Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen Chinazolinderivaten und insbesondere von solchen der folgenden allgemeinen Formel:

In den Verbindungen der obigen allgemeinen Formel (I-a) bezeichnet Alkyl sowohl geradkettige als auch verzweigte aliphatische Kohlenwasserstoffreste, während als niedere Alkylgruppen beispielsweise die folgenden in Frage kommen: Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl- und tert.-Butylgruppen. Als Aralkylgruppen kommen beispielsweise Benzyl-, Phenyläthyl-, Chlorbenzyl- und Fluorbenzylgruppen in Frage. Die niederen Alkoxyreste umfassen beispielsweise die folgenden Gruppen: Methoxy, Äthoxy, n-Propoxy. Isopropoxy, n-Butoxy und tert.-Butoxy.
Die niederen Alkylthiogruppen umfassen beispielsweise Methylthio-, Äthylthio- und Butylthiogruppen, während als niedere Alkanoyloxygruppen beispielsweise Acetoxy- und Propionyloxygruppen in Frage kommen. Die Trihalogenmethylgruppe umfasst beispielsweise die Trichlormethyl- und Trifluormethylreste. Als niedere Alkenylgruppen kommen beispielsweise die folgenden Gruppen in Frage: Vinyl, Allyl,
Methallyl, Butenyl und Crotyl. Als Cycloalkylreste kommen beispielsweise Cyclopropyl-, Cyclobutyl-, Cyclopentylund Cyclohexylreste in Frage. Als Cycloalkenylreste kann man beispielsweise Cyclopentenyl- und Cyclohexenylreste in Betracht ziehen.
Die Alkylengruppe der Formel: CnH2n ist eine geradkettige oder verzweigte Alkylengruppe mit 1 bis 4
Kohlenstoffatomen und umfasst beispielsweise folgende Gruppen: Methylen, Äthylen, 1-Methyläthylen, 2-Methyl äthylen, Trimethylen, l-Methyltrimethylen und 2-Methyltrimethylen. R4 und R5 sowie R; und R7 können zusammen mit dem benachbarten Stickstoffatom einen heterocyclischen Ring bilden, wobei als heterocyclische Gruppen beispielsweise die Pyrrolidino-, Piperidino- und Morpholinoreste und substituierte Derivate davon in Frage kommen.
Die Chinazolinderivate der allgemeinen Formel (I-a), welche neue Verbindungen umfassen, besitzen hervorragende pharmakologische Eigenschaften und sind insbesondere als entzündungshemmende Mittel und Analgetika wirksam. Sie eignen sich als Zwischenprodukte für die Synthese von anderen Arzneien. Die vorliegende Erfindung stellt daher ein neues und wertvolles Verfahren für die Herstellung von wirtschaftlich interessanten Verbindungen dar.
Es sind bereits einige Methoden für die Herstellung einiger dieser Chinazolinderivate beschrieben worden. So kann man beispielsweise Chinazolinderivate herstellen, indem man ein 2-Aminophenylketonderivat mit Harnstoff zum Schmelzen bringt und hierauf gewünschtenfalls das erhaltene Produkt alkyliert. [K. Schofield, 5. Chem. Soc., 4034 (1954)1.
Es wurde indessen überraschenderweise festgestellt, dass man beim Arbeiten gemäss vorliegender Erfindung Chinazolinderivate der Formel (I-a) leicht und in wirtschaftlicher Weise bei hohen Ausbeuten erhalten kann. Ferner lassen sie sich in hohem Reinheitsgrad herstellen, wobei man so vorgeht, dass man ein in 1-Stellung nicht substituiertes Chinazolinderivat der Formel:

worin R1, R2 und R'3 die obigen Bedeutungen haben, mit einem reaktionsfähigen Ester einer Verbindung der Formel: R'-OH (V) worin R' die obige Bedeutung hat, umsetzt.
Die erfindungsgemäss erhältlichen Verbindungen zeichnen sich ferner durch eine niedrige Toxizität aus und eignen sich auch als Zwischenprodukte für die Synthese von anderen Chinazolinderivaten. So besitzen beispielsweise l-Cyclo- propylmethyl.4.(2'.thienyl).6-chlor-2-(lH)-chinazolinon und 1 -Cyclopropylmethyl-4-cyclohexyl-6-chlor-2-(lH)-chinazolinon eine hervorragende Hemmwirkung für durch isländisches Moos erzeugte Oedeme bei Ratten, während keine toxischen Symptome beobachtet werden können. Die Prüfung der Faeces auf okkulte Blutungen bei Ratten nach peroraler Verabreichung von 1000 mg/kg (bei Ratten) fällt negativ aus.
