Bei der weiteren Entwicklung der Prostaglandinderivate hat sich inzwischen herausgestellt, dass insbesondere der neue Methylester von 4,5-cis-Didehydro-16,16-dimethyl-PGF1α und die entsprechende freie Säure in bezug auf ihre pharmakologischen Eigenschaften von besonderer Bedeutung sind.
Die pharmakologischen Eigenschaften sowie auch die Verwendungsmöglichkeiten werden im Hauptpatent ausführlich beschrieben.
Es wurde nun weiter gefunden, dass die oben genannte Verbindung nach dem Verfahren gemäss Hauptpatent hergestellt werden kann; erfindungsgemäss setzt man in Stufe (3) des Verfahrens gemäss Hauptpatent 3z, Sz-Dihydroxy- -2h-(3 sc-hydroxy-4,4-dimethyl-trans- 1 -octenyl)-1 oc-cyclopen- tanpropionaldehyd-8-lactol der Formel

Im folgenden Beispiel wurden die Infrarotabsorptionsspektren mit einem Spektrophotometer Perkin-Elmer Modell 421 aufgenommen. Falls nichts anderes angegeben, wurden unverdünnte Proben verwendet. Die Massenspektren wurden mit einem Massenspektrometer Atlas CH-4 mit einer TO-4 Quelle (Ionisationsspannung 70 ev) aufgenommen. Die NMR Spektren wurden mit einem Spektrophotometer Varian A-60 in Deuterochloroformlösung aufgenommen, mit Tetramethylsilan als innerem Standard (feldabwärts).
Präparat 1
3α,5α-Dihydroxy-2-(3-hydroxy-4,4-dimethyl-trans-1 -octe- nyl)-1α-cyclopentanproponaldehyd-α-lactol
I. Bezugnahme auf Reaktionsschema B des Hauptpatentes.
Das Ausgangsmaterial der Formel XXXVI, worin R l,l-Di- methylpentyl bedeutet, kann erhalten werden, indem man als Ausgangsmaterial die optische aktive Verbindung der Formel XXVII einsetzt. Das auf diese Weise erhaltene Dimethyl -2-oxo-3 ,3-dimethyl-heptylphosphonat wird verwendet, um das entsprechende Produkt der Formel XXXII zu erhalten: Anschliessend stellt man die Verbindung der Formel XXXVI her, nämlich 3α,5α-Dihydroxy-2ss-(3α-hYdroxy-4,4-dimethyl- -trans- 1 -octenyl)- 1 rj..cyclopentanacetaldehyd.y.lactol 3 ,3'-bis- -(Tetrahydropyranyl)- ther, in welcher Formel Rs den 1,1 -Dimethylpentylrest bedeutet.
Eine Mischung, die aus 2,77 g 59%-igem Natriumhydrid und 40 ml Dimethylsulfoxyd besteht, wird etwa 2 Stunden lang auf eine Temperatur von 65 bis 70"C erhitzt und anschliessend auf eine Temperatur von etwa 15"C abgekühlt.
Zu der klaren Lösung von Natriummethylsulfinylcarbanid, die auf diese Weise erhalten wurde, gibt man unter heftigem Rühren 23,8 g Methoxymethyltriphenylphosphoniumchlorid.
Nach etwa 10 Minuten, indem man kühlt, gibt man 15 g des weiter oben erwähnten Lactols (Verbindung Nr. XXXVI) in 30 ml Dimethylsulfoxyd hinzu, wobei die Zugabe tropfenweise geschieht und man zulässt, dass sich die Mischung auf eine Temperatur von etwa 250C erwärmt. während man etwa 16 Stunden lang rührt. Nach dem Abkühlen ergibt diese Mischung etwa 10 g an Nebenprodukten, die mit Diäthyläther gewaschen werden und welche man dann abfiltriert. Anschliessend wird das Filtrat unter vermindertem Druck konzentriert, wobei man ein ölartiges Produkt erhält. Dieses öl wird anschliessend der Chromatographie unter Verwendung von Silicagel unterworfen, und man eluiert mit Aceton- Skellysolve B (15-85).
Diejenigen Fraktionen, in welchen durch Dünnschichtchromatographie nachgewiesen werden konnte, dass sie Zwischenverbindungen der Formel XXXVII enthalten, werden vereinigt und konzentriert, wobei man dann den Enol-Äther erhält.
