Die Verbindungen der Formel I haben interessante pharmakologische Eigenschaften, die insbesondere bei psychopharmakologischen und kreislaufanalytischen Untersuchungen ermittelt werden konnten.
Die Cyclopenta(c)pyrane der Formel II lassen sich, wenn R1 eine Acyloxygruppe ist, aus Didrovaltratum und seinen Homologen durch Überführung der Epoxide in die entsprechenden Hydrine darstellen.
Cyclopenta(c)pyrane, in welchen R1 eine p-D.Glycosido- gruppe ist, kommen in Wurzeln und Rhizomen von Valerianarten vor und können daraus mit polaren Lösungsmitteln, wie Wasser, Alkoholen, Aceton, Essigester oder Gemischen davon oder mit Gemischen von halogenierten Kohlenwasserstoffen und Alkoholen extrahiert werden, wobei gegebenenfalls zur Reindarstellung der Substanzen die Extrakte an Kieselgel oder Aluminiumoxid unter Verwendung von Essigester-Alkohol-Gemischen als Elutionsmittel chromatographiert oder die Extrakte einer Gelfiltration an mit organischen Lösungsmitteln quellbaren Molekularsieben unterworfen werden. Im allgemeinen kann man für die vorliegende Synthese auch von den ungereinigten Extrakten der Didrovaltrate und der analogen p-D.Glucoside ausgehen.
Voraussetzung für die im sauren, vorzugsweise alkoholischen Medium durchführbare Ringschlussreaktion der Cyclopenta(c)pyrane ist einerseits die Cis-Stellung des C-7-Hydr oxyis und des Pyranringes am Cyclopentanring und andererseits das Vorhandensein des reaktiven Allylsystems an den C-Atomen C-3, C-4 und C-8. Der Ringschluss erfolgt formal durch Verknüpfung des Hydroxylsauerstoffes am C-7 mit dem C-3 unter Allylumlagerung und Abspaltung von HX.
Da man in Gegenwart von Alkoholen und sauren Katalysatoren wie beispielsweise HCI, p-Toluolsulfonsäure oder einer Lewissäure arbeitet, wird durch Alkoholyse der Austausch der Rl-Gruppe am 0-1 durch die dem eingesetzten Alkohol entsprechende Alkoxy- oder Aralkoxygruppe bewirkt.

Die erfindungsgemäss erhaltenen Verbindungen der Formel I sind meist farblose, destillierbare öle oder gut kristallisierende Verbindungen von erstaunlicher Stabilität. Sie sind chemisch weiter abwandelbar und meist wasserlöslich.
So kann beispielsweise die Acyloxygruppe am C-4 zur Hydroxygruppe verseift werden. Ausserdem kann die Doppelbindung zwischen G10 und C-11 in üblicher Weise katalytisch hydriert werden, so dass z.B. folgende Verbindungen entstehen:

Die gleichen Verbindungen können auch direkt aus Substanzen der Formel III, in welcher R1 eine Alkoxy- oder Aralkoxygruppe, R2 Halogen oder Wasserstoff und R8 eine Acylgruppe bedeuten, erhalten werden, indem man in Alkoholen und in Gegenwart einer starken Base wie Natronlauge oder Kalilauge katalytisch hydriert, wobei Raney-Nickel der bevorzugte Katalysator ist.
Die erfindungsgemäss hergestellten Verbindungen der Formel I wirken schon in Dosen von 31,6 bis 100 mg/kg per os an Mäusen eindeutig zentraldämpfend. Die Verbindungen der Formel Ia wirken nach per os Applikation bereits in einer Dosis, die nur einen Bruchteil der LD50 beträgt, überraschend stark gefässerweiternd auf die arteriellen Gefässe, hypotensiv und zentraldämpfend ohne die Reflexerregbarkeit zu vermindern, im Gegensatz z.B. von Reserpin, das zwar hypotensiv wirkt, aber als unerwünschte Nebenerscheinungen die Reflexerregbarkeit stark herabsetzt. Die LD50 liegt bei 600 bis 2500 mg/kg per os. Daraus ergibt sich eine sehr günstige therapeutische Breite.
Die erfindungsgemäss erhaltenen Verbindungen können daher sowohl als Mittel gegen arteriosklerotische Erkrankungen als auch als Psychoparmaka eingesetzt werden, was insbesondere in der Geriatrie einen grossen therapeutischen Fortschritt bedeutet.
Die erfindungsgemäss erhaltenen Verbindungen können in Form der üblichen pharmazeutischen Zubereitungen in der Humanmedizn eingesetzt werden. Als Trägersubstanzen für die pharmazeutischen Zubereitungen kommen solche Substanzen in Frage, die gegenüber der neuen Verbindung inert sind, beispielsweise Wasser, pflanzliche öle, Gelatine, Milchzucker, Polyäthylenglykole, Stärke, Magnesiumstearat, Talk und dergleichen. Zur parenteralen Applikation eignen sich vorteilhaft Lösungen, vorzugsweise ölige und wässerige Lösungen, aber auch Suspensionen oder Emulsionen der Wirksubstanz. Für die enterale Applikation können Tabletten, Kapseln oder Dragees dienen, welche die üblichen Zusatz- und Hilfsstoffe, wie Konservierungs-, Stabilisierungsoder Netzmittel enthalten können.