Die entzündungshemmende Wirkung dieser Verbindungen ist wesentlich stärker als jene von 1,2-Diphenyl -3,5-dioxo-4-n-butylpyrazolidin (Phenylbutazon), während überdies die akute, subakuten und chronischen Toxizitätswerte wesentlich niedriger als jene von Phenylbutazon sind.
Zweck der vorliegenden Erfindung ist daher die Schaffung einer neuen Methode für die Herstellung von wirtschaftlich sehr wertvollen Chinazolinderivaten mit hervorragenden entzündungshemmenden Wirkungen.
Die erfindungsgemäss erhältlichen, der Formel (I-a) entsprechenden Verbindungen lassen sich in andere Verbindungen überführen, welche den Verbindungen der Formel (I-a) entsprechen. So kann man beispielsweise der Formel (I-a) entsprechende Verbindungen, bei denen Rl und/oder R2 Wasserstoff bedeuten, nitrieren. Verbindungen, bei denen R1 oder R2 eine Nitrogruppe darstellt, lassen sich reduzieren, wobei man die entsprechenden Verbindungen enthält, welche einen Aminosubstituenten tragen, worauf man diese Aminogruppe durch einen Halogensubstituenten ersetzen kann.
Ferner kann man Verbindungen, bei denen R ein Wasserstoffatom darstellt, in die entsprechenden Verbindungen überführen, bei denen das Wasserstoffatom durch einen der folgenden Reste ersetzt ist, nämlich niederes Alkyl, niederes Alkenyl, Aralkyl, Haloalkyl, Trihalomethylalkyl, Äthoxyalkyl, Cycloalkylalkyl, Alkylthioalkyl, Alkanoyloxyalkyl, Dialkylaminoalkyl oder Dialkylaminocarbonylalkyl, indem man alkyliert, alkenyliert, aralkyliert, haloalkyliert, tribalogenmethylalkyliert, alkoxyalkyliert, cycloalkylalkyliert, alkanoyloxyalkyliert, dialkylaminoalkyliert oder dialkylaminocarbonylalkyliert.
Beispiele von reaktionsfähigen Estern der Formel:
R'OH sind die Hydrohalogensäureester, wie z.B. die Chloride, Bromide und Jodide, sowie Sulfonsäureester, wie z.B. p-Toluolsulfonat, Methansulfonat und Trichlormethansulfonat. Die Umsetzung kann in Gegenwart eines alkalischen Mittels erfolgen, um das entsprechende Metallsalz zu erhalten, worauf man dieses Metallsalz mit einem reaktionsfähigen Ester einer Verbindung der Formel V behandelt.
Beispiele von alkalischen Mitteln sind Alkalimetallhydride, wie z.B. Natriumhydrid oder Kaliumhydrid, Alkalimetallamide, wie z.B. Natriumamid oder Kaliumamid, organische Lithiumverbindungen, wie z.B. Butyllithium oder Phenyllithium, Alkalimetallalkoxyde, wie z.B. Natriummethoxyd, Kaliummethoxyd, Natriumäthoxyd, Kaliumäthoxyd oder Kalium-tert.-butoxyd usw. Die Reaktion erfolgt im allgemeinen in einem organischen Lösungsmittel oder einem Lösungsmittelgemisch. Geeignete Lösungsmittel sind Benzol, Toluol, Xylol, Monochlorbenzol, Dimethylacetamid, Di äthylacetamid, Dimethylformamid, Dioxan und Dimethylsulfoxid.
Die Umsetzung kann in einem Temperaturbereich, wel cher zwischen Zimmertemperatur und dem Siedepunkt des Lösungsmittels liegt, durchgeführt werden.
Die Umsetzung ist häufig von der Bildung von Chinazolinderivaten der Formel:

Das Abtrennen der gewünschten Chinazolinderivate der Formel (I-a) von den Chinazolinderivaten der Formel VI kann in an sich bekannter Weise, beispielsweise durch Chromatographie, erfolgen. Beim Arbeiten nach den obigen Methoden kann man beispelsweise die folgenden Chi- nazolinderivate herstellen.

Die vorliegende Erfindung wird nun ausführlich anhand der folgenden Beispiele erläutert.
