Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Triazolderivaten und ihren Säureadditionssalzen.
Die erfindungsgemäss erhältlichen neuen Triazolderivate entsprechen der allgemeinen Formel I,

in welcher
R, Wasserstoff, Niederalkyl, gegebenenfalls verestertes oder veräthertes Hydroxymethyl, gegebenenfalls acetalisiertes Formyl, verestertes Carboxy, Cyano, gegebenenfalls monooder disubstituiertes Carbamoyl oder mono- oder disubstituiertes Aminomethyl, die Ringe A und B unabhängig voneinander unsubstituiert oder substituiert sein können.
Zum Gegenstand der Erfindung gehören auch die Additionssalze der Triazolderivate der allgemeinen Formel I mit anorganischen und organischen Säuren.
In den Triazolderivaten der allgemeinen Formel I ist R, als Niederalkyl z. B. Aethyl, Propyl, Butyl, Isobutyl, Pentyl, Isopentyl, Neopentyl (2,2-Dimethylpropyl), Hexyl oder Isohexyl und vor allem Methyl, als verestertes Hydroxymethyl z. B.
Niederalkanoyloxymethyl, wie Formyloxymethyl, Propionyloxymethyl, Butyryloxymethyl, Pivaloyloxymethyl und vor allem Acetoxymethyl, als veräthertes Hydroxymethyl z. 13. Niederalkoxymethyl, wie Methoxy-, Aethoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-, Sek-butoxy-, Pentyloxy-, Isopentyloxy-, Neopentyloxy-, Hexyloxy- oder Isohexyloxy-methyl und als acetalisiertes Formyl z. B. Di-(niederalkoxy)-methyl, wie Dimethoxymethyl, Diäthoxymethyl, Dipropoxymethyl oder Dibutoxymethyl. Als verestertes Carboxy ist R1 insbesondere Niederalkoxycarbonyl, wie Propoxycarbonyl, Butoxycarbonyl, Isobutyloxycarbonyl, Pentyloxycarbonyl, Isopentyloxycarbonyl, Hexyloxycarbonyl und vor allem Methoxycarbonyl oder Aethoxycarbonyl.
Substituenten von Carbamoylgruppen R1 wie auch von Aminomethylgruppen R1 sind einerseits vor allem einwertige Kohlenwasserstoffreste mit höchstens 10 Kohlenstoffatomen, wie Niederalkyl, z. B. Propyl, Isopropyl, Butyl, Isobutyl, Sek.butyl, Pentyl, Isopentyl, Neopentyl, Hexyl oder Heptyl, und insbesondere Methyl oder Aethyl, oder araliphatische Kohlenwasserstoffreste mit 7 bis 10 Kohlenstoffatomen, z. B. Benzyl, Phenäthyl, cr-, o-, m- oder p-Methylbenzyl, 3 Phenylpropyl, 2-Phenylpropyl, xx-Methylphenäthyl, 4-Phenylbutyl oder p-Isopropylbenzyl; und anderseits zwei unter sich direkt oder in ss- oder y-Stellung auch über Sauerstoff, Schwefel, den Iminorest oder einen Niederalkyliminorest verbundene Niederalkylreste, die zusammen mit dem anliegenden Stickstoffatome z.
B. die 1-Aziridinyl, 1-Azetidinyl-, 1-Pyrrolidinyl-, Piperidino-, Hexahydro-1H-azepin-1-yl-, Morpholino-, Thio morpholino-, 1-Piperazinyl- oder Hexahydro-1H-1,4-diazepin- 1-yl-gruppe bilden, wobei die beiden letztgenannten Gruppen in 4-Stellung, d.h. in der Iminogruppe z.B. durch Methyl, Aethyl, Propyl, Isopropyl, Butyl oder Isobutyl, und alle vorgenannten cyclischen Gruppen an Kohlenstoffatomen noch durch Aethyl, Propyl oder insbesondere Methyl substituiert sein können.
Soweit vor- und nachstehend von niederen Gruppen die Rede ist, werden darunter solche mit höchstens 7 Kohlenstoffatomen und vorzugsweise höchstens 4 Kohlenstoffatomen verstanden.