II. Die entstandene Zwischenverbindung der Formel XXXVII wird dann stufenweise in die im Titel erwähnte Verbindung der Formel XXXVIII umgewandelt, wobei das Verfahren nachfolgend beschrieben ist. 9.91 g des Enol-Äthers behandelt man in 500 ml Methanol mit 200 ml einer Pufferlösung, die einen pH-Wert von 2 aufweist, wobei man dann anschliessend den Methyläther des Lactols der Formel XXXVIII isoliert. Für diese Zwecke wird das Methanol teilweise entfernt, man gibt Kochsalzlösung hinzu und extrahiert die erhaltene Mischung mit Chloroform. Die Chloroformextrakte werden gewaschen, getrocknet und zu einem öl konzentriert, dessen Menge 6,72 g beträgt.
Die im Titel genannte Verbindung wird erhalten, indem man den weiter oben genannten Methyläther, der sich in 250 ml Tetrahydrofuran befindet, mit 125 ml einer Pufferlösung, die einen pH-Wert von 1,0 aufweist, bei einer Temperatur von etwa 25"C behandelt. Wenn die Reaktion vollständig ist, was durch Dünnschichtchromatographie nachgewiesen werden kann, etwa 27 Stunden, gibt man Kochsalzlösung und festes Natriumchlorid zu der Reaktionsmischung. Die organische Schicht wird gewaschen. getrocknet und konzentriert, wobei man 6,0 g des Lactols der Formel XXXVIII erhält. Die erhaltene Verbindung weist NMR-Peaks bei 5,55 und 0,89 8, sowie Ergebnisse der Infrarotabsorption bei 3380, 1665 und 975 cm-l auf.
Beispiel I
Aus 156,8 g Triphenylphosphin und 100 g 4-Brombuttersäure in 125 ml Methanol wird unter 18-stündigem Kochen am Rüdcfluss die Verbindung 3-Carboxypropyltriphenylphosphoniumbromid dargestellt. Das kristalline Produkt wird abfiltriert, mit Benzol gewaschen und aus xthanol/Acetoni- tril/ < ther umkristallisiert. Die Ausbeute beträgt 150 g, Schmelzpunkt 247-249 CC.
10,6 g des obigen Phosphoniumbromids werden zu Natriummethylsulfinylcarbanid (dargestellt aus 2,08 g Natriumhydrid [57%] und 30 ml Dimethylsulfoxyd) zugegeben und das resultierende Wittig-Reagens wird mit 0,5029 des Lactols von Präparat 1 in 20 ml Dimethylsulfoxyd vereinigt. Das Gemisch wird über Nacht gerührt, mit etwa 200 ml Benzol verdünnt und mit Kaliumbisulfatlösung gewaschen. Die beiden unteren Phasen werden mit Methylenchlorid gewaschen, die organischen Phasen werden vereinigt, mit gesättigter Natriumchloridlösung gewaschen, getrocknet und bei vermindertem Druck eingeengt. Der Rückstand wird an mit Säure gewaschenem Silikagel chromatographiert unter Eluierung mit Äthylacetat/isomeren Hexanen (3:1).
Die gemäss Dünnschichtenchromatogramm das gewünschte Produkt enthaltenden Fraktionen werden vereinigt und eingeengt und man erhält 0,856 g des 4,5-cis-Didehydro-16,16-dimethyl-PGF,, in Form eines öls.
Beispiel 2
0,717 g des nach Beispiel 1 erhaltenen sauren Produktes werden in Dichlormethan-Äthyläther mit Diazomethan umgesetzt. Man konzentriert die erhaltene Reaktionsmischung, und der Rückstand wird der Chromatographie an Silicagel unterworfen, wobei man dann mit Aceton-Dichlormethan (1:1) eluiert. Man erhält dabei 0,721 g des Methylesters der im Beispiel 1 genannten Verbindung, die die folgenden Massenspektral-Peaks aufweist (für das Trimethylsilyl-Derivat) bei 581, 522, 513, 423 und 217): die Infrarot-Absorption liegt bei 3380,.1740 und 970 cm-l; und NMR-Peaks bei 5,43, 4,393,60, 3,67, 2,34 und 0,88 6.

II.
