Die vorliegende Erfindung bezieht sich auf Verfahren zur Herstellung von in ihrer Struktur einen Indolkern enthaltenden tetrazyklischen Verbindungen sowie ihre Verwendung zur Herstellung ihrer Salze.
Es ist bekannt, dass es bis zur heutigen Zeit unter den kondensierten Indolderivaten, darunter auch den tetrazyklischen Verbindungen, die einen Indolkern enthalten, eine Reihe von sowohl synthetischen als auch natürlichen Stoffen gibt, welche hohe physiologische Aktivität zeigen (Derivate der Lysergsäure, Reserpin und andere). Im Zusammenhang damit sind die genannten Stoffe von grossem praktischem Interesse.
Zweck der vorliegenden Erfindung war es, eine neue Gruppe tetrazyklischer Indolkern enthaltender Verbindungen zu synthetisieren, welche für die Entwicklung neuer wirksamer Arzneimittel von Interesse sind.
Es werden neue in ihrer Struktur Indolkern enthaltende tetrazyklische Verbindungen erhalten, welche die folgende allgemeine Formel I besitzen

Die Verbindungen der allgemeinen Formel I stellen feste Stoffe dar, welche basische Eigenschaften aufweisen, in den meisten organischen Lösungsmitteln, verdünnten Säuren leicht löslich und in Wasser schwer löslich sind. Die genannten Verbindungen bilden mit organischen oder anorganischen Säuren in Wasser leicht lösliche Salze, die durch die folgende Formel dargestellt werden können

worin R, R', m und n die oben angegebenen Bedeutungen haben und K für eine organische oder anorganische Säure steht.
Unter den genannten Verbindungen sind Stoffe mit hoher psychotroper Aktivität gefunden worden, beispielsweise die Verbindung 3-Methyl-8-methoxy-3H- 1,2,5 ,6-tetrahydropyra- zino-[1,2,3-ab]-ss-karbolinhydrogenchlorid, welche zur Behandlung depressiver und anderer psychischer Erkrankungen verwendet werden kann.
Diese Verbindung übt im Experiment einen ausgeprägten Einfluss auf das Zentral- und das peripherische Nervensystem aus. Unter dem Einfluss der genannten Verbindung werden die Effekte der Einwirkung von Phenamin und 5-Hydroxytryptophan auf das Zentralnervensystem und die Einwirkung von Adrenalin, Phenyläthylamin, Tyramin, Serotonin und Tryptamin auf das peripherische Nervensystem verstärkt. Die oben genannte Verbindung schwächt die deprimierenden Effekte von Reserpin und Tetrabenazin sowie die kataleptische Aktivität von Phenothiazinderivaten ab. Nach diesen pharmakologischen Eigenschaften steht sie den antidepressiven Stoffen nahe.
Im Unterschied zu anderen antidepressiven Stoffen übt sie keine cholinolytische Wirkung aus, verstärkt nicht den schlafmachenden Effekt von Hexenal, die analgetische Wirkung von Promedol, die lokalanästhesierende Aktivität von Novokain, verstärkt wirksamer als das Imizin die Effekte von Phenyl äthylamin, Tyramin und Tryptamin. Nach den Angaben der biochemischen Untersuchungen bewirkt der Stoff in für die Erzielung eines eindeutigen pharmakologischen Effektes ausreichenden Dosen eine mässige Hemmung der Monoaminoxydase-Aktivität nur in Nierengeweben, weshalb angenommen werden kann, dass die Antimonoaminoxydase-Wirkung dem pharmakologischen Effekt dieser Verbindung nicht zugrundeliegt.
Die toxikologische Untersuchung, durchgeführt an Mäusen, Ratten und Kaninchen bei einmaliger und mehrfacher Einführung, ergab, dass dieser Stoff in den die empfohlenen Dosen um 30- bis 50mal übersteigenden Dosen an Tieren keine toxische Wirkung ausübt. Die klinische Untersuchung dieser Verbindung bestätigte, dass sie eine antidepressive Wirkung besitzt, die vom stimulierenden Effekt begleitet wird.
Nach den Angaben der klinischen Untersuchung ist diese Verbindung indiziert für die Behandlung schlaffer apathischer, adynamischer, neurotischer und nichttiefer hypochondriseher depressiver Zustände. Im Zusammenhang mit unbedeutenden Nebenwirkungen kann sie zur Behandlung von Kranken sowohl im Krankenhaus als auch ambulant empfohlen werden.
Der genannte Stoff kann auch zur Behandlung von psychischen Erkrankungen in Kombination mit Neuroleptika verwendet werden. Er kann in Form von Tabletten, Pulver, Injektionslösungen verwendet werden.
Das erfindungsgemässe Verfahren zur Herstellung von Verbindungen der Formel I oder deren Hydrogenchlorid ist dadurch gekennzeichnet, dass man ein Laktam der allgemeinen Formel II

