Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von Chinazolinonverbindungen der Formel:

worin R1 Wasserstoff, Halogen, z.B. Chlor, Brom, Jod oder Fluor, Niederalkyl, z.B. Methyl, Äthyl, Propyl, Isopropyl oder Butyl, Niederalkoxy, z.B. Methoxy, Äthoxy, Propoxy, Isopropoxy oder Butoxy, Trifluormethyl oder Nitro und R2 Niederalkyl, z.B. Methyl, Äthyl, Propyl, Isopropyl oder Butyl, oder unsubstituiertes oder mit Halogen, z.B. Die Bezeichnung nieder , wie sie weiter oben und im nachstehenden verwendet wird, soll Reste mit 1 bis 5 Kohlenstoffatomen bedeuten. Das Symbol R1, welches ein Atom oder eine Gruppe ist, kann sich in beliebiger Stellung im Ben zofring der Verbindung der Formel (I) oder einer Verbindung einer weiter unten erwähnten Formel befinden.
Die Chinazolinonverbindungen der Formel I sind als wertvolle Antiphlogistika bekannt. Sie eignen sich auch als wertvolle Zwischenprodukte für die Herstellung von anderen Chemikalien, z.B. Antiphlogistika und Mittel zur Dämpfung des zentralen Nervensystems.
Bisher wurden die Chinazolinonverbindungen der Formel I dadurch erhalten, dass man eine Aminoketonverbindung der Formel:

worin R1 und R2 die obigen Bedeutungen haben, beispielsweise mit Urethan kondensiert. Da sich aber diese Aminoketonverbindungen nur schwierig synthetisieren lassen, hat man danach getrachtet, eine geeignetere Methode zu entwikkeln.
Es wurde nun festgestellt, dass man die Chinazolinonverbindungen der Formel I über Indoldicarbonimidverbindungen, die man leicht herstellen kann, unter Verwendung einer neuen Reaktionsart erhalten kann.
Die gesamte Synthese und das erfindungsgemässe Verfahren (letzte Stufe derselben) lassen sich durch das folgende Reaktionsschema wiedergeben:

Bei der ersten Stufe der erwähnten Synthese können Indolcarbonsäureverbindungen der Formel II mit einer Urethanverbindung der Formel:
H2NCOOR3 worin R3 einen niederen Alkylrest, z.B. den Methyl-, Äthyl-, Propyl-, Isopropyl- oder Butylrest, bedeutet, umgesetzt werden.
Für diese Umsetzung kann man die Indolcarbonsäureverbindungen der Formel II in Form von reaktionsfähigen Derivaten davon, z.B. in Form von Säurehalogeniden oder Säureestern, verwenden, sofern dies erwünscht ist. Die Umsetzung erfolgt im allgemeinen in Gegenwart oder in Abwesenheit eines Lösungsmittels, wie z.B. eines Alkohols, z.B. Methanol oder Äthanol, eines aromatischen Kohlenwasserstoffes, z.B.
Benzol, Toluol oder Xylol, eines Ketons, z.B. Aceton, oder eines Äthers, z.B. Diäthyläther, wobei man vorzugsweise erhitzt.
Bei der zweiten Synthesestufe können die so erhaltenen Indoldicarbonimidverbindungen der Formel III durch Oxydation in die Ausgangsprodukte des Verfahrens (IV) übergeführt werden.
Diese Oxydation kann dadurch geschehen, dass man eine Indoldicarbonimidverbindung der Formel III mit einem Oxydationsmittel bei Zimmertemperatur oder unter Erhitzen oder unter Kühlen behandelt. Beispiele von Oxydationsmitteln sind Ozon, Wasserstoffperoxyd, Persäuren, z.B. Peressigsäure, Perameisensäure oder Perbenzoesäure, Chromsäure, Permanganate, z.B. Kaliumpermanganat, Mangandioxyd usw. Die Behandlung erfolgt vorzugsweise in Gegenwart eines inerten Lösungsmittels, das man je nach der Art des Oxydationsmittels in geeigneter Weise auswählen wird. Im allgemeinen verwendet man Wasser, Aceton, Tetrachlorkohlenstoff, Essigsäure, Ameisensäure, Schwefelsäure oder dergleichen.
Verwendet man Ozon als Oxydationsmittel, so erhält man eine Imidazolidinverbindung, d.h eine Ozonidverbindung

