Die Erfindung betrifft ein Verfahren zur Herstellung neuer Imidoester der Formel I (siehe Formelblatt), worin R fir niederes Alkyl und n fir eine ganze Zahi von 1 bis 6 stehen.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I, indem man eine Verbindung der Formel II, worin n obige Bedeutung besitzt, mit einer Verbindung der Formel III, worin R obige Bedeutung besitzt, umsetzt.
Das Verfahren kann vorzugsweise in dem an der Reaktion beteiligten Alkohol der Formel III als Lösungsmittel aber auch unter Verwendung eines zusitzlichen, unter den Reaktionsbedingungen inerten Lösungsmittels ausgeftihrt werden.
Die durch R symbolisierten niederen Alkylgruppen besitzen vorzugsweise 1 bis 4 Kohlenstoffatome und bedeuten insbesondere die Athylgruppe.
Die Verbindungen der Formel I können in ihre Säure- additionssalze aberftibrt werden und umgekehrt.
Die als Ausgangsprodukte benötigten Verbindungen der Formel II können erhalten werden, indem man die Verbindung der Formel IV mit einer Verbindung der Formel V, worin n obige Bedeutung besitzt und X fur den Säurerest eines reaktionsfihigen Esters steht, reagieren list.
Soweit die Herstellung der Ausgangsprodukte nicht be schrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre pharmakologisch vertriglichen Siureadditionssalze besitzen bei geringer Toxizitit wertvolle pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Sie entfalten eine antivirale Wirksamkeit, wie sich durch Untersuchungen in vitro in Plaque-Reduktionstests sowie im Hämagglutinin-Zunahme- Test unter Venvendung verschiedener Viren zeigen list. Diese antivirale Wirksamkeit zeigt sich durch Hemmung des Wachstums, die bei einer Konzentration von ca. 0,003 bis 3 g/ml festgestellt wurde. Daher können die erfindungsgemissen Substanzen als Mittel gegen Virusinfektionen, vor allem als Prophylaktika gegen Grippe, vex vendee werden.
Fir die Dauer der Infektionsgefuhrdung betrigt die Tagesdosis fur Erwachsene etwa 100 bis 400 mg Wirkstoff, vorzugsweise verabreicht in Teilmengen zwischen etwa 35 bis 200 mg 2- bis 3mal täglich.
Die nach dem erfindungsgemissen Verfahren erhiltlichen Verbindungen kdnnen bei der antiviralen Behandlung auf jede Art verabreicht werden, welche einen Kontakt der aktiven Verbindung mit dem Sitz der Virusinfektion im Körper bewirkt. Damit ist die Behandlung der Infektionsstelle sowohl vor der Ausbildung der Infektion als auch nachher zu verstehen. Beispielsweise kann die Verabreichung parenteral, das ist subcutan, intravenös, intramuskulir oder intraperitonal erfolgen.
Alternativ oder gleichzeitig können die Verbindungen wirksam oral verabreicht werden. Da die Verbindungen besonders gegen Infektionen der Atmungsorgane, wie virale Influenza oder virale Pneumonie, wirksam sind, kann die Ver abreichung mittels Dampf oder Spray durch die Mund- oder Nasenginge erfolgen.
Die aktive Verbindung kann in entsprechenden Zusammensetzungen in Form von Tabletten, Kapseln, Pulverpackungen oder fitissigen Lösungen, Suspensionen oder Elixieren fur die orale Verabreichung, oder in Form fitissiger Lösungen fur die parenterale Venvendung, und in gewissen Fallen in Form von Suspensionen fur parenteralen Gebrauch (ausgenommen intravenös) angewendet werden. In solchen Zusammensetzungen ist der aktive Bestandteil gewöhnlich in Mengen von wenigstens 0,0001 Gew.%, bezogen auf das Gesamtgewicht der Zusammensetzung, und in Mengen von nicht mehr als 99 Gew. % zugegen.
Neben dem aktiven Bestandteil enthält die antivirale Zusammensetzung einen festen oder flissigen, nichttoxischen pharmazeutischen Trigger fur den aktiven Bestandteil.
Der feste Triger kann eine Kapsel sein, z. B. eine übliche Gelatinetype. Ferner kann der aktive Bestandteil mit oder ohne Hilfsmittel tablettiert oder in Pulverpackungen eingebracht werden.
Wie bereits erwähnt wurde, kann der pharmazeutische Trigger eine sterile Flüssigkeit, wie Wasser oder ein Ol sein.
Gewöhnlich stellen Wasser, Salzlösungen sowie wissrige Dextroselösungen (Glucose) und ähnliche Zuckerlösungen vorzugsweise flissige Trigger dar, insbesondere fur Injektionslösungen.
Die orale Verabreichung kann in Form einer geeigneten Suspension oder eines Sirups erfolgen, wobei der aktive Bestandteil gewöhnlich in einer Menge von etwa 0,5 bis 10 Gew. %, vorzugsweise etwa 2 bis 5 Gew. %, vorhanden ist.
In dem nachfolgenden Beispiel, das die Erfindung niter erliutern, ihren Umfang aber in keiner Weise einschrinken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel
N-(1-Adamantyl)amino-acetimidoäthylester
Eine Lösung von 23,19 g N-(1-Adamantyl)aminoacetonitril in 20 ml absolutem Athanol wird mit 80 ml gesättigter äthanolischer Salzsiure 24 Stunden bei Zimmertemperatur stehengelassen. Die Lösung wird anschliessend im Vakuum eingedampft und mit anther gefillt. Nach dem Absaugen, Waschen mit anther und Trocknen erhilt man die Titelverbindung mit Schmp. 260 .
D as als Ausgangsprodukt bendtigte N-(1 -Adamantyl)- aminoacetonitril kann folgendermassen erhalten werden:
Eine Lösung von 10,75 g 1-Aminoadamantan in 200 ml Dimethylformamid wird mit 4,5 ml Chloracetonitril und 4,88 g Kaliumkarbonat 4 Tage bei Zimmertemperatur ge rihrt. Man filtriert vom ausgefallenen Salz ab und dampft im Vakuum bei 30 und 10-J Torr ein. Der Eindampfungsrück- stand wird in anther gelöst, nochmals von Salzresten filtriert und schliesslich eingedampft. Man erhält ein weisses Kristallisat vom Schmp. 54-56 .