Es sind zwei Varianten der Umwandlung der Zwischenverbindungen der Formel III in Endverbindungen bevorzugt:
1. Entweder durch die Behandlung der Verbindungen der
Formel III mit einem alkalischen Mittel oder
2. durch die Behandlung des Zwischenproduktes III mit einem niederen aliphatischen Alkohol.
Praktisch wird das Verfahren in der Regel wie folgt durchgeführt:
1. Laktame der allgemeinen Formel II zyklisiert man mit Phosphor(V)-oxychlorid zweckmässigerweise unter Erhitzen.
Es bildet sich dabei eine Zwischenverbindung der Formel III.
Das überschüssige Phosphor(V)-oxychlorid wird abdestilliert und der Rückstand mit wässriger Lösung von Alkalimittel an der Kälte behandelt. Das erhaltene Produkt der Formel I wird mit einem organischen Lösungsmittel, vorzugsweise mit Benzol oder Chloroform, extrahiert. Erforderlichenfalls kann das Produkt der Formel I unter der Einwirkung von organischen oder anorganischen Säuren in das entsprechende Salz übergeführt werden.
2. Laktame der allgemeinen Formel II zyklisiert man unter Erhitzen mit Phosphor(V)-oxychlorid. Das gebildete Zwi schenprodukt der Formel II trennt man ab, behandelt zweckmässigerweise unter Erhitzen mit niederem aliphatischem Alkohol, beispielsweise mit Äthylalkohol, und trennt nach der Abkühlung das Endprodukt ab.
Als Ausgangsverbindungen dienen in der vorliegenden Erfindung Laktame der allgemeinen Formel II, von denen die einen bekannte Stoffe und die anderen neue Stoffe sind. Diese Ausgangsverbindungen können aus den Derivaten von Karbolin und Azepino-[3,4-b]-indol in an sich bekannter Weise erhalten werden.
Zur besseren Illustration der vorliegenden Erfindung werden folgende Beispiele für die Durchführung des Verfahrens zur Herstellung der angemeldeten Verbindungen angeführt.
Das Beispiel 1 illustriert die Herstellung der Ausgangslaktame und ihrer Hydrogenchloride, die der Formel II entsprechen.
Beispiel 1 a) Herstellung von 1-Oxo-10-(3!-dimethylaminoäthyl)- lH-2,3,4,5-tetrahydroazepino-[3,4-b]-indol der Formel II, worin m für 2; n für 3; R für H; R' für CH3 steht.
Einer Suspension von 20 g 1-Oxo-lH,2,3,4,5-tetrahydro azepino-[3,4-b]-indol in 120 ml trockenem Dimethylformamid gibt man bei einer Temperatur von 25 C portionsweise 3 g Natriumhydrid zu und erhitzt bei 30 C 2 Stunden und bei 50 C 30 Minuten. Der Reaktionsmasse tropft man 14 g P-Dimethyl aminoäthylehlorid zu, erhitzt bei 50 bis 55OC 4 Stunden und lässt über Nacht stehen. Das Gemisch giesst man in 100 ml 50 %ige Essigsäure, filtriert, alkalisiert die Lösung mit 15 %igem wässrigem Ammoniak auf einen pH-Wert von etwa 9 und extrahiert mit Benzol. Der Extrakt wird mit Wasser gewaschen, mit glühender Pottasche getrocknet, eingedampft, der Rückstand mit Äther verrieben und getrocknet. Man erhält 14,2 g der Verbindung der Formel II.
Die Ausbeute beträgt 52,4%, der Schmelzpunkt 122 bis 1230C (Äthylazetat).

Die Reaktionsmasse wird im Eis abgekühlt (1 bis 1,5 Stunden), der ausgefällte Niederschlag abfiltriert, mit kaltem absolutem Äthylalkohol gewaschen und getrocknet. Man erhält 3 Methyl-8-methoxy-3H, 1,2,5,6-tetrahydropyrazino-(1,2,3-ab)- ss-karbolin-hydrogenchlorid, worin m für 2; n für 2; R für 8 Methoxy; R' für CH3 steht.
Die Ausbeute beträgt 130,5 g (95 % der Theorie), der Schmelzpunkt 306 bis 3090C.

UNTERANSPRÜCHE
1. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass als niederer Alkohol Äthanol eingesetzt wird.
2. Verfahren nach Patentanspruch I, dadurch gekennzeichnet, dass die Umwandlung des Zwischenproduktes der Formel III in die Verbindung der Formel I durch deren Abtrennung aus der Reaktionsmasse und Behandlung mit niederem aliphatischem Alkohol vorgenommen wird.
3. Verfahren nach Patentanspruch I und Unteranspruch 2 zur Herstellung von 3-Methyl-8-methoxy-3H,1,2,5,6-tetra hydropyrazino-(1,2,3-ab)-ss-karbolin-hydrogenchlorid, dadurch gekennzeichnet, dass man mit Phosphor(V)-oxychlorid 9-ss-(N, M-Dimethylamino)-äthyl-6-methoxy-1-oxo-1,2,3,4- tetrahydro-ss-karbolin-hydrogenchlorid zyklisiert, das gebildete Zwischenprodukt 3 -Methyl-8-methoxy-3H, 1,2,5,6- tetrahydropyrazino-(1,2,3-ab)-ss-karbolin-dichlorphos- phonat abtrennt und mit Äthylalkohol umsetzt.

