Es wurde gefunden, dass die O-AryIoxime von unterschiedlich substituierten 3-Ketosteroiden, in denen der Aryhing seinerseits in o- und/oder p-Stellung mit Nitrogruppen substituiert ist, anabolische und androgene Mittel sind. Darüber hinaus sind zahlreiche dieser Verbindungen sehr wirksame Antikonzeptionsmittel, besonders wenn sie oral postkoital verabreicht werden.

Eine starke kontrazeptive Wirkung haben aus dieser Klasse die Verbindungen, in denen R1 und R2 = H oder NO2, wobei wenigstens einer der Reste R1 und R2 für NO2 steht und in Fällen, in denen
Z eine C-C-Einfachbindung ist, R2 für NO2 und R1 für H steht; R3 = H oder CH3, wobei für den Fall, in dem R1 für H steht, R3 eine CH3-Gruppe ist; R4 = H oder CH3 oder CH2 - CH3, wobei für den Fall, in dem R1 für H steht, R4 ebenfalls Wasserstoff ist; Rs = H oder CH3 R6 = Hydroxy oder deren Ester und R7 = H.
Die Vorstufenketone dieser Antikonzeptionsmittel sind als solche im allgemeinen umwirksam, und die Ausgangsoxime sind wesentlich weniger wirksam oder unwirksam.
Die Ausgangsketone sowie Verfahren zu ihrer Herstellung sind allgemein bekannt.
Wenn eine Verbindung wie 3-(p-Nitrophenoxy)-imino-5aandrostan-17-on hergestellt werden soll, in der der Ring A gesättigt ist, ist eine Umsetzung des als Ausgangsprodukt eingesetzten 3,20-Dions mit dem O-Arylhydroxylamin unwirksam, da Substitution sowohl in 3- als auch in 17-Stellung stattfindet. In einem solchen Fall muss zunächst eine Verbindung wie 3-(p-Nitrophenoxy) -imino-5 a-androstan- 17ss-ol nach einem der oben genannten Verfahren hergestellt und anschliessend die 17B-Hydroxygruppe vorzugsweise unter Verwendung von Jones-Lösung zur entsprechenden 17-on Verbindung oxydiert werden.
Herstellung von:
3 -(2',4'-Dinitrophenoxy)iminoverbindungen
Beispiel 1
Unter Ausschluss von Licht wurden 2 Tropfen konzentrierte Salzsäure zu einer Lösung von 2,5 g Methyltestosteron und 1,65 g 0-(2',4'-Dinitrophenyl)-hydroxylamin (hergestellt gemäss J. Het. Chem., 4, 413 [1967j) in 200 ml Äthanol gegeben. Die Lösung wurde 4 Stunden gerührt, eingeengt und aus Äthanol kristallisiert, wobei 3,0 g (75%) 3-(2',4'-Dinitro phenoxy)-imino-17a-methyl-androst-4-en-17ss-ol vom Schmelzpunkt 180-183"C erhalten wurden.
Ultraviolettmaximum (CH2Cl2) bei 317 (E 20300), 245 (e 2800); [e]2r > 4.9 + 134,4 (Äthylenchlorid).

Unter Ausschluss von Licht wurden 2 Tropfen konzentrierte Salzsäure zu einer Lösung von 137 g 19-Nortestosteron und 1,0 g 0-(2,4-Dinitrophenyl)-hydroxylamin in 300 ml Äthanol gegeben. Die Lösung wurde 4 Stunden gerührt, eingeengt und aus Äthanol kristallisiert, wobei 1,67 g (73,2%) 3-(2',4'-Dini trophenoxy)-imino-östr-4-en-17ss-ol vom Schmelzpunkt 107 147 C erhalten wurden. Ultraviolettmaximum (CH2Ck) bei 317 (e 19800), 245 (E20000); [a]236 + 123,7 (Äthylenchlo- rid).