The invention relates to a process for the preparation of new imido esters of the formula I (see formula sheet), in which R is lower alkyl and n is a whole number from 1 to 6.
According to the invention, the compounds of the formula I are obtained by reacting a compound of the formula II, in which n is as defined above, with a compound of the formula III, in which R is as defined above.
The process can preferably be carried out in the alcohol of the formula III involved in the reaction as the solvent, but also using an additional solvent which is inert under the reaction conditions.
The lower alkyl groups symbolized by R preferably have 1 to 4 carbon atoms and are in particular the ethyl group.
The compounds of the formula I can be converted into their acid addition salts and vice versa.
The compounds of the formula II required as starting materials can be obtained by reacting the compound of the formula IV with a compound of the formula V in which n has the above meaning and X stands for the acid residue of a reactive ester.
If the production of the starting products is not described, they are known or can be produced by processes known per se or analogously to those described here or analogously to processes known per se.
The compounds of the formula I and their pharmacologically acceptable acid addition salts have valuable pharmacodynamic properties with low toxicity and can therefore be used as medicaments. They develop an antiviral effectiveness, as shown by studies in vitro in plaque reduction tests and in the hemagglutinin increase test using various viruses. This antiviral activity is shown by inhibition of growth, which was found at a concentration of approx. 0.003 to 3 g / ml. Therefore, the substances according to the invention can be used as agents against viral infections, especially as prophylactic agents against influenza.
For the duration of the risk of infection, the daily dose for adults is about 100 to 400 mg of active ingredient, preferably administered in partial amounts between about 35 to 200 mg 2 to 3 times a day.
The compounds obtainable by the process according to the invention can be administered in the antiviral treatment in any way which brings about contact of the active compound with the location of the viral infection in the body. This means that the treatment of the infection site is to be understood both before the infection develops and afterwards. For example, the administration can take place parenterally, that is subcutaneously, intravenously, intramuscularly or intraperitoneally.
Alternatively, or simultaneously, the compounds can be effectively administered orally. Since the compounds are particularly effective against infections of the respiratory organs, such as viral influenza or viral pneumonia, the administration can be carried out by means of steam or spray through the oral or nasal gingiva.
The active compound can be in appropriate compositions in the form of tablets, capsules, powder packs or ready-to-use solutions, suspensions or elixirs for oral administration, or in the form of ready-to-use solutions for parenteral use, and in certain cases in the form of suspensions for parenteral use (except intravenous). In such compositions, the active ingredient is usually present in amounts of at least 0.0001% by weight based on the total weight of the composition and in amounts of no more than 99% by weight.
In addition to the active ingredient, the antiviral composition contains a solid or liquid, non-toxic pharmaceutical trigger for the active ingredient.
The solid trigger can be a capsule, e.g. B. a common type of gelatin. In addition, the active ingredient can be tabletted with or without an adjuvant or incorporated in powder packs.
As mentioned earlier, the pharmaceutical trigger can be a sterile liquid, such as water or an oil.
Usually, water, saline solutions and aqueous dextrose solutions (glucose) and similar sugar solutions are preferably liquid triggers, especially for injection solutions.
Oral administration can be in the form of a suitable suspension or syrup, the active ingredient usually being present in an amount of about 0.5 to 10% by weight, preferably about 2 to 5% by weight.

A solution of 23.19 g of N- (1-adamantyl) aminoacetonitrile in 20 ml of absolute ethanol is left to stand with 80 ml of saturated ethanolic hydrochloric acid for 24 hours at room temperature. The solution is then evaporated in vacuo and filled with anther. After filtering off with suction, washing with anthers and drying, the title compound with melting point 260 is obtained.
The N- (1-adamantyl) - aminoacetonitrile finished as the starting product can be obtained as follows:

White crystals with a melting point of 54-56 are obtained.