Die Ringe A und B können je für sich mehrfach substituiert sein, doch ist Ring A vorzugsweise monosubstituiert und Ring B vorzugsweise unsubstituiert oder disubstituiert und vor allem monosubstituiert. Bei den Substituenten handelt es sich beispielsweise um Halogen bis Atomnummer 35, Trifluormethyl, Nitro, Niederalkyl qder Niederalkoxy. Ein Substituent des Ringes A befindet sich vorzugsweise in 4-Stellung zum Triazolring, und der bzw. die Substituenten des Ringes B in der bzw. den beiden ortho-Stellungen zur Carbonylgruppe.
Halogenatome als Substituenten der Ringe A und B sind Fluor-, Chlor- oder Bromatome, während als Niederalkyl beispielsweise Methyl, Aethyl, Propyl, Isopropyl, Butyl, Isobutyl, Tert.butyl, Pentyl, Isopentyl, Neopentyl, Tert.pentyl, Hexyl oder Heptyl und als Niederalkoxy z. 13. Methoxy, Aethoxy, Propoxy, Isopropoxy, Butoxy, Isobutoxy, Pentyloxy, Isopentyloxy, Hexyloxy oder Heptyloxy in Betracht kommen. Ein vorzugsweise in 4-Stellung zum Triazolring befindlicher Substituent des Ringes A ist insbesondere eines der genannten Halogenatome, vor allem Chlor, im weiteren Nitro oder Trifluormethyl. Der Ring B ist vorzugsweise unsubstituiert oder durch Fluor, Chlor, Brom oder Trifluormethyl in beliebiger Stellung, insbesondere jedoch durch Fluor oder Chlor in o- Stellung substituiert.
Die Triazolderivate der allgemeinen Formel I und ihre Additionssalze mit anorganischen und organischen Säuren besitzen wertvolle pharmakologische Eigenschaften. Insbesondere sind sie antikonvulsiv wirksam, wie z. B. an der Maus im Pentetrazolkrampf-Test nach Verabreichung von oralen Dosen ab ca. 0,3 mg/kg, sowie im Strychninkrampf-Test und im Elektroschock-Test nach Verabreichung von oralen Dosen ab je ca. 1 mg/kg, beispielsweise von N,N-Dimethyl-1- [2-(ofluorbenzoyl)- 4-chlor-phenyl]- 5-[(dimethylamino)-methyl] 1H-1,2,4-triazol-3-carboxamid, feststellbar ist. Überdies besitzen die Triazolderivate der allgemeinen Formel I und ihre Säureadditionssalze auch mässige zentraldämpfende Wirksamkeit.
Die genannten und weitere Wirkungsqualitäten, welche durch ausgewählte Standardversuche [vgl. W. Theobald und H.A. Kunz, Arzneimittelforsch.13, 122 (1963) sowie W.
Theobald et al., Arzneimittelforsch. 17, 561 (1967)] erfasst werden können, charakterisieren die Triazolderivate der allgemeinen Formel I und ihre pharmazeutisch annehmbaren Additionssalze mit anorganischen und organischen Säuren als Wirkstoffe für Antikonvulsiva und Psychosedativa (Tranquilizers), die z.B. zur Behandlung der Epilepsie sowie von Spannungs- und Erregungszuständen anwendbar sind. Ferner eignen sich verschiedene Triazolderivate der allgemeinen Formel I auch als Zwischenprodukte zur Herstellung von weiteren unter diese allgemeine Formel fallenden Verbindungen.
Die Erfindung betrifft insbesondere solche Triazolderivate der allgemeinen Formol I, in denen R1 die unter dieser Formel angegebene Bedeutung haben, Ring A in 4-Stellung zum Triazolring durch Halogen bis Atomnummer 35, insbesondere Chlor, oder durch Nitro oder Trifluormethyl, und Ring B unsubstituiert oder in beliebiger Stellung durch Halogen bis Atomnummer 35 oder Trifluormethyl, vorzugsweise aber durch Fluor oder Chlor in ortho-Stellung substituiert ist.
Im Rahmen der allgemeinen Formel 1 - als Triazolderivate der allgemeinen Formel Ia - wie auch innerhalb der vorstehend definierten engeren Verbindungsgruppe sind hinsichtlich ihrer pharmakologischen Eigenschaften Triazolderivate von besonderer Bedeutung, in denen R1 eine Gruppe der Teilformel Iaa ist,

Triazolderivate der allgemeinen Formel I, in denen R1 Hydroxymethyl, Carboxy oder Niederalkoxycarbonyl bedeutet, und die Ringe A und B, wie unter der Formel I angegeben, vorzugsweise jedoch in der weiter oben spezifizierten Weise substituiert sein können, sind vor allem wichtig als Zwischenprodukte für die Herstellung von anderen, unter die allgemeine Formel I fallenden sowie von weiteren pharmakologisch wertvollen Verbindungen.
