Die Erfindung betrifft ein Verfahren zur Herstellung eines neuen Riechstoffes. Bei diesem Riechstoff handelt es sich um ein Gemisch von isomeren Nitrilen der Formel

Das neue Gemisch der isomeren Nitrile der Formel I besitzt besondere Riechstoffeigenschaften. Diese können als blumig (rosenartig). holzig, animalisch, castoreumähnlich und erdig bezeichnet werden. Von besonderem Interesse ist eine originelle Ledernote. Der Geruchscharakter ist des weitern von demjenigen des entsprechenden Aldehydgemisches, also des 6-Isobutyl-2,6-dimethyl-l(und 2)cyclohexen-1aldehyds überraschendenveise völlig verschieden; letzterer ist fruchtig, medizinisch , terpenartig (borneolartig, camphrig). Gegenüber dem Geruchscharakter des offenen Nitrils, des 3,7,9-Trimethyl-2,6-decadiennitrils, zeichnet er sich anderseits dadurch aus, dass dessen Citrusnote ihm völlig abgeht.
Das neue Gemisch von Nitrilen I kann demgemäss auf Grund seiner interessanten, aus einer ganzen Reihe ähnlich gebauter Nitrile bzw. entsprechender Aldehyde herausragenden olfaktorischen Eigenschaften als Riechstoff Venvendung finden, z. B. in der Parfümerie zur Herstellung von Riechstoffkompositionen (insbesondere von solchen mit holzigen, grünen und Leder-Noten) wie Parfüms bzw. zur Parfümierung von Produkten aller Art, wie z. B. Seifen, Waschmitteln, Detergentien, Aerosolen, oder anderen kosmetischen Produkten, wie Salben, Gesichtsmilch, Schminke, Badesalzen etc., dienen.
Die Konzentration des Gemisches I kann je nach dem Verwendungszweck innerhalb weiter Grenzen variieren, beispielsweise zwischen etwa 1 und etwa 15 Gew. %. Für Riechstoffkompositionen liegen die bevorzugten Konzentrationen im Bereich von etwa 3 bis etwa 10 Gel. %.
Das Gemisch von Nitrilen der Formel I wird dadurch erhalten, dass man 3,7,9-Trimethyl-2,6-decadiennitril mit einem sauren Cyclisierungsmittel behandelt.
Die Cyclisierung des 3,7,9-Trimethyl-2,6-decadiennitrils kann nach an sich bekannten Methoden durchgeführt werden.
Als Cyclisierungsmittel eignen sich insbesondere starke Mineralsäuren, wie Phosphorsäure, Polyphosphorsäure, konzentrierte Schwefelsäure, konzentrierte Bromwasserstoffsäure etc.; organische Säuren und deren Anhydride, z. B. Ameisensäure, Essigsäure, Essigsäureanhydrid, p-Toluolsulfonsäure etc. Die Umsetzung kann in einem inerten Verdünnungsmittel erfolgen, z. B. in einem aromatischen Kohlenwasserstoff, wie Toluol, in Dibutyläther, in Dioxan oder in einem chlorierten Kohlenwasserstoff, wie Chloroform oder Trichlor äthan. Es ist jedoch in der Regel vorteilhafter, ohne Verdünnungsmittel zu arbeiten.
Die Reaktion kann bei Raumtemperatur oder auch bei erhöhter Temperatur durchgeführt werden, z. B. bei 40 bis 50ob.
Zur Isolierung des Reaktionsproduktes aus dem Reaktionsgemisch geht man vorzugsweise so vor, dass das Reaktionsgemisch auf Eis gegossen und das Produkt mit einem geeigneten wasserunlöslichen Lösungsmittel, wie z. B. Petrol äther, Toluol usw., extrahiert wird. Die organische Phase kann dann neutral gewaschen und getrocknet, das Lösungsmittel abgedampft und das zurückbleibende Produkt einer Vakuumdestillation unterworfen werden.
Das als Ausgangsmaterial verwendete 3,7,9-Trimethyl2,6-decadiennitril kann nach Schema A erhalten werden: S¯c¯hema A

Durch Behandlung des 3,7,9-Trimethyl-2,6-decadien- 1- als IV (siehe z. B. die schweizerische Patentschrift Nr. 337 826) mit einem Hydroxylammoniumsalz, z. B. dem Chlorid, Sulfat, Acetat, Monosulfonat usw., nach an sich bekannten Methoden (siehe z. B. Houben-Weyl, Methoden der organischen Chemie, Stickstoffverbindungen I, Teil IV, 56-66, G. Thieme-Verlag Stuttgart [1968]) erhält man das Oxim III. Aus dem Oxim III kann das Nitril II durch Dehydratisierung auf an sich bekannte Weise gewonnen werden, z. B. unter Verwendung von Essigsäure, Acetanhydrid, Thionylchlorid, Phosphoroxychlorid oder einem Gemisch von Ameisensäure/Alkaliformiat, z. B. Natriumformiat, als Dehydratisierungsmittel (siehe z. B.
