Eine Acyloxygruppe enthält vorzugsweise den Rest einer gestättigten oder ungesättigten aliphatischen oder cycloaliphatischen, einer araliphatischen oder aromatischen Carbonsäure mit bis zu 20, vorzugsweise bis zu 12 C-Atomen. Beispiele solcher Säuren sind, Ameisensäure, Essigsäure, Pivalinsäure, Propionsäure, Buttersäure, Capronsäure, Oenanthsäure, folsäure, Palmitinsäure, Stearinsäure, Bernsteinsäure, Malonsäure, Fumarsäure, Zitronensäure, Cyclohexylpropionsäure, Phenylessigsäure und Benzoesäure.

Die entsprechenden in 6-Stellung halogenierten Ausgangsverbindungen der Formel II erhält man z. B. durch Halogenierung der Verbindungen der Formel IV in bekannter Weise.
Die Ausgangsstoffe der Formel III können durch mikrobielle Hydroxylierung entsprechender, in 11-Stellung unsubstituierter Steroide hergestellt werden. Geeignete Mikroorganismen für diesen Zweck sind z. B.
Absidia regnieri und A. glauca Aspergillus niger Blakeslea trispora Calonectria decora Colletotrichum phomoides Coniothyrium helleborine Cunninghamella blakesleeana Curvularia lunata und C. falcata Fusarium solani, bulbigenum Helicostylum piriforme Helmintosporium sativum Mucor hiemalis und M. parasiticus Neurospora crassa Nocardia lurida Petalotia foetans Rhizopus arrhizus, Rh. nigricans, Rh. nidulans Phycomyces blakesleeanus Streptomyces fradiae Trichothecium roseum
Die mikrobiologische Hydroxylierung kann beispielsweise wie folgt durchgeführt werden:
Schrägagar der Zusammensetzung 10% Rohrzucker in
100 ml Malzkeim-Extrakt und 2,2% Agar wird mit Aspergillus ochraceus NRRL 405 beimpft und 5 Tage bei 28 qC inkubiert.
Ab dieser Kultur werden vier mit Schikanen versehene Erlenmeyerkolben die 100 ml eines Nährmediums der Zusammensetzung 5% Glucose, 2% Edamin und 0;15% Cornsteep in entionisiertem Wasser enthalten, mit einer Sporensuspension beimpft. Die Glucose wird vor dem Beimpfen getrennt vom Medium sterilisiert. Der pH-Wert wird auf 6,5 mit NaOH eingestellt. Diese Schüttelkulturen werden 36 Stunden bei 28 "Cinkubiert.
Ein Kleinfermenter mit 8 Liter Nährlösung der gleichen Zusammensetzung wie bei der Schüttelkultur verwendet, wird sterilisiert; 400 g Glucose in einem Liter entionisiertem Wasser werden vor dem Beimpfen steril zugesetzt. Mit dem Inhalt der 4 Schüttelkolben wird beimpft. Man inkubiert 36 Stunden unter Rühren (400 RpM) bei 28 qC und belüftet mit 6 Liter Luft pro Minute. Auftretende Schaumbildung wird mit Maisöl bekämpft.
Danach werden 8 g 17,21-Dihydroxy-15ss,16ss- methylenpregn-4-en-3,20-dion in 60 ml Dimethylformamid gelöst, der Kulturlösung beigesetzt. Der Verlauf der Fermentation wird kontrolliert, indem Proben der Fermentationslösung mit Essigester extrahiert und dünnschichtchromatographisch analysiert werden. Nach etwa 37 Stunden ist die Fermentation beendet.
Durch Filtration der Gärbrühe über Hyflo -Supercel wird die Mycelmasse von der Gärlösung getrennt und das Mycel 6 mal mit einem Liter Wasser nachgewaschen. Das steroidhaltige Kulturfiltrat und das Waschwasser werden viermal mit gleichem Volumen Methylenchlorid 15 Minuten extrahiert. Die vereinigten Extrakte werden über Na2SO4 getrocknet und i. V.
bei 45 "C eingedampft. Der Rückstand wurde auf der 30fachen Menge Silicagel chromatographiert. Mit Aether Methanol 9:1 konnten 5,9 g reines 11a,17,21-Trihydroxy 15ss,1 6ss.methylen-pregn-4-en-3 20-dion eluiert werden; Smp.
204-205 (aus Aceton-Isopropyläther); [a] D5 = + 810.
Die Steroide der Formel I sind endocrin, insbesondere anti infiammatorisch wirksam. Beispielsweise zeigten das 11ss,17a, 21-Trihydroxy-15,16ss-methylen-pregna-1,4-dien-3,20-dion bei oraler Verabreichung von 3 mg, das 11ss,17ex,21-Trihy- droxy-15,16ss-methylen-pregn-4-en-3,20-dion bei subcutaner Verabreichung von 10 mg im Filz-Pellet-Test an der Ratte granulationshemmende Wirkung.
Beispiel
Zu einer Lösung von 6,7 g 11ss,17-dihydroxy-15ss,16ss- methylen-pregn-4-en-3,20-dion in 50 ml Tetrahydrofuran und 30 ml Methanol gab man unter Rühren 10 g Calciumoxid. Zu dieser auf O"abgekühlten Mischung liess man innert einer Stunde eine Lösung von 6,15 g Jod in 17 ml Tetrahydrofuran, 7 ml Methanol und 2,0 ml 50% wässeriger CaCl2-Lösung so zutropfen, dass das Jod fortlaufend verbraucht wurde. Anschliessend wurde noch 4 Stunden bei O"gerührt. Zur Aufarbeitung wurde abgenutscht, der Filterrückstand gut mit Methylenchlorid nachgewaschen. Das Filtrat wurde auf Eiswasser gegossen und mit Methylenchlorid extrahiert. Diesen Extrakt wusch man mit Natriumthiosulfatlösung und Wasser, trocknete mit Na2SO4 und dampfte im Vakuum ein.
Man erhielt 9,2 g rohes 21-Jodid, das mit einem Gemisch von 200 ml Aceton, 60 ml Triäthylamin und 40 ml Essigsäure 2 Stunden unter Rückfluss erhitzt wurde. Das Gemisch wurde dann auf Eiswasser gegossen, mit Methylenchlorid extrahiert, der Extrakt mit Wasser gewaschen, mit Na2SO4 getrocknet und im Vakuum eingedampft. Man erhielt 8,1 g rohes 21-Acetat.
