Die bekannten Benzodiazepine sind wertvolle Sedativa, Muskelrelaxantien und Tranquilizer. Diese Verbindungen können jedoch bisher nur mittels Verfahren hergestellt werden, die zum Teil sehr teuer sind und in unbefriedigenden Ausbeuten verlaufen.
Aufgabe der Erfindung war es daher, Verfahren zur Herstellung neuer Benzodiazepine und ihrer Salze zu schaffen, die in technischem Massstab durchführbar sind und in hohen Ausbeuten verlaufen. Diese Aufgabe wird durch die Erfindung gelöst.
Gegenstand der Erfindung ist somit ein Verfahren zur Herstellung von Benzodiazepinen der allgemeinen Formel:

inderR1,R4,R5, R6 und n die angegebene Bedeutung haben und p den Wert 0 oder 1 hat, mit einem Oxydationsmittel behandelt.
Beispiele für verwendbare Oxydationsmittel sind Chromsäure, Salpetersäure, Wasserstoffperoxid, organische Persäuren, wie Perameisensäure, Peressigsäure, Perbenzoesäure oder m-Chlorperbenzoesäure, Natriumperjodat, Kaliumperjodat, Kaliumpersulfat, Selendioxid, Bleitetraacetat, Mangandioxid oder Rutheniumtetroxid. Die Umsetzung wird vorzugsweise in Gegenwart eines Lösungsmittels durchgeführt.
Die Art des verwendeten Lösungsmittels hängt von der Art des Oxydationsmittels ab. Beispiele für geeignete Lösungsmittel sind Wasser, Chloroform, Tetrachlorkohlenstoff, Aceton, Essigsäure, Ameisensäure, Schwefelsäure, Pyridin, Di oxan, Benzol, Toluol, Diäthyläther, Äthylacetat, Methanol und Äthanol sowie deren Gemische. Die Reaktionstemperatur hängt von der Art des verwendeten Oxydationsmittels ab. Zur Herstellung von Verbindungen der allgemeinen Formel I-a, in der m den Wert 1 hat, wird die Umsetzung vorzugsweise bei oder oberhalb Raumtemperatur durchgeführt.
Die erfindungsgemäss herstellbaren Verbindungen sind wertvolle Arzneimittel. Sie sind insbesondere ausgezeichnete Tranquilizer, Sedativa, Muskelrelaxantien, Spasmolytika und Hypnotika.
Zur Herstellung der Ausgangsprodukte der Formel VII kann man sich verschiedener Methoden bedienen.
Insbesondere kann man dadurch vorgehen, dass man (a) ein Benzodiazepin oder dessen Salz der allgemeinen Formel:

