Vorliegende Erfindung betrifft ein Verfahren zur Herstellung eines 1 1P-Dihydro-l-subst. 1-subst.-4-oxo-3-chinolincarbonsäure- derivats der Formel I

Das Verfahren zur Herstellung eines 1,4-Dihydro-1-subst.4-oxo-3-chinolincarbonsäurederivats zeichnet sich erfindungsgemäss dadurch aus, dass ein 4-Hydroxy-3-chinolincarbonsäurederivat der Formel II

worin Rl und n die vorstehend in Formel I genannte Bedeutung haben, mit dem Ester eines Alkohols ROH einer starken Säure in Reaktion gebracht und der so erhaltene 1,4-Dihydro 1 -subst.-4-oxo-3-chinolincarbonsäureester erwünschtenfalls hydrolysiert wird.
Die nach dem erfindungsgemässen Verfahren hergestellte 1 ,4-Dihydro- 1 -subst.-4-oxo-3-chinolincarbonsäure und deren Carbonsäureester sowie ihre Salze bieten den Vorteil, eine nützliche Wirksamkeit zur Bekämpfung gramnegativer Bakterien bei etwa 0,0001 bis 1,0 mg/ccm betragenden Probekonzentrationen zu besitzen. Das gemäss dem beschriebenen Verfahren hergestellte 1 ,4-Dihydro- 1-subst.-4-oxo-3-chinolin- carbonsäurederivat der Formel I kann somit mit bestem Erfolg als Bakterienbekämpfungsmittel verwendet werden.
Zur erfindungsgemässen Herstellung des 1,4-Dihydro-1subst.-4-oxo-3-chinolincarbonsäurederivats der Formel I wird ein 4-Hydroxy-3-chinolincarbonsäurederivat der Formel II