Das erfindungsgemässe Verfahren besteht darin, dass man besagte Verbindung der Formel IV durch Pyrolyse bzw. Hydrolyse in die Verbindung der Formel I überführt.
Die Pyrolyse kann dadurch bewirkt werden, dass man eine Imidazolidinverbindung der Formel IV zum Schmelzen oder zum Zersetzen bringt, was im allgemeinen in Gegenwart oder in Abwesenheit eines inerten Lösungsmittels von hohem Siedepunkt, z.B. Dimethylformamid, Dimethylsulfoxyd oder Diäthylenglycoldimethyläther, geschieht. Die Hydrolyse kann dadurch bewirkt werden, dass man eine Imidazolidinverbindung der Formel IV mit Wasser, gewünschtenfalls in Gegenwart einer Säure oder einer Base, behandelt. Als Säure kann man hierfür beispielsweise eine Mineralsäure, z.B.
Salzsäure, Schwefelsäure, Salpetersäure oder Phosphorsäure, verwenden. Basen können beispielsweise Alkalimetallhydroxyd, z.B. Natriumhydroxyd oder Kaliumhydroxyd, Alkalimetallcarbonate, z.B. Kaliumcarbonat, Erdalkalimetallhydroxyde, z.B. Bariumoxyd oder Calciumhydroxyd, und Ammoniumverbindungen, z.B. Ammoniumhydroxyd, sein. Die Verwendung von Alkalimetall- und Erdalkalimetallhydroxyden wird bevorzugt.
Beispiele von auf diese Weise erhältlichen Chinazolinonverbindungen der Formel I sind die folgenden: 4-Phenyl-6-chlor-2(1H)-chinazolinon 4-Phenyl-2(1 H)-chinazolinon 4-(o-Chlorphenyl) -6-chlor-2(1H)-chinazolinon 4-(o-Tolyl)-7-nitro-2(1H)-chinazolinon 4-(p-Bromphenyl)-6-brom-2(1H)-chinazolinon 4-Phenyl-6-nitro-2(1H)-chinazolinon 4-(o-Fluorphenyl)-6-trifluormethyl-2(1H)-chinazolinon 4(0- Nitrophenyl)-7-methyl-2(l H)-chinazolinon 4-Phenyl-6-methoxy-2(1H)-chinazolinon 4. Methyl-6- chlor- 2(1 H)-chinazollnon 4-Athyl-6-nitro-2(1H)-chinazolinon 4-Isopropyl-6-chlor-2(1H)-chinazolinon, usw.
Wie bereits erwähnt, sind die Chinazolinonverbindungen der Formel I bekannte und wertvolle Antiphlogistika. Die Imidazolidinverbindungen der Formel IV sind neue Verbindungen und wertvoll als Zwischenprodukte für die Herstellung der Chinazolinonverbindungen der Formel I. Ausserdem sei darauf hingewiesen, dass die Imidazolidinverbindungen der Formel IV als solche als Antiphlogistika und überdies auch als Beschwichtigungsmittel für das zentrale Nervensystem wertvoll sind.
Die Erfindung sei durch die folgenden Beispiele erläutert.
Herstellung der Ausgangsprodukte (1V) a) Eine Mischung von 30 g 3-(o-Fluorphenyl)-5-chlorin dol-2-carbonsäure und 60 ml Thionylchlorid wird während 2 Stunden unter Rückfluss erhitzt. Das überschüssige Thionylchlorid wird unter vermindertem Druck abdestilliert. Dann wird das verbleibende Säurechlorid mit 30 g Urethan versetzt und das Gemisch unter Rühren erhitzt, wobei das als Nebenprodukt anfallende Äthanol durch Destillation beseitigt wird.
Nach beendeter Umsetzung wird das überschüssige Urethan unter vermindertem Druck abdestilliert. Der Rückstand wird hierauf mit Äther gewaschen und durch Filtrieren gesammelt, wobei man 29,6 g 3 - (o-Fluorphenyl) -5-chlorindol- 1 ,2-dicar- bonimid erhält. Durch Umkristallisieren aus Aceton erhält man Kristalle vom Schmelzpunkt 253 bis 254"C.
b) In eine Suspension von 1,0 g 3-(o-Fluorphenyl)-5-chlorindol-1,2-dicarbonimid in 25 ml Essigsäure führt man unter Rühren eine Mischung aus Ozon und Sauerstoff ein. Nach ungefähr 30 Minuten bildet sich eine Lösung, worauf das Ausfällen von Kristallen beginnt. Dann leitet man eine Ozon Sauerstoff-Mischung während weiteren 3 Stunden ein. Die ausgefällten Kristalle werden durch Filtrieren gesammelt und getrocknet, wobei man 0,43 g 3-(o-Fluorphenyl)-5-chlorindol- -1,2dicarbonimid-ozonid erhält. Schmelzpunkt 136,5 bis 137"C (unter Zersetzung). Weitere 0,10 g dieses Produktes werden aus dem Filtrat gewonnen.