in welcher R1, R und R6 die obige Bedeutung haben, mit einem reaktionsfähigen Ester einer Verbindung der allgemeinen Formel: amid, Alkylalkahverbindungen, wie Butyllithium, Phenylalkaliverbindungen, wie Phenyllithium, sowie Alkalimetallalkoholate, wie Natriummethylat, Natriumäthylat und Kalium-tert.-butylat. Die Umsetzung wird vorzugsweise auch in Gegenwart eines organischen Lösungsmittels oder Lösungsmittelgemisches durchgeführt. Beispiele für geeignete Lösungsmittel sind Benzol, Toluol, Xylol, Dimethylformamid, Dimethylacetamid, Diphenyläther, Äthylenglykoldimethyläther, Dimethylsulfoxid, Methyläthylketon, N- Methyl- pyrrolidon und deren Gemische.
Die Umsetzung kann bei Temperaturen von etwa Raumtemperatur bis zum Siedepunkt des verwendeten Lösungsmittels durchgeführt werden.
Die Variante (b) wird vorzugsweise ebenfalls in Gegenwart eines Lösungsmittels oder Lösungsmittelgemisches durchgeführt. Beispiele für geeignete Lösungsmittel sind Chloroform, Tetrachlorkohlenstoff, Methylenchlorid, Äthylenchlorid, Diäthyläther, Diisopropyläther, Tetrahydrofuran, Dioxan, Wasser, Methanol, Äthanol, Dimethylformamid, Dimethylsulfoxid oder deren Gemisch. Die Umsetzung wird im allgemeinen in Gegenwart einer Säure durchgeführt. Beispiele für verwendbare Säuren sind Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phospohrsäure, Polyphosphorsäure, Bortrifluorid und p-Toluolsulfonsäure. Die Umsetzung wird bei Temperaturen von -250C bis etwa 1200C, vorzugsweise bei etwa 0 bis 300C durchgeführt.
Temperaturen oberhalb und unterhalb des vorgenannten Bereiches können ebenfalls angewendet werden, sind jedoch weniger bevorzugt. In den meisten Fällen kann die Umsetzung bei Raumtemperatur oder darunter durchgeführt werden. Das Verfahren kann bei Atmosphärendruck, Überdruck oder Unterdruck durchgeführt werden. Gegebenenfalls wird das Verfahren unter einem Schutzgas, wie Stickstoff oder Argon, durchgeführt.
Das Molverhältnis des 2,5-Oxazolidindions zum Aminophenylketon ist nicht kritisch. Vorzugsweise werden die Reak- tionsteilnehmer in mindestens stöchiometrischen Mengen eingesetzt. In den meisten Fällen wird das Oxalidindion vorzugsweise im Überschuss verwendet. Die Umsetzung kann erforderlichenfalls durch Erhitzen in einem Lösungsmittel, wie Dimethylsulfoxid oder Dimethylformamid, erfolgen.
Beispiele für die Schutzgruppen Z in der Variante (c) sind die Phthaloylgruppe, niedere Alkylidengruppen, wie die Isopropylidengruppe, die Benzaigruppe oder aber eine Carbobenzoxygruppe und ein Wasserstoffatom zusammen. Die

sein. Die Abspaltung der Schutzgruppe wird nach üblichen hydrolytischen oder hydrogenolytischen Methoden durchgeführt. Wenn z.B. der Rest Z in der Verbindung der allgemeinen Formel VIII eine Phthaloyigruppe ist, wird die Schutzgruppe durch Umsetzung mit Hydrazin oder Hydrazinderivaten, wie Hydrazinhydrat oder Phenylhydrazin, abgespalten. Man erhält die entsprechenden Verbindungen der allgemeinen Formel VII. Die Umsetzung wird vorzugsweise in Gegenwart eines Lösungsmittels oder Lösungsmittelgemisches durchgeführt.
Beispiele für geeignete Lösungsmittel sind Methanol, Äthanol, Isopropanol, Wasser und deren Gemische. Die Umsetzung wird bei Temperaturen von etwa Raumtemperatur bis zum Siedepunkt des verwendeten Lösungsmittels durchgeführt.
Wenn das System Z in den Verbindungen der allgemeinen Formel VIII aus einer Carbobenzoxygruppe als Schutzgruppe und einem Wasserstoffatom besteht, wird die Ab HO¯CnH2n¯S(O)p¯R4 (III) in welcher R4, n und p die obige Bedeutung haben, kondensiert, oder (b) ein 2-Aminobenzophenol-Derivat der allgemeinen Formel:

in welcher Rl, R4, R5, R6, n und p die obige Bedeutung haben und Z eine leicht abspaltbare Schutzgruppe darstellt, die Schutzgruppe abspaltet.
Beispiele für die in der Variante (a) eingesetzten reaktionsfähigen Ester von Verbindungen der allgemeinen Formel III sind die Ester der Halogenwasserstoffsäuren, wie die Chloride, Bromide und Jodide, sowie die Sulfonsäurerester, wie das Methansulfonat, p-Toluolsulfonat, p-Naphthalinsul- fonat und Trichlormethansulfonat. Die Umsetzung der Verbindung der allgemeinen Formel II mit dem Reaktionsfähigen Ester einer Verbindung der allgemeinen Formel III wird vorzugsweise in Gegenwart eines basischen Kondensationsmittels durchgeführt. Zunächst kann aus der Verbindung der allgemeinen Formel II mit der Base das entsprechende Salz hergestellt werden, das anschliessend mit dem reaktionsfähigen Ester der Verbindung der allgemeinen Formel III kondensiert wird.
Beispiele für basische Kondensationsmittel sind Alkalimetallhydride, wie Natriumhydrid und Lithiumhydrid, Alkalimetallhydroxide, wie Kaliumhydroxid, Alkalimetallamide, wie Natriumamid, Kaliumamid und Lithium spaltung der Schutzgruppe mit Hilfe von Halogenwasserstoffsäuren, wie Bromwasserstoffsäure und Chlorwasserstoffsäure, ereicht. Die bevorzugte Halogenwasserstoffsäure ist Bromwasserstoffsäure. Die Umsetzung wird vorzugsweise in einem Lösungsmittel oder Lösungsmittelgemisch durchgeführt. Beispiele für geeignete Lösungsmittel sind Methanol, Äthanol, Essigsäure, Wasser und deren Gemische. Die Umsetzung wird bei Temperaturen von etwa Raumtemperatur bis zum Siedepunkt des verwendeten Lösungsmittels durchgeführt.
Das Verfahren kann entweder unmittelbar zu den entsprechenden Verbindungen der allgemeinen Formel VII führen oder über Zwischenprodukte der allgemeinen Formel:

Diese Verbindungen können anschliessend zu den entsprechenden Verbindungen der allgemeinen Formel VII cyclisiert werden. Das Zwischenprodukt der allgemeinen Formel IX kann als Salz einer Halogenwasserstoffsäure beim Arbeiten unter milden Reaktionsbedingungen, z.B. bei der entsprechenden Reaktionstemperatur und während einer geeigneten Reaktionszeit, isoliert werden. Der Ringschluss der Verbindungen der allgemeinen Formel Ix zu den Verbindungen der allgemeinen Formel VII erfolgt beim Stehen bei Raumtemperatur oder bei erhöhter Temperatur in einem geeigneten Lösungsmitte?, wie Essigsäure, Methanol, Äthanol, Pyridin oder Dimethylsulfoxid, und gegebenenfalls unter Behandlung mit einer Base.