20 g des so hergestellten Didrovaltratumjodhydrins

werden in 25 ml Methanol gelöst und mit 10 ml Methanol, welches 19 g HCi gelöst enthält, unter Eisaussenkühlung versetzt. Die Lösung färbt sich allmählich dunkelgrün-blau.
Nach einstündigem Stehen bei Raumtemperatur wird mit Wasser verdünnt, mit Äther erschöpfend extrahiert und die Ätherphase mit Natriumbicarbonat neutralisiert. Nach dem Trocknen über Natriumsulfat, dem Klären mit etwaa Kohle und dem Einengen im Vakuum (bei 7 mm Hg und 50"C) bis zur Gewichtskonstanz bzw. bis zum Verschwinden des Geruches nach Isovaleriansäuremethylester werden 13,8 g eines hellbraunen Öls erhalten, das in wenig Metha nol/Äther aufgenommen, beim Stehen in der Kälte allmählich kristallisiert.
Das erhaltene 4-Acetoxy-3-jodmethyl-l0-methylen-8 -methoxy-2,9-dioxatricyclo-(4,3,1,03XT)-decan liegt in Form von weissen Prismen vor und hat folgende physikalische Kenndaten: C1l7 sJ M: 380,19

Wird Didrovaltratum mit Kaliumrhodanid in Eisessig/ Natriumacetat umgesetzt, so entsteht Didrovaltratuunrho- danohydrin, das sich durch analoge Behandlung mit HC1 in Methanol unter Ringschlussbildung in 4-Acetoxy-4-rhodanomethyl-10-methylen-8-methoxy-2,9-dioxatricyclo-(4,3 ,1,- 03X7)decan mit folgenden physikalischen Konstanten umwandeln lässt: Cl4HlTOsNS M : 311,34

Durch Ersatz des Methanols durch einen anderen Alkohol, wie z.B. Äthanol, Propanol, Butanol, Benzylalkohol konnten die entsprechenden 4-Acetoxy-3 -halogenmethyl- 10- -methylen-8-alkoxy-(bzw. -aralkoxy)-2,9-dioxatricyclo-(4,3- 1,0")decane hergestellt werden.
B) Herstellung von 4-Hydroxy-3-methyl-10-methylen-8- -methoxy-2,9-dioxatricyclo-(4,3,103,7)-decan:

Zu einer Suspension von 0,08 g Raney-Nickel in 20 ml Methanol wird eine Lösung von 0,7 g 4-Acetoxy-3-jodmethyl-10-methylen-8-methoxy-2,9-dioxatricyclo-(4,3,13,7)- decan, hergestellt wie in Beispiel 1 beschrieben, in 20 ml Methanol, welches 0,5 ml Diäthylamin enthält, zugegeben und der Ansatz unter Schütteln bei Raumtemperatur und Atmosphärendruck mit Wasserstoff hydriert. Innerhalb von 90 Minuten wird ein Äquivalent H2 verbraucht. Nach Filtration über Asbest wird das Filtrat bis zur Trockene eingeengt und der Rückstand mit Chloroform extrahiert.
Aus dem mit Wasser gewaschenen Chioroformextrakt resultieren nach dem Trocknen über Magnesiumsulfat und dem Einengen im Vakuum (bei 7 mm Hg und 50 C) 0,426 g 4-Aectoxy-3-methyl-1 0-methylen-8-methoxy-2,9-dioxatri- cyclo-(4,3,1,03X7)-decan als farbloses öl, das sind 90,5% der Theorie.
Anschliessend wird das Öl in 25 ml Methanol gelöst und die Lösung mit 5 ml 1 n Natronlauge versetzt. Nach einstündigem Stehen bei Raumtemperatur wird mit Wasser verdünnt und mit Essigester extrahiert. Aus der neutralisierten und über Magnesiumsulfat getrockneten Essigesterphase können nach dem Einengen im Vakuum bei (7 mm Hg und 50"C) 0,292 g 4-Hydroxy-3-methyl-10-methylen-8-methoxy-2,9- -dioxatricyclo-(4,3,1,0327)-decan als farbloses öl gewonnen werden: CllHl6Oi M: 212,25 (a)D20 +420 (in Methanol) nD20:

Beispiel 2
Herstellung von 4-Hydroxy-3,10-dimethyl-8-methoxy-2,9- -dioxatricyclo-(4,3,1,03s?)-decan aus 4-Acetoxy-3-jodmethyl- -IO-methylen- 8-methoxy-2,9-ioxatricyclo-(4,3 ,1,033-decan:
3,8 g 4-Acetoxy-3 -jodmethyl- 1 0-methylen-8-methoxy-2,9- -dioxatricyclo-(4,3,1,03 > T)-decan werden in 25 ml Methanol gelöst, welches 0,95 g NaOH enthält.
Cirka 0,4 g methanolfeuchtes Raney-Nickel werden in 20 ml Methanol suspendiert und in die Hydrierbirne gefüllt.
Nach Absättigung des Katalysators mit Wasserstoff wird die Substanzlösung zu der Katalysatorsuspension gegeben und bei Normalbedingungen (Zimmertemperatur und 760 mm Hg Barometerstand) mit Wassertoff hydriert. Innerhalb von 45 Minuten werden insgesamt 450 mi Wasserstoff aufgenommen.
Der Ansatz wird nun über Asbest filtriert und das Filtrat im Vakuum bei 7 mm Hg und 50"C) bis zur Gewichtskonstanz eingeengt. Nach Aufnahme mit Wasser und Aussalzen mit Ammonsulfat wird mit Äther extrahiert. Die Ätherphase wird über Magnesiumsulfat getrocknet, filtriert und bis zur Trockene eingeengt. Es werden erhalten:
1,6825 g 4-Hydroxy-3,10-dimethyl-8-methoxy-2,9-dioxa- tricyclo-(4,3 1 ,03")-decan als farbloses, hochviskoses Öl, das sind 78,9% der Theorie.
Molekulargewicht: 214,26 (OC)D21 = -120 (in Methanol).
Die Substanz ist gut äther-, alkohol- und zu 2% wasserlöslich. Das IR-Spektrum ist in der Zeichnung wiedergegeben.
Wird als Ausgangsmaterial eine Verbindung gewählt, die in 8-Stellung mit einer Äthoxy-, bzw. Propoxy-, bzw.
Butoxygruppe substituiert ist, so werden die entsprechenden Alkoxyverbindungen erhalten.
Beispiel 3
Herstellung von 4-Hydroxy-3-methyl-10-methylen-8-alk- oxy-2,9-dioxatricyclo-(4,3,1,03sT)-decanen unter Verwendung von l-p-D-Glucosido-4-isovaleroxymethyl-6,7-dihydroxy-7- -methyl-l ,4a,5,6,7,7a-hexahydro-cyclopenta(c)pyran: 1 -fi-D-Glucosido-4-isovaleroxy-methyl-6,7-dihydroxy-7- -methyl-1,4a,5,6,7,7a-hexahydro-cyclopenta(c)-pyran wurde auf folgende Weise gewonnen:
6 kg gemahlene Wurzeln und Rhizome von Valeriana Wallichii wurden in einem Perkolator zunächst mit n-Hexan unter Zusatz von 1 % Essigsäure erschöpfend perkoliert.
Dieses erste Perkolat wurde verworfen. Anschliessend erfolgte eine zweite Perkolation mit Chloroform-Methanol im Verhältnis 5: 2. Das etwas eingeengte Perkolat wurde dreimal mit je 10 Liter Wasser gewaschen, die organische Phase verworfen und die Wasserphase nach Sättigung mit Ammonsulfat viermal mit je 5 Liter Essigester extrahiert. Die Esterphase wurde dann über Magnesiumsulfat getrocknet und bis zur Trockene eingeengt. Der Gesamtrückstand betrug
199 g, das sind 3,310/0 bezogen auf die getrocknete Droge.
Die vornehmlich aus wasserlöslichen Esterglykosiden bestehende Fraktion wurde zur weiteren Reinigung an der Säule chromatographiert:
10 g Essigesterrückstand wurden in cirka 100 ml feuchtem
Essigester aufgenommen und nach einigem Stehen von Un löslichem filtriert. Das Filtrat wurde auf eine Kieselgelsäule aufgetragen, welche mit 250 g Gel - Korngrösse 0,2 bis 0,5 mm - gefüllt war. Die Aufschlämmung des Kieselgels und zunächst die Elution wurden mit feuchtem Essigester durchgeführt, nachher wurde die Elution mit Essigester unter Zusatz von Methanol fortgesetzt. Fraktionen zu 100 ml wurden aufgefangen. Die erfindungsgemässe Verbindung 1 3-D-Glucosido-4-isovaleroxymethyl-6,7-dihydroxy-7-me- thyl-1,4a,5,6,7,7a-hexahydro-cyclopenta(c)pyran befand sich in den Fraktionen 17 bis 27.
Insgesamt wurden aus diesen Fraktionen nach dem Einengen 3,65 g des Glucosides als amorphes gelbliches Pulver isoliert.
Zur Ringschlussbildung wurde 1 g 1 .p-D-Glucosido-4- -isovaleroxy-methyl.6,7-dihydroxy-7-methyl-1 ,4a,5,6,7,7a- -hexahydro-cyclopenta(c)pyran in 20 ml Alkanol gelöst und die Lösung mit 50 mg p-Toluolsulfosäure versetzt. Nach kurzem Anwärmen auf 40"C wurde mit 60 ml Wasser verdünnt, mit Natriumbicarbonat neutralisiert und mit Essigester extrahiert. Der Gesamtextrakt wurde über Natriumsulfat getrocknet. Nach dem Einengen im Vakuum resultierten hellbraun gefärbte öle, die durch präparative Dickschichtchromatographie an Kieselgelplatten gereinigt wurden. Das Fliessmittelgemisch bestand aus 24 Teilen Essigester, 60 Teilen n-Hexan und aus 6 Teilen n-Propanol.
Bei grösseren Ansätzen erfolgte die Reinigung durch Säulenchromatographie an Al203 mit Diäthyläther als Elutionsmittel bzw. durch Vakuum-Destillation.
Je nach dem verwendeten Alkanol wurden folgende Verbindungen hergestellt:

3) 4-Hydroxy-3-methyl-10-methylen-8-(n-butoxy)-2,9- -dioxatricyclo-(4,3,1,03s7)-decan: C14H2sO4 M: 254,33 ( X)D20: +390 (in Methanol > nD20:

Durch einfache Hydrierung mit Palladium/Aktivkohle lassen sich aus diesen Verbindungen die in 10,11-Stellung hydrierten entsprechenden 4-Hydroxy-3,1 O-dimethyl-8-alk- oxy-2,9-dioxatricyclo-(4,3,1,03S7)-decane herstellen.

2. Verfahren nach Patentanspruch I und Unteranspruch 1, dadurch gekennzeichnet, dass als Alkohole aliphatische Alkohole mit 1 bis 4 Kohlenstoffatomen eingesetzt werden.