Wird das Filtrat unter vermindertem Druck bei Zimmertemperatur zur Trockne eingedampft, so erhält man 1-[2-(o -Fluorbenzoyl)-4-chlorphenyl]-parabansäure als weisses Pulver (0,60 g).
Massenspektrum: 346 (21%, M+), 275 (16), 223 (8)

IR: Nmu3-Ol 3550, 3220, 3080, 1784, 1746, 1665, 1608, c) Zu einer Suspension von 2,0 g 3-(o-Fluorphenyl)-5 -chlorindol-1,2-dicarbonimid in 30 ml Essigsäure gibt man eine Lösung von 3,0 g Chromsäureanhydrid in 3 ml Wasser hinzu und rührt dann das Gemisch während 5 Stunden bei 650C. Die Essigsäure wird unter vermindertem Druck abdestilliert. Der Rückstand wird hierauf mit 100 ml Wasser vermischt und das Gemisch mit Äther extrahiert. Der ätherische Extrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft. Der Rückstand wird hernach mit n-Pentan gewaschen, wobei man 1,74 g 1-[2-(o-Fluorbenzoyl)-4-chlorphenyl]-parabansäure als Pulver erhält.
Bei der obigen Extraktion mit Äther erhält man 0,05 g einer unlöslichen Substanz, welche aus 4-(o-Fluorphenyl)-6 -chlor-2(1H)-chinazolinon besteht.
Beispiel 1
Eine Suspension von 100 mg 3-(o-Fluorphenyl)-5-chlor indol-1,2-dicarbonimid-ozonid in 3 ml Wasser wird während einer Stunde auf dem Wasserbade erhitzt. Nach dem Kühlen werden die ausgefällten Kristalle gesammelt und getrocknet, wobei man 70 mg 4-(o-Fluorphenyl)-6-chlor-2(1H)-chinazol non erhält. Der Schmelzpunkt liegt über 3000C.
Beispiel 2
Zu einer Lösung von 200 mg 1-[2-(o-Fluorbenzoyl)-4 -chlorphenyl]-parabansäure in 6 ml Äthanol gibt man 0,8 ml einer 20%igen wässrigen Natriumhydroxydlösung hinzu, worauf man das Gemisch während 45 Minuten unter Rückfluss erhitzt. Das Äthanol wird unter vermindertem Druck abdestilliert. Der Rückstand wird mit 5 ml Wasser vermischt und das Gemisch mittels verdünnter Salzsäure sauer gestellt. Die ausgefällten Kristalle werden durch Filtrieren gesammelt und getrocknet, wobei man 150 mg 4-(o-Fluorphenyl)-6-chlor-2(1H) -chinazolinon erhält. Der Schmelzpunkt liegt bei über 300"C.
Beispiel 3 - In einem Erlenmeyerkolben werden 200 mg 3-(o-Fluor phenyl)-5-chlorindol-1,2-dicarbonimid-ozonid bei 1 800C in einem Olbade zum Schmelzen gebracht, wobei man 4(o -Fluorphenyl)-6-chlor-2(1H)-chinazolinon erhält. Der Schmelz punkt liegt oberhalb 300 C.
Beispiel 4
In 1 ml Dimethylformamid löst man 200 mg 1-[2-(o-Fluor benzoyl)-4-chlorphenyl]-parabansäure, worauf man die Lö sung auf 1500C erhitzt und anschliessend kühlt. Die ausge- fällten Kristalle werden durch Filtrieren gesammelt, wobei man man 4-(o-Fluorphenyl)-6-chlor-2(1H)-chinazolinon erhält. Der
Schmelzpunkt liegt oberhalb 3000C.