Zur Herstellung der Salze werden die Benzodiazepine mit pharmakologisch verträglichen anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwe felsäure, Phosphorsäure, Salpetersäure, Essigsäure, Maleinsäure, Fumarsäure, Weinsäure, Bernsteinsäure, Citronensäure, Kampfersulfonsäure, Äthansulfonsäure, Ascorbinsäure oder Milchsäure umgesetzt.
Wie bereits erwähnt, sind die Benzodiazepine der allgemeinen Formel I und insbesondere der allgemeinen Formel I-a und ihre pharmakologisch verträglichen Salze mit Säuren wertvolle Sedativa, Muskelrelaxantien, Hypnotika und Krampfmittel. Beispielsweise sind -(p-Methylsulfonyläthyl)- -5-(o-fluorphenyl)-7-chlor- 1,3 -dihydro-2H- 1 ,4-benzodiazepin -2-on und 1 -(,8-Methylsulfinyläthyl)-5-phenyl-7-chlior- 1,3- -dihydro-2H-1,4-benzodiazepin-2-on Krampfmittel und Muskelrelaxantien, und sie potensieren die Wirkung von Hexobarbital. Gleichzeitig haben sie eine sehr geringe Toxizität.
Die pharmakologische Aktivität der erfindungsgemäss herstellbaren Verbindungen ist stärker als die chemisch ähnlicher Verbindungen, wie Chlordiazepoxid, d.h. 2-Methylamino-5-phenyl-7-chlor-3H-1,4-benzodiazepin-4-oxid,
Die erfindungsgemäss herstellbaren Benzodiazepine und ihre Salze können parenteral oder oral in üblichen Verabreichungsformen verabfolgt werden, z.B. als Tabletten, Dragees, Kapseln, Suspensionen, Lösungen und Elixiere.
Herstellung der Ausgangsprodukte a) Eine Lösung von 1 g 5-Phenyl-7-chlor-1,3-dihydro -2H- 1 ,4-benzodiazepin-2-on in 15 ml N,N-Dimethylformamid wird zu einer Suspension von 0,3 g Natriummethylat in 15 ml N,N-Dimethylformamid gegeben. Das Gemisch wird 1 Stunde auf 50 bis 60"C erwärmt, danach abgekühlt und unterhalb 100C mit einer Lösung von 2 ml p-Methyl- sulfinyläthylchlorid in 10 ml Toluol versetzt. Das erhaltene Gemisch wird 30 Minuten bei Raumtemperatur und anschliessend 5 Stunden bei 80 bis 90"C gerührt. Hierauf wird das Reaktionsgemisch in Eiswasser eingegossen und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der Rückstand bildet beim Stehen Kristalle.
Nach Umkristallisation aus Äthylacetat schmelzen die farblosen Kristalle von 1-(P-M ethylsulfinyläthyl)-5-phe- nyl-7-chlor- 1 ,3-dihydro-2H- 1 ,4-benzodiazepin-2-on bei 166 bis 167"C.
b) Eine Lösung von 0,5 g 2-(,3-Methylsulfinyläthyl)-ami- no-5-chlorbenzophenon in 20 ml wasserfreiem Methylenchlorid wird mit 0,5 g Oxazolidin-2,5-dion versetzt. Hierauf wird das Gemisch mit 20 ml einer Ätherlösung von Chlorwasserstoff unter Kühlung versetzt. Das Gemisch wird bei Raumtemperatur gerührt. Nach beendeter Umsetzung wird das Gemisch in Eiswasser gegossen, mit wässriger Ammoniaklösung alkalisch gemacht und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der Rückstand wird mit Isopropanol vesetzt. Hierbei bildet sich Kristalle, die aus Aceton umkristallisiert werden.
Das 1 -(ss-Methylsulfinyläthyl)-5-phenyl-7-chlor- 1,3 die hydro-2H-1,4-benzodiazepin-2-on kristallisiert und schmilzt dann bei 166 bis 1670C.
c) Ein Gemisch aus 1 g 2-[N1-B-Methylsulfinyläthyl)- -N1-(N2-carbob enzoxyglycyl) -amido] -5-chlor-benzophenon und 20 ml einer 30prozentigen Lösung von Bromwasserstoff in Essigsäure wird bei Raumtemperatur gerührt. Nach beendeter Umsetzung wird das Gemisch in Wasser eingegossen, mit wässriger Ammoniaklösung alkalisch gemacht und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus Isopropanol umkristallisiert. Nach noch- maliger Umkristallisation aus Aceton schmilzt das erhaltene 1-(,(3-Methylsulfinyläthyl)-5-phenyl-7-chlor-1,3-dihydro-2H- -1,4-benzodiazepin-2-on bei 166 bis 1670C.
d) Ein Gemisch aus 1 g 2-(N-Phthalimidoacetyl-N-p -methylsulfinyläthyl)-amino-5-chlorbenzophenon, 15 ml Methylenchlorid und 15 ml Äthanol wird mit einer Lösung von 2 g Hydrazinhydrat in 2 ml Wasser versetzt und bei Raumtemperatur gerührt. Nach beendeter Umsetzung wird das Reaktionsgemisch unter vermindertem Druck eingedampft, der Rückstand mit Wasser verdünnt, mit wässriger Ammoniaklösung alkalisch gemacht und mit Äther extrahiert. Der Ätherextrakt wird über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird aus wenig Diisopropyläther umkristallisiert. Nach nochmaliger Umkristallisation aus einer Mischung von Diisopropyläther und Methylenchlorid schmelzen die farblosen Kristalle von l-(ss-Methylsulfinyläthyl)-5- -phenyl-7-chlor-1,3-dihydro-2H-1,4-benzodiazepin-2-on bei 166 bis 167"C.
Beispiel I
10 ml 1 Sprozentige wässrige Wasserstoffperoxidlösung werden unter Eiskühlung zu einem Gemisch aus 1,5 g 1-(ss- -Methylthioäthyl)-5-phenyl-7-chlor-1,3-dihydro-2H-1,4-ben- zodiazepin-2-on und 20 ml Essigsäure gegeben. Das Gemisch wird 30 Minuten bei 0 C und anschliessend 1 Stunde bei Raumtemperatur gerührt. Danach wird das Reaktionsgemisch in Eiswasser eingegossen, mit wässriger Ammoniaklösung alkalisch gemacht und mit Chloroform extrahiert.
Der Chloroformextrakt wird über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Der kristalline Rückstand wird aus Äthylacetat umkristallisiert. Man erhält 1 -(p-Methylsulfinyläthyl)-5-phenyl-7-chlor 1 ,3-dihy- dro-2H- 1 ,4-benzodiazepin-2-on vom F. 166"1670C.
Beispiel 2
15 ml einer 30prozentigen wässrigen Wasserstoffperoxidlösung werden unter Rühren zu einem Gemisch aus 1,7 g 1 -(p-Methylthioäthyl)-5-phenyl-7-chlor- 1,3-dihydro-2H-1,4 -benzodiazepin-2-on und 5 ml Essigsäure gegeben, und 3 Stunden auf 35 bis 400C erwärmt. Nach dem Abkühlen wird das Reaktionsgemisch 16 bis 18 Stunden stehengelassen, sodann in Eiswasser eingegossen, mit wässriger Ammoniaklösung alkalisch gemacht und mit Methylenchlorid extrahiert. Der Methylenchloridextrakt wird über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft.
Der Rückstand wird aus Isopropanol umkristallisiert. Nach nochmaliger Umkristallisation aus Aceton schmilzt das 1-(B- -Methylsulfonyläthyl)-5-phenyl-7-chlor-1,3-dihydro-2H-1,4- -benzodiazepin-2-on bei 160 bis 161"C.