Wie bei allen tautomeren Systemen sind dabei die Umwandlungsgeschwindigkeit II= IIa und das jeweilige II/iIa-Ver- hältnis von der jeweils vorliegenden thermodynamischen Umgebung abhängig.
Zur Bildung eines mit Erfolg verwendbaren pharmazeutischen Präparats zur Bekämpfung von Bakterien kann ein pharmazeutisch wirksamer Anteil vom 1,4-Dihydro-1-subst.4-oxo-3-chinolincarbonsäurederivat der Formel I in ein pharmazeutisch zulässiges Träger- oder Verdünnungsmedium oder -mittel gegeben werden.
Bevorzugte Beispiele für mit Erfolg verwendbare Alkylradikale R und Rl sind Alkylradikale mit höchstens 4 C-Atomen, wie Methyl-, Äthyl-, n-Propyl-, n-Butyl-, Isopropyl- oder sekundäre Butylradikale. Bevorzugte Beispiele für R mit Erfolg verwendbare Alkenylradikale sind Vinyl-, Allyl- und Butenylradikale, für Cycloalkylradikale Cyclopropyl-, Cyclobutyl-, Cyclopentyl- und Cyclohexylradikale, für Cycloalkylalkylradikale Cyclopropylmethyl-, Cyclobutylmethyl- und Cyclopentylmethylradikale, für Hydroxyalkylradikale Hydroxymethyl-, Hydroxyäthyl-, Hydroxypropyl- und Hydroxybutylradikale, für Alkoxyalkylradikale Methoxymethyl-, Methoxyäthyl Äthoxymethyl-, Äthoxyäthyl- und Äthoxypropylradikale, für Arylaklylradikale Benzyl- und Phenäthylradikale, für Halogene Chlor und Brom, für Alkylthioradikale sind Methylthio-, Äthylthio-, n-Propylthio-, Iso-propylthio-,
n-Butylthio- und Benzylthioradikale.
Der im erfindungsgemässen Verfahren verwendete Ester eines Alkohols ROH einer starken Säure weist mit Vorteil die Formel V
RY (V) auf, worin R die vorstehend in Formel I genannte Bedeutung hat und Y der Anionteil einer starken anorganischen Säure oder einer organischen Säure HY, wie einer Sulfonsäure, ist; beispielsweise für die erstere kann genannt werden Chlorid, Bromid, Jodid oder Sulfat und für die letzteren Benzolsulfonat, Paratoluolsulfonat, Diäthyloxoniumfluorborat oder eine andere ähnliche Verbindung. Dabei sind Chlorid, Bromid, Jodid oder Sulfat bevorzugt, da sie leichter erhältlich sind.
Die Reaktion wird vorzugsweise in Gegenwart eines Säureakzeptors durchgeführt. Ein Säureakzeptor ist eine basische Substanz, die vorzugsweise frei wasserlösliche Nebenprodukte, die vom Reaktionsprodukt leicht trennbar sind, bildet, wie Natriumhydroxid, Kaliumhydroxid, Natriumcarbonat, Kaliumcarbonat, Natriumalkoxid, Kaliumalkoxid, Natriumamid, Natriumhydrid und andere ähnliche Verbindungen. Der Säureakzeptor ist zur Aufnahme der Säure (HY), die im Verlaufe der Reaktion ausgeschieden wird, bestimmt. Die Reaktion kann entweder in Gegenwart oder in Abwesenheit eines geeigneten Lösungsmittels, vorzugsweise jedoch in Gegenwart eines Lösungsmittels, wie Wasser, einem niederen Alkanol, Aceton, Dioxan, Dimethylformamid oder einer Mischung dieser Substanzen, beispielsweise einer Mischung von Wasser und eines niederen Alkanols, durchgeführt werden.
Die Reaktion kann beispielsweise bei einer weniger als 150oC betragenden Temperatur mit Erfolg durchgeführt und durch Kühl- oder Wärmebehandlung gesteuert werden.
Der erfindungsgemäss erhaltene 1,4-Dihydro-1-subst.-4oxo-3-chinolincarbonsäureester der Formel I, worin R Alkyl bedeutet, kann erwünschtenfalls zwecks Bildung einer entsprechenden freien 1,4-Dihydro-1-subst.-4-oxo-3-chinolincarbonsäure der Formel I, worin Rl Wasserstoff bedeutet, hydrolysiert werden. Die Hydrolyse wird gemäss einem zur Hydrolyse von Estern üblichen herkömmlichen Verfahren durchgeführt.
Der Ester kann mit Wasser in Reaktion gebracht und die Hydrolyse vorzugsweise in Gegenwart einer sauren Verbindung, beispielsweise einer anorganischen Säure, wie Chlorwasserstoffsäure, Schwefelsäure usw., oder einer alkalischen Verbindung, wie Alkalimetallhydroxide usw., durchgeführt werden. Bei Durchführung der Reaktion des 4-Hydroxy-3-chinolincarbonsäureesters der Formel II, worin Rl Alkyl bedeutet, mit einem Ester der Formel V unter Rückflussbehandlung eines wässrigen Lösungsmittels in Gegenwart von Kaliumoder Natriumhydroxid kann nebst dem 1,4-Dihydro-1-subst.4-oxo-3-chinolincarbonsäureester auch die entsprechende Chinolin-3-carbonsäure erhalten werden.
Wenn das Endprodukt in Form eines Esters, beispielsweise eines Äthylesters, erwünscht ist, kann das Äthyl-1,4-Dihydro-1-hydroxy-4-oxo- 3-chinolincarboxylat Zwischenprodukt, wie vorstehend erläutert, in Form eines Salzes, beispielsweise eines Kalium- oder Natriumsalzes, unter Verwendung eines wässrigen Lösungsmittels in Reaktion gebracht werden. Selbstverständlich kann die Hydrolyse nach der Isolierung oder ohne Isolierung des 1,4 Dihydro- 1 -subst. -4-oxo-3 -chinolincarbonsäureesters durchgeführt werden.
Beispiel 1
Eine 2,77 g 4-Hydroxy-3-äthoxycarbonyl-6,7-methylendioxychinolin-1-oxid, 50 ccm Dimethylformamid und 0,48 g Natriumhydrid enthaltende Lösung von einer 50% betragenden Reinheit wurde bei 70 bis 90oC während 30 Minuten rührbehandelt. Dann wurde eine 25 ccm Dimethylformamid und 4,0 g Diäthylsulfat enthaltende Mischung bei der gleichen Temperatur eingegeben. Die resultierende Mischung wurde bei 70 bis 90 C während weiterer 16 Stunden rührbehandelt.
Nach Vollendung der Reaktion wurde das Lösungsmittel durch Destillation unter reduziertem Druck entfernt, wobei ein gelber Feststoff erhalten wurde. Rekristallisation aus Äthylacetat ergab Äthyl-1,4dihydro-1-äthoxy-6,7-methylendioxy-4-oxo-chinolin-carboxylat in Form bei 130 bis 131ob schmelzender farbloser Prismen.