Beispiel I
Zu einer Lösung von 2,74 g 4-(2'-Thienyl)-2(1H)-chinazolinon in 60 cm8 Dimethylformamid gibt man portionsweise 0,50 g 63 %ges Natriumhydrid hinzu. Dann wird das Gemisch während 30 Minuten unter Rühren auf 55 bis 600C erwärmt und hierauf auf Zimmertemperatur abgekühlt.
Hierauf wird das Gemisch mit 3,6 g 90%iges Cyclopropylmethylbromid versetzt und das anfallende Gemisch unter Rühren während 6 Stunden auf 100"C erhitzt. Nach dem Kühlen wird das Reaktionsgemisch in 300"C Wasser gegossen und zweimal mit Chloroform extrahiert. Die Chloroformextrakte werden vereinigt, mit Wasser gewaschen und über Natriumsulfat getrocknet. Hierauf wird das Lösungsmittel unter vermindertem Druck entfernt, wobei man einen braunen öligen Rückstand erhält, welcher mittels Silicium dioxydgel unter Verwendung von Chloroform als Eluiermittel chromatographiert wird.
Auf diese Weise erhält man 2,0 g l-Cyclopropylmethyl-4-(2'-thienyl)-2(1H)-chinazolinon und 1,0 g 2-Cyclopropylmethoxy-4- (2'-thienyl)-chinazolin. Die erstere Verbindung wird aus einer Mischung von Äthanol und Isopropyläther umkristallisiert, wobei man hellgelbe Nadeln vom Schmelzpunkt 89,0 bis 91,00C erhält. Die letztere Verbindung wird aus einer Mischung von Äthanol und Petroläther umkristallisiert, wobei man hellgelbe, feine Kristalle vom Schmelzpunkt 83,0 bis 84,0 C erhält.
Beispiel 2
Zu einer Suspension von 3,15 g 4-(2'-Thienyl)-6-chlor -2(1H)-chinazolinon in 80 cm8 Dimethylformamid werden portionsweise 0,50 g 63%iges Natriumhydrid hinzugegeben.
Das Gemisch wird während 30 Minuten unter Rühren auf 55"C erhitzt und hierauf auf Zimmertemperatur gekühlt.
Anschliessend wird das Gemisch mit 3,6 g 90%iges Cyclopropylmethylbromid versetzt, worauf man das Gemisch in ähnlicher Weise wie in Beispiel 1 behandelt. Auf diese Weise erhält man 2,05 g l-Cyclopropylmethyl-4-(2'4hlenyl)-6- -chlor-2(1H)-chinazolinon, das man aus einer Mischung von Äthanol, Äther und Leichtbenzin umkristallisiert. Auf diese Weise erhält man gelbe Prismen vom Schmelzpunkt 140,0 bis 141,0 C und 1,65 g 2-Cyclopropylmethoxy-4-(2' -thienyl)-6-chlor-2(1H)-chinazolinon das man aus einer Mischung von Äthanol und Leichtbenzin umkristallisiert, wobei man hellgelbe Nadeln vom Schmelzpunkt 97,0 bis 98,00C erhält.
Beispiel 3
Man arbeitet in ähnlicher Weise wie in Beispiel 1, jedoch ersetzt man das 4-(2'-Thienyl)-2(1H)-chinazolinon durch 2,96 g 4-(2'-furyl).6-chlor-2(1H)-chinazolinon. Dabei erhält man 1,4 g 1-Cyclopropylmethyl-4-(2'-furyl)-6-chlor -2(1H)-chinazolinon und 1,3 g 2-Cyclopropylmethoxy-4-(2'- -furyl)-6-chlorchinazolin. Die erstere Verbindung wird aus einer Mischung von Äthanol und Isopropyläther umkristallisiert, wobei man gelbe Prismen vom Schmelzpunkt 160,0 bis 161,00C erhält. Die leztere Verbindung wird aus Isopropyläther umkristallisiert, wobei man gelbe Nadeln vom Schmelzpunkt 109,0 bis 1 10,00C erhält.
Man arbeitet in ähnlicher Weise wie in Beispiel 1, ersetzt aber das 4-(2'-Thienyl)-2(1H)-chinazolinon durch 3,94 g 4-Cyclohexyl-6-chlor-2(1H)-chinazolinon. Dabei erhält man 1,9 g 1-Cydopropylmethyl-4-cyclohexyl-6-chlor-2(1H)-chi- nazolinon und 1,62 g 2-Cyclopropylmethoxy-4-cyclohexyl6- -chlorchinazolin.