A. 3-(2',4'-Dinitrophenoxy)-imino andrasta-1,4-dien-17ss-ol
Unter Ausschluss von Licht wurden 1,40 g 1-Dehydrotestosteron und 1,0 g 0-(2',4'-Dinitrophenyl)-hydroxylamin in 350 ml Äthanol gelöst. Zur Lösung wurden einige Tropfen konzentrierte Salzsäure gegeben. Das Gemisch wurde 2 Stunden bei Raumtemperatur gerührt. Die Lösung wurde zur Trockene eingedampft und der hellgelbe Rückstand aus Äthanol kristallisiert. Schmelzpunkt 106-1 12oC, 154-1570C; W (CH2C12) bei 325 (E 22300), 240 (E (18000).

Unter Verwendung von Testosteron als Ausgangsmaterial und auf die in Beispiel 1 beschriebene Weise wird 3-(2',4' Dinitrophenoxy)-imino-4-androsten-17ss-ol als hellgelber Feststoff vom Schmelzpunkt 170,5-174,00C hergestellt. Die Lösung wurde 3 Stunden gerührt, eingeengt und aus Äthanol kristallisiert, wobei 3,75 g (86%) 17ss-Önanthyloxy-3-(2 ' ,4' -dinitrophenoxy)-iminoan- drost-4-en vom Schmelzpunkt 118 bis 1230C erhalten wurden.

Unter Ausschluss von Licht wurden 1 bis 2 Tropfen konzentrierte Salzsäure zu einer Lösung von 1,86 g 6a-Methyl-17aacetoxy-pregn-4-en-3,20-dion und 1,00 g 0-(2,4-Dinitrophenyl)-hydroxylamin in 300 ml Äthanol gegeben. Die Lösung wurde 18 Stunden bei Raumtemperatur gerührt und unter vermindertem Druck eingeengt. Der Rückstand wurde aus Äthanol kristallisiert, wobei 1,81 g (63%) 3-(2',4'-Dinitrophenoxy)-imino- 1 8a-acetoxy-6a-methyl-pregn-4-en-20-on erhalten wurden, das anschliessend zur Entfernung von etwaigem Hexan am Rückfluss erhitzt wurde. Durch Abdampfen des Hexans wurde die Verbindung vom Schmelzpunkt 129 bis 1330C erhalten.

Unter Ausschluss von Licht wurden 1,48 g 17-Äthyl-19nortestosteron und 1,0 g 0-(2,4-Dinitrophenyl)-hydroxylamin in 350 ml Äthanol gelöst. Zur Lösung wurden einige Tropfen konzentrierte HCI gegeben. Die Lösung wurde dann 1,5 Stunden bei Raumtemperatur gerührt und zur Trockene eingedampft. Der erhaltene blassgelbe Rückstand wurde'aus Äthanol kristallisiert, wobei 0,63 g (26%) der gewünschten Verbindung vom Schmelzpunkt 168 bis 176 C (Zers.) erhalten wurden. Unter Ausschluss von Licht wurde ein Gemisch von 1,49 g (0,005 Mol) Noräthindron, 1 g 0-(2,4-Dinitrophenyl)-hydroxylamin (0,005 Mol) und 500 ml Äthylalkohol zur Auflösung der Verbindungen erhitzt. Zur warmen Lösung wurden 5 Tropfen konzentrierte Salzsäure gegeben. Die Lösung wurde eine Stunde gerührt, worauf weitere 50 mg 0-(2,4-Dinitrophenyl)-hydroxylamin zugesetzt wurden. Die Lösung wurde unter vermindertem Druck eingeengt und der Rückstand aus Äthanol kristallisiert, wobei 1,61 g gelbe Kristalle vom Schmelzpunkt 179 bis 184"C erhalten wurden. UV-Maximum (CH2Ch) bei 427 (E 20500), 408 (E 19500).

