US-Patentschrift 3 867398).
The present invention relates to a process for the preparation of 3-hydroxy-1,2,4-triazole derivatives of the formula I.
EMI1.1
in which R, and R2 each represent an alkyl group with 1 to 6 carbon atoms.
The 3-hydroxy-1,2,4-triazole derivatives of the formula I are valuable intermediates for the preparation of phosphoric acid esters which can be used as pesticides, in particular as insecticides and nematicides. Such phosphoric acid esters and their use are described in US Pat. No. 3,867,398.
It is known that 3-hydroxy-1,2,4-triazole derivatives of the formula I can be prepared by reacting corresponding 1-alkyl-3-hydroxy-5-chloro-1,2,4-triazoles with alkyl mercaptides (cf. US patent specification 3 867398). The 1-alkyl-3-hydroxy-5-chloro-1,2,4-triazoles are in turn obtained by reacting a 1-alkyl semicarbazide with formic acid to give the corresponding 1-alkyl-3-hydroxy-1,2,4-triazole (Ber . German Chem.
Ges. 26, 2613, [1893]) and this is then chlorinated in the 5th position (see US Pat. No. 3,867,396). This known process not only requires a large number of reaction stages, but it is also unsatisfactory in terms of the yields that can be achieved.
It has now been found that the 3-hydroxy-1,3,4-triazole derivatives of the formula I can be prepared in a simple manner if a 1-alkyl-1-cyanohydrazine of the formula II
EMI1.2
in which Rl has the meaning given under formula I, an alkyl mercaptan of the formula III
R2-SH (III), in which R2 has the meaning given under formula I, is added on and the addition product is then added by reaction with a compound of formula IV
EMI1.3
in which R3 and R4 are independently chlorine or an alkoxy group having 1 to 4 carbon atoms, cyclizes.
The process according to the invention is advantageously carried out in an inert solvent, for example in a lower alkanol or in water. Particularly suitable lower alkanols are those having 1 to 4 carbon atoms, such as methanol, ethanol, propanol, isopropanol and butanol. The reaction can also be carried out in a two-phase system consisting of water and an inert water-immiscible solvent, in particular chlorinated hydrocarbons, such as methylene chloride, chloroform, carbon tetrachloride and tetrachloroethane.
The reaction of a 1-alkyl-1-cyanohydrazine of the formula II with an alkyl mercaptan of the formula III can be carried out in the presence of acids, for example hydrogen chloride, or in the presence of bases, for example alkali metal hydroxides such as sodium hydroxide or potassium hydroxide.
The process according to the invention is preferably carried out in such a way that a solution of the alkyl mercaptide is initially introduced and the 1-alkyl-1-cyanohydrazine is added dropwise to this solution at temperatures between 0 and 50 ° C., preferably 15 to 30 ° C. The addition product formed in this way is advantageously cyclized immediately afterwards without isolation by reaction with phosgene or a lower alkyl chloroformate or a lower carbonic acid ester. The cyclization is preferably carried out at temperatures between 20 and 100 "C., the reactants first being combined at low temperatures of about 20 to 50" C. and the reaction being brought to an end by subsequent heating.
The 3-hydroxy-1,2,4-triazole derivatives of the formula I can be isolated from aqueous reaction mixtures after acidification either by crystallization or by extraction. The end products can either be precipitated from alcoholic reaction mixtures by adding glacial acetic acid or, after the alcohol has been evaporated off, obtained by extracting the residue. Chlorinated hydrocarbons, such as methylene chloride, chloroform, carbon tetrachloride and tetrachloroethane, are particularly suitable solvents both for the extraction from aqueous reaction mixtures and for the extraction of the residue obtained after the alcohol has been evaporated off.
The 1-alkyl-1-cyanohydrazines of the formula II used as starting material can be obtained by reacting a corresponding alkylhydrazine with cyanogen chloride in the presence of an acid-binding agent. The reaction is advantageously carried out in a two-phase reaction medium consisting of water and a water-immiscible organic solvent, in particular lower, halogenated hydrocarbons, in particular methylene chloride, being suitable solvents. The reaction is advantageously carried out at temperatures between 0.degree. C. and 30.degree. Particularly suitable acid-binding agents are alkali metal hydroxides, carbonates and, in particular, hydrogen carbonates.
The reaction is carried out, for example, by adding an aqueous solution of the acid-binding agent to a solution of a corresponding alkylhydrazine in methylene chloride and adding or introducing cyanogen chloride in drops with vigorous stirring.
After the reaction has ended, the 1-alkyl-1-cyanohydrazine formed can be isolated by separating off the aqueous phase and evaporating the organic solvent in vacuo.
The process according to the invention makes it possible to prepare the 3-hydroxy-1,3,4-triazole derivatives of the formula I in a particularly simple manner in very good yields.
The process according to the invention is explained in more detail by the following examples: Example 1
19.8 g (0.2 mol) of 1-1sopropyl-1-cyanohydrazine are added dropwise to a solution of 16.0 g (0.4 mol) of sodium hydroxide and 12.4 g (0.2 mol) of ethyl mercaptan in 75 ml of water .
19.8 g (0.2 mol) of phosgene are passed into the resulting white emulsion at room temperature, an exothermic reaction taking place. The reaction mixture, cooled to room temperature, from which a white-yellow precipitate had separated out, is extracted with methylene chloride and the extract is dried with sodium sulfate -50 "C. 1-1sopropyl-3-hydroxy-5-ethylthio-1,2,4-triazole with a melting point of 109 to 114 ° C. is obtained.
Example 2
19.8 g (0.2 mol) of 1-1sopropyl-1-cyanohydrazine are added dropwise to a solution of 8.0 g (0.2 mol) of sodium hydroxide and 12.4 g (0.2 mol) of ethyl mercaptan in 150 ml of water , with a weak exothermic reaction taking place. Thereafter, 21.7 g (0.2 mol) of ethyl chloroformate are added dropwise at 20 to 25 ° C. and the mixture is then refluxed for 1112 hours. When the reaction mixture cools to room temperature, an oil separates out. The reaction mixture is mixed twice with 150 ml of chloroform extracted, the extract dried over sodium sulphate and the solvent evaporated in vacuo. The white crystalline residue is dissolved in 75 ml of methanol, treated with animal charcoal and crystallized at -70 ° C.
1-Isopropyl-3-hydroxy-5-ethylthio-1,2,4-triazole with a melting point of 109 to 113 ° C. is obtained.