Die erstere Verbindung wird aus Äthanol umkristallisiert, wobei man farblose Nadeln vom Schmelzpunkt 157,0 bis 157,5"C erhält. Die letztere Verbindung wird aus Isopropyläther umkristallisiert, wobei man farblose Nadeln vom Schmelzpunkt 108,5 bis 109,00C erhält.
Beispiel 4
Unter Anwendung einer ähnlichen Methode wie jener gemäss Beispiel 1, jedoch durch Ersatz des 4-(2'-Thienyl) -2(1H)-chinazolinons durch 4-Methyl-6-chlor-2(1H)-chinazolinon erhält man l-Cyclopropylmethyl-$methyl-6-chlor- -2(1H)-chinazolinon in Form von bräunlichgelben Nadeln vom Schmelzpunkt 164 bis 165"C, sowie 2-Cydopropyl- methoxy-4-methyl-6-chlorchinazolin in Form eines orangenen öIs.
Ferner kann man nach den Angaben von Beispiel 1 auch die 4 folgenden Verbindungen herstellen: 1 -Cyclopropylmethyl-4-(2'-thlenyl)-6-nitro-2(1H)-chi- nazolinon, Smp. 199,0-199,50C, 2-Cydopropylmethoxy-4-(2'-thienyl)-6-nitrochinazolin, Smp. 157,5-158,00C,

The quinazoline derivatives represented by the general formula (I-a), which comprise novel compounds, have excellent pharmacological properties and are particularly effective as anti-inflammatory agents and analgesics. They are suitable as intermediates for the synthesis of other medicines. The present invention therefore represents a new and valuable process for the preparation of economically interesting compounds.
Some methods for the preparation of some of these quinazoline derivatives have already been described. For example, quinazoline derivatives can be prepared by melting a 2-aminophenyl ketone derivative with urea and then, if desired, alkylating the product obtained. [K. Schofield, 5. Chem. Soc., 4034 (1954) 1.
However, it has surprisingly been found that, when working according to the present invention, quinazoline derivatives of the formula (I-a) can be obtained easily and economically at high yields. Furthermore, they can be produced in a high degree of purity, the procedure being that a quinazoline derivative of the formula that is not substituted in the 1-position is obtained:

The compounds obtainable according to the invention are also distinguished by a low toxicity and are also suitable as intermediates for the synthesis of other quinazoline derivatives. For example, 1-cyclopropylmethyl.4. (2'.thienyl) .6-chloro-2- (1H) -quinazolinone and 1-cyclopropylmethyl-4-cyclohexyl-6-chloro-2- (1H) -quinazolinone have one excellent inhibitory effect on edema caused by Icelandic moss in rats, while no toxic symptoms can be observed. The examination of the faeces for occult bleeding in rats after oral administration of 1000 mg / kg (in rats) turns out negative.
The anti-inflammatory effect of these compounds is much stronger than that of 1,2-diphenyl -3,5-dioxo-4-n-butylpyrazolidine (phenylbutazone), while moreover the acute, subacute and chronic toxicity values are much lower than those of phenylbutazone.
The purpose of the present invention is therefore to provide a new method for the production of economically very valuable quinazoline derivatives with excellent anti-inflammatory effects.
The compounds corresponding to formula (I-a) obtainable according to the invention can be converted into other compounds which correspond to the compounds of formula (I-a). For example, compounds corresponding to formula (I-a) in which R1 and / or R2 are hydrogen can be nitrated. Compounds in which R1 or R2 represents a nitro group can be reduced, in which case the corresponding compounds which carry an amino substituent are obtained, whereupon this amino group can be replaced by a halogen substituent.
Furthermore, compounds in which R represents a hydrogen atom can be converted into the corresponding compounds in which the hydrogen atom has been replaced by one of the following radicals, namely lower alkyl, lower alkenyl, aralkyl, haloalkyl, trihalomethylalkyl, ethoxyalkyl, cycloalkylalkyl, alkylthioalkyl, alkanoyloxyalkyl , Dialkylaminoalkyl or dialkylaminocarbonylalkyl, by alkylating, alkenylating, aralkylating, haloalkylating, tribalomethylalkylated, alkoxyalkylated, cycloalkylalkylated, alkanoyloxyalkylated, dialkylaminoalkylated or dialkylaminocarbonylalkylated.
Examples of reactive esters of the formula:

Examples of alkaline agents are alkali metal hydrides, e.g. Sodium hydride or potassium hydride, alkali metal amides, e.g. Sodium amide or potassium amide, organic lithium compounds, e.g. Butyllithium or phenyllithium, alkali metal alkoxides, e.g. Sodium methoxide, potassium methoxide, sodium ethoxide, potassium ethoxide or potassium tert-butoxide, etc. The reaction generally takes place in an organic solvent or a mixture of solvents. The present invention will now be explained in detail with reference to the following examples.