auch Weyand/Hilgetag, organisch-chemische Experimentierkunst, J. A. Barth-Verlag Leipzig [1970], 527). Als Lösungsmittel kann im allgemeinen das Dehydratisierungsmittel dienen. Wird als Dehydratisierungsmittel Thionylchlorid oder Phosphoroxychlorid verwendet, empfiehlt sich jedoch zusätzlich die Verwendung von, beispielsweise, Pyridin. Es ist von Vorteil, die Reaktion bei erhöhter Temperatur, beispielsweise bei der Rückflusstemperatur des Reaktionsgemisches, durchzuführen. Das Reaktionsprodukt II kann aus dem Reaktionsgemisch z. B. durch Destillation isoliert werden.
Im nachfolgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
Zu 472 g Eisessig werden 774 g konz. Schwefelsäure so zugegeben, dass die Temperatur 35 nicht übersteigt. Bei einer Temperatur von 40 gibt man anschliessend zum Reaktionsgemisch 248,5 g (1,3 Mol) 3,7,9-Trimethyl-2,6-decadiennitril unter Kühlung zu. Alsdann wird während 30 Minuten weitergerührt. Das Reaktionsgemisch wird auf 780 g Eis gegeben und dreimal mit je 150 ml Petroläther (40-60) extrahiert. Die vereinigten Petrolätherextrakte werden mit 5% Natriumbicarbonatlösung, hierauf mit Wasser gewaschen, über Magnesiumsulfat getrocknet und eingeengt.
Die Destillation des 222 g betragenden Rohproduktes im Hochvakuum ergibt 194,5 g (78,2% Ausbeute) 6-Isobutyl-2,6 dimethyl- 1 -(und 2)-cyclohexen- 1 -carbonsäurenitril vom Siedepunkt 80-82 /0,01 mm Hg. Das Isomerengemisch kann mittels präparativer Gaschromatographie in die Komponenten aufgetrennt werden.
Das als Ausgangsmaterial verwendete 3,7,9-Trimethyl2,6-decadiennitril kann wie folgt erhalten werden:
144 g (2,07 Mol) Hydroxylamin-hydrochlorid werden in 1,3 Liter Wasser gelöst und 117 g Soda zugegeben. Zur klaren Lösung gibt man 450 ml Methanol und 385 g (1,98 Mol) 3 ,7,9-Trimethyl-2,6-decadien- 1 -al. Die sich leicht erwärmende Lösung wird 4 Stunden bei Raumtemperatur gerührt und anschliessend dreimal mit je 150 ml Petroläther (40-60) extrahiert. Die vereinigten Petrolätherextrakte werden mit Wasser gewaschen, über Natriumsulfat getrocknet und eingeengt. Die erhaltenen 390 g (94% Ausbeute) 3,7,9-Trimethyl2,6-decadien-1-aldoxim werden direkt für die nächste Stufe verwendet.
250 ml eines Gemisches von 350 g (1,68 Mol) rohem 3,7,9-Trimethyl-2,6-decadien-1-aldoxim und 306 g (3 Mol) Acetanhydrid werden vorsichtig zum Sieden erhitzt (¯ 135 ).
Nach Erreichen der Rückflusstemperatur tritt eine heftige Reaktion ein und der Rest des Gemisches wird nun so zugegeben, dass das Reaktionsgemisch leicht weiter siedet. Während 30 Minuten wird bei dieser Rückflusstemperatur weitergerührt. Nach dem Abkühlen wird das überschüssige Acetanhydrid am Wasserstrahlvakuum abdestilliert. Die 260 g (81,2% Ausbeute) reinen 3,7,9-Trimethyl-2,6-decadiennitrils destillieren bei 108-110 /0,2 mm Hg. Geruch: blumig, grün, fettig, Zimtsäure- und Citral-Note.

The new mixture of isomeric nitriles of the formula I has special odor properties. These can be considered floral (rose-like). woody, animal, castoreum-like and earthy. Of particular interest is an original leather note. Furthermore, the odor character is surprisingly completely different from that of the corresponding aldehyde mixture, ie of 6-isobutyl-2,6-dimethyl-1 (and 2) cyclohexene-1aldehyde; the latter is fruity, medicinal, terpene-like (borneol-like, camphor-like). Compared to the odor character of the open nitrile, the 3,7,9-trimethyl-2,6-decadiene nitrile, on the other hand, it is characterized by the fact that it completely lacks its citrus note.
The new mixture of nitriles I can accordingly be used as a fragrance due to its interesting olfactory properties which are outstanding from a whole range of similarly constructed nitriles or corresponding aldehydes, e.g. B. in perfumery for the production of fragrance compositions (especially those with woody, green and leather notes) such as perfumes or for perfuming products of all kinds, such as. B. soaps, detergents, detergents, aerosols, or other cosmetic products such as ointments, facial milk, make-up, bath salts, etc., are used.
