Die Erfindung betrifft ein Verfahren zur Herstellung von gegebenenfalls deuterohaltigem 3-Fluor-D-alanin, das ein wirksames antibakterielles Mittel, das das Wachstum sowohl grampositiver als auch gramnegativer pathogener Bakterien hemmt, ist.
Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man ein N-acyliertes Derivat eines gegebenenfalls deuterohaltigen 3^Fluor-DL-alanins der enzymatischen Wirkung von Renalacylase I aussetzt, wodurch das L-Isomer desacyliert wird, dieses von dem unveränderten gegebenenfalls deuterohaltigen N-Acyl-3-fluor-D-alanin abtrennt und dieses mit einem Hydrolysierungsmittel umsetzt.
Als bevorzugtes deuterohaltiges N-Acyl-3-Fluor-DL-alanin wird N-Chloracetyl-3-DL-alanin eingesetzt, und bei der Durchführung des erfindungsgemässen Verfahrens findet dann eine selektive asymmetrische Hydrolyse des L-Isomers unter Bildung eines Gemisches aus 3-Fluor-L-alanin und N -Chloracetyl-3-fluor-D-alanin bzw. der Deutero-Analoga statt.
Die Auftrennung dieses Gemisches kann durch fraktionierte Kristallisation aus Äthanol mit anschliessender Hydrolyse des N-Chloracetyl-3-fluor-D-alanins ausgeführt werden.
Als Ausgangsmaterial wird im erfindungsgemässen Verfahren vorzugsweise N-Chloracetyl-3-fluor-DL-alanin, N -Chloracetyl-2-deutero-3-fluor-DL-alanin, N-Chloracetyl -2,3,3-trideutero-3-fluor-D-alanin und dgl. verwendet.
Das für die Deacetylierung verwendete Enzym, die Schweinerenalacylase I, ist von Greenstein und Winitz in aChemistry ob tee Amino Acide, Band 3, Seite 1831(1961) beschrieben. Die enzymatische Desacetylierung wird gewöhnlich bei 37"C für etwa 24 Stunden durchgeführt, wonach das L-Isomer praktisch vollständig desacetyliert ist und leicht von dem unveränderten N-Chloracetyl-3-fluor-D-alanin-isomer abgetrennt werden kann.
Das folgendeBeispiel veranschaulicht eineDurchführungsform des Verfahrens gemäss der Erfindung.
Beispiel
Eine Lösung von 5 Mol N-Chloracetyl-DL-3-fluor-alanin in 5 Liter Wasser, mit konzentriertem wässrigem Ammoniak auf pH 7,0 eingestellt, wird mit 150 mg gereinigter Schweinerenalacylase I vereinigt und 24 Stunden bei 370C bebrütet.
Das dabei erhaltene Produkt (digest), das freies 3-Fluor-L -alanin und N-Chloracetyl-3-fluor-D-alanin enthält, wird deproteinisiert und entfärbt, indem man nacheinander Essigsäure bis zu einer Konzentration von 0,2 m und Aktivkohle (Norit) bis zu einer Menge von 2 mg/ml zugibt und dann filtriert. Das Filtrat wird im Vakuum bis zu 10% seines Anfangsvolumens eingedampft, und 6 Volumen Äthanol werden zugesetzt, worauf die Hauptmenge des 3-Fluor-L-alanins kristallisiert und abfiltriert werden kann. Der Rest des 3-Fluor-L-alanins wird selektiv abgetrennt, indem man die verdünnte Mutterlauge über ein Ionenaustauscherharzbett (Dowex-50-H+) mit einer Austauscherkapazität, die 20% der anfänglich in dem Digestionsprodukt anwesenden Aminosäure gleichkommt, führt.
Das Eluat der Säule wird mit 2n wässriger Salzsäure 2 Stunden auf 100"C erhitzt. Das pH des Hydrolysats wird mit wässrigem Ammoniak auf 4,8 eingestellt. Ein gleiches Volumen Äthanol wird zugesetzt, und das ausgefallene Material wird abfiltriert und getrocknet, wobei man das 3-Fluor-D -alanin erhält.
PATENTANSPRUCH
Verfahren zur Herstellung von gegebenfalls deuterohaltigem 3-Fluor-D-alanin, dadurch gekennzeichnet, dass man ein N-acyliertes Derivat eines gegebenenfalls deuterohaltigen 3 -Fluor-DL-alanins der enzymatischen Wirkung von Renalacylase I aussetzt, wodurch das L-Isomer desacyliert wird, dieses von dem unveränderten gegebenenfalls deuterohaltigen N-Acyl-3-fluor-D-alanin abtrennt und dieses mit einem Hydrolysierungsmittel umsetzt.
UNTERANSPRUCH
Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man das N-acylierte Derivat N-Chloracetyl-3-fluor-DL -alanin verwendet und das bei der enzymatischen Umsetzung erhaltene Gemisch aus 3-Fluor-L-alanin und N-Chloracetyl -3-fluor-D-alanin durch fraktionierte Kristallisation aus wässrigem Äthanol auftrennt.