Example I.
To a solution of 2.74 g of 4- (2'-thienyl) -2 (1H) -quinazolinone in 60 cm8 of dimethylformamide, 0.50 g of 63% saturated sodium hydride is added in portions. The mixture is then heated to 55 to 60 ° C. for 30 minutes with stirring and then cooled to room temperature.
3.6 g of 90% strength cyclopropylmethyl bromide are then added to the mixture, and the resulting mixture is heated to 100 ° C. for 6 hours with stirring. After cooling, the reaction mixture is poured into 300 ° C. water and extracted twice with chloroform. The chloroform extracts are combined, washed with water and dried over sodium sulfate. The solvent is then removed under reduced pressure to give a brown oily residue which is chromatographed on silicon dioxide gel using chloroform as the eluent.
In this way, 2.0 g of 1-cyclopropylmethyl-4- (2'-thienyl) -2 (1H) -quinazolinone and 1.0 g of 2-cyclopropylmethoxy-4- (2'-thienyl) -quinazoline are obtained. The former compound is recrystallized from a mixture of ethanol and isopropyl ether to give pale yellow needles with a melting point of 89.0 to 91.00C. The latter compound is recrystallized from a mixture of ethanol and petroleum ether, pale yellow, fine crystals with a melting point of 83.0 to 84.0 ° C. being obtained.
Example 2
To a suspension of 3.15 g of 4- (2'-thienyl) -6-chloro -2 (1H) -quinazolinone in 80 cm8 of dimethylformamide, 0.50 g of 63% sodium hydride are added in portions.
The mixture is heated to 55 ° C. for 30 minutes with stirring and then cooled to room temperature.
3.6 g of 90% strength cyclopropylmethyl bromide are then added to the mixture, whereupon the mixture is treated in a manner similar to that in Example 1. In this way, 2.05 g of l-cyclopropylmethyl-4- (2'4hlenyl) -6- -chlor-2 (1H) -quinazolinone are obtained, which is recrystallized from a mixture of ethanol, ether and mineral spirits. In this way, yellow prisms with a melting point of 140.0 to 141.0 ° C. and 1.65 g of 2-cyclopropylmethoxy-4- (2'-thienyl) -6-chloro-2 (1H) -quinazolinone are obtained, which are obtained from a mixture recrystallized from ethanol and mineral spirits, giving pale yellow needles with a melting point of 97.0 to 98.00C.
Example 3
The procedure is similar to that of Example 1, except that the 4- (2'-thienyl) -2 (1H) -quinazolinone is replaced by 2.96 g of 4- (2'-furyl) .6-chloro-2 (1H ) -quinazolinone. This gives 1.4 g of 1-cyclopropylmethyl-4- (2'-furyl) -6-chloro -2 (1H) -quinazolinone and 1.3 g of 2-cyclopropylmethoxy-4- (2'-furyl) -6 -chlorquinazoline. The former compound is recrystallized from a mixture of ethanol and isopropyl ether, yellow prisms with a melting point of 160.0 to 161.00C being obtained. The last compound is recrystallized from isopropyl ether, yellow needles with a melting point of 109.0 to 10.00C being obtained.
The procedure is similar to that in Example 1, but the 4- (2'-thienyl) -2 (1H) -quinazolinone is replaced by 3.94 g of 4-cyclohexyl-6-chloro-2 (1H) -quinazolinone. This gives 1.9 g of 1-cyclopropylmethyl-4-cyclohexyl-6-chloro-2 (1H) -quinazolinone and 1.62 g of 2-cyclopropylmethoxy-4-cyclohexyl6-chloroquinazoline.
The former compound is recrystallized from ethanol, giving colorless needles with a melting point of 157.0 to 157.5 "C. The latter compound is recrystallized from isopropyl ether, giving colorless needles with a melting point of 108.5 to 109.00C.
Example 4
Using a method similar to that of Example 1, but replacing the 4- (2'-thienyl) -2 (1H) -quinazolinone with 4-methyl-6-chloro-2 (1H) -quinazolinone, l-cyclopropylmethyl is obtained - $ methyl-6-chloro--2 (1H) -quinazolinone in the form of brownish-yellow needles with a melting point of 164 to 165 "C, and 2-cydopropyl methoxy-4-methyl-6-chloroquinazoline in the form of an orange oil.