Die vorstehenden Ausführungen beziehen sich ebenfalls auf die Additionssalze der genannten, unter die allgemeine Formel I fallenden Triazolderivate mit anorganischen und organischen Säuren, vor allem auf die pharmazeutisch annehmbaren Säureadditionssalze.
Die neuen Triazolderivate der allgemeinen Formel I und ihre Säureadditionssalze stellt man erfindungsgemäss her, indem man eine Verbindung der allgemeinen Formel II,

in welcher RX die unter der Formel I angegebene Bedeutung hat und die Ringe A und B, wie dort angegeben, substituiert sein können, mit Formaldehyd und Ameisensäure umsetzt, und ein erhalten es Triazolderivat der allgemeinen Formel I als freie Base oder als Additionssalz einer anorganischen oder organischen Säure isoliert.
Bei der Spaltung des Diazepinringes mittels Formaldehyd und Ameisensäure wird nicht nur die zuvor zum Diazepinring gehörige Aminogruppe dimethyliert, sondern es wird auch ein gegebenenfalls vorhandenes Aminomethyl oder monosubsti tuiertes Aminomethyl R1 dimethyliert bzw. monomethyliert.
Die verfahrensgemässe Spaltung wird bei erhöhter Temperatur, vorzugsweise bei ca. 80" C bis Siedetemperatur des Reaktions gemisches durchgeführt. Obwohl gemäss der Gesamtbilanz der
Reaktion nicht unbedingt notwendig, ist ein beträchtlicher
Wassergehalt des Reaktionsgemisches vorteilhaft, deshalb kann sowohl wässrige, z. B. 30 bis 36%-ige Formaldehydlösung, als auch wasserhaltige, z.B. 85 bis 95 %-ige Ameisensäure, verwen det werden. Vorzugsweise wird der Formaldehyd in einem erheblichen Überschuss, z. B. dem zwei- bis fünffachen der pro
Mol Ausgangsstoff der allgemeinen Formal 112 bis 4 Mol betragenden theoretischen Menge, und die Ameisensäure in noch grösserem Überschuss, z.B. der zwei- bis fünffach molaren
Menge, bezogen auf den Formaldehyd, eingesetzt.
Von den
Ausgangsstoffen der allgemeinen Formel II sind solche mit
Wasserstoff, Carboxy oder Niederalkoxycarbonyl R1 in den
Deutschen Offenlegungsschriften 2 159 527 und 2 215 993, solche mit Niederalkyl oder Wasserstoff als R1 in den Deut schen Offenlegungsschriften 2 055 889 und 2 159 527, solche mit gegebenenfalls mono-oder disubstituiertem Aminomethyl oder mit Hydroxymethyl Rt in der Deutschen Offenlegungs schrift 2 234 652, und solche mit gegebenenfalls mono- oder disubstituiertem Carbamoyl oder mit Formyl R1 in der Deut schen Offenlegungsschrift 2 304 307 beschrieben. Weitere
Verbindungen der allgemeinen Formel IV lassen sich analog den in den vorgenannten Offenlegungsschriften beschriebenen herstellen.
Zu Ausgangsstoffen der allgemeinen Formel II mit einer acetalisierten Formylgruppe, insbesondere Di-(niederalkoxy) methyl als R1 gelangt man beispielsweise durch Umsetzung von entsprechenden Verbindungen mit Formyl als R1, von denen einzelne Vertreter als Zwischenprodukte in der Deutschen
Offenlegungsschrift 2 304 307 beschrieben wurden und weitere analog zu diesen herstellbar sind, mit der mindestens doppelt molaren Menge einer Monohydroxyverbindung, insbesondere eines Niederalkanols, vorzugsweise in einem grösseren Über schuss desselben als Reaktionsmedium in Gegenwart eines sauren Katalysators, z.B. von Schwefelsäure, Phosphorsäure, p
Toluolsulfonsäure oder Bortrifluorid bei Temperaturen zwischen ca. 200 und 1700 C, vorzugsweise bei Siedetempera tur des Reaktionsgemisches.
