlachttieren beeinflussende oder in anderer Weise für den Organismus nützliche Stoffe enthalten. Auch können sie mit anderen Anthelmintika kombiniert werden, wodurch ihre Wirkung verbreitert und an gegebene Umstände angepasst wird.
Als solche sind zu nennen: Nematodenmittel, z. B. Absomal, Alcopar, Anthelcide, Ascaridol, Banmith II, Bephenium, Bradosol, Cambcudasol, Calorophos, Chlorthion, Coumaphos, Cyanin, Destomycin, Diäthylcarbamazin, Dichlorophen, DDVP, 1,4-Di-(D-glyconyl)-piperazin, Dithiazonin, Dow ET/70 Douco 132, Dymanthin HCl, Egressin, Gainex, Hexachlorophen, Hexylresorcin, Tonit, Levamisol, Meparin, Methylenviolett, 1-Methyl-1-tridecyl-piperazinium- 4-carbonsäure-äthylester, Methyridin, Monopar, Marlene, Weguvon, Nematodin, Nemuarl, Nidanthel, Parbendazol, Parvex Phenothiazin, Piperazin, Polymethylenpiperazin, Promethanzin, Pyrantel, Pyranthiazin, Pyrvininmembonat, Rametin, Ronnel, Santonin Shell 1808, Stilbazium, Tetramisol, Thenium, Thiabendazol, Thymolan, Vermella, Nebendazol, Fenbendazol; Trematodenmittel, z. Die Herstellung anthelmintischer Mittel kann in an sich bekannter Weise durch inniges Vermischen und Vermahlen von Wirkstoffen der allgemeinen Formel I mit geeigneten Trägerstoffen, gegebenenfalls unter Zusatz von gegenüber den Wirkstoffen inerten Dispersions- oder Lösungsmitteln erfolgen
Die Wirkstoffe können in den folgenden Aufarbeitungsformen vorliegen und angewendet werden: Feste Aufarbeitungsformen:
Stäubemittel, Granulate, Umhüllungsgranulate, Imprägnie rungsgranulate und Homogengranulate.
In Wasser dispergierbare Wirkstoffkonzentrate:
Spritzpulver (wettable powder).
Flüssige Aufarbeitungsformen:
Lösungen, Pasten (Emulsionen).
Die Korngrösse der Trägerstoffe beträgt für Stäubemittel und Spritzpulver zweckmässig bis ca. 0,1 mm und für Granulate 10 bis 500,u (0,01 bis 0,5 mm).
Die Wirkstoffkonzentrationen in den festen Aufarbeitungsformen betragen 0,5 bis 80%, in den flüssigen Aufarbeitungsformen 0,5 bis 15%.
Diesen Gemischen können ferner den Wirkstoff stabilisierende Zusätze und/oder nichtionische, anionaktive und kationenaktive Stoffe zugegeben werden, die beispielsweise eine bessere Benetzbarkeit (Netzmittel) sowie Dispergierbarkeit (Dispergatoren) gewährleisten.
In Wasser dispergierbare Pulvermischung:
Zusammensetzung: 25 Teile Wirksubstanz der Formel 1,
3 Teile einer Mischung von Polyoxyäthylen-Tallölester
Harnstoff,
7 Teile Polyvinylpyrrolidon, 31,5 Teile Kieselsäure, hochdispers, 33,5 Teile Bolus alba.
Der Wirkstoff wird zusammen mit der Polyoxyäthylen Tallölester-Harnstoff-Mischung und dem Polyvinylpyrrolidon unter Zusatz von ca. 30% des Kieselsäureanteils in einem Planeten-Mischer homogen gemischt. Anschliessend setzt man den restlichen Anteil Kieselsäure und Bolus alba zu und vermischt in geeigneten Mischern bis zur Homogenität. Auf einer Scheiben-Stift-Mühle wird dann die Mischung auf eine Partikelgrösse von weniger als 20 Mikron vermahlen.
Die anthelmiPtische Wirksamkeit der Verbindungen der Formel I wird anhand folgender Versuche demonstriert:
1. Versuche an von Oxyuren befallenen Mäusen
Die Wirkstoffe werden in Form einer Suspension per Magensonde weissen Mäusen verabreicht, die mit Mäuseoxyuren infiziert sind. Pro Versuch werden 5 Tiere verwendet. Jeder Tiergruppe werden die Wirkstoffe während drei aufeinanderfolgenden Tagen einmal täglich verabreicht. Die Tagesdosis pro Tier beträgt 750 mg Wirkstoff pro kg Körpergewicht.