The concentration of the mixture I can vary within wide limits depending on the intended use, for example between about 1 and about 15% by weight. For fragrance compositions, the preferred concentrations are in the range from about 3 to about 10 gels. %.
The mixture of nitriles of the formula I is obtained by treating 3,7,9-trimethyl-2,6-decadiene nitrile with an acidic cyclizing agent.
The cyclization of the 3,7,9-trimethyl-2,6-decadienonitrile can be carried out by methods known per se.
Strong mineral acids such as phosphoric acid, polyphosphoric acid, concentrated sulfuric acid, concentrated hydrobromic acid, etc .; are particularly suitable as cyclizing agents; organic acids and their anhydrides, e.g. B. formic acid, acetic acid, acetic anhydride, p-toluenesulfonic acid, etc. The reaction can be carried out in an inert diluent, e.g. B. ethane in an aromatic hydrocarbon such as toluene, in dibutyl ether, in dioxane or in a chlorinated hydrocarbon such as chloroform or trichloro. However, it is usually more advantageous to work without a diluent.
The reaction can be carried out at room temperature or at elevated temperature, e.g. B. at 40 to 50ob.
To isolate the reaction product from the reaction mixture, the procedure is preferably such that the reaction mixture is poured onto ice and the product is treated with a suitable water-insoluble solvent, such as. B. petroleum ether, toluene, etc. is extracted. The organic phase can then be washed neutral and dried, the solvent evaporated and the product which remains can be subjected to vacuum distillation.

By treating the 3,7,9-trimethyl-2,6-decadiene-1- as IV (see e.g. Swiss Patent No. 337,826) with a hydroxylammonium salt, e.g. B. the chloride, sulfate, acetate, monosulfonate, etc., by methods known per se (see, for example, Houben-Weyl, Methods of Organic Chemistry, Nitrogen Compounds I, Part IV, 56-66, G. Thieme-Verlag Stuttgart [ 1968]) one obtains the oxime III. The nitrile II can be obtained from the oxime III by dehydration in a manner known per se, e.g. B. using acetic acid, acetic anhydride, thionyl chloride, phosphorus oxychloride or a mixture of formic acid / alkali metal formate, e.g. B. sodium formate, as a dehydrating agent (see e.g.
also Weyand / Hilgetag, organic-chemical experimental art, J. A. Barth-Verlag Leipzig [1970], 527). The dehydrating agent can generally serve as the solvent. If thionyl chloride or phosphorus oxychloride is used as the dehydrating agent, the use of, for example, pyridine is also recommended. It is advantageous to carry out the reaction at an elevated temperature, for example at the reflux temperature of the reaction mixture. The reaction product II can from the reaction mixture, for. At a temperature of 40, 248.5 g (1.3 mol) of 3,7,9-trimethyl-2,6-decadienonitrile are then added to the reaction mixture with cooling. The mixture is then stirred for a further 30 minutes. The reaction mixture is poured onto 780 g of ice and extracted three times with 150 ml of petroleum ether (40-60) each time. The combined petroleum ether extracts are washed with 5% sodium bicarbonate solution, then washed with water, dried over magnesium sulfate and concentrated.
Distillation of the crude product totaling 222 g in a high vacuum gives 194.5 g (78.2% yield) of 6-isobutyl-2,6-dimethyl-1- (and 2) -cyclohexen-1-carboxonitrile with a boiling point of 80-82 / 0, 01 mm Hg. The isomer mixture can be separated into the components by means of preparative gas chromatography.
The 3,7,9-trimethyl2,6-decadiene nitrile used as the starting material can be obtained as follows:
144 g (2.07 mol) of hydroxylamine hydrochloride are dissolved in 1.3 liters of water and 117 g of soda are added. 450 ml of methanol and 385 g (1.98 mol) of 3,7,9-trimethyl-2,6-decadien-1-al are added to the clear solution. The solution, which warms up slightly, is stirred for 4 hours at room temperature and then extracted three times with 150 ml of petroleum ether (40-60) each time. The combined petroleum ether extracts are washed with water, dried over sodium sulfate and concentrated. The 390 g (94% yield) of 3,7,9-trimethyl2,6-decadiene-1-aldoxime obtained are used directly for the next step.
250 ml of a mixture of 350 g (1.68 mol) of crude 3,7,9-trimethyl-2,6-decadiene-1-aldoxime and 306 g (3 mol) of acetic anhydride are carefully heated to the boil (¯ 135).
After the reflux temperature has been reached, a vigorous reaction occurs and the remainder of the mixture is now added in such a way that the reaction mixture continues to boil slightly. Stirring is continued at this reflux temperature for 30 minutes. After cooling, the excess acetic anhydride is distilled off in a water jet vacuum. The 260 g (81.2% yield) of pure 3,7,9-trimethyl-2,6-decadiene nitrile distill at 108-110 / 0.2 mm Hg. Smell: flowery, green, greasy, notes of cinnamic acid and citral .