17a-Acetoxy-3 -(2',41-dinitrophenoxy)- imino-5a-androstan
Unter Ausschluss von Licht wurde 1 Tropfen konzentrierte Salzsäure zu einer Lösung von 2,98 g Dihydrotestosteronacetat und 1,8 g 0-(2,4-Dinitrophenyl)-hydroxylamin in 20 ml Methanol gegeben. Die Lösung wurde eine Stunde unter Rühren am Rückfluss erhitzt, eingeengt, mit Wasser gewaschen und aus Äthanol kristallisiert, wobei 3,25 g (71%) der gewünschten Verbindung vom Schmelzpunkt 175 bis 177oC erhalten wurden. Beispiel 10 3 -(p-Nitrophenoxy)-iminoverbindungen
Zu einer Lösung von 1,33 g tert.-Butyl-N-hydroxycarbamat und 0,66 g Kaliumhydroxyd (85%) in 20 ml Äthanol wurde tropfenweise eine Lösung von 1,41 g p-Fluornitrobenzol in 20 ml Äthanol gegeben. Die Lösung wurde 18 Stunden gerührt und auf Eiswasser gegossen. Der gebildete Feststoff wurde abfiltriert und mit Wasser gut gewaschen. Durch Kristallisation aus Benzol-Hexan wurden 2,15 g (84,5%) tert. Butyl-N-(p-nitrophenoxy)-carbamat in Form von weissen Kristallen vom Schmelzpunkt 106 bis 1080C erhalten.

Eine Lösung von 12 g tert.-Butyl-N-(p-nitrophenoxy)carbamat in 45 ml Trifluoressigsäure wurde 15 Minuten gerührt, auf 3QOf ml Eiswasser gegossen und filtriert. Der erhaltene Feststoff wurde getrocknet. Die Mutterlauge wurde mit Kaliumcarbonat neutralisiert, wobei zusätzliches Produkt erhalten wurde. Die Gesamtausbeute an vereinigtem Produkt betrug 5,49 g (75,5%). Dieses Produkt wurde anschliessend aus Äthanol kristallisiert, wobei reines O-(p-Nitrophenyl)hydroxylamin vom Schmelzpunkt 126,5 bis 128 C erhalten wurde. W-Maximum (CH2Ck) bei 308 my (E 11400).

Die Lösung wurde 24 Stunden gerührt, eingeengt und aus Äthanol kristallisiert, wobei 1,99 g (68,2%) der oben genannten Verbindung erhalten wurden. Schmelzpunkt 111 bis 159oC. UV-Maximum (CH2Ck) bei 327 mF (E 20400), 258 mF (E 13300), 232 mF (E 17500).

Zu einer Lösung von 1 g 17ss-Acetoxy-2a-brom-Sa-andro- stan-3-on in Äthanol wurden 0,370 g O-(p-Nitrophenyl)hydroxylamin und 1 Tropfen konzentrierte Salzsäure gegeben.
Die Lösung wurde 2 Stunden gerührt, worauf das Lösungsmittel entfernt wurde. Durch Kristallisation aus Äthanol wurden 1,14 g (84%) weisse Kristalle vom Schmelzpunkt 180 bis 1830C erhalten. UV-Maximum (CH2Ck) bei 313 m (E 16200).

Zu einer in einem Eisbad gehaltenen Lösung von 5 g 3-(p Nitrophenoxy)-imino-5a-androstan-17ss-ol und 150 ml Aceton wurden tropfenweise 4,5 ml Jones-Lösung gegeben. Die Lösung wurde 3 Stunden gerührt, in Eiswasser gegossen, abfiltriert und mit Wasser geaschen. Durch Umkristallisation aus einer Benzol-Hexan-Lösung wurden 4,3 g der oben genannten Verbindung erhalten. Schmelzpunkt 170 bis 172 C. UV-Maximum (CH2Ck) bei 318 my (e 15900).

Zu einer Lösung von 2 g 17ss-Acetoxy-2a-chlor-5a-andro- stan-3-on in Äthanol gab man 0,84 g O-(p-Nitrophenyl)- hydroxylamin sowie einen Tropfen konzentrierter Chlorwasserstoffsäure. Man rührte die Mischung bei Nacht und filtrierte. Durch Kristallisation aus Äthanol erhielt man 2,1 g (77 %) weisser Kristalle mit einem Schmelzpunkt von 170 bis 1720C.