Je nach Verfahrensbedingungen und Ausgangsstoffen erhält man die Endstoffe in freier Form oder in der ebenfalls in der
Erfindung inbegriffenen Form ihrer Säureadditionssalze oder gegebenenfalls auch als Hydrate der letzteren. Die Säureaddi tionssake der neuen Verbindungen der allgemeinen Formel I können in an sich bekannter Weise in die freien Basen überge führt werden, z.B. mit basischen Mitteln, wie Alkalien oder
Ionenaustauschern. Andererseits können die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I gewünschtenfalls in üblicher Weise in ihre
Additionssalze mit anorganischen oder organischen Säuren übergeführt werden. Beispielsweise versetzt man eine Lösung einer Verbindung der allgemeinen Formel I in einem organi schen Lösungsmittel mit der als Salzkomponente gewünschten 1Säure.
Vorzugsweise wählt man für die Umsetzung organische
Lösungsmittel, in denen das entstehende Salz schwer löslich ist, damit es durch Filtration abgetrennt werden kann. Solche
Lösungsmittel sind z.B. Aethylacetat, Methanol, Aether,
Aceton, Methyläthylketon, Aceton-Aehter, Aceton-Aethanol,
Methanol-Aether oder Aethanol-Aether.
Zur Verwendung als Arzneistoffe können anstelle freier
Basen pharmazeutisch annehmbare Säureadditionssalze eingesetzt werden, d.h. Salze mit solchen Säuren, deren
Anionen bei den in Frage kommenden Dosierungen nicht toxisch sind. Ferner ist es von Vorteil, wenn die als Arzneistoffe zu verwendenden Salze gut kristalüsierbar und nicht oder wenig hygroskopisch sind. Zur Salzbildung mit Verbindungen der allgemeinen Formel I können z. B. die Chlorwasserstoffsäure,
Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methan sulfonsäure, Aethansulfonsäure, 2-Hydroxyäthansulfonsäure,
Essigsäure, Milchsäure, Bernsteinsäure, Fumarsäure, Malein säure, Aepfelsäure, Weinsäure, Citronensäure, Benzoesäure,
Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure verwendet werden.
Die neuen Verbindungen können je nach der Wahl der Aus gangsstoffe und Arbeitsweisen als optische Antipoden oder Racemate oder, sofern sie mindestens zwei asymmetrische
Kohlenstoffatome enthalten, auch als Isomerengemische (Racematgemische) vorliegen. Erhaltene Isomerengemische (Racematgemische) können aufgrund der physikalisch chemischen Unterschiede der Bestandteile in bekannter Weise in die beiden stereoisomeren (diastereomeren) reinen Racemate aufgetrennt werden, beispielsweise durch Chromatographie und/oder fraktionierte Kristaffisation.
Erhaltene Racemate lassen sich nach bekannten Methoden, beispielsweise durch Umkristallisation aus einem optisch aktiven Lösungsmittel, mit Hilfe von Mikroorganismen oder durch Umsetzen mit einer mit der racematischen Verbindung Salze bildenden optisch aktiven Säure und Trennung der auf diese Weise erhaltene Salze, z. B. auf Grund ihrer verschiedenen Löslichkeiten, in die Diastereomeren, aus denen die Antipoden durch Einwirkung geeigneter Mittel freigesetzt werden können, zerlegen. Besonders gebräuchliche optisch aktive Säuren sind z. 13. die D- und L-Formen von Weinsäure, Di-o-Toluylweinsäure, Aepfelsäure, Mandelsäure, Camphersulfonsäure oder Chinasäure. Vorteilhaft isoliert man den wirksameren der beiden Antipoden.
Die neuen Wirkstoffe werden peroral, rektal oder parenteral verabreicht. Die Dosierung hängt von der Applikationsweise, der Spezies, dem Alter und von dem individuellen Zustand ab. Die täglichen Dosen der freien Basen oder von pharmazeutisch annehmbaren Salzen derselben bewegen sich zwischen 0,1 mg/kg und 3 mg/kg für Warmblüter. Geeignete Doseneinheitsformen, wie Dragees, Tabletten, Suppositorien oder Ampullen, enthalten vorzugsweise 0,5-50 mg eines erfindungsgemässen Wirkstoffes.