Das Verfahren von Beispiel 1 wurde unter den in Tabelle 1 gezeigten und sonst gleichen Bedingungen wiederholt, wobei, wie weiter unten schematisch gezeigt ist, Äthyl 1,4-Dihydro-1-subst.-6,7-methylendioxy-4-oxo-3-chinolin- carboxylatderivat erhalten wurde.

Beispiel 6
Eine 2,77 g 4-Hydroxy-3-äthoxycarbonyl-6,7-methylendioxychinolin-1-oxid, 30 ccm 10% wässriges Natriumhydroxid und 8,0 g Diäthylsulfat enthaltende Mischung wurde bei 20 bis 400C während vier Stunden und bei 80 bis 90oC während weiterer fünf Stunden rührbehandelt.
Nach Ablauf der Reaktion wurde die Lösung mit Aktivkohle behandelt und filtriert. Das Filtrat wurde mit konzentrierter Chlorwasserstoffsäure bis zu einem pH von 1 behandelt. Das resultierende weisse Präzipitat wurde filtriert und mit Wasser gewaschen. Umkristallisation aus Dimethylformamid ergab Äthyl-1,4-dihydro-1-äthoxy-6,7-methylendioxy-4-oxo- 3 -chinolincarboxylat.

Präzipitat wurde filtriert und mit Wasser gewaschen. Umkristallisation aus Dimethylflormamid ergab, 1,4-Dihydro-1-methoxy-6,7-methylendioxy-4-oxo-3-chinolincarbonsäure in quantitativer Ausbeute.

Eine 1,7 g Äthyl-1,4-dihydro-1-äthoxy-6,7-methylen- dioxy-4-oxo-3-chinolincarboxylat und 40 ccm N-Chlorwasserstoffsäure enthaltende Mischung wurde während einer Stunde rückflussbehandelt. Die ausgeschiedenen farblosen Prismen wurden filtriert und mit Wasser gewaschen. Rekristallisation aus Dimethylformamid ergab 1,49 g (94%) bei 258 bis 259 schmelzende 1,4-Dihydro-1-äthoxy-6,7-methylendioxy-4oxo-3-chinolincarbonsäure.
Beispiel 18
Eine 1 g Äthyl-1,4-dihydro-1-methoxy-6,7-methylendioxy4-oxo-3-chinolincarbonsäure und 15 ccm 5% wässriges Natriumhydroxid enthaltende Mischung wurde während einer Stunde rückflussbehandelt und durch Eingabe von konzentrierter Chlorwasserstoffsäure angesäuert. Das weisse Präzipitat wurde filtriert und mit Wasser gewaschen. Rekristallisation aus Dimethylformamid ergab 1,4-Dihydro-1-methoxy-6,7- methylendioxy-4-oxo-3-chinolincarbonsäure.

To form a successfully usable pharmaceutical preparation for combating bacteria, a pharmaceutically effective portion of the 1,4-dihydro-1-substituted 4-oxo-3-quinolinecarboxylic acid derivative of the formula I can be added to a pharmaceutically acceptable carrier or diluent medium or agent will.