Zu einer Lösung von 0,5 g 17ss-Acetoxy-2a-chlor-5a- androstan-3-on in 250 ml Methanol gab man 2 ml konzentrierter Chlorwasserstoffsäure. Nachdem man 11/2 Stunden lang am Rückfluss gekocht hatte, wurde die Lösung konzentriert, in Methylenchlorid gelöst, mit Natriumbicarbonat gewaschen und getrocknet. Durch Kristallisation aus Äthanol erhielt man 0,33 g (75 %) weisser Kristalle, die einen Schmelzpunkt von 190 bis 1930C hatten.

2a-Chlor-3-(p-nitrophenoxy)-imino Sa-androstan-i7P-ol
Zu einer Lösung von 5 g 2a-Chlor-Sa-androstan-17ss-ol-3- on in Äthanol fügte man 2,37 g O-(p-Nitrophenyl)-hydroxyl- amin und einen Tropfen konzentrierter Chlorwasserstoffsäure hinzu. Man rührte über Nacht und filtrierte. Durch Kristallisation aus Äthanol erhielt man 3 g (44%) weisser Kristalle mit einem Schmelzpunkt von 181 bis 184,5 C.

Ebenfalls verwendete man hier 10 ml der Jones-Lösung anstelle von 4,5 ml. Durch Kristallisation aus Äthanol erhielt man 2 g (80%) weisse Kristalle mit einem Schmelzpunkt von 202-203"C.

Nach dem Verfahren von Beispiel 21 und unter Verwendung äquivalenter Mengen der entsprechenden 2a-Brom Ausgangsverbindung und der Hydroxylaminverbindung erhielt man 2a-Brom-3-(p-nitrophenoxy)-imino-Sa-androstan - 17ss- ol. Durch Kristallisation aus Äthanol konnten 2 g (83 %) weisser Kristalle mit einem Schmelzpunkt von 165,5 bis 166"C erhalten werden.

Nach dem Verfahren gemäss Beispiel 22 und unter Verwendung des Produktes von Beispiel 23 als Ausgangsmaterial erhielt man 2ot-Brom-3-(p-nitrophenoxy)-imino-5a-andro- stan-17-on. Durch Kristallisation aus Äthanol erhielt man 1,1 g (75 %) weisser Kristalle mit einem Schmelzpunkt von 184,5 bis 185"C.

Wie bereits erwähnt, sind zahlreiche Verbindungen gemäss der Erfindung hochwirksame Antikonzeptionsmittel. Die kontrazeptive Aktivität wird wie folgt gemessen:
Eine Testgruppe von Ratten erhält eine berechnete Menge einer zu prüfenden Substanz im Futter während einer Zeit von 7 Tagen. Während dieser Zeit werden männliche und weibliche Tiere getrennt gehalten. Beide Geschlechter erhalten die zu prüfende Substanz. Nach dieser Zeit werden männliche und weibliche Tiere zusammengegeben, worauf weitere 15 Tage das das gleiche Futter verabreicht wird. Nach dieser Zeit werden die Geschlechter wieder getrennt, und die zu prüfende Substanz wird dem Futter nicht mehr zugesetzt.
Die weiblichen Tiere werden dann während einer Zeit von 21 Tagen beobachtet. Man lässt sie die Jungen werfen und aufziehen. Eine Kontrollgruppe von Ratten wird in der gleichen Zeit genau in der gleichen Weise behandelt mit dem Unterschied, dass ihr Futter nicht die zu prüfende Substanz enthält.
In der folgenden Tabelle ist der Prozentsatz der Ratten angegeben, die bei der genannten Dosis Junge warfen.