Doseneinheitsformen können in an sich bekannter Weise hergestellt werden.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I sowie von bisher nicht bekannten Ausgangsstoffen näher, sollen jedoch den Umfang der Erfindung in keiner Weise beschränken. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu der Lösung von 6,0 g (0,0203 Mol) 6-Phenyl-8-chlor4H-s-triazolo [1,5-a] [1,4]-benzodiazepin (siehe Deutsche OffenlegungsschriftNr. 2 159 527) in 27 ml 85 %-iger Amei- sensäure gibt man 13,7 ml 36%-ige wässrige Formaldehydlösung und erwärmt das Gemisch unter Rühren 2 Stunden auf 100 ". Hierauf lässt man die Reaktionslösung auf Raumtemperatur abkühlen, giesst sie auf Eiswasser und fügt konz. Natriumhydroxidlösung zu, bis der pH-Wert 11 erreicht. Das ausgefallene Rohprodukt wird in Aether aufgenommen. Die organische Phase wäscht man zweimal mit Wasser und einmal mit gesättigter Natriumchloridlösung, trocknet sie über Natriumsulfat und dampft sie im Vakuum ein. Den Rückstand löst man in 100 ml Aethylacetat und fügt ätherische Salzsäurelösung zu, bis der pH-Wert 3 erreicht.
Das Hydrochlorid fällt kristallin an. Nach 2 Stunden wird abgenutscht und mit Aether und Hexan gewaschen. Man erhält das 1-(2-Benzoyl-4-chlorphenyl)- 5-[(dimethylamino)-methyl]- 1H-1,2,4-triazol-hydrochlorid vom Smp. 182-185 " unter Zersetzung.
In analoger Weise erhält man den 1-(2-Benzoyl-4-chlorphenyl)-5- [(dimethylamino)methyl]- 1H-1,2,4-triazol-3- carbonsäure-methylester vom Smp.118-121 ; den 1-[2-(o-Chlorbenzoyl)-4-chlorphenyl)-5 - [(dimethylamino)-methyl]- 1H-1,2,4-triazol-3- carbonsäure-methylester vom 131-134" die rohe 1 -[2-(o-Chlorbenzoyl)-4-chlorphenyl] -5- [(dimethyl amino)-methyl]- 1H-1,2,4-tnazol-3 lH-1,2,4-triazol-3-carbonsäure;
Zur Überführung in das Hydrochlorid wird eine Probe der Carbonsäure in Aethanol mit einer gesättigten Lösung von Chlorwasserstoff in Aether versetzt, bis der pH-Wert des Gemisches nach Zusatz von Wasser 3 erreicht.
Nach Zusatz von Aethylacetat kristaffisiert das Salz aus. Es wird abgenutscht und mit Aethylacetat und Hexan gewaschen. Nach dem Trocknen schmilzt das erhaltene 1-[2-(o-Chlorbenzoyl)-4-chlorphenylj- 5-(dimethylamino-methyl)-1H- 1,2,4-triazol-3-carbonsäure- hydrochlorid bei 220" unter Zersetzung; das rohe 1-(2-Benzoyl-4-chlorphenyl)-5- [(dimethylamino)methyl] 1H-1,2,4-triazol- 3-methanol als erstarrten Schaum;
Eine Probe des Rohproduktes wird in Aether gelöst und mit ätherischer Chlorwasserstoff-Lösung versetzt, bis der pH-Wert 3 erreicht ist. Das kristallin anfallende Hydrochlorid wird abgenutscht und schmilzt nach dem Trocknen bei 198-200 unter Zersetzung.
den rohen 1-(2-Benzoyl-4-chlorphenyl)-5- [(dimethylamino)methyl]- 1H-1,2,4-triazol- 3-carboxaldehyd als erstarrten Schaum;
Eine Probe des Aldehyds wird in Aceton gelöst und mit einer gesättigten Lösung von Oxalsäure in Aceton versetzt, bis der pH-Wert 3 erreicht ist. Das Oxalat fällt kristallin aus. Es wird abgenutscht, gut mit Aceton und Aether gewaschen und Vakuum getrocknet. Smp. 178-181" unterAufschäumen; das rohe [[1-(2-Benzoyl-4-chlorphenyl)-5 [(dimethylamino)methyl] -1H-1,2,4-triazol-3-yl]- methyl]-acetat als schwach gelbes Ö1;
Eine Probe des Rohproduktes wird in Aether gelöst und mit ätherischer Chlorwasserstoff-Lösung versetzt, bis der pH-Wert 3 erreicht ist.
