Gegenstand der vorliegenden Anmeldung ist ein Verfahren zur Herstellung von neuen 21-Fluor-l9-norpregnadien-Ver- bindungen der allgemeinen Formel

worin X für Niederalkyl steht und Rl und R2 je einzeln ein Wasserstoffatom oder einen acyclischen, carbocyclischen oder carbocyclisch-acyclischen Kohlenwasserstoffrest bedeuten oder beide zusammen mit dem benachbarten Kohlenstoffatom einen cycloaliphatischen Ring bilden.
Die Substituenten Rl und R2 können gleich oder verschieden sein. Ein acyclischer Kohlenwasserstoffrest ist insbesondere ein gesättigter oder ungesättigter aliphatischer Rest mit höchstens 15 Kohlenstoffatomen, z.B. ein Alkyl-, Alkenyloder Alkinylrest. Ein carbocyclischer Kohlenwasserstoffrest ist insbesondere ein monocyclischer, cycloaliphatischer oder aromatischer Rest; ein carbocyclisch-acyclischer Rest ist insbesondere ein cycloaliphatisch-aliphatischer oder araliphatischer monocyclischer Rest. Ein cycloaliphatischer Ring ist insbesondere ein gesättigter 5- oder 6-gliedriger Ring, der auch durch Alkylreste, z.B. die unten genannten, substituiert sein kann, vor allem der Cyclopentan- und Cyclohexanring.
Als Alkylreste sind insbesondere Niederalkylreste, z.B.
Äthyl-, n-Propyl-, i-Propyl-, n-Butyl-, i-Butyl-, sec.-Butyl-, tert.-Butyl-, gerade und verzweigte Pentyl- und Hexylreste, und vor allem der Methylrest zu nennen. Als Niederalkylreste, die für X stehen, kommen insbesondere solche der genannten Reste in Betracht, welche höchstens 4 Kohlenstoffatome und vorzugsweise eine gerade Kette aufweisen, in erster Linie ein Äthyl- und vor allem ein Methylrest. Als Alkenyl- bzw. Alkinylreste kommen insbesondere diejenigen in Betracht, die den oben genannten Niederalkylresten entsprechen und eine ungesättigte Bindung tragen, z.B. Vinyl-, Allyl-, Methallyl-, Äthinyl- und Propargylreste.
Als cycloaliphatische und cycloaliphatisch-aliphatische Reste sind insbesondere diejenigen zu nennen, die sich von einem 5- oder 6-gliedrigen gesättigten Ring, der durch die oben genannten Niederalkylreste substituiert sein kann, ableiten, z.B. niedere Cycloalkylreste, wie Cyclopentyl-, 2- oder 3-Methylcyclopentyl-, 2,5- oder 3,4-Dimethylcyclopentyl-; Cyclohexyl-; 2-, 3- oder 4-Methylcyclohexylreste, und niedere Cycloalkyl-alkylreste, wie Cycloalkylmethyl und 2-(Cycloalkyl)-äthylreste, in welchen der cyclische Teil durch die obgenannten niederen Cycloalkylreste gebildet wird. Die bevorzugten aromatischen und araliphatischen Reste sind z.B. die Phenyl-, Benzyl-, 1- und 2-Phenyläthylreste, die in den Stellungen 2 bis 6 durch Niederalkylreste, z.B. die obgenannten, substituiert sein können.
Die neuen Verbindungen sind insbesondere 16,17-Ketale des entsprechenden 16a,17a-Dihydroxysteroids, die sich von Ketonen der Formel Rl-CO-R2 ableiten, worin Rl und R2 für kohlenstoffhaltige Reste der oben genannten allgemeinen und bevorzugten Bedeutungen stehen. Besonders bevorzugte Ketonkomponenten dieser Ketale sind gesättigte niederaliphatische Ketone, z.B. 2-Butanon, 2-Pentanon, 3-Pentanon, 4- Methyl-2-pentanon, 4-Heptanon, 5-Nonanon, und vor allem Aceton; gesättigte cycloaliphatische Ketone, z.B. Cyclopentanon und Cyclohexanon, und Phenylketone, z.B. Acetophenon oder Benzophenon. Das entsprechende 16a,l7a-Dihydroxy- steroid ist insbesondere das 21-Fluor-16a,17a-dihydroxy-6methyl- 19-nor-4,6-pregnadien-3,20-dion.
Die neuen Verbindungen der vorliegenden Erfindung besitzen wertvolle pharmakologische Eigenschaften. So weisen sie eine ausgeprägte gestagene und olvulationshemmende Wirkung, insbesondere bei peroraler Applikation, auf, wie sich im Tierversuch nachweisen lässt. Eine progestative Wirkung im Clauberg-Test am Kaninchen tritt bei peroraler Applikation bereits mit Dosen im Bereich von 0.01 bis 0,1 mg/kg und im Ovulationstest an der Ratte mit Dosen im Bereich von 0,01 bis 0,1 mg/kg peroral auf.