Zwar sind alle erhaltenen Verbindungen wenigstens schwache anabolische Mittel, jedoch hat eine Anzahl von ihnen eine ungewöhnlich hohe anabolische Wirksamkeit. With the exclusion of light, 1.40 g of 1-dehydrotestosterone and 1.0 g of 0- (2 ', 4'-dinitrophenyl) hydroxylamine were dissolved in 350 ml of ethanol. A solution of 1.41 g of p-fluoronitrobenzene in 20 ml of ethanol was added dropwise to a solution of 1.33 g of tert-butyl N-hydroxycarbamate and 0.66 g of potassium hydroxide (85%) in 20 ml of ethanol. The solution was stirred for 18 hours and poured onto ice water. The solid formed was filtered off and washed well with water. By crystallization from benzene-hexane, 2.15 g (84.5%) of tert. 0.370 g of O- (p-nitrophenyl) hydroxylamine and 1 drop of concentrated hydrochloric acid were added to a solution of 1 g of 17ss-acetoxy-2a-bromo-sa-androstan-3-one in ethanol.

To a solution of 5 g of 3- (p-nitrophenoxy) -imino-5a-androstan-17ss-ol and 150 ml of acetone kept in an ice bath, 4.5 ml of Jones solution was added dropwise. The solution was stirred for 3 hours, poured into ice water, filtered off and ashed with water. Recrystallization from a benzene-hexane solution gave 4.3 g of the above-mentioned compound. 0.84 g of O- (p-nitrophenyl) hydroxylamine and a drop of concentrated hydrochloric acid were added to a solution of 2 g of 17ss-acetoxy-2a-chloro-5a-androstan-3-one in ethanol. The mixture was stirred overnight and filtered. Crystallization from ethanol gave 2.1 g (77%) of white crystals with a melting point of 170 to 1720C.

2.37 g of O- (p-nitrophenyl) hydroxylamine and a drop of concentrated hydrochloric acid were added to a solution of 5 g of 2a-chloro-sa-androstan-17ss-ol-3-one in ethanol. The mixture was stirred overnight and filtered. Crystallization from ethanol gave 3 g (44%) of white crystals with a melting point of 181 to 184.5 C.

5a-androstane-17-one
Using 2.5 g of 2a-chloro-3- (p-nitrophenoxy) imino-5cr-androstan-17P-ol instead of the imino compound unsubstituted in the 2-position, the procedure of Example 18 was repeated. 10 ml were also used here the Jones solution instead of 4.5 ml. Crystallization from ethanol gave 2 g (80%) of white crystals with a melting point of 202-203 "C.

Example 23
Following the procedure of Example 21 and using equivalent amounts of the corresponding 2a-bromine starting compound and the hydroxylamine compound, 2a-bromo-3- (p-nitrophenoxy) -imino-sa-androstane-17ss-ol was obtained. By crystallization from ethanol, 2 g (83%) of white crystals with a melting point of 165.5 to 166 "C could be obtained.

Example 24
Following the process according to Example 22 and using the product from Example 23 as starting material, 2ot-bromo-3- (p-nitrophenoxy) -imino-5a-androstan-17-one was obtained. Crystallization from ethanol gave 1.1 g (75%) of white crystals with a melting point of 184.5 to 185.degree.

As already mentioned, numerous compounds according to the invention are highly effective contraception agents. Contraceptive activity is measured as follows:
A test group of rats receives a calculated amount of a substance to be tested in the feed for a period of 7 days. During this time, male and female animals are kept separate. Both sexes receive the substance to be tested. After this time, male and female animals are given together, whereupon the same feed is administered for a further 15 days. After this time, the sexes are separated again and the substance to be tested is no longer added to the feed.
The females are then observed for a period of 21 days. They let the youngsters throw them and raise them. A control group of rats is treated in exactly the same way at the same time, with the difference that their feed does not contain the substance to be tested.
The table below shows the percentage of rats that gave birth at the stated dose.

While all the compounds obtained are at least weak anabolic agents, a number of them have unusually high anabolic potency. The compound according to Example 2 has a ventral prostate potency of 0.15 compared to methyltestosterone and a levator-ani potency of 2.1 compared to methyltestosterone. The ratio of anabolic to androgenic effects is therefore very favorable.
This also applies to the compound of Example 6, which has an anabolic to androgenic effect ratio of 12.5 with a ventral prostate potency of 0.18 and a Levatorani potency of 2.24 compared to methyltestosterone, and for the compound of Example 9, which has a ventral prostate potency of 1.9 and a levator ani potency of 4.6.