In analoger Weise wird das 1-(ss-Methylsulfonyläthyl)-5- -(o-fluorphenyl)-7-chlor-1 ,3-dihydro-2H-1 4-benzodiazepin- -2-on vom F. 155 bis 156"C hergestellt.

The well-known benzodiazepines are valuable sedatives, muscle relaxants and tranquilizers. However, up to now these compounds can only be produced by means of processes which are in some cases very expensive and have unsatisfactory yields.
The object of the invention was therefore to create processes for the preparation of new benzodiazepines and their salts which can be carried out on an industrial scale and which proceed in high yields. This object is achieved by the invention.
The invention therefore relates to a process for the preparation of benzodiazepines of the general formula:

Examples of oxidizing agents that can be used are chromic acid, nitric acid, hydrogen peroxide, organic peracids such as performic acid, peracetic acid, perbenzoic acid or m-chloroperbenzoic acid, sodium periodate, potassium periodate, potassium persulfate, selenium dioxide, lead tetraacetate, manganese dioxide or ruthenium tetroxide. The reaction is preferably carried out in the presence of a solvent.
The type of solvent used depends on the type of oxidizing agent. Examples of suitable solvents are water, chloroform, carbon tetrachloride, acetone, acetic acid, formic acid, sulfuric acid, pyridine, Di oxane, benzene, toluene, diethyl ether, ethyl acetate, methanol and ethanol and mixtures thereof. The reaction temperature depends on the type of oxidizing agent used. To prepare compounds of the general formula I-a in which m is 1, the reaction is preferably carried out at or above room temperature.
The compounds which can be prepared according to the invention are valuable medicaments. In particular, they are excellent tranquilizers, sedatives, muscle relaxants, antispasmodics, and hypnotics.
Various methods can be used to prepare the starting materials of the formula VII.
In particular, one can proceed in that (a) a benzodiazepine or its salt of the general formula:

in which R1, R and R6 have the above meaning, with a reactive ester of a compound of the general formula: amide, alkyl alkali compounds such as butyl lithium, phenyl alkali compounds such as phenyl lithium, and alkali metal alcoholates such as sodium methylate, sodium ethylate and potassium tert-butylate. The reaction is preferably also carried out in the presence of an organic solvent or solvent mixture. Examples of suitable solvents are benzene, toluene, xylene, dimethylformamide, dimethylacetamide, diphenyl ether, ethylene glycol dimethyl ether, dimethyl sulfoxide, methyl ethyl ketone, N-methyl pyrrolidone and mixtures thereof.
The reaction can be carried out at temperatures from about room temperature to the boiling point of the solvent used.
Variant (b) is preferably also carried out in the presence of a solvent or solvent mixture. Examples of suitable solvents are chloroform, carbon tetrachloride, methylene chloride, ethylene chloride, diethyl ether, diisopropyl ether, tetrahydrofuran, dioxane, water, methanol, ethanol, dimethylformamide, dimethyl sulfoxide or a mixture thereof. The reaction is generally carried out in the presence of an acid. Examples of acids that can be used are hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, polyphosphoric acid, boron trifluoride and p-toluenesulfonic acid. The reaction is carried out at temperatures from -250C to about 1200C, preferably at about 0 to 300C.
Temperatures above and below the aforementioned range can also be used, but are less preferred. In most cases, the reaction can be carried out at room temperature or below. The process can be carried out at atmospheric pressure, overpressure or underpressure. If necessary, the process is carried out under a protective gas such as nitrogen or argon.
The molar ratio of the 2,5-oxazolidinedione to the aminophenyl ketone is not critical. The reactants are preferably used in at least stoichiometric amounts. In most cases the oxalidinedione is preferably used in excess. The reaction can be carried out by heating in a solvent such as dimethyl sulfoxide or dimethylformamide, if necessary.
Examples of the protective groups Z in variant (c) are the phthaloyl group, lower alkylidene groups, such as the isopropylidene group, the benzene group, or a carbobenzoxy group and a hydrogen atom together. The

be. The protective group is split off by customary hydrolytic or hydrogenolytic methods. If e.g. the radical Z in the compound of the general formula VIII is a phthaloyl group, the protective group is split off by reaction with hydrazine or hydrazine derivatives, such as hydrazine hydrate or phenyl hydrazine. The corresponding compounds of the general formula VII are obtained. The reaction is preferably carried out in the presence of a solvent or solvent mixture.
Examples of suitable solvents are methanol, ethanol, isopropanol, water and mixtures thereof. The reaction is carried out at temperatures from about room temperature to the boiling point of the solvent used.

Examples of the reactive esters of compounds of the general formula III used in variant (a) are the esters of hydrohalic acids such as the chlorides, bromides and iodides, and also the sulfonic acid esters such as methanesulfonate, p-toluenesulfonate, p-naphthalene sulfonate and Trichloromethanesulfonate. The reaction of the compound of the general formula II with the reactive ester of a compound of the general formula III is preferably carried out in the presence of a basic condensing agent. First, the corresponding salt can be prepared from the compound of general formula II with the base, which is then condensed with the reactive ester of the compound of general formula III.
Examples of basic condensing agents are alkali metal hydrides such as sodium hydride and lithium hydride, alkali metal hydroxides such as potassium hydroxide, alkali metal amides such as sodium amide, potassium amide and lithium cleavage of the protective group with the aid of hydrohalic acids such as hydrobromic acid and hydrochloric acid. The preferred hydrohalic acid is hydrobromic acid. The reaction is preferably carried out in a solvent or solvent mixture. Examples of suitable solvents are methanol, ethanol, acetic acid, water and mixtures thereof. The reaction is carried out at temperatures from about room temperature to the boiling point of the solvent used.
The process can either lead directly to the corresponding compounds of the general formula VII or via intermediates of the general formula:

To prepare the salts, the benzodiazepines are reacted with pharmacologically acceptable inorganic or organic acids such as hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, nitric acid, acetic acid, maleic acid, fumaric acid, tartaric acid, succinic acid, citric acid, camphor sulfonic acid, ethanesulfonic acid, ascorbic acid or lactic acid.

The pharmacological activity of the compounds which can be prepared according to the invention is stronger than that of chemically similar compounds such as chlordiazepoxide, i. 2-methylamino-5-phenyl-7-chloro-3H-1,4-benzodiazepine-4-oxide,
The benzodiazepines and their salts which can be prepared according to the invention can be administered parenterally or orally in conventional administration forms, e.g. as tablets, dragees, capsules, suspensions, solutions and elixirs.
Preparation of the starting products a) A solution of 1 g of 5-phenyl-7-chloro-1,3-dihydro -2H- 1, 4-benzodiazepin-2-one in 15 ml of N, N-dimethylformamide becomes a suspension of 0, 3 g of sodium methylate are added to 15 ml of N, N-dimethylformamide. The mixture is heated to 50 to 60 ° C. for 1 hour, then cooled and a solution of 2 ml of p-methylsulfinylethyl chloride in 10 ml of toluene is added below 100 ° C. b) A solution of 0.5 g of 2- (3-methylsulfinylethyl) -amino-5-chlorobenzophenone in 20 ml of anhydrous methylene chloride is mixed with 0.5 g of oxazolidine-2,5-dione. The 1 - (ss-methylsulfinylethyl) -5-phenyl-7-chloro-1,3-hydro-2H-1,4-benzodiazepin-2-one crystallizes and then melts at 166 to 1670C.
c) A mixture of 1 g of 2- [N1-B-methylsulfinylethyl) - -N1- (N2-carbobenzoxyglycyl) -amido] -5-chlorobenzophenone and 20 ml of a 30 percent solution of hydrogen bromide in acetic acid is stirred at room temperature. After the reaction has ended, the mixture is poured into water, made alkaline with aqueous ammonia solution and extracted with methylene chloride. The methylene chloride extract is dried over sodium sulfate and evaporated. The residue is recrystallized from isopropanol. After another recrystallization from acetone, the 1 - (, (3-methylsulfinylethyl) -5-phenyl-7-chloro-1,3-dihydro-2H-1,4-benzodiazepin-2-one obtained melts at 166 ° to 1670 ° C. .
d) A mixture of 1 g of 2- (N-phthalimidoacetyl-N-p -methylsulfinylethyl) amino-5-chlorobenzophenone, 15 ml of methylene chloride and 15 ml of ethanol is mixed with a solution of 2 g of hydrazine hydrate in 2 ml of water and stirred at room temperature. After the reaction has ended, the reaction mixture is evaporated under reduced pressure, the residue is diluted with water, made alkaline with aqueous ammonia solution and extracted with ether. Example I.
10 ml of 1 percent aqueous hydrogen peroxide solution are added to a mixture of 1.5 g of 1- (ss- -methylthioethyl) -5-phenyl-7-chloro-1,3-dihydro-2H-1,4-benzodiazepine while cooling with ice. 2-one and 20 ml acetic acid added. Example 2
15 ml of a 30 percent aqueous hydrogen peroxide solution are stirred into a mixture of 1.7 g of 1 - (p-methylthioethyl) -5-phenyl-7-chloro-1,3-dihydro-2H-1,4-benzodiazepin-2-one and added 5 ml of acetic acid, and heated to 35 to 40 ° C. for 3 hours.