Besonders hervorzuheben ist das 21-Fluor-16a,17a-di hydroxy-6-methyl- 19-nor-4,6-pregnadien-3,20-dion- 16,17- acetonid, das in den oben erwähnten Tierversuchen eine deutliche Wirksamkeit schon bei der unteren Grenze des angegebenen Dosenbereichs zeigt.
Dank diesen Eigenschaften finden die erfindungsgemässen neuen Verbindungen ihre Anwendung in der Human- sowie Tiermedizin und als Wirkungskomponenten in Antikonzeptionsmitteln für Menschen und Säugetiere.
Die neuen Verbindungen der vorliegenden Erfindung sind aber zudem Zwischenprodukte zur Herstellung anderer nützlicher Stoffe, insbesondere pharmakologisch wirksamer Verbindungen.
Die erfindungsgemässen neuen Verbindungen können in an sich bekannter Weise hergestellt werden.
Gemäss einem allgemeinen Verfahren kann man z.B. so vorgehen, dass man eine Verbindung der Formel

worin R1, R2 und X die oben genannten Bedeutungen haben, und X in a- oder ss-Konfiguration stehen kann, oder einen 3- Enoläther davon, in der 6,7-Stellung dehydriert, wobei die gegebenenfalls vorhandene 3-Äthergruppe gleichzeitig gespalten wird.
Dies gelingt beispielsweise gemäss bekannten Dehydrierungsmethoden dadurch, dass man die Verbindung der Formel II mit einem dehydrierend wirkenden Chinon, z.B. Chloranil oder insbesondere 2,3-Dichlor-5,6-dicyan- 1,4-benzochinon, behandelt. Bei der Anwendung des ersteren arbeitet man vorzugsweise bei Siedehitze in organischen Lösungsmitteln, z.B. aromatischen Kohlenwasserstoffen, wie Benzol oder Xylol, niederaliphatischen Alkoholen, wie Äthanol, Propanol oder tert.-Butylalkohol, niederaliphatischen Ketonen, wie Aceton oder 2-Butanon, aliphatischen Estern, wie Essigester, oder cyclischen Äthern, wie Dioxan oder Tetrahydrofuran.
Bei Anwendung des Dichlordicyanbenzochinons arbeitet man vorzugsweise in Anwesenheit von Salzsäure bei oder unterhalb Zimmertemperatur in einem mit Wasser mischbaren organischen Lösungsmittel, z.B. einem der oben genannten.
Anstelle der Verbindung der Formel II kann man auch einen entsprechenden 3-Enoläther, vorzugsweise einen Niederalkyl, wie Methyl- oder Äthylenoläther, einsetzen. Der 3- Enoläther ist aus einer Verbindung der Formel II nach allgemein bekannten Methoden, vorzugsweise durch Behandeln mit einem entsprechenden Ameisensäure-orthoester unter Säurekatalyse, erhältlich. Den 3-EnoloÅather kann man ebenfalls mit einem dehydrierend wirkenden Chinon, z.B. wie oben beschrieben, behandeln, oder aber durch Einwirkung von Mangandioxid, vorzugsweise in einem halogenierten Kohlenwasserstoff, wie Chloroform oder Dichlormethan, zum gewünschten Endstoff dehydrieren.
Die Ausgangsstoffe für das Verfahren der vorliegenden Erfindung sind bekannt oder können in an sich bekannter Weise, z.B. gemäss der im Durchführungsbeispiel gezeigten Methode. hergestellt werden.
Die vorliegende Erfindung ist insbesondere für die Herstellung von pharmazeutischen Präparaten zur Anwendung in der Human- oder Veterinärmedizin sowie von antikonzeptionellen Mitteln für Menschen und Säugetiere, welche die neuen oben beschriebenen pharmakologisch wirksamen Stoffe der vorliegenden Erfindung als aktive Substanzen zusammen mit einem pharmazeutischen Trägermaterial enthalten, von Bedeutung.
Die Verbindungen der vorliegenden Erfindung können auch als Futterzusatzmittel verwendet werden.
Im folgenden Beispiel sind dieTemperaturen in Celsiusgraden angegeben.
Beispiel
Zu einer siedenden Mischung von 1,3 g 21-Fluor-16a,17isopropylidendioxy-6-methylen- 1 9-norpregn-4-en-3.20-dion, 455 mg 5 %iger Palladiumkohle und 65 ml Äthanol gibt man 9,75 ml einer 5 %igen Lösung von Benzylalkohol in Äthanol.