It is known that among the condensed indole derivatives, including the tetracyclic compounds containing an indole nucleus, there are a number of both synthetic and natural substances that show high physiological activity (derivatives of lysergic acid, reserpine and others ). In connection with this, the substances mentioned are of great practical interest.
The purpose of the present invention was to synthesize a new group of compounds containing tetracyclic indole nucleus which are of interest for the development of new, effective medicaments.

The compounds of general formula I are solid substances which have basic properties, are easily soluble in most organic solvents, dilute acids and are sparingly soluble in water. Among the compounds mentioned, substances with high psychotropic activity have been found, for example the compound 3-methyl-8-methoxy-3H-1,2,5,6-tetrahydropyrazino- [1,2,3-ab] -ss- carboline hydrogen chloride, which can be used to treat depressive and other mental illnesses.
In the experiment, this connection exerts a pronounced influence on the central and peripheral nervous systems. Under the influence of the compound mentioned, the effects of the action of phenamine and 5-hydroxytryptophan on the central nervous system and the action of adrenaline, phenylethylamine, tyramine, serotonin and tryptamine on the peripheral nervous system are increased. The above compound weakens the depressing effects of reserpine and tetrabenazine, as well as the cataleptic activity of phenothiazine derivatives. According to these pharmacological properties, it is close to antidepressant substances.
In contrast to other antidepressant substances, it does not exert a cholinolytic effect, does not increase the sleep-inducing effect of Hexenal, the analgesic effect of Promedol, the local anesthetic activity of novocaine, and more effectively than imicine increases the effects of phenylethylamine, tyramine and tryptamine. According to the information from the biochemical investigations, in doses sufficient to achieve a clear pharmacological effect, the substance only moderately inhibits monoamine oxidase activity in kidney tissues, which is why it can be assumed that the antimonoamine oxidase effect is not the basis for the pharmacological effect of this compound.
The toxicological investigation, carried out on mice, rats and rabbits after single and multiple introductions, showed that this substance does not exert any toxic effects on animals in doses which are 30 to 50 times higher than the recommended doses. The clinical examination of this compound confirmed that it has an antidepressant effect, which is accompanied by the stimulatory effect.
According to the clinical investigation, this compound is indicated for the treatment of flaccid apathetic, adynamic, neurotic, and non-deep hypochondriacal depressive states. In connection with insignificant side effects, it can be recommended for the treatment of sick both in hospital and outpatient.
The substance mentioned can also be used in combination with neuroleptics for the treatment of mental illnesses. It can be used in the form of tablets, powders, solutions for injection.

The product of the formula I obtained is extracted with an organic solvent, preferably with benzene or chloroform. 2. Lactams of the general formula II are cyclized with heating with phosphorus (V) oxychloride. The intermediate product of the formula II formed is separated off, advantageously treated with heating with lower aliphatic alcohol, for example with ethyl alcohol, and the end product is separated off after cooling.
The starting compounds used in the present invention are lactams of the general formula II, some of which are known substances and the others are new substances. These starting compounds can be obtained in a manner known per se from the derivatives of carboline and azepino- [3,4-b] indole.
To better illustrate the present invention, the following examples are given for carrying out the process for the preparation of the registered compounds.
Example 1 illustrates the preparation of the starting lactams and their hydrogen chlorides, which correspond to formula II.
Example 1 a) Preparation of 1-oxo-10- (3! -Dimethylaminoethyl) -lH-2,3,4,5-tetrahydroazepino- [3,4-b] indole of the formula II, in which m is 2; n for 3; R for H; R 'stands for CH3.
A suspension of 20 g of 1-oxo-1H, 2,3,4,5-tetrahydro azepino- [3,4-b] -indole in 120 ml of dry dimethylformamide is added in portions at a temperature of 25 C and 3 g of sodium hydride heated at 30 C for 2 hours and at 50 C for 30 minutes. 14 g of P-dimethylaminoethyl chloride are added dropwise to the reaction mass, the mixture is heated at 50 to 55 ° C. for 4 hours and left to stand overnight. The mixture is poured into 100 ml of 50% acetic acid, filtered, the solution is made alkaline with 15% aqueous ammonia to a pH value of about 9 and extracted with benzene. The extract is washed with water, dried with red hot potash, evaporated, the residue triturated with ether and dried. 14.2 g of the compound of the formula II are obtained.