Das kristallin anfallende Hydrochlorid wird abgenutscht und zeigt nach dem Trocknen einem Smp. von 166170.
Beispiel 2
3,4 g (0,001 Mol) 6-Phenyl-8-chlor-4H-s- triazolo[1,5-a] [1,4]-benzodiazepin-2-carbonsäure (siehe Deutsche Offen legungsschriften Nr. 2 159 527 und 2 215 943) werden versetzt mit 13,5 ml 85 %-iger Ameisensäure und 6,7 ml 36%-iger wässriger Formaldehydlösung. Anschliessend erwärmt man das Reaktionsgemisch unter Rühren 2 Stunden auf 100 , wobei das Ausgangsprodukt klar in Lösung geht. Nach dem Abkühlen gibt man konz. Natriumhydroxidlösung zu, bis der pH-Wert 5 erreicht, und dampft das Gemisch im Vakuum zur Trockne ein.
Der Rückstand wird zur totalen Trocknung noch dreimal mit einem Gemisch von Toluol-Methanol (1:1) im Vakuum bei 60 eingedampft. Der Rückstand wird dreimal mit warmem Methylenchlorid digeriert. Die organische Phase wird im Vakuum eingedampft und der Rückstand in 300 mlAethylacetat gelöst. Die trübe Lösung wird mit Kohle behandelt und über Hyflo filtriert. Das klare Filtrat wird bis zur kongosauren Reaktion mit einer Lösung von Chlorwasserstoff in Aether versetzt. Die ausgefallenen Kristalle werden abgenutscht und mit Aethylacetat und Aether gewaschen. Man erhält das 1-(2 Benzoyl-4-chlorphenyl)-5 - [(dimethylamino)-methyl]- 1H- 1,2,4-triazol-3- carbonsäure-hydrochlorid vom Smp. 160-165" unter Zersetzung.

Zu der Lösung von 19,2 g (0,050 Mol) N,N-Dimethyl-6- (o.
fluorphenyl)- 8-chlor-4H-s-triazolo [1 ,5-a] [1,4]benzo- diazepin-2-carboxamid (siehe Deutsche Offenlegnngsschrift Nr. 2 304 307) in 68 ml 85%-iger Ameisensäure gibt man 35 ml 36%-ige wässrige Formaldehydlösung und erwärmt das Gemisch unter Rühren 2 Stunden auf 1000. Hierauf kühlt man die Reaktionslösung auf Raumtemperatur ab, giesst sie auf Eiswasser und fügt konz. Natriumhydroxidlösung zu, bis der pH Wert 11 erreicht. Das ausgefallene Rohprodukt wird in Methylenchlorid aufgenommen. Die Methylenchlondlösung wäscht man zweimal mit Wasser und einmal mit gesättigter Natriumchloridlösung, trocknet sie über Natriumsulfat und dampft sie im Vakuum ein.
Den Rückstand kristallisiert man aus Isopropanol um und erhält nach dem Trocknen im Vakuum das N,N-Dimethyl-1 - [2-(o-fluorbenzoyl)- 4-chlorphenyl]-5 [(dimethylamino)-methyl]- 1H- 1,2,4-triazol-3- carboxamid vom Smp. 140-142".
In analoger Weise erhält man das 1-(2-Benzoyl-4-chlorpheny -5- [(dimethylamino)methyl]- lH-1,2,4-triazol-3- carboxamid-hydrochlorid vom Smp. 250-255" unter Zersetzung; das 1-(2-Benzoyl-4-nitrophenyl)-5- [(dimethylamino)-methyl]- 1H-1,2,4-triazol-3- carboxamid vom Smp.187-190 ; das 1-(2-Benzoyl-4-chlorphenyl)-5 [(dimethylamino)- methyl]- lH-1,2,4-triazol-3- carboxamid-hydrochlorid vom Smp. 250" unter Zersetzung; das 1-[2-(o-Chlorbenzoyl)-4-chlorphenyl]-5- [(dimethylamino)-methyl]- 1H-1,2,4-triazol-3- carboxamid vom Smp.