33/4 Stunden später wird mit einer Aufschlämmung von 455 mg 5 %iger Palladiumkohle in 65 ml Äthanol und anschliessend erneut mit 9,75 ml einer 5 %igen Lösung von Benzylalkohol in Äthanol versetzt. Nach einer Gesamtkochzeit von 5 Stunden wird durch Celite filtriert und mit Methylenchlorid nachgewaschen, das Filtrat wird mit gesättigter Kochsalzlösung gewaschen, getrocknet und im Vakuum eingedampft. Anschliessend filtriert man den Rückstand in Methylenchloridlösung durch 84,5 g Kieselgel unter Nachwaschen mit 6,4 Liter Methylenchlorid. Der Rückstand des ersten 1,6 Liters wird verworfen. Nach Eindampfen der nachfolgenden 4,8 Liter in Vakuum verbleiben 1,08 g eines kristallinen Rückstandes. Dieser wird zu 108 ml einer 2N-Lösung von Salzsäure in Dioxan gegeben, worauf man unter Rühren mit 540 mg 2,3 Dichlor-5,6-dicyan-benzochinon versetzt.
Nach 1l/2 Stunden wird abgenutscht und mit Toluol nachgewaschen. Das Filtrat wird mit Wasser, 1 %iger Natronlauge und Wasser gewaschen, getrocknet und im Vakuum eingedampft. Dann chromatographiert man den Rückstand an 30 g Kieselgel unter Verwendung von Fraktionen à 500 ml Methylenchlorid. Der Rückstand der Fraktionen 2-4 wird an 6 Platten Kieselgel à 1 m unter Verwendung eines Toluol-Aceton-(19: 1)-Gemisches als Fliessmittel aufgetrennt. Man erhält so reines 21-Fluor 16cd, 1 7-isopropylidendioxy-6-methyl- 1 9-norpregna-4,6dien-3,20-dion vom Smp. 226-2280C.
Das als Ausgangsmaterial verwendete 21 -Fluor- 1 6a, 17- isopropylidendioxy-6-methylen- 1 9-norpregn-4-en-3,20-dion wird wie folgt hergestellt:
6,56 g des in der Schweizerischen Patentschrift Nr. 577 000 beschriebenen 21 -Fluor-1 6a, 17-isopropylidendioxy-1 9-nor pregna-4,6-dien-3,20-dion, 545 mg 10%ges Palladium auf Calciumcarbonat und 327 ml Dimethylformamid werden in Wasserstoffatmosphäre bis zur Aufnahme von 1 Moläquivalent Wasserstoff geschüttelt. Nach Abnutschen vom Katalysator wird das Filtrat mit Wasser verdünnt, worauf man erneut abnutscht.
Das so erhaltene 21-Fluor-16cr,l7-isopropylidendi- oxy-19-norpregn-4-en-3,20-dion schmilzt nach Umlösen aus Methylenchlorid-Äther bei 258-261 C. 400 mg dieser Verbindung, 40 mg p-Toluolsulfonsäure, 10 ml Dioxan und 0,68 ml Orthoameisensäure-methylester werden während 16 Stunden bei Zimmertemperatur in Stickstoffatmosphäre gerührt. Dann versetzt man mit 0,32 ml Pyridin und giesst auf 40 ml Wasser und 4,8 ml gesättigte Natriumhydrogencarbonatlösung. Nach dreistündigem Rühren wird abgenutscht, mit Wasser gewaschen und im Vakuum über Phosphorpentoxid getrocknet.
Man erhält 422 mg rohes 21-Fluor-16a,17-isopropylidendi- oxy-3-methoxy- 1 9-norpregna-3,5-dien-20-on. Dieses kann durch Chromatographie an Aluminiumoxid und Kristallisation aus Äther-Methanol gereinigt werden, Smp. 176-180 C. Zu einer Mischung von 20 ml Methylenchlorid und 2,6 ml Dimethylformamid gibt man unter Rühren und Kühlen mit einer Eis-Methanol-Mischung 2,2 ml Phosphoroxychlorid. 15 Minuten später werden 3,91 g des oben erhaltenen Methoxydiens unter Nachspülen mit 5 ml Methylenchlorid zugegeben. Nach zweistündigem Rühren unter Eis-Wasserkühlung wird das Kühlbad entfernt und eine weitere Stunde gerührt.
Dann ersetzt man wieder unter Eis-Wasser-Kühlung mit einer Mischung von 8,8 g Natriumacetat und 28 ml 90%igem Methanol, 30 Minuten später wird mit Methylenchlorid verdünnt und mit 2N-Sodalösung und Wasser gewaschen. Den nach Trocknen und Eindampfen im Vakuum erhaltenen Rückstand filtriert man in Toluollösung durch 40 g Kieselgel unter Nachwaschen mit 3 Liter Toluol. Aus dem Rückstand dieses Filtrates werden durch Kristallisation aus Methylenchlorid Äther 3,5 g 21-Fluor-6-formyl-16a,17-isopropylidendioxy-3- methoxy-19-norpregna-3,5-dien-20-on erhalten. E220 = 7800; ±323 = 11 700 (Feinsprit).