The term lower, as used above and below, is intended to mean radicals with 1 to 5 carbon atoms. The symbol R1, which is an atom or a group, can be in any position in the benzofring of the compound of the formula (I) or a compound of a formula mentioned below.
The quinazolinone compounds of the formula I are known to be valuable anti-inflammatory drugs. They are also suitable as valuable intermediates for the manufacture of other chemicals, e.g. Anti-inflammatory drugs and agents for depressing the central nervous system.
So far, the quinazolinone compounds of the formula I have been obtained by using an aminoketone compound of the formula:

It has now been found that the quinazolinone compounds of formula I can be obtained via indole dicarbonimide compounds which are easy to prepare using a new type of reaction.

In the first stage of the synthesis mentioned, indolecarboxylic acid compounds of the formula II can be mixed with a urethane compound of the formula:
H2NCOOR3 where R3 is a lower alkyl radical, e.g. the methyl, ethyl, propyl, isopropyl or butyl radical, means, are implemented.
For this reaction the indolecarboxylic acid compounds of formula II can be used in the form of reactive derivatives thereof, e.g. in the form of acid halides or acid esters, if so desired. In general, water, acetone, carbon tetrachloride, acetic acid, formic acid, sulfuric acid or the like is used.

The process according to the invention consists in converting the said compound of the formula IV into the compound of the formula I by pyrolysis or hydrolysis.
The pyrolysis can be effected by melting or decomposing an imidazolidine compound of Formula IV, generally in the presence or absence of an inert, high boiling point solvent, e.g. Dimethylformamide, dimethyl sulfoxide or diethylene glycol dimethyl ether happens. The hydrolysis can be effected by treating an imidazolidine compound of the formula IV with water, if desired in the presence of an acid or a base. The acid that can be used for this purpose is, for example, a mineral acid, e.g.
Use hydrochloric acid, sulfuric acid, nitric acid or phosphoric acid. Bases can for example be alkali metal hydroxide, e.g. Sodium hydroxide or potassium hydroxide, alkali metal carbonates, e.g. Potassium carbonate, alkaline earth metal hydroxides, e.g. Barium oxide or calcium hydroxide, and ammonium compounds, e.g. Ammonium hydroxide. The use of alkali metal and alkaline earth metal hydroxides is preferred.
Examples of quinazolinone compounds of the formula I obtainable in this way are the following: 4-phenyl-6-chloro-2 (1H) -quinazolinone 4-phenyl-2 (1H) -quinazolinone 4- (o-chlorophenyl) -6-chloro -2 (1H) -quinazolinone 4- (o-Tolyl) -7-nitro-2 (1H) -quinazolinone 4- (p-bromophenyl) -6-bromo-2 (1H) -quinazolinone 4-phenyl-6-nitro -2 (1H) -quinazolinone 4- (o-fluorophenyl) -6-trifluoromethyl-2 (1H) -quinazolinone 4 (0-nitrophenyl) -7-methyl-2 (1 H) -quinazolinone 4-phenyl-6-methoxy -2 (1H) -quinazolinone 4. Methyl-6-chloro-2 (1H) -quinazolinone 4-ethyl-6-nitro-2 (1H) -quinazolinone 4-isopropyl-6-chloro-2 (1H) -quinazolinone , etc.
As already mentioned, the quinazolinone compounds of the formula I are known and valuable anti-inflammatory drugs. The imidazolidine compounds of the formula IV are new compounds and valuable as intermediates for the preparation of the quinazolinone compounds of the formula I. It should also be noted that the imidazolidine compounds of the formula IV are valuable as such as anti-inflammatory agents and also as sedatives for the central nervous system.
The invention is illustrated by the following examples.
Preparation of the starting products (1V) a) A mixture of 30 g of 3- (o-fluorophenyl) -5-chlorine dol-2-carboxylic acid and 60 ml of thionyl chloride is refluxed for 2 hours. The excess thionyl chloride is distilled off under reduced pressure. Then the remaining acid chloride is mixed with 30 g of urethane and the mixture is heated with stirring, the ethanol obtained as a by-product being removed by distillation.
When the reaction has ended, the excess urethane is distilled off under reduced pressure. The residue is then washed with ether and collected by filtration, 29.6 g of 3 - (o-fluorophenyl) -5-chloroindole-1,2-dicarbonimide being obtained. Crystallization from acetone gives crystals with a melting point of 253 to 254 ° C.
b) A mixture of ozone and oxygen is introduced into a suspension of 1.0 g of 3- (o-fluorophenyl) -5-chloroindole-1,2-dicarbonimide in 25 ml of acetic acid while stirring. After about 30 minutes a solution forms and crystals begin to precipitate. An ozone-oxygen mixture is then passed in for a further 3 hours. The precipitated crystals are collected by filtration and dried, whereby 0.43 g of 3- (o-fluorophenyl) -5-chloroindole-1,2dicarbonimide-ozonide is obtained. If the filtrate is evaporated to dryness under reduced pressure at room temperature, 1- [2- (o -fluorobenzoyl) -4-chlorophenyl] parabanic acid is obtained as a white powder (0.60 g).