The ester of an alcohol ROH of a strong acid used in the process according to the invention advantageously has the formula V
RY (V), where R has the meaning given above in formula I and Y is the anion part of a strong inorganic acid or an organic acid HY, such as a sulfonic acid; for example, chloride, bromide, iodide or sulphate can be mentioned for the former, and benzene sulphonate, paratoluene sulphonate, diethyloxonium fluoroborate or another similar compound for the latter. Chloride, bromide, iodide or sulfate are preferred because they are more readily available.
The reaction is preferably carried out in the presence of an acid acceptor. An acid acceptor is a basic substance which preferably forms freely water-soluble by-products which are easily separable from the reaction product, such as sodium hydroxide, potassium hydroxide, sodium carbonate, potassium carbonate, sodium alkoxide, potassium alkoxide, sodium amide, sodium hydride and other similar compounds. The acid acceptor is intended to take up the acid (HY) that is excreted in the course of the reaction. The reaction can be carried out either in the presence or in the absence of a suitable solvent, but preferably in the presence of a solvent such as water, a lower alkanol, acetone, dioxane, dimethylformamide or a mixture of these substances, for example a mixture of water and a lower alkanol .
The reaction can, for example, be carried out successfully at a temperature less than 150 ° C. and can be controlled by cooling or heat treatment.
The 1,4-dihydro-1-substituted-4oxo-3-quinolinecarboxylic acid ester of the formula I, in which R is alkyl, obtained according to the invention can, if desired, be used for the purpose of forming a corresponding free 1,4-dihydro-1-substituted-4-oxo- 3-quinolinecarboxylic acid of the formula I, in which Rl is hydrogen, are hydrolyzed. The hydrolysis is carried out according to a conventional method customary for hydrolysis of esters.
The ester can be reacted with water and the hydrolysis can be carried out preferably in the presence of an acidic compound such as an inorganic acid such as hydrochloric acid, sulfuric acid, etc., or an alkaline compound such as alkali metal hydroxide, etc. When carrying out the reaction of the 4-hydroxy-3-quinolinecarboxylic acid ester of the formula II, wherein Rl is alkyl, with an ester of the formula V under reflux treatment of an aqueous solvent in the presence of potassium or sodium hydroxide, in addition to the 1,4-dihydro-1-subst. 4-oxo-3-quinoline-carboxylic acid ester, the corresponding quinoline-3-carboxylic acid can also be obtained.
If the end product is desired in the form of an ester, for example an ethyl ester, the ethyl 1,4-dihydro-1-hydroxy-4-oxo-3-quinoline carboxylate intermediate, as discussed above, can be in the form of a salt, for example potassium - or sodium salt, can be reacted using an aqueous solvent. A solution of 50% purity containing 2.77 g of 4-hydroxy-3-ethoxycarbonyl-6,7-methylenedioxyquinoline-1-oxide, 50 cc of dimethylformamide and 0.48 g of sodium hydride was treated with stirring at 70 to 90 ° C. for 30 minutes. Then a mixture containing 25 cc of dimethylformamide and 4.0 g of diethyl sulfate was charged at the same temperature. After the completion of the reaction, the solvent was removed by distillation under reduced pressure to obtain a yellow solid. Recrystallization from ethyl acetate gave ethyl 1,4-dihydro-1-ethoxy-6,7-methylenedioxy-4-oxo-quinoline-carboxylate in the form of 130 to 131ob melting colorless prisms.

Example 6
A mixture containing 2.77 g of 4-hydroxy-3-ethoxycarbonyl-6,7-methylenedioxyquinoline-1-oxide, 30 cc of 10% aqueous sodium hydroxide and 8.0 g of diethyl sulfate was heated at 20 to 40 ° C. for four hours and at 80 to 90 ° C. treated with stirring for a further five hours.
After the reaction had ended, the solution was treated with activated charcoal and filtered. The filtrate was treated with concentrated hydrochloric acid to pH 1. A mixture containing 1.7 g of ethyl 1,4-dihydro-1-ethoxy-6,7-methylenedioxy-4-oxo-3-quinolinecarboxylate and 40 cc of N-hydrochloric acid was refluxed for one hour. The precipitated colorless prisms were filtered and washed with water.