Zu einer Mischung von 3,66 g dieser Verbindung und 37 ml Methanol gibt man unter Rühren eine Lösung von 460 mg Natriumborhydrid in 4,6 ml 1N Natronlauge, all/2 Stunden später lässt man unter Eis-Wasser Kühlung langsam eine Mischung von 3 ml konzentrierter Salzsäure und 1,5 ml Wasser zufliessen. Nach einer halben Stunde wird mit 4,6 g Natriumacetat und Wasser versetzt, worauf man abnutscht, mit Wasser wäscht und im Vakuum über Phosphorpentoxid trocknet. Der so erhaltene Rückstand wird mit 82,5 ml Aceton und 20,5 ml Methylenchlorid versetzt, worauf man unter Eiskühlung langsam 3,25 ml Kiliany Lösung zutropfen lässt. Nach 13/4 stündigem Rühren bei 06C wird erneut mit 0,8 ml Kiliany-Lösung versetzt. 15 Minuten später gibt man 825 ml einer 10%igen Natriumacetatlösung zu und extrahiert mit Methylenchlorid.
Nach Waschen mit Natriumhydrogencarbonatlösung und Wasser wird getrocknet und im Vakuum eingedampft. Der Rückstand wird an 90 g Kieselgel chromatographiert. Aus Toluol-Essigester-(19: 1)- Gemisch wird das 21-Fluor-16a, 17-isopropylidendioxy-6 methylen- 19-norpregn-4-en-3,20-dion eluiert. Man erhält nach Umlösen aus Methylenchlorid-Äther 1,6 g; E267 = 11 100 (Feinsprit).

The new compounds of the present invention have valuable pharmacological properties. They have a pronounced gestagenic and olvulation-inhibiting effect, especially when administered orally, as can be demonstrated in animal experiments. 422 mg of crude 21-fluoro-16a, 17-isopropylidenedioxy-3-methoxy-19-norpregna-3,5-dien-20-one are obtained. This can be purified by chromatography on aluminum oxide and crystallization from ether-methanol, melting point 176-180 C. To a mixture of 20 ml of methylene chloride and 2.6 ml of dimethylformamide, while stirring and cooling with an ice-methanol mixture 2, 2 ml of phosphorus oxychloride. 15 minutes later, 3.91 g of the methoxydiene obtained above are added while rinsing with 5 ml of methylene chloride. After stirring for two hours with ice-water cooling, the cooling bath is removed and the mixture is stirred for a further hour.
Then it is replaced again with a mixture of 8.8 g of sodium acetate and 28 ml of 90% methanol, while cooling with ice and water, and 30 minutes later it is diluted with methylene chloride and washed with 2N soda solution and water. The residue obtained after drying and evaporation in vacuo is filtered in toluene solution through 40 g of silica gel, washing with 3 liters of toluene. 3.5 g of 21-fluoro-6-formyl-16a, 17-isopropylidenedioxy-3-methoxy-19-norpregna-3,5-dien-20-one are obtained from the residue of this filtrate by crystallization from methylene chloride ether. E220 = 7800; ± 323 = 11 700 (fine spirit).
To a mixture of 3.66 g of this compound and 37 ml of methanol, a solution of 460 mg of sodium borohydride in 4.6 ml of 1N sodium hydroxide solution is added with stirring, every / 2 hours later a mixture of 3 ml is slowly allowed to cool with ice-water concentrated hydrochloric acid and 1.5 ml of water. After half an hour, 4.6 g of sodium acetate and water are added, whereupon it is suction filtered, washed with water and dried in vacuo over phosphorus pentoxide. The residue obtained in this way is mixed with 82.5 ml of acetone and 20.5 ml of methylene chloride, whereupon 3.25 ml of Kiliany solution is slowly added dropwise while cooling with ice. After stirring for 13/4 hours at 06C, 0.8 ml of Kiliany solution is added again. 15 minutes later, 825 ml of a 10% strength sodium acetate solution are added and the mixture is extracted with methylene chloride.
After washing with sodium hydrogen carbonate solution and water, it is dried and evaporated in vacuo. The residue is chromatographed on 90 g of silica gel. The 21-fluoro-16a, 17-isopropylidenedioxy-6-methylen-19-norpregn-4-ene-3,20-dione is eluted from toluene-ethyl acetate (19: 1) mixture. After redissolving from methylene chloride-ether, 1.6 g are obtained; E267 = 11 100 (fine spirit).