IR: Nmu3-Ol 3550, 3220, 3080, 1784, 1746, 1665, 1608, c) To a suspension of 2.0 g of 3- (o-fluorophenyl) -5-chloroindole-1,2-dicarbonimide in 30 ml of acetic acid a solution of 3.0 g of chromic anhydride in 3 ml of water is added and the mixture is then stirred for 5 hours at 650.degree. The acetic acid is distilled off under reduced pressure. The residue is then mixed with 100 ml of water and the mixture is extracted with ether. The ethereal extract is washed with water, dried over anhydrous sodium sulfate and evaporated to dryness. The residue is then washed with n-pentane, 1.74 g of 1- [2- (o-fluorobenzoyl) -4-chlorophenyl] parabanic acid being obtained as a powder.
The above extraction with ether gives 0.05 g of an insoluble substance which consists of 4- (o-fluorophenyl) -6-chloro-2 (1H) -quinazolinone.
example 1
A suspension of 100 mg of 3- (o-fluorophenyl) -5-chloroindole-1,2-dicarbonimide-ozonide in 3 ml of water is heated on the water bath for one hour. After cooling, the precipitated crystals are collected and dried, whereby 70 mg of 4- (o-fluorophenyl) -6-chloro-2 (1H) -quinazolnon are obtained. The melting point is above 3000C.
Example 2
0.8 ml of a 20% strength aqueous sodium hydroxide solution is added to a solution of 200 mg of 1- [2- (o-fluorobenzoyl) -4-chlorophenyl] -parabanic acid in 6 ml of ethanol, whereupon the mixture is refluxed for 45 minutes heated. The ethanol is distilled off under reduced pressure. The residue is mixed with 5 ml of water and the mixture is made acidic using dilute hydrochloric acid. The crystal precipitated is collected by filtration and dried to give 150 mg of 4- (o-fluorophenyl) -6-chloro-2 (1H) -quinazolinone. The melting point is over 300 "C.
Example 3 - In an Erlenmeyer flask, 200 mg of 3- (o-fluorophenyl) -5-chloroindole-1,2-dicarbonimide-ozonide are melted at 1,800C in an oil bath, 4 (o -fluorophenyl) -6- chlor-2 (1H) -quinazolinone is obtained. The melting point is above 300 C.
Example 4
200 mg of 1- [2- (o-fluoro-benzoyl) -4-chlorophenyl] -parabanic acid are dissolved in 1 ml of dimethylformamide, whereupon the solution is heated to 150.degree. C. and then cooled. The precipitated crystals are collected by filtration, whereby 4- (o-fluorophenyl) -6-chloro-2 (1H) -quinazolinone is obtained. Of the