Zu der Lösung der aus 4,30 g (0,010 Mol) 2-[(Methylamino)-methyl]- 6-(o-chlorphenyl)- 8-chlor-4H-s-triazolo [1,5-a] [1,4]benzodiazepin-fumarat-(2: 1) (siehe Deutsche Offenlegungsschrift 2 234 652) freigesetzten Base in 13,5 ml (ca. 0,25 Mol) 85 %-iger Ameisensäure gibt man 8,35 ml (ca.
0,10 Mol) 36%-ige wässrige Formaldehydlösung und erwärmt das Gemisch unter Rühren 2 Stunden auf 100 . Hierauf kühlt man die Reaktionslösung auf Raumtemperatwxr ab, giesst sie auf Eiswasser und fügt konz. Natriumhydroxidlösung zu, bis der pH-Wert 11 erreicht. Das ausgefallene Rohprodukt wird in Aether aufgenommen. Die organische Phase wäscht man zweimal mit Wasser und einmal mit gesättigter Natriumchloridlösung, trocknet sie über Natriumsulfat und dampft sie im Vakuum ein. Den öligen Rückstand löst man in 100 ml Aethanol und fügt ätherische Chlorwasserstofflösung zu, bis den pH-Wert 2 erreicht Hierauf destilliert man das Aethanol im Vakuum vollständig ab und versetzt den Rückstand mit Aethyl acetat. Das kristallin anfallende Produkt wird abgenutscht und mit Aethylacetat und Aether gewaschen.
Nach dem Trocknen im Vakuum erhält man das 1-[2-(o-Chlorbenzoyl)-4-chlorphenyl]- 3,5-bis-[(dimethylamino)- methyl]-iH-1,2,4-triazol dihydrochlorid vom Smp. 100-105" unter Zersetzung.
In analoger Weise erhält man das 1-(2-Benzoyl-4-chlorphenyl)-3,5-bis [(dimethylamino)methyl]- 1H-1,2,4-triazol-dihydrochlorid vom Smp. 900 (unter allmählicher Zersetzung); das 1 -[2- (o-Chlorbenzoyl)-4-chlorphenyl] -5 - [(dimethylamino)-methyl]- 1H-1,2,4-tnazol-3-carbonitril vom Smp.

The present invention relates to a process for the preparation of new triazole derivatives and their acid addition salts.

The triazole derivatives of the general formula I and their addition salts with inorganic and organic acids have valuable pharmacological properties. In particular, they are anticonvulsant, such. Theobald et al., Arzneimittelforsch. 17, 561 (1967)] characterize the triazole derivatives of the general formula I and their pharmaceutically acceptable addition salts with inorganic and organic acids as active ingredients for anticonvulsants and psychosedativa (tranquilizers), which e.g. for the treatment of epilepsy as well as states of tension and excitement are applicable. Furthermore, various triazole derivatives of the general formula I are also suitable as intermediates for the preparation of further compounds falling under this general formula.
The invention relates in particular to those triazole derivatives of the general formula I in which R1 has the meaning given under this formula, ring A in the 4-position to the triazole ring by halogen up to atom number 35, in particular chlorine, or by nitro or trifluoromethyl, and ring B unsubstituted or is substituted in any position by halogen up to atomic number 35 or trifluoromethyl, but preferably by fluorine or chlorine in the ortho position.

To the solution of 4.30 g (0.010 mol) of 2 - [(methylamino) methyl] - 6- (o-chlorophenyl) - 8-chloro-4H-s-triazolo [1,5-a] [1, 4] benzodiazepine fumarate (2: 1) (see German Offenlegungsschrift 2 234 652) released base in 13.5 ml (approx. 0.25 mol) of 85% formic acid is added 8.35 ml (approx.
0.10 mol) of 36% aqueous formaldehyde solution and the mixture is heated to 100 for 2 hours while stirring. The reaction solution is then cooled to room temperature, poured into ice water and conc. Sodium hydroxide solution until the pH reaches 11. The precipitated crude product is taken up in ether. The organic phase is washed twice with water and once with saturated sodium chloride solution, dried over sodium sulfate and evaporated in vacuo. The oily residue is dissolved in 100 ml of ethanol and ethereal hydrogen chloride solution is added until the pH value is 2. The ethanol is then completely distilled off in vacuo and the residue is treated with ethyl acetate. The crystalline product is filtered off with suction and washed with ethyl acetate and ether.