Die Tiere werden dann am 4. Tag nach Beginn der Behandlung getötet und seziert. Die Auswertung erfolgt nach Sektion der Versuchstiere durch Vergleich der im Darm verbliebenen Anzahl Mäuseoxyuren mit einer unbehandelten gleichartig und gleichzeitig infizierten Kontrolle.
2. Versuche an von Nippostrongylus brasiliensis befallenen Mäusen
Die Wirkstoffe werden in Form einer Suspension per Magensonde weissen Mäusen verabreicht, die mit Nippostrongylus brasiliensis infiziert sind. Pro Versuch werden 5 Tiere verwendet. Jeder Tiergruppe werden die Wirkstoffe während drei aufeinanderfolgenden Tagen einmal täglich verabreicht. Die Tagesdosis pro Tier beträgt 750 mg Wirkstoff pro kg Körpergewicht.
Die Tiere werden dann am 4. Tag nach Beginn der Behandlung getötet und seziert. Die Auswertung erfolgt nach Sektion der Versuchstiere durch Vergleich der im Darm verbliebenen Anzahl Nippostrongylus brasiliensis mit einer unbehandelten gleichartig und gleichzeitig infizierten Kontrolle.
3. Versuche an durch Hymenolepis nana befallenen Mäusen
Die Wirkstoffe werden in Form einer Suspension per Magensonde weissen Mäusen verabreicht, die künstlich mit Hymenolepis nana infiziert sind. Pro Versuch werden 5 Tiere verwendet. Jeder Tiergruppe werden die Wirkstoffe während drei aufeinanderfolgenden Tagen einmal täglich verabreicht. Die Tiere werden dann am 4. Tag nach Beginn der Behandlung getötet und seziert.
Die Auswertung erfolgt nach Sektion der Versuchstiere durch Vergleich der sich im Darm befindlichen Bandwürmer mit einer unbehandelten, gleichzeitig und gleichartig infizierten Kontrollgruppe.
In diesen Versuchen zeigten Wirkstoffe, die der Formel I entsprechen, eine gute anthelmintische Wirkung.

In a mixture of 45.6 g of 6-bromo-3-t-butylphenol, 24 g of soda, 0.5 g of copper powder and 200 ml of acetonitrile, 40.5 g of O-ethyl-S -propylthiophosphoric chloride added dropwise. The compounds of formula I are particularly active against nematodes, such as ascarids, trichostrongylidene, ancylostomatids, strongylids and cestodes, such as anoplocephalids and taenioids.
The anthelmintic effect of the compounds of formula I makes them suitable for the treatment of worm diseases in domestic and farm animals. The compounds show excellent activity in very low doses and are also characterized by low toxicity and good tolerance.
Agents containing compounds of formula I as active component can be used to control parasitic helminths in domestic and farm animals such as cattle, sheep, goats, horses, pigs, cats, dogs and poultry. They can be administered to the animals both as a single dose and repeatedly, the individual doses preferably being between 5 and 1000 mg of active ingredient per kg of body weight, depending on the animal species. A protracted administration can achieve a better effect in some cases or you can manage with low total doses. The active compounds of the formula I or the mixtures they contain can also be added to the feed or the drinkers. The animals are then sacrificed and dissected on the 4th day after the start of treatment. The evaluation is carried out according to the section of the test animals by comparing the number of mouse oxyurs remaining in the intestine with an untreated control of the same type and simultaneously infected.
2. Trials on mice infested with Nippostrongylus brasiliensis
The active substances are administered in the form of a suspension by gastric tube to white mice infected with Nippostrongylus brasiliensis. 5 animals are used per experiment. The active ingredients are administered to each group of animals once a day for three consecutive days. The daily dose per animal is 750 mg of active ingredient per kg of body weight.
The animals are then sacrificed and dissected on the 4th day after the start of treatment. The evaluation is carried out after sectioning the test animals by comparing the number of Nippostrongylus brasiliensis remaining in the intestine with an untreated control of the same type and at the same time infected.
3. Trials on mice infested with Hymenolepis nana
The active ingredients are administered in the form of a gastric tube suspension to white mice artificially infected with Hymenolepis nana. 5 animals are used per experiment. The active ingredients are administered to each group of animals once a day for three consecutive days. The animals are then sacrificed and dissected on the 4th day after the start of treatment.
The evaluation is carried out after sectioning the test animals by comparing the tapeworms in the intestine with an untreated, simultaneously and similarly infected control group.
In these tests, active ingredients that correspond to Formula I showed a good anthelmintic effect.
